Synthesis and antimicrobial activity of novel benzoxazoles.

A series of 2-(p-substituted-benzyl)-5-[[4-(p-chloro/fluoro-phenyl)piperazin-1-yl]acetamido]-benzoxazoles were synthesized in need of new compounds for the fight against microbial pathogens. Their structures were elucidated by spectral techniques. These new derivatives, along with previously synthesized 2-(p-substituted-benzyl)-5-substituted-benzoxazoles, were evaluated for their antibacterial and antifungal activities against standard strains and drug-resistant isolates in comparison with ampicillin, gentamicin sulfate, ofloxacin, vancomycin, fluconazole, and amphotericin B trihydrate. Rapid identification of pathogens from positive blood cultures by multiplex polymerase chain reaction using the FilmArray system.

Sepsis is a leading cause of death. Rapid and accurate identification of pathogens and antimicrobial resistance directly from blood culture could improve patient outcomes. The FilmArray® (FA; Idaho Technology, Salt Lake City, UT, USA) Blood Culture (BC) panel can identify >25 pathogens and 4 antibiotic resistance genes from positive blood cultures in 1 h. We compared a development version of the panel to conventional culture and susceptibility testing on 102 archived blood cultures from adults and children with bacteremia. Of 109 pathogens identified by culture, 95% were identified by FA. Amon

Probiotic potential and safety assessment of bacteriocinogenic Enterococcus faecalis CAUM157.

INTRODUCTION: Over the past few years, the genus Enterococcus has been implicated as both probiotic and pathogenic bacteria, making it challenging to introduce enterococcal probiotic candidates. Based on rigid case-by-case analysis, some enterococcal strains have been developed as probiotics, exhibiting various beneficial activities that promote the health of the host. In this study, the probiotic potential of Enterococcus faecalis CAUM157 (KACC81148BP), isolated from raw cow's milk, was examined, leveraging its bacteriocin production for potential control of pathogenic and spoilage-associated

Microbial keratitis at a single tertiary eye care in Andhra Pradesh: A 12-year analysis of microbiological profile and in vitro susceptibility of bacterial isolates.

CONTEXT: Microbial keratitis is a potentially vision-threatening emergency that demands prompt diagnosis and treatment to ensure better vision recovery. Empirical use of broad-spectrum antimicrobial therapies is often the primary treatment modality, which is then tailored according to clinical response, cultured organisms, and their sensitivities. Since the spectrum of microbial agents associated with corneal ulcers is wide and varies from one geographical location to another, it is imperative that local microbial trends and susceptibility patterns are analyzed to enable better care in the man

Bacteriophage and antibiotic combination therapy for recurrent Enterococcus faecium bacteremia.

UNLABELLED: Enterococcus faecium is a member of the human gastrointestinal (GI) microbiota but can also cause invasive infections, especially in immunocompromised hosts. Enterococci display intrinsic resistance to many antibiotics, and most clinical E. faecium isolates have acquired vancomycin resistance, leaving clinicians with a limited repertoire of effective antibiotics. As such, vancomycin-resistant E. faecium (VREfm) has become an increasingly difficult to treat nosocomial pathogen that is often associated with treatment failure and recurrent infections. We followed a patient with recurr

Identification of drivers of mycobacterial resistance to peptidoglycan synthesis inhibitors.

Beta-lactams have been excluded from tuberculosis therapy due to the intrinsic resistance of Mycobacterium tuberculosis (Mtb) to this antibiotic class, usually attributed to a potent beta-lactamase, BlaC, and to an unusually complex cell wall. In this pathogen, the peptidoglycan is cross-linked by penicillin-binding proteins (PBPs) and L,D-transpeptidases, the latter resistant to inhibition by most beta-lactams. However, recent studies have shown encouraging results of beta-lactam/beta-lactamase inhibitor combinations in clinical strains. Additional research on the mechanisms of action and res

In Vitro and In Vivo Studies on the Antibacterial Activity and Safety of a New Antimicrobial Peptide Dermaseptin-AC.

