Ameloblastic carcinoma in horses: case report and literature review.

Ameloblastic carcinoma is a malignant odontogenic neoplasm that has been reported only rarely in veterinary species. A 16-y-old Arabian crossbred mare was presented for evaluation of a hard mass on the body of the mandible, with evidence of osteolysis on radiographs. Incisional biopsies revealed an invasive neoplasm comprised of spindloid epithelial cells with a high mitotic count and partial dual cytokeratin-vimentin immunoreactivity. The horse was euthanized because of rapid tumor progression 3 mo after presentation. Postmortem evaluation revealed partial obliteration of the mandible by a la

Aggressive ameloblastic fibro-odontoma assessment with CBCT and treatment.

BACKGROUND: Ameloblastic fibro-odontomas (AFO) are rare lesions and defined by the World Health Organisation as a tumour composed of a proliferation of odontogenic epithelium immersed in ectomesenchyme reminiscent of the interdental papilla. It also presents inductive processes leading to formation of enamel and dentine, confusing, histopathologically, with odontoma. Despite numerous efforts, there is still considerable confusion over its controversial aetiopathogenesis and treatment. A brief review of the literature on the clinical, pathological and therapeutic features of this lesion is repo

Delayed tooth eruption associated with an ameloblastic fibro-odontoma.

Delayed eruption of a single primary tooth is an uncommon event. Excluding a previous traumatic insult, the presence of a pericoronal odontogenic cyst or neoplasm is the primary cause for this abnormality. This case report describes the clinical and radiographic features of an ameloblastic fibro-odontoma in a young child, who presented with delayed eruption of the primary mandibular canine and prominent buccal expansion. A differential diagnosis for mixed, radiolucent and radiopaque lesions of the jaws will be discussed.

A histochemical study of apoptosis in the reduced ameloblasts of erupting mouse molars.

Apoptotic reactions were demonstrated with a commercial detection kit in the reduced ameloblasts around the tip of the cusp just before and after exposure to the mouth but, as tooth eruption advanced, they were no longer seen in the remainder of the reduced ameloblasts. Although some of the reduced ameloblasts appear to be expelled as a consequence of apoptosis, the remainder appear to constitute the junctional epithelium.

Ultrastructural changes of secreting rat-incisor ameloblasts following administration of vincristine and vinblastine.

Ultrastructural changes of secreting rat-incisor ameloblasts after administration of vincristine and vinblastine were observed. Polarity of nuclei and cytoplasmic organellae was remarkably disturbed; consequently, the regular secreting orientation of cellular products was altered greatly, leading to heterotopic deposition of large amounts of enamel matrix.

Alpl ablation in dental epithelium disrupts ameloblasts and incisor enamel mineralization in male mice.

Tissue-nonspecific alkaline phosphatase (gene: ALPL; protein: TNAP) is a mineralization-associated enzyme that catalyzes several physiological substrates and is widely expressed in skeletal and nonskeletal tissues indicating diverse physiological actions. Loss-of-function mutations in ALPL cause hypophosphatasia (HPP), an inherited skeletal dysplasia featuring rickets and osteomalacia. Hypophosphatasia is also associated with dentoalveolar abnormalities, including dentin defects and acellular cementum hypoplasia, resulting in premature tooth loss. Some reports indicate HPP causes enamel defect

Claudin-16 Deficiency Impairs Tight Junction Function in Ameloblasts, Leading to Abnormal Enamel Formation.

Claudin-16 protein (CLDN16) is a component of tight junctions (TJ) with a restrictive distribution so far demonstrated mainly in the kidney. Here, we demonstrate the expression of CLDN16 also in the tooth germ and show that claudin-16 gene (CLDN16) mutations result in amelogenesis imperfecta (AI) in the 5 studied patients with familial hypomagnesemia with hypercalciuria and nephrocalcinosis (FHHNC). To investigate the role of CLDN16 in tooth formation, we studied a murine model of FHHNC and showed that CLDN16 deficiency led to altered secretory ameloblast TJ structure, lowering of extracellula

Ultracytochemical demonstration of ATP-dependent calcium pump in ameloblasts of rat incisor enamel organ.

The enamel organ of the growing rat incisor was fixed with a mixture of formaldehyde and glutaraldehyde and processed for ultracytochemical demonstration of Ca- and Mg-activated membrane ATPase by a one-step lead technique at alkaline pH. To inhibit nonspecific alkaline phosphatase, 5 mM levamisole was added to the incubation media. Intense Ca- and Mg-ATPase activity was demonstrated in the cell surfaces of the secretory ameloblasts, except at the proximal and distal junctional complexes and the gap junctions in the lateral and basal cell surfaces. Deep plasma membrane invaginations at the pro

Ameloblastic Fibroma: Clinical Perspectives on a Rare Odontogenic Lesion.

