Abscisic acid and hydraulic conductivity of maize roots: a study using cell- and root-pressure probes.

Using root- and cell-pressure probes, the effects of the stress hormone abscisic acid (ABA) on the water-transport properties of maize roots (Zea mays L.) were examined in order to work out dose and time responses for root hydraulic conductivity. Abscisic acid applied at concentrations of 100-1,000 nM increased the hydraulic conductivity of excised maize roots both at the organ (root Lp(r): factor of 3 4) and the root cell level (cell Lp: factor of 7-27). Effects on the root cortical cells were more pronounced than at the organ level. From the results it was concluded that ABA acts at the plas

Alleviation of Drought Stress by Hydrogen Sulfide Is Partially Related to the Abscisic Acid Signaling Pathway in Wheat.

Little information is available describing the effects of exogenous H2S on the ABA pathway in the acquisition of drought tolerance in wheat. In this study, we investigated the physiological parameters, the transcription levels of several genes involved in the abscisic acid (ABA) metabolism pathway, and the ABA and H2S contents in wheat leaves and roots under drought stress in response to exogenous NaHS treatment. The results showed that pretreatment with NaHS significantly increased plant height and the leaf relative water content of seedlings under drought stress. Compared with drought stress

Abscisic acid enhances glucose disposal and induces brown fat activity in adipocytes in vitro and in vivo.

Abscisic acid (ABA) is a plant hormone also present in animals, where it is involved in the regulation of innate immune cell function and of glucose disposal, through its receptor LANCL2. ABA stimulates glucose uptake by myocytes and pre-adipocytes in vitro and oral ABA improves glycemic control in rats and in healthy subjects. Here we investigated the role of the ABA/LANCL2 system in the regulation of glucose uptake and metabolism in adipocytes. Silencing of LANCL2 abrogated both the ABA- and insulin-induced increase of glucose transporter-4 expression and of glucose uptake in differentiated

The role of abscisic acid in the response of a specific vacuolar invertase to water stress in the adult maize leaf.

Among the numerous molecular and physiological modifications induced by water deficit, one of the earliest events observed in maize mature leaves subjected to water deprivation was a strong enhancement of acid vacuolar invertase activity, which occurred before the classical reduction in gas exchange due to stomatal closure. The increase in invertase activity coincided with the rapid accumulation of glucose and fructose that reached 8-fold the control leaf value. In addition, acid vacuolar invertase activity appeared to be highly correlated with xylem sap ABA concentration. In order to investig

Neither endogenous abscisic acid nor endogenous jasmonate is involved in salicylic acid-, yeast elicitor-, or chitosan-induced stomatal closure in Arabidopsis thaliana.

Salicylic acid (SA), yeast elicitor (YEL), and chitosan (CHT) induced stomatal closure in Arabidopsis wild-type and aba2-2 plants, induced stomatal closure in fluridon-treated wild-type plants, and induced stomatal closure in aos mutants. These results suggest that neither endogenous abscisic acid nor endogenous jasmonic acid is involved in SA-, YEL-, or CHT-induced stomatal closure.

Integrated Transcriptomic Profiling Reveals Distinct and Overlapping Transcriptional Responses and Regulatory Pathways Mediated by Salicylic Acid, Jasmonic Acid, and Abscisic Acid in Lotus (Nelumbo nucifera).

Sacred lotus (Nelumbo nucifera) is an important aquatic crop whose growth and productivity are severely constrained by environmental stresses. Salicylic Acid (SA), Jasmonic Acid (JA), and Abscisic Acid (ABA) are key plant growth regulators (PGRs) involved in stress responses, but their regulatory mechanisms in lotus remain unclear. In this study, transcriptome sequencing was performed in lotus seedlings treated with exogenous SA, JA, and ABA to characterize hormone-responsive regulatory networks. SA predominantly resulted in transcriptional repression, with responsive genes mainly associated w

The miR1866-OsUBP7 regulatory module promotes grain yield and seed dormancy through abscisic acid signaling in rice.

Higher grain weight and stronger seed dormancy are key objectives for improving rice (Oryza sativa) yield and inhibiting pre-harvest sprouting. Therefore, identifying genes that coordinately regulate grain weight and seed dormancy is an urgent priority. Here, we report that knocking out miR1866 (KO1866) increased grain weight and reinforced dormancy. We identified the transcript of ubiquitin-specific processing protease 7 (OsUBP7), which encodes a protein with deubiquitination activity in vitro, as the primary target of miR1866. Consistent with miRNA-directed repression, OsUBP7 transcript abun

Deciphering highly potent natural abscisic acid agonists for binding to pyrabactin resistance 1 receptor through computational approaches.

