In Silico Analysis of ACE Inhibitory Peptides from Chloroplast Proteins of Red Alga Grateloupia asiatica.

Inhibition of angiotensin I-converting enzyme (ACE) is one of the key factors to repress high blood pressure. Although many studies have been reported that seaweed protein hydrolysates showed the ACE inhibitory activity, the comprehensive understanding of the relationship was still unclear. In this study, we employed chloroplast genome for in silico analysis and compared it with in vitro experiments. We first extracted water-soluble proteins (WSP) from red alga Grateloupia asiatica, which contained mainly PE, PC, APC, and Rbc, and prepared WSP hydrolysate by thermolysin, resulting that the hyd

Crystal structure and functional characterization of the microalgal carotenoprotein AstaP-pink1.

The recently discovered family of microalgal water-soluble astaxanthin-binding proteins (AstaPs) functions in carotenoid sequestration and transfer. We present the crystal structure and characterization of AstaP-pink1, a homolog from Scenedesmus sp. We show that despite low sequence identity with the known AstaP-orange1, AstaP-pink1 also binds various xanthophylls, but in contrast to AstaP-orange1, induces a pronounced bathochromic shift and a near-UV spectral bump upon binding ketocarotenoids, which is reminiscent of photoactivated Orange Carotenoid Protein (OCP). Mutagenesis and domain-swap

Disruption of microtubular cytoskeleton induced by cryptogein, an elicitor of hypersensitive response in tobacco cells.

The dynamics of microtubular cytoskeleton were studied in tobacco (Nicotiana tabacum cv Xanthi) cells in response to two different plant defense elicitors: cryptogein, a protein secreted by Phytophthora cryptogea and oligogalacturonides (OGs), derived from the plant cell wall. In tobacco plants cryptogein triggers a hypersensitive-like response and induces systemic resistance against a broad spectrum of pathogens, whereas OGs induce defense responses, but fail to trigger cell death. The comparison of the microtubule (MT) dynamics in response to cryptogein and OGs in tobacco cells indicates tha

Cadmium detoxification strategies in two phytoplankton species: metal binding by newly synthesized thiolated peptides and metal sequestration in granules.

The aim of this study was to evaluate whether intracellular detoxification mechanisms could explain, at least partially, the different sensitivity to Cd of two freshwater green algae, Chlamydomonas reinhardtii and Pseudokirchneriella subcapitata. Subcellular Cd distribution and the synthesis of metal-binding thiolated peptides were thus examined in both algae exposed to a range of free [Cd(2+)] from 0.7 to 253 nM. Cadmium partitioning among five subcellular fractions (cellular debris, granules, organelles, heat-denaturable proteins - HDP, and heat-stable proteins - HSP) was determined after di

Designing an eco-efficient sonocatalytic process: iron-modified bamboo biochar for cyanobacteria elimination and algal organic matter valorization potential.

Cyanobacterial blooms have emerged as a persistent threat to global aquatic ecosystem stability and drinking water safety. Conventional algae removal technologies (e.g., coagulation, photocatalysis) generally face two long-standing bottlenecks: the trade-off between removal efficiency and secondary pollution from cell lysis-triggered microcystin release, and the lack of valorization pathways for hazardous algal residues. In this study, a low-cost iron/potassium hydroxide co-modified bamboo biochar (BC-900-2, prepared via one-step pyrolysis at 900 °C with a Fe/KOH molar ratio of 2:1) was develo

Toxic effects and mechanisms of nanoplastics and sulfonamide antibiotics on Scenedesmus obliquus.

The prevalence of nanoplastics (NPs) and sulfonamide antibiotics (SAs) in the aquatic environment is potentially harmful to the environment, and these pollutants are often present in the environment in the form of composite ones, thereby introducing more complex effects and hazards to the environment. Therefore, it is crucial to investigate the toxic effects of the individual target pollutants and their mixtures. In this study, we used Scenedesmus obliquus as the test organisms, two types of NPs: polystyrene (PS) and amine-modified (NH2-PS), four SAs: sulfapyridine (SPY), sulfamethazine (SMR),

Computational structural insights into alternative proteins: The impact of AlphaFold and AI-based prediction.