Ameloblastic fibroma (AF) is a rare non-cancerous growth in the jaw that involves abnormal development of the epithelial and connective tissue parts of a tooth. A 33-year-old female presented with bleeding gums, mobile anterior mandibular teeth, and persistent halitosis. Initial oral examination and orthopantomogram revealed a unilocular radiolucent lesion with partially sclerotic borders, associated with an unerupted supernumerary tooth in the mandibular ramus. Computed tomography demonstrated cortical plate thinning around the lesion. Given the clinical and radiographic presentation, a diffe

Solitary ameloblastic fibroma with impacted teeth: A case report.

This case report aimed to describe a rare benign mandibular tumour and assess the outcomes of the most recent reviews, between January 2017 and August 2023. Presenting a detailed clinical case, this study advances our understanding of the diagnostic and therapeutic aspects, ultimately improving the management of similar cases in clinical practice. Orthopantomogram (OPG) revealed a well-defined unilocular radiolucency extending from the midline of the ramus and teeth 47 and 48 were submerged at the base of the mandible. A rare case of secondary ameloblastic carcinoma in a young Man.

Ameloblastic carcinoma is a rare and aggressive malignant odontogenic tumour that can arise de novo or from a preexisting benign lesion. It most frequently involves the mandible, and its clinical course is aggressive with extensive local destruction. Although rare, these lesions have been known to metastasize, mostly to regional lymph nodes or lungs. Surgical therapy, eventually followed by radiotherapy, is the treatment modality most frequently used, while the role of chemotherapy remains unclear. Here we present a case of secondary ameloblastic carcinoma of the mandible in a 33-year-old male

MMP20 Overexpression Disrupts Molar Ameloblast Polarity and Migration.

Ameloblasts responsible for enamel formation express matrix metalloproteinase 20 (MMP20), an enzyme that cleaves enamel matrix proteins, including amelogenin (AMELX) and ameloblastin (AMBN). Previously, we showed that continuously erupting incisors from transgenic mice overexpressing active MMP20 had a massive cell infiltrate present within their enamel space, leading to enamel mineralization defects. However, effects of MMP20 overexpression on mouse molars were not analyzed, although these teeth more accurately represent human odontogenesis. Therefore, MMP20-overexpressing mice ( Mmp20+/+Tg+)

TEM observations on the ameloblast/enamel interface in the rat incisor.

The structure of rat incisor enamel is established at the topographically complex interface between secretory ameloblasts and forming enamel. The aim of this study was to gain additional information on this interface by sectioning parallel with the rows and the long axis of Tomes' processes and prisms. Rats were sacrificed and fixed by glutaraldehyde/paraformaldehyde perfusion. After dissection, demineralization and embedding transverse jaw/incisor segments were cut, reembedded, and reoriented. Sections were prepared for and observed in the transmission electron microscopy (TEM). The intraenam

Ameloblastic carcinoma: a clinicopathologic study and assessment of eight cases.

The term ameloblastic carcinoma is differentiated from the term malignant ameloblastoma and is defined as an ameloblastoma in which there is histologic evidence of malignancy in the primary tumor or the recurrent tumor (or metastasis), regardless of whether it has metastasized. Eight cases of ameloblastic carcinoma from the Armed Forces Institute of Pathology (AFIP) are reported. The mean age of patients was 30.1 years, with no sex predilection noted. Seven cases involved the mandible and one involved the maxilla, with the posterior regions favored. The most common sign was swelling, although

G protein-coupled receptor Gpr115 (Adgrf4) is required for enamel mineralization mediated by ameloblasts.

Dental enamel, the hardest tissue in the human body, is derived from dental epithelial cell ameloblast-secreted enamel matrices. Enamel mineralization occurs in a strictly synchronized manner along with ameloblast maturation in association with ion transport and pH balance, and any disruption of these processes results in enamel hypomineralization. G protein-coupled receptors (GPCRs) function as transducers of external signals by activating associated G proteins and regulate cellular physiology. Tissue-specific GPCRs play important roles in organ development, although their activities in tooth

Carbonic anhydrase II regulates differentiation of ameloblasts via intracellular pH-dependent JNK signaling pathway.

Differentiation of ameloblasts from undifferentiated epithelial cells is controlled by diverse growth factors, as well as interactions between epithelium and mesenchyme. However, there is a considerable lack of knowledge regarding the precise mechanisms that control ameloblast differentiation and enamel biomineralization. We found that the expression level of carbonic anhydrase II (CAII) is strongly up-regulated in parallel with differentiation of enamel epithelium tissues, while the enzyme activity of CA was also increased along with differentiation in ameloblast primary cultures. The express
