Fluazuron orally administered to guinea pigs: pharmacokinetic and efficacy against Amblyomma sculptum.

BACKGROUND: Brazilian spotted fever (BSF), the most lethal tick-borne disease in the Western Hemisphere, is caused by the bacterium Rickettsia rickettsii and transmitted by the bite of Amblyomma sculptum. Capybaras are considered primary hosts of this tick and amplifier hosts of R. rickettsii, generating new infected lineages of A. sculptum in BSF-endemic areas. To define a possible treatment regimen for controlling the tick A. sculptum in capybaras, the aim of this study was to establish an effective fluazuron (FLU) dose to control A. sculptum larvae in artificially infested guinea pigs. In vitro acaricidal activity of different ectoparasiticide classes against Amblyomma sculptum larvae.

Zoonoses are major causes of morbidity and mortality worldwide. Among them, Brazilian Spotted Fever (BSF) is an important one that occurs in some regions of South America and can be transmitted by the "star tick" Amblyomma sculptum. Application of acaricides against the larval stage is important as strategy of population control. However, there is still a deficiency of studies on chemical control of A. sculptum and the present work aims to evaluate the in vitro acaricidal activity of cypermethrin, flumethrin, deltamethrin, fipronil, coumaphos and chlorpyrifos against A. sculptum larvae. Perianal tick-bite lesion caused by a fully engorged female Amblyomma testudinarium.

A perianal tick and the surrounding skin were surgically excised from a 73-year-old man residing in a southwestern costal area of the Korean Peninsula. Microscopically a deep penetrating lesion was formed beneath the attachment site. Dense and mixed inflammatory cell infiltrations occurred in the dermis and subcutaneous tissues around the feeding lesion. Amorphous eosinophilic cement was abundant in the center of the lesion. The tick had Y-shaped anal groove, long mouthparts, ornate scutum, comma-shaped spiracular plate, distinct eyes, and fastoons. It was morphologically identified as a fully

Repellent efficacy of DEET and KBR 3023 against Amblyomma hebraeum (Acari: Ixodidae).

The bont tick, Amblyomma hebraeum Koch, is the principal vector of Rickettsia africae, the agent of African tick bite fever, in southern Africa. In contrast to most other hard ticks, members of the genus Amblyomma exhibit a characteristic attack host-seeking behavior. The efficacy of topically applied 20% lotions of DEET (diethyl-3-methylbenzamide), the currently considered reference repellent substance, and KBR 3023, a recently developed piperidine compound, was evaluated against laboratory-reared A. hebraeum nymphs on treated fingers. Both substances repelled >85% of nymph attacks at 0 and 1

Dissection and internal anatomy of the giant tropical bont tick Amblyomma variegatum.

The tropical bont tick, Amblyomma variegatum, is endemic in sub-Saharan and southern Africa, as well as several Caribbean islands. This tick is deleterious for the general health of ruminants and is the primary vector of Ehrlichia ruminantium, the causative agent of heartwater. Understanding the ecology and biology of A. variegatum is thus crucial to describe tick-host-bacteria interactions and thus develop effective control methods. To better understand vector competence, detailed functional characterization and precise isolation of tick organs is needed. All procedures described in this stud

Detection of a Borrelia species in questing Gulf Coast ticks, Amblyomma maculatum.

Borrelia spp. are agents of Lyme disease and relapsing fever, diseases which use Ixodes hard ticks and Ornithodoros soft ticks, respectively, as primary vectors. Some relapsing fever spirochetes, such as B. miyamotoi, are also found in hard ticks. To date, no Borrelia sp. is known to use the hard tick, Amblyomma maculatum, as a vector. However, both B. burgdorferi and B. lonestari were recently detected in A. maculatum removed from hosts. In our study, DNA extracts from 306 questing adult A. maculatum collected in Mississippi in 2009 and 2010 were tested for Borrelia spp. DNA by PCR amplificat

Efficacy of local essential oils against Amblyomma variegatum tick from Burkina Faso.

The native tick Amblyomma variegatum remains one of the most important tick species affecting cattle in West Africa. This hinders animal production by negatively impacting the health and reproduction of animals infested with the tick. Given the negative consequences on production quality and environmental health, the use of chemical products for tick control is increasingly being discouraged. Therefore, this study aimed to assess the acaricidal activity of essential oils such as Ageratum conyzoïdes, Cymbopogon citratus, Cymbopogon giganteus, Lippia multiflora and Ocimum gratissimum against spe

Rickettsia bellii in Amblyomma rotundatum ticks parasitizing Rhinella jimi from northeastern Brazil.

This study evaluated rickettsial infection in Amblyomma rotundatum ticks collected from toads (Rhinella jimi) in the Brazilian Caatinga biome, an unique semiarid region of South America. Tick infestations were observed in 57.8% toads (26/45); mean infestation: 1.6 ticks/toad. DNA extraction from 42 ticks (6 larvae, 22 nymphs and 11 female adults) was tested by polymerase chain reaction (PCR) targeting Rickettsia organisms, which were detected in 100% of the ticks. Amplicons' DNA sequences were identical to each other and 99% identical to Rickettsia bellii from GenBank. DNA samples extracted fr

Cross-reactivity between antigens from Amblyomma cajennense and A. hebraeum (Acari: Ixodidae).