Proteins are fundamental biomolecules composed of amino acids linked by peptide bonds, forming complex macromolecules that determine cellular structure and biological function. The three-dimensional conformation of proteins plays a crucial role in defining their biochemical activity, stability, and interactions. In the emerging field of alternative proteins, such as microbial, algal, plant-based, and precision-fermented proteins, understanding structural organization is essential for improving functional properties including digestibility, nutritional value, texture, and enzymatic performance.

Curcumin loading in Camellia seed oil body and multi-compartment emulsion fabrication: effect of high-pressure homogenization pretreatment.

In this study, multi-compartment emulsion based on camellia seed oil bodies (CSO) were engineered for co-encapsulation of lipophilic bioactive compounds. High-pressure homogenization (HPH) was employed to modulate CSO size distribution and interfacial properties (surface hydrophobicity and protein conformation) of CSO. Results demonstrated that HPH treatment at 90 MPa dramatically reduced CSO-90 diameter from 1498.00 nm to 280.87 nm. This processing concurrently induced CSO interface protein unfolding, leading to significant increase in surface hydrophobicity and remarkable enhancement in curc

Algal and Cyanobacterial Hydrolysates as Serum-Reducing Supplements Across Mammalian, Avian and Fish Cells with Identification of Bioactive Chromopeptides.

Maintaining sustainable production of healthy and nutritious food represents a major global challenge. Cultivated meat could reduce the environmental impact of livestock production, but it remains costly, largely due to the expense of fetal bovine serum, a common cell culture media supplement. Algal and cyanobacterial proteins, particularly phycobiliproteins, represent promising sustainable sources of potentially bioactive peptides that may contribute to the development of serum-reduced or serum-free culture media. This study investigated the potential of hydrolysates prepared using the protei

Evaluation of membrane fouling at elevated temperature impacted by algal organic matter.

Membrane distillation (MD) is a thermally driven technology applied in desalination and water reuse with utilisation of sustainable energy. However, algal organic matter (AOM) could foul membrane critically and plague MD's long-term operational stability. In this study, the soluble extracellular polymeric substance (sEPS) and intracellular organic matter with bound extracellular polymeric substance (IOM + bEPS) of two algal species (Amphora coffeaeformis and Navicula incerta) were exposed to 60 °C, 70 °C and 80 °C for 8 h with polypropylene hydrophobic membrane, simulating heated AOMs contacte

Temperature effects on productivity, respiration, and dissolved organic matter degradation in subtropical Florida waters following hurricane-driven riverine runoff and brownification.

Increasing ocean warming and storm-driven terrestrial inputs are reshaping subtropical coastal ecosystems. This study investigated how elevated temperature and humic acid enrichment, under both pre- and post-hurricane (Idalia, Debby, Helene and Milton) in situ and manipulated conditions, affect the metabolic rates of phytoplankton communities on the West Florida Shelf (WFS). We hypothesized that warming, combined with hurricane-driven humic inputs, would decrease primary production rates while enhancing respiration and dissolved organic matter (DOM) degradation, shifting system toward net hete

Photolysis triggers multiple microbial responses: New insights of phosphorus compensation for algal blooms.

Photolysis and microbial degradation enabling the rapid mineralization of organic phosphorus constitute the crucial mechanism for phosphorus compensation during algal bloom outbreaks in shallow lakes. This study explored the key pathways of microbial degradation of algae-derived organic phosphorus (ADOP) exacerbated by photolysis through molecular biology techniques. The results showed that photolysis could exacerbate microbial degradation, and the effects on microbial degradation were multifaceted. The photolysis process changes the composition of dissolved organic matter (DOM) in the environ

Effects of cyclin-dependent kinase activity on the coordination of growth and the cell cycle in green algae at different temperatures.

