Glucose and alanine metabolism in children with maple syrup urine disease.

In vitro studies have suggested that catabolism of branched chain amino acids is linked with alanine and glutamine formed in, and released from, muscle. To explore this possibility in vivo, static and kinetic studies were performed in three patients with classical, and one patient with partial, branched chain alpha-ketoacid decarboxylase deficiency (maple syrup urine disease, MSUD) and compared to similar studies in eight age-matched controls. The subjects underwent a 24-30-h fast, and a glucose-alanine flux study using stable isotopes. Basal plasma leucine concentrations were elevated (P <0.0

Coupling of glycine and alanine transport to respiration in cells of Escherichia coli.

Energy coupling for uptake of glycine and alanine in glycerol grown cells of Escherichia coli differs from that of the aromatic amino acids. Respiration and uptake of glycine and alanine show similar inactivations in cells exposed to high intensity violet light or to various concentrations of cyanide. In contrast, uptake of phenylalanine, tyrosine, and tryptophan is resistant to effects of light or cyanide. Anoxia largely inhibits uptake of glycine and alanine while that of the aromatic amino acids is only partially affected. Furthermore, ferricyanide (but not ferrocyanide) completely restores

Mimicry of Canonical Translation Elongation Underlies Alanine Tail Synthesis in RQC.

Aborted translation produces large ribosomal subunits obstructed with tRNA-linked nascent chains, which are substrates of ribosome-associated quality control (RQC). Bacterial RqcH, a widely conserved RQC factor, senses the obstruction and recruits tRNAAla(UGC) to modify nascent-chain C termini with a polyalanine degron. However, how RqcH and its eukaryotic homologs (Rqc2 and NEMF), despite their relatively simple architecture, synthesize such C-terminal tails in the absence of a small ribosomal subunit and mRNA has remained unknown. Here, we present cryoelectron microscopy (cryo-EM) structures

Exogenous alanine and/or glucose plus kanamycin kills antibiotic-resistant bacteria.

Multidrug-resistant bacteria are an increasingly serious threat to human and animal health. However, novel drugs that can manage infections by multidrug-resistant bacteria have proved elusive. Here we show that glucose and alanine abundances are greatly suppressed in kanamycin-resistant Edwardsiella tarda by GC-MS-based metabolomics. Exogenous alanine or glucose restores susceptibility of multidrug-resistant E. tarda to killing by kanamycin, demonstrating an approach to killing multidrug-resistant bacteria. The mechanism underlying this approach is that exogenous glucose or alanine promotes th

Biochemical and structural characterization of alanine racemase from Bacillus anthracis (Ames).

BACKGROUND: Bacillus anthracis is the causative agent of anthrax and a potential bioterrorism threat. Here we report the biochemical and structural characterization of B. anthracis (Ames) alanine racemase (AlrBax), an essential enzyme in prokaryotes and a target for antimicrobial drug development. We also compare the native AlrBax structure to a recently reported structure of the same enzyme obtained through reductive lysine methylation. RESULTS: B. anthracis has two open reading frames encoding for putative alanine racemases. We show that only one, dal1, is able to complement a D-alanine aux

Preparation and comprehensive characterization of [Hg6(alanine)4(NO3)4].H2O.

A new mercury-alanine complex has been isolated from reaction mixtures of mercurous nitrate dihydrate and alanine (L and D enantiomers). The solid-state structure contains mercury(I) and mercury(II) associated by alanine ligands in a polymeric array. The disproportionation of mercury(I) to mercury(II) and mercury(0) was facilitated by alanine and is evidenced by the appearance of mercury(0) in reactions of mercury(I) with the 20 common amino acids. This complex is the first mercury(I)-amino acid complex characterized in the solid state. The compounds have been comprehensively characterized usi

Unfolding of alanine-based peptides using electron spin resonance distance measurements.

We describe a scheme for tagging an alanine-based peptide with a Cu(II) and a nitroxide to measure unfolding transitions. The enhancement in longitudinal relaxation rate of the nitroxide due to the presence of Cu(II) was measured at physiological temperatures by pulsed electron spin resonance (ESR). The change in relaxation rate provided the average interspin distance between the Cu(II) and the nitroxide. Control experiments on a proline-based peptide verify the robustness of the method. The change in interspin distances with temperature for the alanine-based peptide is in accord with the chan

Effect of beta-alanine administration on carbon tetrachloride-induced acute hepatotoxicity.

Mice were supplemented with beta-alanine (3%) in drinking water for one week. beta-Alanine intake reduced hepatic taurine levels, but elevated cysteine levels significantly. Hepatotoxicity of CCl4 in mice fed with beta-alanine was decreased as determined by changes in serum enzyme activities. Hepatic glutathione and taurine concentrations after CCl4 challenge were increased markedly by beta-alanine intake. The enhanced availability of cysteine for synthesis of glutathione and/or taurine appears to account for the hepatoprotective effects of beta-alanine against CCl4-induced liver injury.

Mycobacterium bovis BCG vaccines exhibit defects in alanine and serine catabolism.

