Effect of pH on sulfite oxidation by Thiobacillus thiooxidans cells with sulfurous acid or sulfur dioxide as a possible substrate.

The oxidation of sulfite by Thiobacillus thiooxidans was studied at various pH values with changing concentrations of potassium sulfite. The optimal pH for sulfite oxidation by cells was a function of sulfite concentrations, rising with increasing substrate concentrations, while that by the cell extracts was unaffected. The sulfite oxidation by cells was inhibited at high sulfite concentrations, particularly at low pH values. The results from kinetic studies show that the fully protonated form of sulfite, sulfurous acid or sulfur dioxide, is the form which penetrates the cells for the oxidatio

Biological removal of H2S gas in a semi-pilot scale biotrickling filter: Optimization of various parameters for efficient removal at high loading rates and low pH conditions.

In this study, a semi-pilot scale biotrickling filter (BTF) was operated in a continuous co-current mode to remove high concentration of hydrogen sulfide (H2S) at optimum operational conditions. The early startup period of 6 days was needed, and then stable removal of H2S gas at inlet concentrations up to about 2000 ppm was successfully obtained at gas retention time (GRT) of 15 min and liquid recirculation rate (LRR) of 120 ml/min. The elimination capacities (ECs) increased linearly with increase in H2S loading rates (HLRs up to 38.5 g/m3 h), but a gradual decrease in removal efficiency was o

An electrochemical sensing approach for scouting microbial chemolithotrophic metabolisms.

The present study was aimed to test an electrochemical sensing approach for the detection of an active chemolithotrophic metabolism (and therefore the presence of chemolithotrophic microorganisms) by using the corrosion of pyrite by Acidithiobacillus ferrooxidans as a model. Different electrochemical techniques were combined with adhesion studies and scanning electron microscopy (SEM). The experiments were performed in presence or absence of A. ferrooxidans and without or with ferrous iron in the culture medium (0 and 0.5 g L-1, respectively). Electrochemical parameters were in agreement with

Novel insights into the kinetics and mechanism of arsenopyrite bio-dissolution enhanced by pyrite.

Arsenopyrite and pyrite often coexist in metal deposits and tailings, thus simultaneous bioleaching of both sulfides has economic (as well as environmental) significance. Important targets in bio-oxidation operations are high solubilization rates and minimized accumulation of Fe(III)/As-bearing secondary products. This study investigated the role of pyrite bioleaching in the enhancement of arsenopyrite dissolution. At a pyrite to arsenopyrite mass ratio of 1:1, 93.6% of As and 93.0% of Fe were solubilized. The results show that pyrite bio-oxidation can promote arsenopyrite dissolution, enhance

Hexavalent chromium remediation based on the synergistic effect between chemoautotrophic bacteria and sulfide minerals.

Hexavalent chromium (Cr(VI)) is an environmental concern due to the carcinogenic and mutagenic effect on living organisms. Sulfide minerals based Cr(VI) reduction is an economical and efficient strategy for Cr(VI) remediation. In this study, Cr(VI) reduction through the synergistic effect between chemoautotrophic bacteria and sulfide mineral is systematically investigated. Sulfide minerals dissolution and Cr(VI) reduction performance highly depends on mineral acid soluble property. Cr(VI) reduction capacity of pyrrhotite, pyrite, marcasite and sphalerite was 50, 104, 104 and 44 mg/g (Cr(VI)/mi

Fulvic acid promotes amorphous FeAsO4 formation and arsenic immobilization under high sulfate during microbe-pyrite/arsenopyrite interaction.

Microbe-pyrite/arsenopyrite (M-Py/APy) interactions in acid mine drainage (AMD) are the primary driver of arsenic (As) release into the environment. Fulvic acid (FuA), with strong Fe/As affinity, actively participates in Fe(III)-secondary mineral transformations and As adsorption, thus influencing As immobilization. Understanding the behavior of FuA-mediated As in M-Py/APy interactions is vital, but the mechanisms remain unclear. Herein, we integrate quartz crystal microbalance with dissipation and multiple microanalytical techniques to elucidate the way surface-adsorbed FuA mediates microbial

Metagenomic analysis reveals enhanced sludge dewaterability through acidified sludge inoculation: Regulation of Fe (II) oxidation electron transport pathway.