The progression of the cell cycle in green algae dividing by multiple fission is, under otherwise unlimited conditions, affected by the growth rate, set by a combination of light intensity and temperature. In this study, we compared the cell cycle characteristics of Desmodesmus quadricauda at 20 °C or 30 °C and upon shifts between these two temperatures. The duration of the cell cycle in cells grown under continuous illumination at 20 °C was more than double that at 30 °C, suggesting that it was set directly by the growth rate. Similarly, the amounts of DNA, RNA, and bulk protein content per c

Photoresponse in the heterotrophic marine dinoflagellate Oxyrrhis marina.

Expressed rhodopsins were detected by proteomic analysis in an investigation of potential signal receptors in the cell membrane of the marine heterotrophic dinoflagellate Oxyrrhis marina (CCMP604). We inferred these to be sensory rhodopsins, a type of G-protein-coupled receptor trans-membrane signaling molecule. Because phototactic behavior based on sensory rhodopsins has been reported in other protists, we investigated the photosensory response of O. marina. This dinoflagellate exhibited strongest positive phototaxis at low levels (2-3 μE/m(2)/s) of white light when the cells were previously

Physiological, biochemical, and ultrastructural responses of the green macroalga Urospora penicilliformis from Arctic Spitsbergen to UV radiation.

Exposure of the filamentous turf green alga Urospora penicilliformis to ambient and artificial ultraviolet radiation (UVR) revealed a considerable resilient species. This explains the ability of this alga to thrive in the middle-upper intertidal zones of the Arctic sea where it is periodically exposed to environmental extremes. A transient UVR effect on photosynthesis under photosynthetically active radiation (PAR) + UV-A and PAR + UV-A + UV-B was found, but dynamic recovery of photoinhibition was observed immediately after reduction of the photon fluence rate of PAR in the absence or presence

A Natural qE- and qZ-Deficient Alga Retains Functional Plant-Like and Chlorophycean Violaxanthin De-Epoxidases.

Photosynthetic organisms protect themselves from excess light through non-photochemical quenching (NPQ), a process that safely dissipates excess light energy as heat. Current understanding of NPQ is largely drawn from a few model systems, leaving it unclear how universal NPQ mechanisms are across the green lineage. We characterize the slow, sustained NPQ of the green alga, Auxenochlorella protothecoides x symbiontica (UTEX 250-A). The Auxenochlorella genome lacks Light-Harvesting Complex Stress Related (LHCSR), one of the two proteins required for the fast energy-dependent quenching (qE) compo

Anticoagulant activity of a sulfated galactan: serpin-independent effect and specific interaction with factor Xa.

An algal sulfated galactan has high anticoagulant and antithrombotic activities. Its serpin-dependent anticoagulant action is due to promoting thrombin and factor (F)Xa inhibition by antithrombin and heparin cofactor II. Here, we evaluated the anticoagulant effect of the algal sulfated galactan using serpin-free plasma. In contrast to heparin, the sulfated galactan is still able to prolong coagulation time and delay thrombin and FXa generation in serpin-free plasma. We further investigated this effect using purified blood coagulation proteins, discovering that sulfated galactan inhibits the in

Interactions between 2-methylisoborneol and C-phycocyanin: Binding site analysis and effects of complexation on 2-methylisoborneol oxidation.

2-Methylisoborneol (2-MIB), predominantly produced by cyanobacteria in drinking water sources, is a major off-flavor compound responsible for earthy-musty odors at extremely low concentrations. Recent evidence suggests that a substantial fraction of 2-MIB exists in a bound state associated with algal-derived proteins, yet the mechanism governing this association and its influence on 2-MIB removal during advanced oxidation processes (AOPs) remain poorly understood. In this study, the influence of protein binding on 2-MIB degradation was systematically investigated in UV/H2O2 and UV/Cl2 processe
