Atypical Actinobacillus pleuropneumoniae serotype 12 strains with a higher virulence potential.

Actinobacillus pleuropneumoniae is the causative agent of porcine pleuropneumonia, a disease of major economic impact. Serotype 12 is generally considered to be of low virulence and is typically associated with subclinical infections. However, we describe four atypical serotype 12 field strains recovered from severe clinical outbreaks in Chile. These strains exhibited an unusual toxin gene profile (apxIICA, apxIIICA, apxIBD, apxIIIBD), suggesting the ability to produce both ApxII and ApxIII toxins. Comparative genomic analyses revealed that these atypical strains carry the capsule genes of ser

High-dimensional analysis reveals an immune atlas and novel neutrophil clusters in the lungs of model animals with Actinobacillus pleuropneumoniae-induced pneumonia.

Due to the increase in bacterial resistance, improving the anti-infectious immunity of the host is rapidly becoming a new strategy for the prevention and treatment of bacterial pneumonia. However, the specific lung immune responses and key immune cell subsets involved in bacterial infection are obscure. Actinobacillus pleuropneumoniae (APP) can cause porcine pleuropneumonia, a highly contagious respiratory disease that has caused severe economic losses in the swine industry. Here, using high-dimensional mass cytometry, the major immune cell repertoire in the lungs of mice with APP infection wa

Field efficacy of a combined use of Mycoplasma hyopneumoniae and Actinobacillus pleuropneumoniae vaccines in growing pigs.

The effectiveness of simultaneous administration of commercial Mycoplasma hyopneumoniae and Actinobacillus pleuropneumoniae vaccines was tested in an indoor commercial piggery which had experienced continuing respiratory-disease problems confirmed as due to both of these pathogens. Piglets were randomly assigned in equal numbers to vaccination and control groups, and each vaccine was administered at a separate site to assigned piglets at two and four weeks of age. Live weight of vaccinates immediately prior to slaughter was 2.49 kg higher (p = 0.04) than for controls at equal mean slaughter ag

Identification and characterization of serovar-independent immunogens in Actinobacillus pleuropneumoniae.

Despite numerous actions to prevent disease, Actinobacillus pleuropneumoniae (A. pleuropneumoniae) remains a major cause of porcine pleuropneumonia, resulting in economic losses to the swine industry worldwide. In this paper, we describe the utilization of a reverse vaccinology approach for the selection and in vitro testing of serovar-independent A. pleuropneumoniae immunogens. Potential immunogens were identified in the complete genomes of three A. pleuropneumoniae strains belonging to different serovars using the following parameters: predicted outer-membrane subcellular localization; ≤ 1 t

Update on Actinobacillus pleuropneumoniae-knowledge, gaps and challenges.

Porcine pleuropneumonia, caused by the bacterial porcine respiratory tract pathogen Actinobacillus pleuropneumoniae, leads to high economic losses in affected swine herds in most countries of the world. Pigs affected by peracute and acute disease suffer from severe respiratory distress with high lethality. The agent was first described in 1957 and, since then, knowledge about the pathogen itself, and its interactions with the host, has increased continuously. This is, in part, due to the fact that experimental infections can be studied in the natural host. However, the fact that most commercia

Effect of epinephrine on growth characteristics of Actinobacillus pleuropneumoniae field isolates.

Based on empirical observations, stressors in swine populations may contribute to outbreaks due to Actinobacillus pleuropneumoniae (App) in colonized pigs. Stress hormones as catecholamines were found to impact immune mechanism but also bacterial protein expression in vivo, so it can be hypothesized that they are key molecules in disease pathogenesis. Culturing methods to isolate App from tonsillar tissue of carrier pigs for further typing are mostly not successful. In this study the effect of epinephrine in culturing media on App growth behavior was tested. Starting cultures of different age

JMM Profile: Actinobacillus pleuropneumoniae: a major cause of lung disease in pigs but difficult to control and eradicate.

The Gram-negative bacterium Actinobacillus pleuropneumoniae is the causative agent of pleuropneumonia in pigs, its only known natural host. Typical symptoms of peracute disease include fever, apathy and anorexia, and time from infection to death may only be 6 h. Severe lung lesions result from presence of one or two of the ApxI-III toxins. Control is through good husbandry practice, vaccines and antibiotic use. Culture and presence of the species-specific apxIV gene by PCR confirms diagnosis, and identification of serovar, of which 19 are known, informs on appropriate vaccine use and epidemiol

Degraded neutrophil extracellular traps promote the growth of Actinobacillus pleuropneumoniae.

Actinobacillus pleuropneumoniae (A.pp) causes severe pneumonia associated with enormous economic loss in pigs. Peracute diseased pigs die in <24 h with pneumonia. Neutrophils are the prominent innate immune cell in this infection that massively infiltrate the infected lung. Here we show that neutrophils release neutrophil extracellular traps (NETs) as response to A.pp infection. Numerous NET-markers were identified in bronchoalveolar lavage fluid (BALF) of A.pp-infected piglets in vivo, however, most NET fibers are degraded. Importantly, A.pp is able to enhance its growth rate in the presence

Epidemiological Analysis of Environmental Factors Affecting Porcine Pleuropneumonia in a Herd Endemic for Actinobacillus pleuropneumoniae.

Actinobacillus pleuropneumoniae (APP) is the bacterial agent associated with porcine pleuropneumonia, a severe respiratory disease with significant economic impact due to increased mortality and elevated therapeutic costs, whose progression may be exacerbated by environmental factors. This study evaluates the impact of climatic factors, particularly temperature and relative humidity, on mortality in finishing pigs during the fattening phase within a herd with endemic porcine pleuropneumonia. Data were collected from a commercial swine farm between January 2021 and December 2024, including reco

Otitis in a weaned pig: a new pathological role for Actinobacillus (Haemophilus) pleuropneumoniae.

