The endospore forming bacterium Bacillus anthracis causes lethal anthrax disease in humans and animals. The ability of this pathogen to replicate within macrophages is dependent upon the display of bacterial surface proteins attached to the cell wall by the B. anthracis Sortase A ((Ba)SrtA) enzyme. Previously, we discovered that the class A (Ba)SrtA sortase contains a unique N-terminal appendage that wraps around the body of the protein to contact the active site of the enzyme. To gain insight into its function, we determined the NMR structure of (Ba)SrtA bound to a LPXTG sorting signal analog

Detoxification of xenobiotic compounds and heavy metals is a pivotal capacity of organisms, in which glutathione (GSH) plays an important role. In plants, electrophilic herbicides are conjugated to the thiol group of GSH, and heavy metal ions form complexes as thiolates with GSH-derived phytochelatins (PCs). In both detoxification processes of plants, phytochelatin synthase (PCS) emerges as a key player. The enzyme is activated by heavy metal ions and catalyzes PC formation from GSH by transferring glutamylcysteinyl residues (gamma-EC) onto GSH. In this study with Arabidopsis, we show that PCS

In vitro transcription has proven to be a successful tool for preparation of functional RNAs, especially in the tRNA field, in which, despite the absence of post-transcriptional modifications, transcripts are correctly folded and functionally active. Human mitochondrial (mt) tRNA(Lys) deviates from this principle and folds into various inactive conformations, due to the absence of the post-transcriptional modification m(1)A9 which hinders base-pairing with U64 in the native tRNA. Unavailability of a functional transcript is a serious drawback for structure/function investigations as well as in

Phytochelatin synthase (PCS) catalyzes the final step in the biosynthesis of phytochelatins, which are a family of cysteine-rich thiol-reactive peptides believed to play important roles in processing many thiol-reactive toxicants. A modified Arabidopsis thaliana PCS sequence (AtPCS1) was active in Escherichia coli. When AtPCS1 was overexpressed in Arabidopsis from a strong constitutive Arabidopsis actin regulatory sequence (A2), the A2::AtPCS1 plants were highly resistant to arsenic, accumulating 20-100 times more biomass on 250 and 300 microM arsenate than wild type (WT); however, they were h

Streptococcus gordonii is a commensal oral organism. Harmless in the oral cavity, S. gordonii is an opportunistic pathogen. S. gordonii adheres to body surfaces using surface adhesive proteins (adhesins), which are critical to subsequent formation of biofilm communities. As in most Gram-positive bacteria, S. gordonii surface proteins containing the C-terminal LPXTG motif cleavage sequence are processed by sortase A (SrtA) to become covalently attached to the cell wall. To characterize the functional diversity and redundancy in the family of SrtA-processed proteins, an S. gordonii DL1 markerles

Proteins containing a C-terminal thioester are important intermediates in semisynthesis. Currently there is one main method for the synthesis of protein thioesters that relies upon the use of engineered inteins. Here we report a simple strategy, utilizing sortase A, for routine preparation of recombinant proteins containing a C-terminal (α)thioester. We used our method to prepare two different anthrax toxin cargo proteins: one containing an (α)thioester and another containing a D-polypeptide segment situated between two protein domains. We show that both variants can translocate through protec

Gram-positive pathogenic bacteria display proteins on their surface that play important roles during infection. In Staphylococcus aureus, these surface proteins are anchored to the cell wall by two sortase enzymes, SrtA and SrtB, that recognize specific surface protein sorting signals. The role of sortase enzymes in bacterial virulence was examined using a murine septic arthritis model. Intravenous inoculation with any of the Delta(srtA), Delta(srtB) or Delta(srtAB) mutants resulted in significantly increased survival and significantly lower weight loss compared with the parental strain. Mice

Alzheimer's disease (AD) is the major cause of dementia and one of the most common chronic diseases affecting aging populations. As such, novel and effective therapeutic agents are urgently needed to address this growing public health challenge. Given the involvement of glutaminyl cyclase (QC) and cannabinoid type 2 receptor (CB2R) dysregulation in AD progression, we designed, synthesized, and evaluated a series of biphenyl carboxamide derivatives as dual modulators targeting QC and CB2R. In vitro assays showed that most of the 35 compounds exhibited notable QC inhibitory and CB2R agonistic ac

Corynebacterium striatum and Staphylococcus aureus are opportunistic pathogens that share ecological niches and are frequently co-isolated in healthcare-associated infections. They commonly exist as sessile populations organized within multi-species biofilms associated with medical devices. Sortase A (SrtA) plays a critical role in anchoring surface proteins in S. aureus, thereby influencing adhesion, colonization, and biofilm formation. This study evaluated the influence of C. striatum on the formation and stability of biofilms produced by wild-type and isogenic ΔsrtA mutant S. aureus on hydr

Dental caries is initiated by the adherence of Streptococcus mutans to tooth surfaces, leading to biofilm (plaque) formation and the production of enamel-eroding acids. Sortase A (SrtA) plays a critical role in this process because it anchors many protein adhesins to the bacterial cell wall, making it a promising target for plaque prevention. Our previous research demonstrated that edible polyphenols from maple extracts-nortrachelogenin-8'-O-β-D-glucopyranoside, lariciresinol, isoscopoletin, and (-)-epicatechin gallate (ECG)-inhibit SrtA in Listeria monocytogenes and act as antibiofilm agents.

