A Simple Proteomics-Based Approach to Identification of Immunodominant Antigens from a Complex Pathogen: Application to the CD4 T Cell Response against Human Herpesvirus 6B.

Most of humanity is chronically infected with human herpesvirus 6 (HHV-6), with viral replication controlled at least in part by a poorly characterized CD4 T cell response. Identification of viral epitopes recognized by CD4 T cells is complicated by the large size of the herpesvirus genome and a low frequency of circulating T cells responding to the virus. Here, we present an alternative to classical epitope mapping approaches used to identify major targets of the T cell response to a complex pathogen like HHV-6B. In the approach presented here, extracellular virus preparations or virus-infect

Broadly reactive antibodies against influenza B virus hemagglutinin neutralize and protect through distinct structural mechanisms.

Influenza B viruses contribute substantially to seasonal disease burden; however, the structural basis by which antibodies recognize the major glycoprotein hemagglutinin (HA) and mediate antiviral activity remains incompletely defined. Influenza B virus used to circulate as two antigenically distinct lineages, B/Victoria/2/1987-like and B/Yamagata/16/1988-like, although the latter has not been detected in global surveillance in recent years. Antigenic drift in HA contributes to reduced vaccine effectiveness; however, the structural and functional basis by which antibodies recognize influenza B

Distinct virus-specific regulation of RNA synthesis across genome segments by thogotovirus polymerases: insights from Oz virus and Dhori virus.

Thogotoviruses are a group of tick-borne, six-segmented, negative-sense single-stranded RNA viruses. These viruses encode an RNA-dependent RNA polymerase that recognizes promoter sequences located at the genomic termini to initiate RNA synthesis. The 5' and 3' ends of the genome bind to the polymerase and function as a promoter. Outside the catalytic center, they base-pair with each other to form a double-stranded RNA structure. This structure is referred to as the distal duplex and plays an important role in RNA synthesis. In this study, we investigated how the RNA sequence of the distal dupl

Myelin Basic Protein Cleaves Cell Adhesion Molecule L1 and Improves Regeneration After Injury.

Myelin basic protein (MBP) is a serine protease that cleaves neural cell adhesion molecule L1 and generates a transmembrane L1 fragment which facilitates L1-dependent functions in vitro, such as neurite outgrowth, neuronal cell migration and survival, myelination by Schwann cells as well as Schwann cell proliferation, migration, and process formation. Ablation and blocking of MBP or disruption of its proteolytic activity by mutation of a proteolytically active serine residue abolish L1-dependent cellular responses. In utero injection of adeno-associated virus encoding proteolytically active MB

Novel HBx-interacting host cellular factors regulate dynamic nuclear and cytoplasmic localization of Hepatitis B virus X protein (HBx).

The hepatitis B virus (HBV) X protein (HBx) is a key player in viral transcription, replication, and HBV-associated hepatocarcinogenesis. Several novel HBx-interacting host cellular factors: DDX1, DDX3, MOV10 RNA helicases, INI1/hSNF5 chromatin-remodeling factor, Rad18 DNA repair enzyme, and CPSF6 cleavage factor were identified. DDX1, DDX3, and MOV10 colocalized with HBx in cytoplasmic foci or aggregates. In contrast, INI1/hSNF5 and CPSF6 relocalized with HBx in nuclear speckles, interchromatin granule clusters (IGCs), and/or PML-nuclear bodies. Rad18 sequestered HBx in the nucleoli. Importan

Design, synthesis, and anti-influenza A virus evaluation of 2-aminoquinoline derivatives.

The rapid emergence of drug-resistant influenza A virus (IAV) strains has severely limited the efficacy of current antiviral therapies, highlighting an urgent need for novel agents with distinct mechanisms of action. In this study, a series of 2-aminoquinoline derivatives were synthesized via a trimethylsilyl trifluoromethanesulfonate (TMSOTf) -catalyzed annulation strategy. Evaluation of their anti-influenza virus activity revealed that derivative 3g exhibited potent antiviral efficacy, low cytotoxicity, and a high selectivity index(SI), making it the most promising candidate in this series.

[On the wrong track: Misled by the patient history].

Case report of a young immunocompetent patient presenting with aseptic meningitis due to varicella-zoster virus (VZV) infection. Reactivation of VZV manifesting as meningitis is an uncommon clinical presentation compared to herpes zoster. While a concomitant vesicular rash is frequently observed, it may be absent in some cases. With antiviral therapy initiated promptly, the clinical outcome is generally favorable.

Modulation of IRF7-driven transcription as a strategy to control HIV-1 latency.

BACKGROUND: The persistence of latent HIV-1 reservoirs remains a major barrier to achieving a cure for HIV. While latency-reversing agents (LRAs) have been extensively studied, latency-promoting agents (LPAs) offer a complementary strategy to silence viral transcription and prevent immune activation. Here, we propose that modulation of IRF7-driven transcription may represent a novel approach to control HIV-1 latency, by characterizing the role of the Janus kinase 2 inhibitor (JAK2i) pacritinib as a novel latency-promoting agent (LPA). METHODS: The impact of JAK2i on HIV-1 reactivation, immune

PAV-05 Naphthoquinone Potently Inhibit Zika Virus Replication in Infected Cells.

BACKGROUND: Zika (ZIKV) is a virus transmitted by mosquitoes that can cause Guillain- Barré syndrome and congenital malformations like microcephaly. Given its explosive resurgence and the resulting epidemics in 2016, the search for effective antiviral drugs has become absolutely necessary. METHODS: In this study, we examined the potential of naphthoquinone derivatives that have a sulfonamide or sulfonate group to inhibit ZIKV replication in primary cultured neurons and in Vero cells. RESULTS: In our in vitro studies, we found that PAV05 had low cytotoxicity with a CC50 of 329 μM ±3.6 for Ver

Association of Increased Epstein-Barr Virus Reactivation With Imbalanced Immunity and Myenteric Neuron Loss in Achalasia.

