Electrocatalytic Radical Dichlorination of Alkenes with Nucleophilic Chlorine Sources.

We report a Mn-catalyzed electrochemical dichlorination of alkenes with MgCl2 as the chlorine source. This method provides operationally simple, sustainable, and efficient access to a variety of vicinally dichlorinated compounds. In particular, alkenes with oxidatively labile functional groups, such as alcohols, aldehydes, sulfides, and amines, were transformed into the desired vicinal dichlorides with high chemoselectivity. Mechanistic data are consistent with metal-mediated Cl atom transfer as the predominant pathway enabling dual C-Cl bond formation and contradict an alternative pathway inv

Three common pathways of nephrotoxicity induced by halogenated alkenes.

Glutathione-dependent bioactivation is a common pathway in nephrotoxicity caused by haloalkanes and haloalkenes. Glutathione conjugation forms the link between halogenated hydrocarbons, based on the formation of an episulfonium ion (vicinal halomethanes) or a cysteine conjugate (haloalkenes). Herein, we review the metabolic pathways underlying the nephrotoxic effects of the three well-known haloalkenes trichloroethylene, tetrachloroethylene, and hexachloro-1:3-butadiene to emphasize the role of cysteine-conjugate β-lyase and the oxidative metabolism in renal toxicity. Activation by cysteine-co

Intramolecular aminocyanation of alkenes by N-CN bond cleavage.

A metal-free, Lewis acid promoted intramolecular aminocyanation of alkenes was developed. B(C6F5)3 activates N-sulfonyl cyanamides, thus leading to a formal cleavage of the N-CN bonds in conjunction with vicinal addition of sulfonamide and nitrile groups across an alkene. This method enables atom-economical access to indolines and tetrahydroquinolines in excellent yields, and provides a complementary strategy for regioselective alkene difunctionalizations with sulfonamide and nitrile groups. Labeling experiments with (13)C suggest a fully intramolecular cyclization pattern due to the lack of l

Electrochemical Cascade Cyclization of Sulfoxonium Ylides with Unactivated Alkenes.

Herein, we report an electrochemical cascade cyclization of sulfoxonium ylides with unactivated alkenes under mild conditions. This protocol provides a concise and sustainable route to γ-substituted 1-tetralones. Mechanistically, this oxidant- and metal-free transformation proceeds via the generation of an α-carbonyl carbon radical through the reductive proton-coupled electron transfer of sulfoxonium ylides at the cathode, which subsequently triggers a radical addition/cascade cyclization sequence with unactivated alkenes. The success of this reaction relies on the precise matching of redox po

Nickel-Catalyzed Asymmetric Carboacylation of Alkenes via CO Transfer.

We report a nickel-catalyzed asymmetric carboacylation of alkenes using acyl chlorides as bifunctional carbon and carbonyl sources. The reaction proceeds via a functional group transfer mechanism, enabling efficient access to enantioenriched ketones without the use of toxic CO gas. Precise control of CO extrusion and reinsertion affords broad substrate scope and high enantioselectivity, with good tolerance of functional groups commonly found in drug-like molecules. Mechanistic studies, including radical inhibition experiments, isotopic labeling, and DFT calculations, support a Ni0/NiII/NiIII/N

Difunctionalization of Alkenes via Photoinduced Oxidative Radical-Polar Crossover.

Alkene difunctionalization represents one of the most powerful and straightforward tools for the rapid assembly of structurally diverse organic molecules. This review provides a comprehensive overview of recent developments in the difunctionalization of alkenes enabled by photoinduced oxidative radical-polar crossover (ORPC). By merging the advantages of radical reactivity and polar reaction pathways, this strategy enables the efficient construction of diverse 1,2-difunctionalized skeletons under mild and controllable photochemical conditions. We systematically categorize these advances accord

Rhodium-Catalyzed 1,1-Arylesterification of Unactivated Alkenes with Dinucleophiles.