Streptococcus pneumoniae (S. pneumoniae) is a major Gram-positive opportunistic pathogen that causes pneumonia, bacteremia, and other fatal infections. This bacterium is responsible for more deaths than any other single pathogen in the world. Inexplicably, these symptoms persist despite the administration of effective antibiotics. Targeting pneumolysin (PLY) and sortase A (SrtA), the major virulence factors of S. pneumoniae, this study uncovered a novel resistance mechanism to S. pneumoniae infection. Using protein phenotype assays, we determined that the small molecule inhibitor alnustone is

Increasing attention has been focused on the health of vegetables and grains grown in the contaminated agricultural soil, it is thus meaningful to find ways to reduce the heavy metals (HMs) accumulation in plants. As sulfur is considered to be an essential macronutrient for plant stress defenses, the important role of sulfur assimilation in plants responding to HMs stress has been followed. However, the potential mechanism of the only sulfur-containing gasotransmitter hydrogen sulfide (H2S) and its main endogenously generated substrate, cysteine (Cys), in plant defense is poorly understood. Cinnamaldehyde (CA) from Cinnamomum cassia shows strong antimicrobial activity against MRSA. This study used proteomics and high-resolution mass spectrometry with an MRSA-infected cell model to explore CA's antimicrobial mechanisms. CA covalently modifies Sortase A's active site cysteine (Cys184), a key enzyme for anchoring virulence factors to the MRSA cell wall. This modification inhibits Sortase A's activity, blocking virulence factor anchoring and impairing MRSA's adhesion, infection, and immune evasion. CA also covalently modifies Aconitase's [4Fe-4S] cluster active sites (Cys509, Cys512)

UNLABELLED: Enterococcus faecalis sortase-assembled endocarditis and biofilm-associated pili (Ebp) are virulence factors implicated in enterococcal biofilm-associated infections and gastrointestinal colonization. We previously showed that E. faecalis biofilm metabolism is influenced by extracellular electron transfer (EET) under iron-rich conditions, raising the question of whether Ebp pili also play a role in EET. Here, we report a novel role of Ebp pili in E. faecalis adhesion to the iron oxides magnetite, goethite, and hematite, where the EbpA tip adhesin contributes to this interaction. Ad

OBJECTIVES: Nonencapsulated Streptococcus pneumoniae (NESp) is emerging after the introduction of pneumococcal conjugate vaccines (PCVs). This study aimed to elucidate the genetic characteristics of penicillin-binding proteins (PBPs; PBP1a, 2b, and 2x) associated with penicillin nonsusceptibility in emergent NESp.
METHODS: A total of 71 NESp isolates that were identified in our previous study during the PCV era in Japan (2011-2019) were analyzed for their amino acid sequences of transpeptidase domain in PBP 1a, 2b, and 2x.
RESULTS: Overall, we identified 21 different PBP profiles (1a-2b-2x),

Penicillin-non-susceptible Streptococcus pneumoniae (PNSP) were first detected in the 1960s, and are now common worldwide, predominantly through the international spread of a limited number of strains. Extant PNSP are characterized by mosaic pbp2x, pbp2b and pbp1a genes generated by interspecies recombinations, with the extent of these alterations determining the range and concentrations of β-lactams to which the genotype is non-susceptible. The complexity of the genetics underlying these phenotypes has been the subject of both molecular microbiology and genome-wide association and epistasis a

Bacillus anthracis, the spore-forming agent of anthrax, requires iron for growth and is capable of scavenging heme-iron during infection. We show here that the B. anthracis iron-regulated surface determinants (isd) locus encompasses isdC, specifying a heme-iron binding surface protein. Anchoring of IsdC to the cell wall envelopes of vegetative bacilli requires srtB, which encodes sortase B. Purified sortase B cleaves IsdC between the threonine and the glycine of its NPKTG motif sorting signal. B. anthracis variants lacking either isdC or srtB display defects in heme-iron scavenging, suggesting

Sortase A (SrtA) and Sortase B (SrtB) promote S. aureus adhesion by anchoring a variety of virulence proteins to the surface of the cell wall, which in turn promotes the formation of bacterial biofilms and triggers cytotoxicity and inflammation. Therefore, inhibiting the function of SrtA and SrtB to block the anchoring of virulence proteins is a promising mechanism to alleviate the pathogenicity of S. aureus. In this study, we found that a natural compound named hyperin occupied the active pockets of SrtA and SrtB and inhibited their transpeptide capacity, which resulted in a reduction in bact
