Teratogenicity of acetaldehyde in vitro: relevance to the fetal alcohol syndrome.

Day 10 rat embryos grown in vitro showed significant retardation in growth and development when culture media contained acetaldehyde. A concentration-response range for acetaldehyde-induced embryotoxicity was defined, from no effect at 5 microM to complete lethality at 100 microM. The relative teratogenicity of ethanol and acetaldehyde, and the potential roles of these compounds in producing the Fetal Alcohol Syndrome are discussed. Despite intensive investigation into alcohol teratogenicity, the mechanism that produces the Fetal Alcohol Syndrome (FAS) remains unknown. Observed anomalies may r

Acute effects of acetaldehyde and ethanol on rat heart mitochondria.

The effects of acute exposure of rat heart mitochondria to ethanol (87, 65, and 45 mM) and acetaldehyde (3, 1, and 0.3 mM) were studied using both glutamate and pyruvate/malate substrates. Mitochondria assayed with pyruvate/malate substrate showed no apparent effects of acute exposure to or 30 min preincubation with ethanol at the three concentrations tested. With glutamate substrate, acute ethanol at 87 and 65 mM produced significant decreases in respiratory control ratio (RCR). With 65 and 45 mM acute ethanol, mitochondrial oxygen consumption (QO2) was significantly decreased. Acute acetalde

Preparation and pharmacological activity of the condensation product of adrenaline with acetaldehyde.

In vitro incubation of adrenaline with acetaldehyde resulted in the formation of an amorphous product (MA 3) which gave origin to two spots on chromatography plates. Preparative TLC allowed us to separate the corresponding substances, MA 4 and MA 5. Gas chromatography-mass spectrometry, ultraviolet and infrared spectra of MA 4 agree with the structure corresponding to 1,2-dimethyl-4,6,7-trihydroxy-1,2,3,4-tetrahydroisoquinoline. MA 5 is very unstable and was not further characterized. Pharmacological experiments were carried out with MA 3 and MA 4 (and in some cases, MA 5) on isolated saphenou

Combined acetaldehyde metabolism burden modifies IBD susceptibility to alcohol consumption.

BACKGROUND: The effect of alcohol consumption on IBD remains inconclusive, potentially due to the overlooked role of alcohol metabolism. OBJECTIVE: We aimed to investigate the potential modifying role of alcohol metabolic capacity in the associations. DESIGN: A prospective cohort study (n=455 417) was used for association analysis between alcohol consumption and IBD risk using Cox proportional hazards regression. An acetaldehyde burden score, using genetic variants of alcohol dehydrogenase and aldehyde dehydrogenase validated by expression quantitative trait loci and proteomic data, was cons

Multiple DNA repair pathways prevent acetaldehyde-induced mutagenesis in yeast.

Acetaldehyde is the primary metabolite of alcohol and is present in many environmental sources, including tobacco smoke. Acetaldehyde is genotoxic, whereby it can form DNA adducts and lead to mutagenesis. Individuals with defects in acetaldehyde clearance pathways have increased susceptibility to alcohol-associated cancers. Moreover, a mutation signature specific to acetaldehyde exposure is widespread in alcohol- and smoking-associated cancers. However, the pathways that repair acetaldehyde-induced DNA damage and thus prevent mutagenesis are vaguely understood. Here, we used Saccharomyces cere

Time-Resolved Acetaldehyde-Based Accessibility Profiling Maps Ligand-Target Interactions.

Elucidating ligand-protein interactions is important in understanding the biochemical machinery for given proteins. Previously, formaldehyde (FH)-based labeling has been employed to obtain such structural knowledge, since reactive residues that participate in ligand-target interactions display reduced accessibility to FH-labeling reagents, and thus can be identified by quantitative proteomics. Although being rapid and efficient for probing proteinaceous lysine accessibility, here, we report an acetaldehyde (AcH)-labeling approach that complements with FH for probing ligand-target interactions.

Key role of local acetaldehyde in upper GI tract carcinogenesis.