Recent advances have significantly expanded the scope of alkene functionalization reactions. In contrast to the well-established 1,2-difunctionalization, the 1,1-difunctionalization of alkenes presents greater synthetic challenges. Herein, we disclose a rhodium-catalyzed 1,1-arylesterification of alkenes with two different nucleophiles. By the catalysis of rhodium(III), diaryl tertiary alcohol undergoes β-C elimination, followed by sequential olefin migration insertion, elimination, and reinsertion processes. Subsequent coordination of a second nucleophile to the rhodium center ultimately yiel

Iron-Catalyzed Selective Sulfonylation of Alkenes by Sulfonyl Chlorides.

Herein, we report an iron-catalyzed approach for the generation of sulfonyl radicals from readily available bulk raw material sulfonyl chlorides and selective sulfonylation of alkenes for the synthesis of β-keto sulfones and vinyl sulfones depending on the reaction conditions. Under mild aerobic oxidation conditions in ethanol, β-keto sulfones were obtained via C(sp2)-C bond cleavage, while under argon protection in hexafluoroisopropanol, vinyl sulfones were formed. Mechanism studies reveal that iron salts play the role of hitting two birds with one stone, involving hydrogen atom transfer (HAT

TMSCl-promoted allylic phosphorothiolation and intermolecular difunctionalization of alkenes.

Herein, we describe a TMSCl-promoted, divergent strategy for the synthesis of structurally diverse S-alkyl phosphorothioates from various classes of olefins using N-phosphorothiosuccinimide as the phosphorothiolation reagent. For aromatic trisubstituted and 1,1-disubstituted olefins, the reaction proceeds via regioselective allylic phosphorothiolation. In the case of aliphatic 1,1-disubstituted olefins, an electrophilic intermolecular phosphorothiolation-difunctionalization is achieved. Interestingly, with (1-cyclopropylvinyl)benzene, a 1,5-chlorophosphorothiolation product is obtained via all

Visible-Light-Driven Iron-Catalyzed 1,2-Difluoroalkylthiolation of Alkenes.

The synthesis of medicinally interesting aryldifluoromethylated compounds has drawn significant research attention in recent years. Herein, we report an unprecedented iron-mediated process for the selective defluorination of trifluoromethylarenes to achieve the 1,2-difluoroalkylthiolation of alkenes. Preliminary mechanistic studies revealed that thiolate anion, trifluoromethylarene, and iron cation could form an electron donor-acceptor (EDA) complex, which induced selective defluorination and then difunctionalization of alkenes to obtain aryldifluoromethylated products. The generated aryldiflu

Catalytic Asymmetric Hydrogen Atom Transfer: Enantioselective Hydroamination of Alkenes.

We report a highly enantioselective radical-based hydroamination of enol esters with sulfonamides jointly catalyzed by an Ir photocatalyst, Brønsted base, and tetrapeptide thiol. This method is demonstrated for the formation of 23 protected β-amino-alcohol products, achieving selectivities up to 97:3 er. The stereochemistry of the product is set through selective hydrogen atom transfer from the chiral thiol catalyst to a prochiral C-centered radical. Structure-selectivity relationships derived from structural variation of both the peptide catalyst and olefin substrate provide key insights into

Multicomponent Sulfonylation of Alkenes to Access β-Substituted Arylsulfones.

A novel multicomponent sulfonylation of alkenes is described for the assembly of various β-substituted arylsulfones using cheap and easily available K2S2O5 as a sulfur dioxide source. Of note, the procedure does not need any extra oxidants and metal catalysts and exhibits a relatively wide substrate scope and good functional group compatibility. Mechanistically, an initial arylsulfonyl radical is formed involving the insertion of sulfur dioxide with aryl diazonium salt, followed by alkoxyarylsulfonylation or hydroxysulfonylation of alkenes.

Catalytic intramolecular aminoarylation of unactivated alkenes with aryl sulfonamides.

