An empirical relationship between physicochemical properties and sublingual absorption of the model drugs.

Sublingual drug delivery offers a non-invasive alternative to oral administration, allowing drugs to be rapidly absorbed while bypassing the first-pass effect. The purpose of this study was to compare the pharmacokinetics of model drugs following sublingual and oral administration in healthy human subjects and to explore the empirical relationship between sublingual drug-absorption parameters and the physicochemical properties of the model compounds. Orally disintegrating tablets and oral tablets of five model drugs atenolol, caffeine, naproxen, propranolol, and verapamil were administered to

Development of Sedative Dexmedetomidine Sublingual In Situ Gels: In Vitro and In Vivo Evaluations.

Intravenous dexmedetomidine (DEX) is currently approved by the FDA for the sedation of intubated patients in intensive care units to reduce anxiety and to augment postoperative analgesia. Bradycardia and hypotension are limitations associated with the intravenous administration of DEX. In this study, DEX sublingual in situ gels were developed and assessed for their pH, gelling capacity, viscosity, mucoadhesion and in vitro drug release. The optimized gelling system demonstrated enhanced mucoadhesion, superior gelling capacity, reasonable pH and optimal rheological profile. In vivo, compared to

Pharmacokinetics of repeated doses of misoprostol.

BACKGROUND: Misoprostol is widely used in obstetrics and gynaecology for medical abortion, cervical priming and induction of labour. To aid the design of effective and safe regimens, we have investigated the pharmacokinetic parameters after the vaginal or sublingual administration of repeated doses of 400 microg of misoprostol. METHODS: Women undergoing termination of pregnancy by suction evacuation were randomized to receive 400 microg of sublingual or vaginal misoprostol every 3 h for five doses. Venous blood was taken at 180, 200, 240, 360, 380, 420, 540, 560, 600, 720, 740, 780 and 900 mi

Early abortion with buccal versus sublingual misoprostol alone: a multicenter, randomized trial.

OBJECTIVE: To compare efficacy, safety/side effects and acceptability of buccal versus sublingual administration of a misoprostol-only regimen commonly used for early medical abortion. STUDY DESIGN: We conducted a randomized trial at six clinics in two Latin American countries. We randomized women seeking early abortion to buccal or sublingual administration of three doses of misoprostol 800 mcg repeated every 3 h. At initial follow-up (7-14 days after misoprostol), we offered women without a complete abortion aspiration or additional misoprostol plus waiting 7 more days. The primary outcome

Use of mifepristone and sublingual misoprostol for early medical abortion.

OBJECTIVE: Existing drug-induced abortion techniques involve oral administration of 200 mg of mifepristone, followed by oral administration of 600 microg of misoprostol 48 hours later, but the effects are variable. As revealed by recent research, sublingual and oral administrations of misoprostol are equally efficacious in terms of rapid absorption, but the former lasts longer in serum. Hence, in the near future, sublingual administration of misoprostol may become the most effective way to induce abortion. MATERIALS AND METHODS: Women with intrauterine pregnancy up to 49 gestational days rece

Asenapine for the treatment of manic and mixed episodes associated with bipolar I disorder: from clinical research to clinical practice.

INTRODUCTION: Asenapine is a sublingually administered second-generation antipsychotic with proven efficacy for the treatment of moderate to severe manic episodes associated with bipolar I disorder in adults. Its relatively favorable weight and metabolic profile, as well as the lack of appreciable activity at muscarinic cholinergic receptors and the sublingual administration are of clinical interest. AREAS COVERED: This paper comprises a review and commentary regarding the use of sublingual asenapine in the treatment of acute manic and mixed episodes of bipolar disorder. Basic principles in d

Sublingual Crushed Levodopa for Parkinson's Disease Management of a Lewy Body Dementia Patient With Dysphagia: A Case Report.

Levodopa/carbidopa is the cornerstone treatment for Parkinson's disease (PD), but in patients with dysphagia, there are limited options for the routes of administration. Traditional methods of administering levodopa/carbidopa in these patients, such as rectal administration, have been shown to offer limited symptomatic relief and suboptimal pharmacokinetic outcomes. This case explores sublingual administration of levodopa/carbidopa as a novel solution for managing parkinsonian symptoms in dysphagic patients in the hospital setting, where non-oral options are often limited. We present an 85-yea

Electrospun Nanofiber Oral Thin Film Platform for Sublingual Peptide Delivery: A Promising Alternative to Conventional Semaglutide Formulations.

BACKGROUND: Administration of drugs via the oromucosal route, especially using oral thin films, improves patient adherence compared to conventional oral and injectable methods. However, existing production techniques for oral thin films face considerable challenges in incorporating peptide and protein drugs. This study presents electrospun nanofiber-based oral thin film as a viable platform for the sublingual delivery of semaglutide. This platform was tested in Göttingen minipigs, which showed a greater response to attenuation of weight gain. METHODS: Semaglutide-loaded InStrips were produced

Rational use of sublingual opioids in palliative medicine.

The sublingual administration of opioid analgesics has been a mainstay in the pain management of homebound dying hospice patients who are no longer able to swallow. It is also a potentially useful route of administration in other situations in which the oral route is not available and other routes are impractical or inappropriate. Potential advantages of the sublingual route include rapid analgesic onset and avoidance of hepatic first-pass metabolism. Pharmacokinetic and pharmacodynamic studies have yielded widely disparate data on sublingual morphine. Other opioids have been less studied. Ava