Ethanol is neither genotoxic nor mutagenic. Its first metabolite acetaldehyde, however, is a powerful local carcinogen. Point mutation in ALDH2 gene proves the causal relationship between acetaldehyde and upper digestive tract cancer in humans. Salivary acetaldehyde concentration and exposure time are the two major and quantifiable factors regulating the degree of local acetaldehyde exposure in the ideal target organ, oropharynx. Instant microbial acetaldehyde formation from alcohol represents >70% of total ethanol associated acetaldehyde exposure in the mouth. In the oropharynx and achlorhydr

Pyridoxal phosphate annuls acetaldehyde inhibition of uridine uptake by hepatocytes.

Methods were evaluated for depressing the acetaldehyde (AcH) inhibition of uridine uptake by Chang liver cells and isolated autologous liver cells, obtained by percutaneous liver biopsy from cases of alcoholic hepatitis. Hepatocytes so obtained were significantly more susceptible to AcH-induced inhibition of uridine uptake than hepatocytes from normal liver, alcoholic fatty liver, stable alcoholic cirrhosis and acute viral hepatitis. Benzylamine (an aldehyde buffer) and pyridoxal-5' phosphate (PLP) counteracted the inhibition of uridine uptake by AcH in vitro. We suggest that benzylamine neutr

Ethylene, Ethane, Acetaldehyde, and Ethanol Production By Plants under Stress.

Red pine (Pinus resinosa Ait.) and paper birch (Betula papyrifera Marsh.) seedlings exposed to sulfur dioxide produced acetaldehyde and ethanol, and exhibited increased production of ethylene and ethane. Gas chromatographic measurement of head space gas from incubation tubes containing leaves or seedlings was a simple method of simultaneously measuring all four compounds. Increased ethylene production had two phases, a moderate increase from the beginning of the stress period and a large increase just prior to appearance of leaf lesions. Ethane production in SO(2)-stressed plants did not incre

Electrothermal Tandem Catalysis for Acetaldehyde Synthesis From CO2 and Water.

Acetaldehyde (CH3CHO) is an important intermediate in various market sectors, while its industrial production mainly relies on fossil fuels-derived C2H4 and elevated temperature/pressure, leading to massive CO2 emission. In this study, we proposed a novel electrothermal tandem strategy to efficiently synthesize CH3CHO from CO2 and water under mild conditions, by integrating aqueous CO2 electrolysis and Fe3+-promoted Pd2+-catalyzed Wacker oxidation. Specifically, CO2 is electrochemically reduced to C2H4 over Ga-doped Cu/Cu2O hybrid and water is oxidized to O2 over reconstructed Ni5P4 array simu

Malondialdehyde Acetaldehyde-Adduction Changes Surfactant Protein D Structure and Function.

Alcohol consumption with concurrent cigarette smoking produces malondialdehyde acetaldehyde (MAA)-adducted lung proteins. Lung surfactant protein D (SPD) supports innate immunity via bacterial aggregation and lysis, as well as by enhancing macrophage-binding and phagocytosis. MAA-adducted SPD (SPD-MAA) has negative effects on lung cilia beating, macrophage function, and epithelial cell injury repair. Because changes in SPD multimer structure are known to impact SPD function, we hypothesized that MAA-adduction changes both SPD structure and function. Purified human SPD and SPD-MAA (1 mg/mL) wer

Essential medicines containing ethanol elevate blood acetaldehyde concentrations in neonates.

UNLABELLED: Neonates administered ethanol-containing medicines are potentially at risk of dose-dependent injury through exposure to ethanol and its metabolite, acetaldehyde. Here, we determine blood ethanol and acetaldehyde concentrations in 49 preterm infants (median birth weight = 1190 g) dosed with iron or furosemide, medicines that contain different amounts of ethanol, and in 11 control group infants (median birth weight = 1920 g) who were not on any medications. Median ethanol concentrations in neonates administered iron or furosemide were 0.33 (range = 0-4.92) mg/L, 0.39 (range = 0-72.77

NOX2 amplifies acetaldehyde-mediated cardiomyocyte mitochondrial dysfunction in alcoholic cardiomyopathy.