BACKGROUND AND AIM: Viral infection has been postulated as a potential etiology of achalasia, yet the definitive role of viral involvement remains elusive. This study aims to elucidate virus infections and to analyze their relationships with clinical characteristics, immunological alterations, and neuron loss. METHODS: We investigated viral infections in achalasia patients (n = 288) and controls (n = 71). Propensity score matching (PSM) was employed to mitigate selection bias. Flow cytometry and ELISA were performed to characterize the immunological changes. In situ hybridization and immunofl

Human SLAM-adapted canine distemper virus can enter human peripheral blood mononuclear cells and replicate in mice expressing human SLAM and defective for STAT1 expression.

Canine distemper virus (CDV) is a member of the genus Morbillivirus with a worldwide distribution that causes fatal diseases in canids and marine mammals. In recent years, CDV has demonstrated the remarkable ability of pathogens to cross species barriers. The natural host range of CDV has expanded from Canidae to Primates, presumably attributed to ecological shifts and the emergence of viral variants. Therefore, it is important to investigate whether CDV can infect humans by adapting to the human signalling lymphocyte activation molecule (hSLAM) receptor to cross the species barrier. Through s

Intermittent loss of anti-HBc antibodies in people with multiple sclerosis undergoing disease-modifying therapies.

BACKGROUND: Disease-modifying therapies (DMTs) are widely used in the treatment of multiple sclerosis (MS). A mounting body of evidence suggests that the risk of hepatitis B virus (HBV) reactivation is primary associated with anti-CD20 therapies. HBV infection leads to the development of anti-HBc antibodies, which typically persist for life. However, the existing literature also highlights the intermittent loss of anti-HBc antibodies in certain immunocompromised individuals. The present study aims to gather real-world evidence on the risk of infection in people with MS (pwMS) prior to the init

N-Hydroxypiridinedione: A Privileged Heterocycle for Targeting the HBV RNase H.

Hepatitis B virus (HBV) remains a global health threat. Ribonuclease H (RNase H), part of the virus polymerase protein, cleaves the pgRNA template during viral genome replication. Inhibition of RNase H activity prevents (+) DNA strand synthesis and results in the accumulation of non-functional genomes, terminating the viral replication cycle. RNase H, though promising, remains an under-explored drug target against HBV. We previously reported the identification of a series of N-hydroxypyridinedione (HPD) imines that effectively inhibit the HBV RNase H. In our effort to further explore the HPD s

Sequential activation of ERα-AMPKα signaling by the flavonoid baicalin down-regulates viral HNF-dependent HBV replication.

Baicalin (BA), a natural component found in many traditional Chinese medicines, exerts protective effects against several viruses. Although our previous studies have revealed that the anti-hepatitis B virus (anti-HBV) activity of BA depends on hepatocyte nuclear factor (HNF) signaling, the specific mechanisms remain unclear. The present study explored the potential signaling mechanisms involved in BA-mediated HBV suppression. Transcriptomic analysis suggested that BA significantly modulates the estrogen receptor (ER) and AMPK signaling pathways in HepG2 cells. The ER alpha (ERα) binding affini

Structure-Activity Relationships and Discovery of (S)-6-Isopropyl-2-methoxy-3-(3-methoxypropoxy)-10-oxo-5,10-dihydro-6H-pyrido[1,2-h][1,7]naphthyridine-9-carboxylic Acid (AB-452), a Novel Orally Available HBV RNA Destabilizer.

Approved therapies for hepatitis B virus (HBV) treatment include nucleos(t)ides and interferon alpha (IFN-α) which effectively suppress viral replication, but they rarely lead to cure. Expression of viral proteins, especially surface antigen of the hepatitis B virus (HBsAg) from covalently closed circular DNA (cccDNA) and the integrated genome, is believed to contribute to the persistence of HBV. This work focuses on therapies that target the expression of HBV proteins, in particular HBsAg, which differs from current treatments. Here we describe the identification of AB-452, a dihydroquinolizi

Navigating the complex terrain of hepatitis B virus reactivation in the era of Bruton tyrosine kinase inhibitors.

In this editorial, we offer a summary of the risk associated with hepatitis B reactivation (HBVr) in the setting of both solid and hematologic malignancies treated with Bruton tyrosine kinase (BTK) inhibitors, with insights derived from current studies. Furthermore, we emphasize the critical need for a framework regarding robust risk evaluation in patients undergoing such treatments. This framework is essential for identifying those at increased risk of HBVr, enabling healthcare providers to implement proactive measures to prevent reactivation and ensure the safe administration of BTK inhibito

Discovery of 2-Amide-3-methylester Thiophenes that Target SARS-CoV-2 Mac1 and Repress Coronavirus Replication, Validating Mac1 as an Antiviral Target.

The COVID-19 pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus has made it clear that further development of antiviral therapies will be needed. Here, we describe small-molecule inhibitors for SARS-CoV-2 Mac1, which counters ADP-ribosylation-mediated innate immune responses. Three high-throughput screening hits had the same 2-amide-3-methylester thiophene scaffold. We studied the compound binding mode using X-ray crystallography, allowing us to design analogues. Compound 27 (MDOLL-0229) had an IC50 of 2.1 μM and was selective for CoV Mac1 proteins after prof