The bioleaching utilizing indigenous microbial inoculation can continuously improve the dewaterability of sludge. In this study, metagenomic analysis was innovative employed to identify the key microorganisms and functional genes that affect the dewatering performance of sludge in the bioleaching conditioning process. The results demonstrated that long-term repeated inoculation of acidified sludge resulted in increased abundance of many functional genes associated with the transport of carbohydrate and amino acid. Additionally, genes encoding key iron transport proteins (such as afuA, fhuC, an

Comparison of chromium III and VI toxicities in water using sulfur-oxidizing bacterial bioassays.

The toxicities of Cr (III) and Cr (VI) in water were evaluated using sulfur-oxidizing bacterial (SOB) bioassays both in batch and fed-batch conditions. Two days were enough for a quick buildup of SOB consortium in the master culture reactor (MCR). At concentrations up to 100 mg L(-1), Cr (III) was found to be nontoxic in both conditions, while Cr (VI) at very low concentrations (0.1-2 mg L(-1)) was very toxic to the SOB. Literature review suggested that the nontoxic nature of Cr (III) might be due to the absence of the iron uptake pathway in Acidithiobacillus caldus (the predominant bacteria i

Tetrathionate hydrolase from the acidophilic microorganisms.

Tetrathionate hydrolase (TTH) is a unique enzyme found in acidophilic sulfur-oxidizing microorganisms, such as bacteria and archaea. This enzyme catalyzes the hydrolysis of tetrathionate to thiosulfate, elemental sulfur, and sulfate. It is also involved in dissimilatory sulfur oxidation metabolism, the S4-intermediate pathway. TTHs have been purified and characterized from acidophilic autotrophic sulfur-oxidizing microorganisms. All purified TTHs show an optimum pH in the acidic range, suggesting that they are localized in the periplasmic space or outer membrane. In particular, the gene encodi

Facilitating role of biogenetic schwertmannite in the reduction of Cr(VI) by sulfide and its mechanism.

The efficient conversion of Cr(VI) to Cr(III) has attracted an increasing concern in recent years owing to its threat to the environment. In the present paper, the catalytic role of biogenetic schwertmannite in the reduction of Cr(VI) by sulfide and its mechanism were investigated under different conditions through batch experiments. The results demonstrated that schwertmannite markedly accelerated the removal of Cr(VI) by sulfide, and the rates of the reaction were enhanced by 11, 8 and 6 times, respectively at pH 7.5, 8.0 and 8.8 as compared with control (no schwertmannite). In addition, the

Hybrid process for heavy metal removal from wastewater sludge.

Bioleaching processes have been demonstrated to be effective technologies in removing heavy metals from wastewater sludge, but long hydraulic retention times are typically required to operate these bioprocesses. A hybrid process (coupling biological and chemical processes) has been explored in laboratory pilot-scale experiments for heavy metals (cadmium [Cd], copper [Cu], chromium [Cr], and zinc [Zn]) removal from three types of sludge (primary sludge, secondary activated sludge, and a mixture of primary and secondary sludge). The hybrid process consisted of producing a concentrate ferric ion

Identification of inhibitory substances affecting bioleaching of heavy metals from anaerobically digested sewage sludge.

Significant inhibitory effects of the filtrate medium of anaerobically digested sewage sludge on iron oxidation by Acidithiobacillus ferrooxidans ANYL-1 were observed in our preliminary experiments, indicating the presence of inhibitory substances in anaerobically digested sewage sludge. The objectives of the present study were to identify the possible inhibitory substances and to evaluate their impacts on metal solubilization during bioleaching of sewage sludge. The results showed that the concentrations of total reducing sugars, all tested metal ions, and anions were too low to suppress iron

Comparison of Removal Behavior of Two Biotrickling Filters under Transient Condition and Effect of pH on the Bacterial Communities.

Although biotrickling filters (BTFs) applied under acidic condition to remove H2S from waste gases have been reported, the removal behavior of the acidic BTF under transient condition which was normal in most industry processes, and corresponding bacterial community have not been thoroughly studied. In the present study, two BTFs were run under neutral (BTFn) and acidic (BTFa) conditions, respectively. The results revealed that the removal performance of BTFa under transient condition was superior to that of BTFn; the maximum H2S eliminating capacities (ECs) achieved by BTFa and BTFn were 489.

Carbon dioxide and hydrogen sulfide associations with regional bacterial diversity patterns in microbially induced concrete corrosion.