Alcoholic cardiomyopathy (ACM) resulting from excess alcohol consumption is an important cause of heart failure (HF). Although it is assumed that the cardiotoxicity of the ethanol (EtOH)-metabolite acetaldehyde (ACA) is central for its development and progression, the exact mechanisms remain obscure. Murine cardiomyocytes (CMs) exposed to ACA or EtOH showed increased superoxide (O2(•-)) levels and decreased mitochondrial polarization, both being normalized by NADPH oxidase (NOX) inhibition. C57BL/6 mice and mice deficient for the ACA-degrading enzyme mitochondrial aldehyde dehydrogenase (ALDH-

Acetaldehyde oral self-administration: evidence from the operant-conflict paradigm.

BACKGROUND: Acetaldehyde (ACD), ethanol's first metabolite, has been reported to interact with the dopaminergic reward system, and with the neural circuits involved in stress response. Rats self-administer ACD directly into cerebral ventricles, and multiple intracerebroventricular infusions of ACD produce conditioned place preference. Self-administration has been largely employed to assess the reinforcing and addictive properties of most drugs of abuse. In particular, operant conditioning is a valid model to investigate drug-seeking and drug-taking behavior in rats. METHODS: This study was ai

Generation of protein adducts with malondialdehyde and acetaldehyde in muscles with predominantly type I or type II fibers in rats exposed to ethanol and the acetaldehyde dehydrogenase inhibitor cyanamide.

BACKGROUND: Alcoholic myopathy is known to primarily affect type II muscle fibers (glycolytic, fast-twitch, anaerobic), whereas type I fibers (oxidative, slow-twitch, aerobic) are relatively protected. OBJECTIVE: We investigated whether aldehyde-derived adducts of proteins with malondialdehyde and acetaldehyde are formed in muscle of rats as a result of acute exposure to ethanol and acetaldehyde. The differences between type I muscle, type II muscle, and liver tissue were also assessed. DESIGN: The formation and distribution of malondialdehyde- and acetaldehyde-protein adducts were studied w

Effects of ethanol and acetaldehyde on multi-organ genomic mutation landscapes.

Alcohol consumption is a major risk factor for cancer, particularly of the head and neck. Acetaldehyde, the main ethanol metabolite, is linked to oral carcinogenesis, but the mechanisms are not well understood. While some mutational signatures such as single-base substitution signature SBS16, doublet-base substitution signature DBS4, and indel signature ID11 have been linked to alcohol use, they are not limited to alcohol-related cancers. We examined the genome-wide in vivo mutation landscapes of rats chronically exposed to ethanol or acetaldehyde, focusing on head-and-neck tumors (involving t

A Prebiotic Route to Lactate from Acetaldehyde, Cyanide and Carbon Dioxide.

The atmospheric concentration of carbon dioxide (CO2) has fluctuated throughout Earth's history. However, the role of CO2 in prebiotic chemistry has been predominantly and limitedly postulated as a C1 precursor, which can be reduced to carbon monoxide or methane mimicking the Wood-Ljungdahl pathway. Herein we present neglected roles of CO2 as an active promoter in accessing biologically important C3-builidng blocks such as lactate, via redox-economic reaction cycles starting from cyanide (C1) and acetaldehyde (C2). We verified that Lewis acidic CO2 facilitates the formation of cyanohydrin of a

The mutagenic properties of formaldehyde and acetaldehyde: Reflections on half a century of progress.

Formaldehyde and acetaldehyde are reactive, small compounds that humans are exposed to routinely, variously from endogenous and exogenous sources. Both small aldehydes are classified as human carcinogens. Investigation of the DNA damaging properties of these two compounds began some 50 years ago. In this review, we summarize progress in this field since its inception over half a century ago, distilling insights gained by the collective efforts of many research groups while highlighting areas for future directions. Over the decades, general consensus about aspects of the mutagenicity of formald

Theophylline Acetaldehyde as the Initial Product in Doxophylline Metabolism in Human Liver.

Doxophylline (DOXO) and theophylline are widely used as bronchodilators for treating asthma and chronic obstructive pulmonary disease, and DOXO has a better safety profile than theophylline. How DOXO's metabolism and disposition affect its antiasthmatic efficacy and safety remains to be explored. In this study, the metabolites of DOXO were characterized. A total of nine metabolites of DOXO were identified in vitro using liver microsomes from human and four other animal species. Among them, six metabolites were reported for the first time. The top three metabolites were theophylline acetaldehyd