Antimicrobial resistance has been an increasing public health threat in recent years. Antimicrobial peptides are considered as potential drugs against drug-resistant bacteria because they are mainly broad-spectrum and are unlikely to cause resistance. In this study, a novel peptide was obtained from the skin secretion of Agalychnis callidryas using the "shotgun" cloning method. The amino acid sequence, molecular weight, and secondary structure of Dermaseptin-AC were determined. The in vitro antimicrobial activity, hemolysis, and cytotoxicity of Dermaseptin-AC were evaluated. MICs and minimum b

Modified glove use for contact precautions: Health care workers' perceptions and acceptance.

BACKGROUND: Patients colonized or infected with methicillin-resistant Staphylococcus aureus and or vancomycin-resistant Enterococcus are placed under contact precautions. Contact precautions require patients to be placed in single rooms and their health care workers (HCWs) to wear gowns, aprons and gloves on entry and doffing on exit. Glove use is widely accepted to be associated with poor hand hygiene compliance. We trailed the removal of gloves for contact precautions for contacts not expected to involve body fluids to improve hand hygiene between multiple contacts of the patient and patient

Prevalence and antimicrobial resistance profile of Staphylococcus in dairy farms, abattoir and humans in Addis Ababa, Ethiopia.

BACKGROUND: Staphylococcus species cause mastitis and wound infection in livestock and food poisoning in humans through ingestion of contaminated foods, including meat and dairy products. They are evolving pathogens in that they readily acquire drug resistance, and multiple drug-resistant (MDR) isolates are increasing in human and veterinary healthcare. Therefore, this study was conducted to evaluate the prevalence of Staphylococci and their drug resistance in dairy farms and abattoir settings of Addis Ababa. METHODS: In this cross-sectional study, 193 samples of milk, meat, equipment and hum

Promoting Wound Regeneration Through Targeted Suppression of Chronic Inflammation with Active Molecular Chitosan.

This study aimed to investigate the wound-healing mechanisms of chitosan with a defined molecular weight (MW) and degree of deacetylation (DD), and to explore its potential in hydrogel formulations, optimized for enhanced antibacterial performance. An active molecular chitosan (AMC) was prepared via enzymatic treatment to target a specific MW range with excellent biological activity. The antibacterial, anti-inflammatory, and wound-healing effects of AMC-based hydrogels were evaluated. Given AMC's antibacterial activity against vancomycin-resistant Staphylococcus aureus (VRSA), its anti-inflamm

Proteomic and metabolomic responses of priority bacterial pathogens to subinhibitory concentration of antibiotics.

This study employed a comprehensive proteomic and metabolomic analysis to characterize adaptive cellular mechanisms of priority pathogens-Escherichia coli, Klebsiella pneumoniae, Enterococcus faecium, and Staphylococcus aureus-under sub-inhibitory concentrations of antibiotics. Despite significant metabolomic perturbations, some pathogens had minimal or no significant changes in their proteome. Notably, trimethylamine metabolism was consistently altered across all species, suggesting its role in survival under antibiotic stress. Shared adaptive responses to chloramphenicol in S. aureus and E.

Gut microbiota induces mammary gland sIgA production to alleviate S. aureus mastitis.

Mastitis threatens dairy cow health and public safety. While gut dysbiosis increases mastitis susceptibility, the mechanism is unclear. We hypothesized that gut dysbiosis exacerbates Staphylococcus aureus (S. aureus)-induced mastitis by reducing milk secretory immunoglobulin A (sIgA). In mice, vancomycin-induced gut dysbiosis reduced sIgA and IgA+ B cells in milk, gut, and Peyer's patches, impairing the blood-milk barrier and worsening mastitis, effects reversed by fecal microbiota transplantation. Dysbiosis specifically reduced intestinal M. intestinale abundance and GABA levels. Supplementin
