Ethanol binge drinking exposure affects alveolar bone quality and aggravates bone loss in experimentally-induced periodontitis.

BACKGROUND: Periodontitis is a multifactorial inflammatory disease of tooth supporting tissues caused by oral biofilms, influenced by environmental and genetic factors, among others. Ethanol consumption has been considered a factor that enhances alveolar bone loss, especially in high doses. The present study aims to investigate the changes promoted by ethanol binge drinking per se or associated with ligature-induced periodontal breakdown on alveolar bone loss. MATERIALS AND METHODS: Thirty-two Wistar rats were randomly allocated into four groups: control (C), ethanol (3g/kg/day; 3 days On-4 d

Dietary Flavonols Intake and Alveolar Bone Loss: Insights From a Cross-Sectional Analysis.

BACKGROUND: Alveolar bone resorption is typically associated with aging or chronic inflammation, while flavonols intake is known for its extensive anti-aging and anti-inflammatory properties. However, the relationship between them remains unclear. OBJECTIVE: This study aimed to examine the association between dietary flavonol intake and alveolar bone loss. METHODS: Using 2009-2010 NHANES data, this cross-sectional study analyzed 2-day dietary recalls of flavonols (isorhamnetin, quercetin, and kaempferol) and alveolar bone loss-defined as teeth with bone loss exceeding 5 mm. Analyses included

Deep learning-based automated assessment of alveolar bone loss in CBCT for periodontitis.

OBJECTIVES: This study aimed to develop and validate a CBCT-based automated system for assessing radiographic alveolar bone loss (RBL) to improve the accuracy and efficiency of periodontitis diagnosis. METHODS: A total of 110 patients (2,796 teeth) with Stage I-IV periodontitis from four center were included. The nnU-Net framework was used to segment teeth, alveolar bone, and the cemento-enamel junction (CEJ). RBL was calculated automatically using an edge-constrained shortest path algorithm. The model was trained on data from Center A and externally validated with datasets from Centers B-D.

Modeling chronic periodontitis in rats: Persistent alveolar bone loss mediated by periodontal pathogens.

BACKGROUND: Although widely used in periodontal research, rodents are naturally resistant to periodontitis. Conventional models, such as ligature-induced periodontitis, often fail to sustain defects due to spontaneous tissue regeneration after ligature removal. To address this, we refined a rat ligature-induced model of experimental periodontitis to better mimic the chronic, progressive nature of human periodontitis. METHODS: As a first step, following a split-mouth design, we compared the effectiveness of 3/0 silk thread and 0.008-inch orthodontic wire as ligature materials. Ligatures were a

Atorvastatin Accelerates Alveolar Bone Loss in Type 1 Diabetic Rats Submitted to Periodontitis.

Periodontal bone loss is potentiated by diabetes. Despite the beneficial anti-inflammatory and antiresorptive effects of Atorvastatin (ATV) on periodontitis, it has been reported to increase the risk of diabetes, which may modify the course of periodontal disease. Therefore, this study aimed to evaluate the effect of ATV on alveolar bone in rats with periodontitis and diabetes. For this, 72 Wistar rats were divided into groups: Naïve (N) not submitted to any procedure; Experimental periodontitis (EP) group submitted to ligature-induced periodontitis; diabetes mellitus (DM), submitted to EP and

Autoinducer-2 produced by oral microbial flora and alveolar bone loss in periodontitis.

OBJECTIVE: The aim of this study was to investigate the association between autoinducer-2 (AI-2) of oral microbial flora and the alveolar bone destruction in periodontitis to determine if AI-2 may have the potential that monitor periodontitis and predict bone loss. BACKGROUND: Plaque biofilm was the initiating factor of periodontitis and the essential factor of periodontal tissue destruction. The formation of biofilms depended on the complex regulation of the quorum sensing (QS) system, in which bacteria could sense changes in surrounding bacterial density by secreting the autoinducer (AI) to

Resolvin D2 Restrains Th1 Immunity and Prevents Alveolar Bone Loss in Murine Periodontitis.

Periodontitis is an infectious inflammatory disease of the supporting structures of the teeth. Resolvins are part of a large family of specialized pro-resolving lipid mediators that enhance active resolution of inflammation and return of inflammatory lesions to homeostasis. In this paper, we demonstrate that resolvin D2 (RvD2), a product of docosahexaenoic acid (DHA) metabolism, prevents alveolar bone loss in Porphyromonas gingivalis-induced experimental periodontitis. Investigations of the immune mechanism of RvD2 actions reveal that 6 weeks after infection, the gingiva of RvD2-treated mice e

MicroRNAs: Potential Biomarkers and Therapeutic Targets for Alveolar Bone Loss in Periodontal Disease.

Periodontal disease is an inflammatory disease caused by bacterial infection of tooth-supporting structures, which results in the destruction of alveolar bone. Osteoclasts play a central role in bone destruction. Osteoclasts are tartrate-resistant acid phosphatase (TRAP)-positive multinucleated giant cells derived from hematopoietic stem cells. Recently, we and other researchers revealed that microRNAs are involved in osteoclast differentiation. MicroRNAs are novel, single-stranded, non-coding, small (20-22 nucleotides) RNAs that act in a sequence-specific manner to regulate gene expression at

Oral administration of 5-hydroxytryptophan aggravated periodontitis-induced alveolar bone loss in rats.