Mycobacterium bovis BCG is the only accepted vaccine for the prevention of tuberculosis (TB) in humans. BCG is a live vaccine, and induction of immunity to TB requires productive infection of the host by BCG. However, BCG is not a satisfactory vaccine, because it fails to protect against pulmonary TB in adults. In this study, we found that BCG strains cannot utilize many naturally occurring amino acids as the sole nitrogen source for growth. This defect is caused, at least partially, by the lack of functional metabolic enzymes. All BCG strains are unable to catabolize L-alanine or D-alanine du

L-alanine supplementation in late infantile glycogen storage disease type II.

We report a male with late infantile glycogen storage disease type II (Pompe's disease) who presented at 12 months of age with muscular hypotonia and developmental delay. Oral supplementation with L-alanine has been administered for 5 years. Progression of skeletal myopathy was slow, and cardiomyopathy resolved almost completely. L-alanine may be a valuable supplement for infants with glycogen storage disease type II.

Controversies in transfusion medicine. Alanine aminotransferase screening of blood donors: pro.

In my opinion, independent, carefully conducted scientific studies indicate that an accurate, rapid, relatively sensitive, and inexpensive laboratory test substantially reduces the major long-term risk of blood transfusion in the United States; donor ALT has emerged as one of the most effective laboratory determinants for reducing the incidence of NANB PTH. Despite its nonspecificity and limited predictive value, ALT screening may prevent up to 30 percent of cases, one-half of which would progress to chronic liver disease and then possibly to cirrhosis and hepatocellular carcinoma. Blood donor

Utilization of Alanine Dosimetry for 10 MV Photon Beam Dose Evaluation.

Radiation therapy is a crucial treatment method that delivers a high dose of radiation to localized areas. Therefore, ensuring the accuracy of the radiation dose from the radiation generator is essential. Alanine dosimetry offers the advantage of being equivalent to water and tissue and is thus a valuable tool for estimating radiation doses in the human body. In this study, we aimed to assess the feasibility of utilizing electron paramagnetic resonance (EPR)/alanine dosimetry for quality control of medical linear accelerators (LINACs). The EPR signal and slope of the dose-response curve were c

Preoperative Alanine Aminotransferase and Postoperative ALT Elevation: A Retrospective Cohort Study.

Background: Modestly elevated serum alanine aminotransferase (ALT) levels are associated with increased morbidity and mortality in the general population; however, the perioperative significance of mildly elevated preoperative ALT levels in surgical patients remains unclear. Methods: This retrospective cohort study included adults undergoing non-hepatic, non-cardiac surgery at Samsung Medical Center. Patients with preoperative ALT measured within 3 months and <2 times the upper limit of normal (ULN) were eligible. Preoperative ALT was categorized as lower (men 0-34 U/L; women 0-22 U/L) or high

Tissue-dependent enzymatic control of N-acetyl-β-alanine by PTER.

β-alanine is one of the most abundant β-amino acids in mammals and occupies a central position at the intersection of vitamin, dipeptide, and energy metabolism. In addition to dietary intake, β-alanine availability in mammalian tissues is shaped by endogenous biochemical pathways, including pyrimidine catabolism, transamination reactions, and dipeptide turnover, and contributes to the synthesis of carnosine-related dipeptides that support cellular buffering and stress responses. Beyond these established biochemical fates, β-alanine also gives rise to secondary metabolites, including N-acetyl-β

D-serine and D-alanine supplementation protects against chronic kidney disease.

Novel therapeutic targets for renal protection are imperative due to the escalating prevalence of end-stage kidney disease despite existing treatments for chronic kidney disease (CKD). We explored the reno-protective effects of D-serine and D-alanine, which have previously demonstrated efficacy in acute kidney injury against CKD. Female and male 5/6-nephrectomy mice were used as CKD models, receiving 20 mM D-serine or D-alanine supplementation, and their survival rates, renal function, and transcriptomic changes were examined. The proliferation of human tubular epithelial cells supplemented wi

Chiral Alanine Functionalized Perylene Diimides: Supramolecular Self-Assembly and Optical Properties.

Water-soluble alanine-functionalized perylene diimide derivatives (PDI-Ala) were synthesized by introducing alanine units at the imide positions of the PDI framework. Their photoluminescence properties and supramolecular aggregation behavior were investigated in solvents of varying polarity. Chiral L-PDI-Ala displayed distinct solvent-dependent emission colors and intensities, whereas the achiral PDI-Ala showed negligible emission under the same conditions. Poor solvents such as acetone and ethyl acetate promoted relatively efficient emission with hypsochromically shifted bands, while good sol

Chemotactic study of Escherichia coli towards alanine using a microfluidics platform.

Chemotaxis, the capacity of cells and microorganisms to detect and respond to chemical gradients, is integral to various biological processes, including biofilm formation, environmental decontamination, pathogen identification, and targeted drug delivery. This study involved the fabrication of a Y-shaped microfluidic device to examine the chemotactic migration of Escherichia coli K12 DH5α toward gradient concentrations of alanine, an amino acid that serves as a potent chemoattractant for E. coli. The microfluidic device was designed and fabricated using standard photolithography and soft litho