A weaned pig exhibiting clinical signs characteristic of a vestibular syndrome (abnormal head tilt and abnormal gait including a tendency to circle) was examined as a representative example of a condition which, in a 12-month period, affected approximately 400 other pigs on the farm of origin. The condition caused significant morbidity and financial loss but the directly attributable mortality was negligible. Post mortem examination revealed a severe suppurative, left-sided otitis media and interna. Mixed growths of bacteria, including Actinobacillus pleuropneumoniae, were isolated from the pu

[Clinical and laboratory diagnostic findings in chronic pleuropneumonia of swine].

In the course of an experimental study 12 pigs were infected intrabronchially with the pathogen of infectious porcine pleuropneumonia, Actinobacillus pleuropneumoniae. All animals survived the acute stage of infection due to the application of subtherapeutic doses of antibiotics. After infection all of the animals showed clinical signs of acute pneumonia with an elevation of body temperature to about 41 degrees C. Bronchoalveolar lavage fluid (BALF) was taken every week endoscopically. An increase of neutrophil granulocytes could be observed at day 7 and 14 after infection. In parallel, a clea

Isolation and pathogenicity of Actinobacillus pleuropneumoniae Serovar 19 in pigs.

Porcine contagious pleuropneumonia (PCP), caused by Actinobacillus pleuropneumoniae (APP), is a common and severe respiratory disease of pigs worldwide. To date, 19 serovars have been documented and serovar 19 has recently been detected in strains isolated by several researchers in China. However, its in vivo pathogenicity has not yet been reported. In this study, an APP serovar 19 strain, designated HN0314, was isolated from a pig with respiratory disease. Twenty-four 10-week-old pigs were intranasally infected with high, medium, or low doses of APP strain HN0314, with six pigs in each group,

Actinobacillus ureae may be a critical pathogen in patients with predispositions: A case report and review of the literature.

RATIONALE: Actinobacillus ureae (A. ureae) is an unusual commensal of human respiratory flora, rarely causing human infection. The predisposing factors, identification, clinical features, and antibiotic therapy of A. ureae are seldomly reported. Herein, we present a case of 64-year-old man affected by A. ureae pneumonia after intracranial surgery. PATIENT CONCERNS AND DIAGNOSES: A 64-year-old male was admitted with vomiting, drowsiness, and a severe disturbance of consciousness and was later diagnosed with cerebral hemorrhage by computed tomography images. After a craniocerebral surgery, the

Multifocal granulomatous hepatitis caused by Actinobacillus pleuropneumoniae serotype 2 in slaughter pigs.

In a survey of 66 894 slaughter pigs, 11 animals from three farms were found to have multifocal granulomatous lesions in the liver, caused by Actinobacillus pleuropneumoniae serotype 2. The lesions consisted of epithelioid cells and multinucleated giant cells, with asteroid bodies and discernible gram-negative bacteria. Lymph nodes and spleen were occasionally affected. The results suggested that haematogenous spread had occurred from pre-existing pulmonary infections.

In silico-derived Actinobacillus equuli-specific DNA markers and development of associated PCR assays.

The ability to accurately and rapidly identify causative agents of infectious diseases facilitates precise treatment, improves clinical outcomes, and augments epidemiology studies. For many veterinary and zoonotic pathogens, however, simple molecular tests for species identification are not available. Actinobacillus equuli causes severe diseases, such as sleepy foal disease, septicemia, and meningitis in horses and pigs. A. equuli can also cause severe diseases in humans bitten by infected animals. Existing A. equuli identification methods are biochemical tests, 16S rRNA gene amplification fol

Facial cellulitis because of Aggregatibacter (Actinobacillus) actinomycetemcomitans and Capnocytophaga species in an immunocompetent patient.

The species of Capnocytophaga and Aggregatibacter are normal flora and mostly cause periodontal diseases. The soft tissue infection caused by Aggregatibacter often is associated with Actinomyces species. Beside, most Capnocytophaga infections are described in immunocompromised patients. We identified facial cellulitis caused by Capnocytophaga spp and Aggregatibacter (Actinobacillus) actinomycetemcomitans in a 16-year-old immunocompetent female.

Pneumonia caused by Actinobacillus ureae in an immunocompetent healthy young male: a case report.

BACKGROUND: Actinobacillus ureae is a commensal found in the respiratory tract of humans. It is known to cause infections such as meningitis and septic arthritis in immunocompromised patients and also pneumonia especially in those with underlying chronic respiratory diseases. CASE REPORT: We report a case of right lower lobe pneumonia in a 15-year-old immunocompetent male who did not have any chronic illness including respiratory disease. Sputum analysis showed Gram-negative bacilli initially identified as Haemophilus ducreyi based on conventional biochemical tests. Later the organism was conf

Efficacy of vaccination against Actinobacillus pleuropneumoniae in two Belgian farrow-to-finish pig herds with a history of chronic pleurisy.

The efficacy of an Actinobacillus pleuropneumoniae subunit vaccine based on ApxIA, ApxIIA, ApxIIIA and OMP-2 (Porcilis App, MSD) was investigated in two farrow-to-finish pig herds (A and B) affected by chronic pleurisy. In total, 1161 pigs were included. At three weeks of age, the pigs were randomly allocated to non-vaccinated control (NV; n=580) and vaccinated (V; n=581) groups. At 6 and 10 weeks of age, pigs were injected with Porcilis-APP (V group) or adjuvant (NV group). At slaughter (26 weeks), pleurisy and pneumonia lesions were assessed. All pigs were weighed individually at 6 and 26 we
