Genetic algorithm combined with back-propagation artificial neural network (GA-BP-ANN) to optimize ultrahigh pressure Actinidia arguta juice sterilization conditions to extend shelf life.

In this study, response surface methodology and GA-BP-ANN model were used to optimize the process conditions for ultrahigh pressure (UHP) killing of E. coli in Actinidia arguta juice. The results showed that the GA-BP artificial neural network model predicted the optimal UHP sterilization conditions (holding time 17 min, 350 MPa, and 2 times) with an E. coli death order of magnitude of 7.56. Both UHP and pasteurization had better sterilizing efficacy and mitigated the changes in pH and TSS values of the juice during storage. UHP sterilization preserved the polyphenols, flavonoids and ascorbic

Integrated multi-omics identifies plant hormone signal transduction and phenylpropanoid biosynthesis as key pathways in kiwifruit (Actinidia chinensis var. deliciosa) resistance to Botryosphaeria Dothidea infection.

BACKGROUND: Soft rot, a devastating fungal disease, severely impacts kiwifruit during postharvest storage, causing substantial fruit damage and rendering the fruit inedible. Botryosphaeria dothidea (B. dothidea) is the primary pathogen causing kiwifruit soft rot, but its pathogenic mechanism remains unclear. RESULTS: In this study, morphological observation, transcriptome sequencing, and untargeted metabolomics were employed to analyze the changes in 'Jinkui' kiwifruit after artificial inoculation with B. dothidea. The results showed that obvious lesions began to appear on the fruit surface a

The impact of high-pressure processing on the polyphenol profile and anti-glycaemic, anti-hypertensive and anti-cholinergic activities of extracts obtained from kiwiberry (Actinidia arguta) fruits.

This study analysed the impact of high pressure processing (HHP) on the inhibitory effects (IC50) of kiwiberries (cv. 'Weiki'), on the formation of advanced glycation end-products (AGEs) and the activity of angiotenisn-converting enzyme (ACE) and the enzyme acetylcholinesterase (AChE). The polyphenol profile (HPLC-MS/MS) and antioxidant capacity (PCLACW, ABTS, FRAP) were also studied. HHP-treated 'Weiki' (450 MPa/5 min and 650 MPa/5 min) was the most potent inhibitor of AGEs in the BSA-GLU model (6.52 mg/mL on average) relative to other materials (12.09-7.21 mg/mL). Actinidia arguta Extract Containing Myo-Inositol Suppresses TNF-α-Induced VCAM-1 Expression and Monocyte Adhesion to Endothelial Cells via Inhibition of the PTEN/Akt/GSK-3β and NF-κB Signaling Pathways.

The primary inflammatory process in atherosclerosis, a major contributor to cardiovascular disease, begins with monocyte adhering to vascular endothelial cells. Actinidia arguta (kiwiberry) is an edible fruit that contains various bioactive components. While A. arguta extract (AAE) has been recognized for its anti-inflammatory characteristics, its specific inhibitory effect on early atherogenic events has not been clarified. We used tumor necrosis factor-α (TNF-α)-stimulated human umbilical vein endothelial cells (HUVECs) for an in vitro model. AAE effectively hindered the attachment of THP-1

AcWRKY70-AcAMR1 modulates ascorbic acid accumulation in kiwifruit.

Kiwifruit (Actinidia species) are among the fruits rich in ascorbic acid (AsA), although the AsA content varies significantly across species and cultivars. However, the regulatory mechanisms underlying these differences in AsA content remain largely unexplored. Here, we cloned the AsA negative regulators AeAMR1 and AcAMR1 from Actinidia eriantha 'Maohua' and Actinidia chinensis 'Hongyang', respectively. AcAMR1 exhibited higher expression in 'Hongyang' compared to 'Maohua'. Overexpression of AcAMR1 significantly reduced AsA content by approximately 16.03% in fruits, while silencing AcAMR1 incre

Occurrence and Molecular Variability of the Main Kiwifruit Viruses in the Sichuan Province of China.

Viruses cause important yield losses in kiwifruit. Here, we studied the occurrence and population structure of the major kiwifruit viruses in the Sichuan province of China. RT-PCR results showed the presence of Actinidia virus A (AcVA), Actinidia virus B (AcVB), Actinidia chlorotic ringspot-associated virus (AcCRaV), and the cucumber mosaic virus (CMV). AcCRaV was widely distributed, followed by CMV. These two viruses were often detected in co-infection with AcVA and AcVB. The virus detection rate was positively correlated with vine age. Four phylogenetic groups of AcVA and AcVB were identifie

Mutagenesis of kiwifruit CENTRORADIALIS-like genes transforms a climbing woody perennial with long juvenility and axillary flowering into a compact plant with rapid terminal flowering.