Abscisic acid (ABA) agonists bind to the same receptors as ABA in plant cells and exhibit similar physiological effects. Even though synthetic ABA agonists like Pyrabactin, Quinabactin, Cyanabactin, and Opabactin can bind to the PYR1 receptor, no plant-derived natural ABA agonists have been reported. The present study explored the highly potent natural ABA agonists through the target inhibition efficacy of phytochemicals from the different parts of the Ficus carica plant against the binding of pyrabactin resistance 1 (PYR1) receptor (PDB: 3K3K) using computational approaches. Molecular docking

Anthocyanin accumulation is initiated by abscisic acid to enhance fruit color during fig (Ficus carica L.) ripening.

Fig fruit is well-known for its attractive flavor, color, and nutritional and medicinal value. Anthocyanin contributes to the fruit's color and constitutes a high percentage of the total antioxidant content of the fig fruit. We quantified the major anthocyanins and characterized the expression levels of anthocyanin-biosynthesis and transcription factor genes in fruit treated on-tree with exogenous abscisic acid (ABA) or ethephon, or the ABA inhibitors nordihydroguaiaretic acid (NDGA) or fluridone. The major anthocyanins cyanidin 3-O-glucoside and cyanidin 3-O-rutinoside were found in significa

A basic leucine zipper transcription factor, AabZIP1, connects abscisic acid signaling with artemisinin biosynthesis in Artemisia annua.

Artemisinin is a sesquiterpenoid especially synthesized in the Chinese herbal plant, Artemisia annua, which is widely used in the treatment of malaria. Artemisinin accumulation can be enhanced by exogenous abscisic acid (ABA) treatment. However, it is not known how ABA signaling regulates artemisinin biosynthesis. A global expression profile and phylogenetic analysis as well as the dual-LUC screening revealed that a basic leucine zipper family transcription factor from A. annua (namely AabZIP1) was involved in ABA signaling to regulate artemisinin biosynthesis. AabZIP1 had a higher expression

A novel role of PR2 in abscisic acid (ABA) mediated, pathogen-induced callose deposition in Arabidopsis thaliana.

Pathogenesis-related protein 2 (PR2) is known to play a major role in plant defense and general stress responses. Resistance against the fungal pathogen Leptosphaeria maculans in Arabidopsis requires abscisic acid (ABA), which promotes the deposition of callose, a β-1,3-glucan polymer. Here, we examined the role of PR2 in callose deposition in relation to ABA treatment and challenge with L. maculans and Pseudomonas syringae. Characterization of PR2-overexpressing plants and the knockout line indicated that PR2 negatively affects callose deposition. Recombinant PR2 purified from Pichia pastoris

Jasmonoyl-L-Isoleucine Induces Systemic Photosynthetic Responses to Local Heat Stress by Contributing to Abscisic Acid Accumulation.

Systemic regulation of photosynthesis is crucial for plant survival in variable environments, yet the hormonal mechanisms coordinating photosynthetic responses to local stimuli are not fully elucidated. This study investigates the interplay between jasmonates (JAs) and abscisic acid (ABA) in systemic photosynthetic responses induced by local heat stress in Arabidopsis thaliana. Using phytohormone quantification, chlorophyll fluorescence and gas exchange measurements in wild-type and transgenic plants impaired in JA biosynthesis, this study showed that local heating-induced variation potential

Abscisic acid metabolism to dehydrovomifoliol by Candidatus Liberibacter asiaticus and CLas-associated Flavobacterium strains CGRL1 and CGRL2.

INTRODUCTION: Citrus huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), is a globally devastating plant disease characterized by severe disruption of host physiology. Although alterations in plant hormone homeostasis, including salicylic acid and abscisic acid (ABA), are hallmarks of HLB, the contribution of plant-associated microbiota to these hormonal changes remains poorly understood. Here, we investigated whether ABA depletion in HLB-affected citrus is associated with microbial metabolism. METHODS: ABA and its metabolites were quantified in HLB-affected citrus tissue

Abscisic acid improves non-alcoholic fatty liver disease in mice through the AMPK/NRF2/KEAP1 signaling axis.