Arylethylamines are abundant motifs in myriad natural products and pharmaceuticals, so efficient methods to synthesize them are valuable in drug discovery. In this work, we disclose an intramolecular alkene aminoarylation cascade that exploits the electrophilicity of a nitrogen-centered radical to form a C-N bond, then repurposes the nitrogen atom's sulfonyl activating group as a traceless linker to form a subsequent C-C bond. This photoredox catalysis protocol enables the preparation of densely substituted arylethylamines from commercially abundant aryl sulfonamides and unactivated alkenes un

Cathodic Regioselective Coupling of Unactivated Aliphatic Ketones with Alkenes.

A regioselective coupling of aliphatic ketones with alkenes has been realized by cathodic reduction. This reaction enables the formation of ketyl radicals and the activation of challenging alkenes under mild electrolysis conditions, providing an effective protocol for accessing diverse tertiary alcohols with substrate-dependent regioselectivity. The practicability of this reaction is demonstrated by scale-up experiments. The hydrogen source for the products, the migration isomerization of allylarenes, and the applicability of internal alkenes are demonstrated by control experiments.

Three-component carboacylation of alkenes via cooperative nickelaphotoredox catalysis.

Various commercially available acyl chlorides, aldehydes, and alkanes were exploited for versatile three-component 1,2-carboacylations of alkenes to forge two vicinal C-C bonds through the cooperative action of nickel and sodium decatungstate catalysis. A wealth of ketones with high levels of structural complexity was rapidly obtained via direct functionalization of C(sp2)/C(sp3)-H bonds in a modular manner. Furthermore, a regioselective late-stage modification of natural products showcased the practical utility of the strategy, generally featuring high resource economy and ample substrate sco

Photocatalytic Hydroacylation of Alkenes by Directly Using Acyl Oximes.

Acyl oximes are directly used as the acyl radical precursors in the hydroacylation reactions for the first time. In this work, acyl radicals can be effectively generated via β-scission of a phosphoranyl radical under photocatalytic conditions. As a result, the hydroacylation of alkenes triggered by the resulting acyl radicals leads to facile syntheses of a range of valuable ketones.

Conformational Isomerization as a Process to Determine Selectivity over Reaction Pathways: Effect of Alkene Rotation in Chain Walking via Cis Alkene Intermediates.

In organic reactions, bond-forming and bond-cleaving processes are generally considered to be more important than other processes such as conformational isomerization. We report herein an example where a conformational isomerization process, propeller-like alkene rotation, is considered to determine the selectivity over the reaction pathways. The transition state with the highest energy barrier in some alkylpalladium isomerization (chain walking) events was theoretically indicated to correspond to alkene rotation, while transition states for bond-cleaving β-hydride elimination and bond-forming

Chromium-Catalyzed Direct Conversion of Ethers to Aryl-Substituted Alkenes.

We report a Cr-catalyzed strategy that enables the direct transformation of ethers into aryl-substituted alkenes via a radical-organometallic crossover pathway. This protocol furnishes aryl-substituted homoallylic alcohols with high regioselectivity when tetrahydrofuran is used, while acyclic ethers undergo selective C-H/C-O bond activation to afford styrene derivatives. Mechanistic studies, including radical trapping and deuterium labeling experiments, support the involvement of aryl radicals and organochromium intermediates.

Synergistic Fe/Ni catalysis for electrochemical 1,1-difunctionalization of alkenes.

Alkene 1,1-difunctionalization holds significant importance in organic synthesis due to its ability to effectively enhance the complexity and functionality of molecular frameworks. Herein, we report an electrochemical strategy for 1,1-difunctionalization of halogenated aromatics with unactivated alkenes using synergistic Fe/Ni catalysis. This system integrates redox activity of nickel with Lewis acid functionality of iron: the nickel catalyst governs aryl halide oxidative addition and alkene migration, while iron species activates catalytic sites, stabilizes radical acceptors, and precisely re
