Selection and isolation of bacteria capable of degrading dinoseb (2-sec-butyl-4,6-dinitrophenol).

Dinoseb (2-sec-butyl-4,6-dinitrophenol) has been a widely used herbicide that persists in some contaminated soils, and has been found in groundwaters, causing health and environmental hazards. Persistence in some soils may stem from a lack of dinoseb-degrading organisms. We established a chemostat environment that was strongly selective for aerobic (liquid phase) and anaerobic (sediment phase) bacteria able to degrade dinoseb. The chemostat yielded five taxonomically diverse aerobic isolates that could transform dinoseb to reduced products under microaerophilic or denitrifying conditions, but

Understanding the effects of per- and polyfluoroalkyl substances on early skin development: Role of ciliogenesis inhibition and altered microtubule dynamics.

Per- and polyfluoroalkyl substances (PFAS) are a class of highly stable chemicals, widely used in everyday products, and widespread in the environment, even in pregnant women. While epidemiological studies have linked prenatal exposure to PFAS with atopic dermatitis in children, little is known about their toxic effects on skin development, especially during the embryonic stage. In this study, we utilized human embryonic stem cells to generate non-neural ectoderm (NNE) cells and exposed them to six PFAS (perfluorooctanoic acid (PFOA), undecafluorohexanoic acid (PFHxA), heptafluorobutyric acid

Effects of Per- and Polyfluoroalkyl Compounds (PFASs) on Anaerobic Granular Sludge: Methane Production and Microbial Community Composition.

PFASs are a group of pollutants ubiquitous in the environment, for which their effects on the microbial community composition and activity of anaerobic granular sludge are still poorly understood. With our study, we aimed to provide insight into the impact of four PFASs on the methane yield of anaerobic granular sludge (AGS) using acetate as the substrate. Anaerobic granular sludge was exposed to different concentrations of 1H,1H,2H,2H-Tridecafluorooctan-1-ol (6:2FTOH), 1H,1H,2H,2H-Perfluoro-1-decanol (8:2FTOH), Tridecafluorohexane-1-sulfonic acid (PFHxS) and Perfluorooctanoic acid (PFOA). PFAS in plant-biosolids-soil systems: Distribution, fractionation, and effects on soil microbial communities.

This study examined the behavior of six U.S. Environmental Protection Agency (EPA) regulated per- and polyfluoroalkyl substances (PFAS) compounds in vegetated soils amended with Class A and Class B biosolids. Short-chain PFAS were evenly distributed throughout the 8-cm soil column, while long-chain PFAS accumulated in the lower layers. Sequential extraction showed that short-chain PFAS dominated the water-leachable fraction, while long-chain PFAS were more concentrated in the methanol-extractable fraction. Plant cultivation appeared to promote the redistribution of long-chain PFAS from the wat

Comparative Toxicity of Aquatic Per- and Polyfluoroalkyl Substance Exposure in Three Species of Amphibians.

Per- and polyfluoroalkyl substances (PFAS) are contaminants of concern due to their widespread occurrence in the environment, persistence, and potential to elicit a range of negative health effects. Per- and polyfluoroalkyl substances are regularly detected in surface waters, but their effects on many aquatic organisms are still poorly understood. Species with thyroid-dependent development, like amphibians, can be especially susceptible to PFAS effects on thyroid hormone regulation. We examined sublethal effects of aquatic exposure to four commonly detected PFAS on larval northern leopard frog

Effects of perfluorinated compounds homologues on chemical property, microbial composition, richness and diversity of urban forest soil.

Perfluorinated compounds (PFCs), as an important class of new persistent organic pollutants, are widely distributed in the environment. Yet the effects of different types and concentrations of PFCs on soil microbial community in urban forest ecosystems are remain uncertain. Here, two typical PFCs, perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS), were selected to carry out a pot experiment in greenhouse with singly and joint treatment at different concentrations, to examine their effects on composition and diversity of soil microorganisms and availability of soil macronut

Long-term efficiency of biochar amendment in PFAS-contaminated soil under unsaturated field conditions.

The present study investigates the long-term immobilization efficiency of biochar on target per- and polyfluoroalkyl substances (PFAS) and precursors in well-drained soils contaminated by aqueous film-forming foam (AFFF) (Ʃ27PFAS = 1624 ± 276 µg/kg) over 2 years. The total oxidizable precursor (TOP) assay revealed a large precursor reservoir in the soil. Fifteen outdoor field-scale columns were packed with contaminated soil (48 kg) without (control columns, triplicates) and with biochar amendments: Three sewage sludge-based biochars were homogeneously mixed into the soil at a 1% (w/w) dose in

Molecular mechanism by which perfluorooctane sulfonate regulates the initiation and progression of prostate cancer via the ENTPD5-adenine axis.