Laboratory animals exposed to feeding ticks develop resistance which is reflected by a decline in tick engorgement weight, egg-laying by adults and reduced egg viability. Serum antibodies from these hosts and their reaction with tick antigens have been detected by different methods, including precipitation techniques, immunofluorescent techniques, ELISA and Western blots. However, little is known about the effects of antibodies on ticks that engorge on resistant hosts, or which tissues of the tick body are possibly immunogenic. Some researchers, using immunohistochemistry, have detected host a

Synganglial morphology and neurosecretory centers of adult Amblyomma americanum (L.) (Acari: Ixodidae).

Lone star ticks, Amblyomma americanum (L.), were processed by standard histological means for paraffin embedding, sectioning, and staining by the paraldehyde-fuchsin technique. The synganglion is highly condensed around the esophagus and possesses paired optic, cheliceral, palpal, pedal I-IV nerves, and opisthosomal nerves and a single unpaired esophageal nerve. Although optic nerves were observed leading from the eyes to the protocerebrum, distinct optic ganglia were not seen in any of the preparations examined. The paraldehyde-fuchsin technique revealed 14 neurosecretory centers (11 paired,

Amblyomma maculatum-associated rickettsiae in vector tissues and vertebrate hosts during tick feeding.

Rickettsia parkeri, a causative agent of spotted fever rickettsiosis, is transmitted by Amblyomma maculatum (Gulf Coast tick), a tick that may also carry a non-pathogenic spotted fever group Rickettsia, "Candidatus Rickettsia andeanae". Here, we evaluated R. parkeri and "Candidatus R. andeanae" in tissues from A. maculatum prior to, during, and after blood feeding on rabbits. Using colony-reared A. maculatum that were capillary-fed uninfected cells, R. parkeri, "Candidatus R. andeanae", or both rickettsiae, we detected higher levels of Rickettsia spp. in the respective treatment groups. Efficacy of sarolaner (Simparic™) against induced infestations of Amblyomma cajennense on dogs.

BACKGROUND: Amblyomma cajennense is the main vector of Rickettsia rickettsii which causes Brazilian spotted fever. This adult tick preferably infests horses and capybaras, but has low host specificity during its immature stages, thus posing a threat to humans and dogs. In this study, the efficacy of sarolaner (Simparic™/Simparica®, Zoetis) when administered once orally to dogs at 2 mg/kg was evaluated against induced infestations of A. cajennense nymphs for up to 35 days after treatment. METHODS: Based on pretreatment tick counts, 20 dogs were randomly allocated to treatment with sarolaner (S

Amblyomma rotundatum (Koch, 1844) (Acari: Ixodidae) two-host life-cycle on Viperidae snakes.

Amblyomma rotundatum is an ixodid tick that infests ectothermic animals and reproduces exclusively by parthenogenesis. This tick has been frequently reported to infest reptiles and amphibians, under natural conditions and sometimes in captivity. It was described in Brazil and several other countries of South, Central and North America. Although many studies have reported aspects of its biology, none of them has used regularly either ophidian as hosts, or controlled temperature, humidity and luminosity for parasitic stages. The objective of this experiment was to study the life cycle of A. rotu

Aspects of the life cycle of Amblyomma parvum (Acari: Ixodidae) under natural conditions.

Monthly samplings of the rodent Galea musteloides (Caviidae) were carried out from January 2005 to December 2006 in Quilino, Córdoba Province, Argentina, to determine the seasonal distribution of immature stages of Amblyomma parvum. In the same locality, cattle and goats were also monthly examined to asses the seasonality of the females of this species. Engorged ticks collected on their natural hosts were maintained in the laboratory at 25 degrees C and 83-86% relative humidity to study biological parameters, and groups of larvae and nymphs were also maintained at three different photoperiods

Hosts, seasonality and geographic distribution of the South African tortoise tick, Amblyomma marmoreum.

The tortoise tick Amblyomma marmoreum was collected from large numbers of reptiles and other animals during the course of numerous surveys conducted in South Africa. A total of 1229 ticks, of which 550 were adults, were recovered from 309 reptiles belonging to 13 species, with leopard tortoises, Geochelone pardalis being the most heavily infested. The 269 birds sampled harboured 4901 larvae, 217 nymphs and no adult ticks, and the prevalence of infestation was greatest on helmeted guineafowls, Numida meleagris. Only two larvae were recovered from 610 rodents, including 31 spring hares, Pedetes

Assessing the potential risk of lone star tick, Amblyomma americanum, establishment in California.

The lone star tick (LST), Amblyomma americanum, is an aggressive human biter and important vector of the causative agents of tick-borne diseases. As geographic ranges of multiple Amblyomma species rapidly expand due to habitat changes and human and other animal movement patterns, there are concerns for establishment of LST in US states with suitable climates, such as California, especially given a dearth in gaps in tick surveillance. Importantly, previous niche modeling confirmed habitat in California along the length of the coast and coast range mountains. We sought data on LSTs and LST risk