Non-alcoholic fatty liver disease (NAFLD) has emerged as a global health concern, placing a substantial financial strain on public health systems. Currently, no specific pharmacological treatments are recommended in existing guidelines. Abscisic acid (ABA), a natural plant hormone, is recognized for its promising potential in the healthcare field due to its diverse biological activities. Therefore, this study is aimed at exploring the protective mechanism of ABA against NAFLD. In vitro, experiments were conducted using palmitic acid (PA) to establish a fatty liver cell model, whereas in vivo,

OsVDE, a xanthophyll cycle key enzyme, mediates abscisic acid biosynthesis and negatively regulates salinity tolerance in rice.

OsVDE, a lipocalin-like protein in chloroplasts, negatively regulated the ABA biosynthesis and stomatal closure under salt stress in rice seedlings. Violaxanthin de-epoxidase (VDE) is a key enzyme of xanthophyll cycle. It plays a critical role in abscisic acid (ABA) biosynthesis, growth and stress responses in plants. Although functions of several VDE genes have been characterized, it is largely unknown whether OsVDE regulates the ABA biosynthesis and salt stress tolerance in rice. In this study, we generated the OsVDE overexpressing and CRISPR-Cas9-mediated gene-editing transgenic lines, and

Phytohormone abscisic acid boosts pentobarbital-induced sleep through activation of GABA-A, PPARβ and PPARγ receptor signaling.

BACKGROUND: Sleep disorders induce anxiety and forgetfulness and change habits. The chemical hypnotic drugs currently used have serious side effects and, therefore, people are drawn towards using natural compounds such as plant-based healing agents. Abscisic acid (ABA) is produced in a variety of mammalian tissues and it is involved in many neurophysiological functions. OBJECTIVE: To investigate the possible effect of ABA on pentobarbital-induced sleep and its possible signaling through GABA-A and PPAR (γ and β) receptors, in male Wistar rats. METHODS: The possible effect of ABA (5 and 10 µg

Persulfidation-based Modification of Cysteine Desulfhydrase and the NADPH Oxidase RBOHD Controls Guard Cell Abscisic Acid Signaling.

Hydrogen sulfide (H2S) is a gaseous signaling molecule that regulates diverse cellular signaling pathways through persulfidation, which involves the post-translational modification of specific Cys residues to form persulfides. However, the mechanisms that underlie this important redox-based modification remain poorly understood in higher plants. We have, therefore, analyzed how protein persulfidation acts as a specific and reversible signaling mechanism during the abscisic acid (ABA) response in Arabidopsis (Arabidopsis thaliana). Here we show that ABA stimulates the persulfidation of l-CYSTEI

Interaction of intracellular hydrogen peroxide accumulation with nitric oxide production in abscisic acid signaling in guard cells.

UNLABELLED: Reactive oxygen species and nitric oxide (NO•) concomitantly play essential roles in guard cell signaling. Studies using catalase mutants have revealed that the inducible and constitutive elevations of intracellular hydrogen peroxide (H2O2) have different roles: only the inducible H2O2 production transduces the abscisic acid (ABA) signal leading stomatal closure. However, the involvement of inducible or constitutive NO• productions, if exists, in this process remains unknown. We studied H2O2 and NO• mobilization in guard cells of catalase mutants. Constitutive H2O2 level was higher

Abscisic acid-induced gene expression in the liverwort Marchantia polymorpha is mediated by evolutionarily conserved promoter elements.

Abscisic acid (ABA) is a phytohormone widely distributed among members of the land plant lineage (Embryophyta), regulating dormancy, stomata closure and tolerance to environmental stresses. In angiosperms (Magnoliophyta), ABA-induced gene expression is mediated by promoter elements such as the G-box-like ACGT-core motifs recognized by bZIP transcription factors. In contrast, the mode of regulation by ABA of gene expression in liverworts (Marchantiophyta), representing one of the earliest diverging land plant groups, has not been elucidated. In this study, we used promoters of the liverwort Mar

Foliar abscisic acid content underlies genotypic variation in stomatal responsiveness after growth at high relative air humidity.

BACKGROUND AND AIMS: Stomata formed at high relative air humidity (RH) respond less to abscisic acid (ABA), an effect that varies widely between cultivars. This study tested the hypotheses that this genotypic variation in stomatal responsiveness originates from differential impairment in intermediates of the ABA signalling pathway during closure and differences in leaf ABA concentration during growth. METHODS: Stomatal anatomical features and stomatal responsiveness to desiccation, feeding with ABA, three transduction elements of its signalling pathway (H2O2, NO, Ca(2+)) and elicitors of thes