Perfluorooctane sulfonate (PFOS), a persistent member of the per- and polyfluoroalkyl substances family, has been increasingly associated with adverse carcinogenic effects; however, the molecular basis by which PFOS may contribute to prostate cancer (PCa) initiation and progression remains poorly defined. In this study, we aimed to systematically elucidate the mechanisms underlying PFOS-associated prostate carcinogenesis, with particular emphasis on the identification of actionable molecular targets and metabolically relevant pathways. By integrating network toxicology, transcriptomic differen

An innovative homogeneous electrochemistry coupled with colorimetry dual-model sensing strategy for perfluorooctane sulfonate based on Cu@CuO aerogel nanozyme.

By preparing Cu@CuO aerogel as a nanozyme which exhibits prominent peroxidase-like (POD) activity, an innovative homogeneous electrochemistry (HEC) coupled with the colorimetry dual-model sensing strategy is proposed to detect perfluorooctane sulfonate (PFOS) for the first time. Cu@CuO aerogel accelerates the oxidation process of colorless o-phenylenediamine to form yellow 2,3-diaminophenazinc (DAP), and meanwhile, DAP as an electroactive substance creates a reduction peak current upon the electrochemical measurements. Interestingly, in the presence of PFOS, the POD activity of Cu@CuO aerogel

Combating pharmaceutical folklore: No alkyl-sulfonate impurities formed during the synthesis of sulfonate salts.

Whilst an alcohol can be forced to react with a sulfonic acid, this reaction produces minimal ester conversion even under extreme conditions (anhydrous, very low pH) that bear no resemblance to the mild synthetic procedures typically used for the formation of sulfonate salts of basic drugs. The latter involve the addition of a molar equivalent of pharma-grade sulfonic acid to the base form of a drug substance (pKa ≥3.5), dissolved or suspended in an alcohol solvent, normally ethanol (pKa -2). All added acid is neutralized, and so there is no potential for ester formation. Many drug-substance b

Mobilization of per- and poly-fluoroalkyl substances (PFAS) in soils with different organic matter contents.

There is considerable interest in addressing soils contaminated with per- and polyfluoroalkyl substances (PFAS) because of the PFAS in the environment and associated health risks. The neutralization of PFAS in situ is challenging. Consequently, mobilizing the PFAS from the contaminated soils into an aqueous solution for subsequent handling has been pursued. Nonetheless, the efficiency of mobilization methods for removing PFAS can vary depending on site-specific factors, including the types and concentrations of PFAS compounds, soil characteristics. In the present study, the removal of perfluor

Factors Affecting the Adsorption of Per- and Polyfluoroalkyl Substances (PFAS) by Colloidal Activated Carbon.

The immobilization of per- and polyfluoroalkyl substances (PFAS) by colloidal activated carbon (CAC) barriers has been proposed as a potential in-situ method to mitigate the transport of plumes of PFAS in the subsurface. However, if PFAS are continuously released from a source zone, the adsorptive sites on CAC will eventually become saturated, upon which point the breakthrough of PFAS in the barrier will occur. To predict the long-term effectiveness of CAC barriers, it is important to evaluate the factors that may affect the adsorption of PFAS on CAC. In this study, the adsorption of 7 PFAS on

The vitamin D receptor as a potential target for the toxic effects of per- and polyfluoroalkyl substances (PFASs): An in-silico study.

Due to their persistence and toxicity, perfluoroalkyl and polyfluoroalkyl substances (PFASs) constitute significant hazards to human health and the environment. Their effects include immune suppression, altered hormone levels, and osteoporosis. Recently, the most studied PFAS, perfluorooctanoic acid (PFOA), was shown to competitively binding to the Vitamin D receptor (VDR). VDR plays a crucial role in regulating genes involved in maintaining immune, endocrine, and calcium homeostasis, suggesting it may be a target for at least some of the health effects of PFAS. Hence, this study examined the

Joint effects of per- and polyfluoroalkyl substance alternatives and heavy metals on renal health: A community-based population study in China.

BACKGROUND: Previous studies have indicated that chlorinated polyfluorinated ether sulfonic acids (Cl-PFESAs), when used as an alternative to per- and polyfluoroalkyl substances (PFASs), result in kidney toxicity. However, their co-exposure with heavy metals, has not yet been described. OBJECTIVES: To explore the joint effects of Cl-PFESAs and heavy metal exposure on renal health in Chinese adults, and identify specific pollutants driving the associations. METHODS: Our sample consists of 1312 adults from a cross-sectional survey of general communities in Guangzhou, China. We measured Cl-PFES

Subchronic reproductive effects of 6:2 chlorinated polyfluorinated ether sulfonate (6:2 Cl-PFAES), an alternative to PFOS, on adult male mice.

With a similar structure to perfluorooctane sulfonate (PFOS), 6:2 chlorinated polyfluorinated ether sulfonate (6:2 Cl-PFAES) has been widely used as a mist suppressant in the chromium plating industry in China since the 1970s. After being disregarded for the past 30 years, 6:2 Cl-PFAES has now been detected in environmental matrices and human sera, suggesting potential health concerns. We carried out a subchronic exposure study to investigate the reproductive toxicity of 6:2 Cl-PFAES exposure (0, 0.04, 0.2, and 1.0 mg/kg/d body weight, 56 d) in adult male BALB/c mice.