Annualization of woody perennials has the potential to revolutionize the breeding and production of fruit crops and rapidly improve horticultural species. Kiwifruit (Actinidia chinensis) is a recently domesticated fruit crop with a short history of breeding and tremendous potential for improvement. Previously, multiple kiwifruit CENTRORADIALIS (CEN)-like genes have been identified as potential repressors of flowering. In this study, CRISPR/Cas9- mediated manipulation enabled functional analysis of kiwifruit CEN-like genes AcCEN4 and AcCEN. Mutation of these genes transformed a climbing woody p

NMR resonance assignments of the PR-10 allergens Act c 8 and Act d 8 from golden and green kiwifruit.

Kiwifruits have become one of the most common food sources triggering allergic reactions. In patients suffering from birch pollen related food allergy, reactions result from initial sensitization to the birch (Betula verrucosa) pollen allergen Bet v 1, followed by immunological cross-reactivity to structurally homologous proteins in kiwifruit. Clinical symptoms range from scratching and itching of the oral cavity to more severe immunological reactions such as rhino conjunctivitis. In this work we assigned backbone and side chain 1H, 13C and 15N chemical shifts of the 17 kDa PR-10 allergens Act

Deciphering Kiwifruit Bacterial Canker: Molecular Insights, Host-Pathogen Dynamics, and Innovative Management Strategies.

Kiwifruit bacterial canker, caused by Pseudomonas syringae pv. actinidiae (Psa), severely threatens global kiwifruit production. Psa uses type III secretion, effectors, biofilms, and hormone manipulation to suppress the host immunity and achieve successful colonization. In response, kiwifruit activates multilayered defense systems involving pattern-triggered immunity, effector-triggered immunity, structural barriers, reactive oxygen species signaling, and hormone-mediated defense pathways. However, these immune mechanisms remain insufficiently validated in Actinidia species, limiting the trans

Kiwifruit Bacterial Canker Susceptibility of 86 Accessions and Their Physiological Response to Psa Inoculation.

Kiwifruit bacterial canker, caused by Pseudomonas syringae pv. actinidiae (Psa), severely restricts the sustainable development of the kiwifruit industry. Screening stable resistant germplasm and establishing efficient disease resistance evaluation methods are core prerequisites for breeding resistant cultivars. In this study, 86 Actinidia accessions were systematically assessed for Psa susceptibility over three consecutive years using the reported detached shoot inoculation assay. Seven representative accessions with contrasting resistance phenotypes, namely 'Cuiyu', 'Chuhong', 'Jinmei', 'Hon

Filling in the blanks: In silico prediction of novel type III secreted effectors in the Pseudomonas syringae species complex.

Predicting novel type III secreted effectors (T3SEs) in bacteria remains challenging because of their extensive sequence diversity and our heavy reliance on features derived from previously annotated genes. The available tools for predicting T3SS effectors produce a high number of false positives, which complicates their reliable identification. To address this gap, we developed a new bioinformatics workflow and meta-analysis designed to improve confidence in novel T3SE prediction by integrating multiple complementary approaches, including Effectidor2, Bastion3, EffectiveT3, curated effector d

Targeted Mutagenesis of the Female-Suppressor SyGI Gene in Tetraploid Kiwifruit by CRISPR/CAS9.

Kiwifruit belong to the genus Actinidia with 54 species apparently all functionally dioecious. The sex-determinants of the type XX/XY, with male heterogametic, operate independently of the ploidy level. Recently, the SyGI protein has been described as the suppressor of female development. In the present study, we exploited the CRISPR/Cas9 technology by targeting two different sites in the SyGI gene in order to induce a stable gene knock-out in two tetraploid male accessions of Actinidia chinensis var. chinensis. The two genotypes showed a regenerative efficiency of 58% and 73%, respectively. D

Kiwifruit: our daily prescription for health.

Kiwifruit are unequalled, compared with other commonly consumed fruit, for their nutrient density, health benefits, and consumer appeal. Research into their health benefits has focussed on the cultivars Actinidia deliciosa 'Hayward' (green kiwifruit) and Actinidia chinensis 'Hort 16A', ZESPRI(®) (gold kiwifruit). Compared with other commonly consumed fruit, both green and gold kiwifruit are exceptionally high in vitamins C, E, K, folate, carotenoids, potassium, fibre, and phytochemicals acting in synergy to achieve multiple health benefits. Kiwifruit, as part of a healthy diet, may increase hi

Green and gold kiwifruit peel ethanol extracts potentiate pentobarbital-induced sleep in mice via a GABAergic mechanism.

Kiwifruit is one of the most popular fruits worldwide, and it has various biological properties, including antioxidant, anti-allergic, and cardiovascular protective effects. The peel of kiwifruit, which is a by-product of processing, is a good source of flavonoids; however, its bioactivity has not been widely investigated. In this study, we evaluated the hypnotic effects of green (GRPE, Actinidia deliciosa) and gold (GOPE, Actinidia chinensis) kiwifruit peel ethanol extracts and their solvent fractions, and the possible underlying mechanisms. Oral GRPE and GOPE administration (125-1000mg/kg) p