The microbial communities associated with deteriorating concrete corrosion fronts were characterized in 35 samples taken from wastewater collection and treatment systems in ten utilities. Bacterial communities were described using Illumina MiSeq sequencing of the V1V2 region of the small subunit ribosomal ribonucleic acid (SSU-rRNA) gene recovered from fresh corrosion products. Headspace gas concentrations (hydrogen sulfide, carbon dioxide, and methane), pore water pH, moisture content, and select mineralogy were tested for correlation to community outcomes and corrosion extent using pairwise

Automated Microscopic Analysis of Metal Sulfide Colonization by Acidophilic Microorganisms.

Industrial biomining processes are currently focused on metal sulfides and their dissolution, which is catalyzed by acidophilic iron(II)- and/or sulfur-oxidizing microorganisms. Cell attachment on metal sulfides is important for this process. Biofilm formation is necessary for seeding and persistence of the active microbial community in industrial biomining heaps and tank reactors, and it enhances metal release. In this study, we used a method for direct quantification of the mineral-attached cell population on pyrite or chalcopyrite particles in bioleaching experiments by coupling high-throug

A new bio-oxidation method for removing iron deposits from waterlogged wood of Nanhai I shipwreck, Guangdong, China.

The widespread presence of iron and sulfur compounds such as pyrite in marine waterlogged archeological wood (WAW) can cause irreversible damage to the safety of its preservation. This issue has been a longstanding concern for cultural heritage conservation communities. In this study, we examined the distribution and phase composition of Fe and sulfur compounds in wood samples obtained from the Nanhai I shipwreck using ESEM-EDS, micro-Raman spectroscopy, and an X-ray diffractometer. The removal of iron from WAW samples of the Nanhai I shipwreck using Acidithiobacillus ferrooxidans (A. ferrooxi

Cell-shape assemblage and nanostructure of akaganéite bioformed in FeCl2 solutions.

Akaganéite (β-FeOOH) with a tunnel structure typically occupied by chloride can undergo anion-exchange reactions in aqueous solutions for pollutant removal. In this work, we studied bioformation of akaganéite in FeCl2 solutions with Acidithiobacillus ferrooxidans cells at pH 2.9, during 36-h incubation. The obtained products were analyzed and characterized by XRD, FTIR, EDS, FETEM, and HRTEM. Results showed that in acidic media with pH 2.9, the cells facilitated ferrous biooxidation and ferric precipitation. The resulting ferric precipitates were identified as polycrystalline akaganéite powder

Integrating sulfur, iron(II), and fixed organic carbon for mixotrophic denitrification in a composite filter bed reactor for decentralized wastewater treatment: Performance and microbial community.

Decentralized wastewater treatment in rural areas is an imperative challenge around the world, particularly in developing countries. The composite filter bed reactor is viable for decentralized wastewater treatment, but its performance on nitrogen removal often fluctuates with the unstable influent characteristics and loadings. Here, a composite filter bed reactor integrating sulfur, iron(II), and fixed organic carbon (shaddock peel) was developed and continuously operated under different conditions. The fixed organic carbon source promoted nitrogen removal with an efficiency higher than 90% a

Life in blue: copper resistance mechanisms of bacteria and archaea used in industrial biomining of minerals.

Industrial biomining processes to extract copper, gold and other metals involve the use of extremophiles such as the acidophilic Acidithiobacillus ferrooxidans (Bacteria), and the thermoacidophilic Sulfolobus metallicus (Archaea). Together with other extremophiles these microorganisms subsist in habitats where they are exposed to copper concentrations higher than 100mM. Herein we review the current knowledge on the Cu-resistance mechanisms found in these microorganisms. Recent information suggests that biomining extremophiles respond to extremely high Cu concentrations by using simultaneously

A designed moderately thermophilic consortia with a better performance for leaching high grade fine lead-zinc sulfide ore.

Unwieldy fine sulfide ores are produced during mining; without being appropriately disposed of, they can cause environmental pollution and waste resources. This study investigated the leaching performance of a moderately thermophilic consortia (Leptospirillum ferriphilum + Acidithiobacillus caldus + Sulfobacillus benefaciens) for fine lead-zinc sulfide raw ore. The results showed this microbial community created a low pH, high ORP, and high cell concentration environment for mineral leaching, improving bioleaching efficiency. Under the action of this consortia, the zinc leaching rate reached 9