OBJECTIVE: 5-Hydroxytryptophan (5-HTP) is the precursor of serotonin and 5-HTP has been widely used as a dietary supplement to raise serotonin level. Serotonin has recently been discovered to be a novel and important player in bone metabolism. As peripheral serotonin negatively regulates bone, the regular take of 5-HTP may affect the alveolar bone metabolism and therefore influence the alveolar bone loss induced by periodontitis. The aim of this study was to investigate the effect of 5-HTP on alveolar bone destruction in periodontitis. DESIGN: Male Sprague-Dawley rats were randomly divided in

Cytomegalovirus periodontal presence is associated with subgingival Dialister pneumosintes and alveolar bone loss.

Destructive periodontal disease is associated with human cytomegalovirus (HCMV), Epstein-Barr type 1 virus (EBV-1) and other members of the Herpesviridae family as well as with various gram-negative anaerobic bacteria, including the Dialister pneumosintes species. This study aimed to determine possible interrelationships between periodontal HCMV, EBV-1, herpes simplex virus and D. pneumosintes, and relate the microbiological findings to periodontitis clinical status. Sixteen subjects each contributed paper point samples from two progressing and two stable periodontitis lesions, as determined b

Septic arthritis drives alveolar bone loss in mice through microbiota-dependent functional shifts.

AIMS: Periodontal alterations, including alveolar bone loss (ABL), can be influenced by systemic inflammatory conditions and by changes in oral microbiota. Although periodontal disease (PD) provides an established framework for understanding how dysbiosis-driven inflammation affects the periodontium, the present study did not employ a PD model. Instead, we investigated whether septic arthritis (SA), an infectious arthritis, can modulate periodontal parameters. Specifically, we evaluated changes in the oral microbiota and the contribution of the CCR2 in SA-induced periodontal alterations. METH

Focusing on ferroptosis in alveolar bone loss during periodontitis: From mechanisms to therapies.

Periodontitis is an oral immunoinflammatory disease induced by bacterial infection. During periodontitis, the aggravating destruction of the alveolar bone can result in tooth movement and even tooth loss. Current conventional treatments for periodontitis primarily focus on infection control, but their effectiveness in halting and restoring alveolar bone destruction is limited. To identify additional therapeutic targets, researchers have been dedicated to investigating other pathological mechanisms underlying alveolar bone loss during periodontitis. Recently, findings indicate that ferroptosis

Milk Kefir therapy reduces inflammation and alveolar bone loss on periodontitis in rats.

Periodontitis is a chronic inflammatory disease that affects the tooth-supporting tissues. This study evaluated the anti-inflammatory and antiresorptive effects of milk kefir (MK) on periodontitis in rats. Micro-Raman spectroscopy was performed on MK at different fermentation times to verify the presence of Lactobacillus kefiri. From these results, Wistar rats were divided into the following groups: C (Control); EP (experimental periodontitis); K1 (animals that received MK with one day of fermentation); K1+EP; K4 (animals without EP using MK with four days of fermentation) and K4+EP. MK was ad

Hyperactive lesions of gingiva associated with severe alveolar bone loss: A rare finding.

Pyogenic granuloma (PG) is an inflammatory reactive hyperplasia of connective tissue. It usually arises in response to various stimuli such as low-grade local irritation, traumatic injury, hormonal factors or certain kinds of drugs. It predominantly occurs in the second decade of life in young females and rarely may cause significantly alveolar bone loss. It managed by conservative surgical excision and removal of causative irritants. This paper presents the case of a PG in a 55-year-old male with severe alveolar bone loss in the affected site, managed by surgical intervention.

Inhibition of alveolar bone loss by matrix metalloproteinase inhibitors in experimental periodontal disease.

Periodontal disease is characterized by excessive host collagenase resulting in loss of gingival and periodontal ligament collagen and adjacent alveolar bone. Intragingival endotoxin injection induces a model of periodontal disease characterized by rapid bone loss with biochemical features similar to that of naturally occurring adult periodontitis. CH1766, a peptide with a zinc binding moeity which fits into the active site of the enzyme, and CH6631, a hydroxamic acid derivative with aryl-substituted sulphonamide residues, are inhibitors of matrix metalloproteinases (MMPIs) with differing inhi

The potential role of cfDNA-related innate immune responses in postoperative bone loss after alveolar bone grafting.

The purpose of treating alveolar bone cleft is to restore a normal maxilla structure. Multiple factors have been identified that can affect the success of alveolar bone grafting. However, with consistent treatment modifications, the surgical outcomes have been improved, but alveolar bone loss still exists. Thus, a new aspect should be found to solve this problem. As alveolar bone belongs to the periodontal tissues, the mechanism of the alveolar bone loss after bone grafting in patients with alveolar bone cleft may be similar to the development of alveolar bone loss in periodontitis. Deep Learning for the Assessment of Alveolar Bone Loss on Intraoral Radiographs: A Systematic Review.

Background. Radiographic bone loss is a primary determinant of periodontitis stage. Intraoral radiographs (periapical and bitewing) are the standard projections for assessing interproximal bone levels, yet their interpretation is subjective and poorly reproducible, and previous syntheses have pooled intraoral with panoramic imaging. Objectives. To appraise and synthesise studies developing or validating deep learning (DL) models for detecting, quantifying, staging or classifying alveolar bone loss on intraoral radiographs. Methods. Seven databases and six supplementary sources were searched fr
