Effect of post-harvest ultrasound treatment on phytochemical enhancement in Gynura procumbens leaves and their protection against oxidative stress-induced muscle atrophy.

BACKGROUND: Gynura procumbens is a medicinal plant widely recognized for its antioxidant and anti-inflammatory properties. Despite its traditional use in Southeast Asia, its potential role in promoting muscle health has not been thoroughly investigated. Sarcopenia, characterized by the progressive loss of muscle mass and strength during aging, remains a significant public health concern with limited therapeutic options. This study aimed to enhance the phytochemical content of G. procumbens leaves through ultrasound treatment and to evaluate the biological effects of treated extracts on muscle

Changes in phenolic compounds production as a defensive mechanism against hydrogen sulfide pollution in Scrophularia striata.

Increasing pollutants such as hydrogen sulfide (H2S) from industrial activities is an ecological challenge for plants, which seriously affects their health and productivity. Scrophularia striata is a plant endemic to Iran growing in the province of Ilam, wherein a gas refinery releases toxic agents such as H2S whose detrimental effects on the function and tolerability of medicinal plants in this region have yet to be elucidated. Thus, we initiated a hydroponic study into hormetic effect of sodium hydrogen sulfide (NaHS) concentrations (0, 3 and 7 mM) as H2S-donor at different time points on ox

Ornithine transcarbamylase and arginase I deficiency are responsible for diminished urea cycle function in the human hepatoblastoma cell line HepG2.

A possible cell source for a bio-artificial liver is the human hepatblastoma-derived cell line HepG2 as it confers many hepatocyte functions, however, the urea cycle is not maintained resulting in the lack of ammonia detoxification via this cycle. We investigated urea cycle activity in HepG2 cells at both a molecular and biochemical level to determine the causes for the lack of urea cycle expression, and subsequently addressed reinstatement of the cycle by gene transfer. Metabolic labelling studies showed that urea production from 15N-ammonium chloride was not detectable in HepG2 conditioned m

Current state of the treatment landscape of phenylketonuria.

BACKGROUND: Phenylketonuria (PKU) is an inborn error of amino acid metabolism caused by a deficiency of the L-phenylalanine-4-hydroxylase enzyme or its cofactor tetrahydrobiopterin (BH4) resulting in increased levels of phenylalanine (Phe) in blood and cerebrospinal fluid. Symptoms include hypopigmentation, a musty, mouse-like smell and various neurological complications. MAIN TEXT: Treatment options include (i) dietary restriction of Phe with supplementation of other amino acids and micronutrients through medical mixtures, (ii) daily dosing of large neutral amino acids, (iii) synthetic forms

The Chinese soft-shelled turtle, Pelodiscus sinensis, decreases nitrogenous excretion, reduces urea synthesis and suppresses ammonia production during emersion.

The objective of this study was to examine the effects of 6 days of emersion on nitrogen metabolism and excretion in the Chinese soft-shelled turtle, Pelodiscus sinensis. Despite having a soft shell with a cutaneous surface that is known to be water permeable, P. sinensis lost only ~2% of body mass and was able to maintain its hematocrit and plasma osmolality, [Na(+)] and [Cl(-)] during 6 days of emersion. During emersion, it ameliorated water loss by reducing urine output, which led to a reduction (by 29-76%) in ammonia excretion. In comparison, there was a more prominent reduction (by 82-99%

Long-term outcome of urea cycle disorders: Report from a nationwide study in Japan.

Urea cycle disorders (UCDs) are inherited metabolic disorders with impaired nitrogen detoxification caused by defects in urea cycle enzymes. They often manifest with hyperammonemic attacks resulting in significant morbidity or death. We performed a nationwide questionnaire-based study between January 2000 and March 2018 to document all UCDs in Japan, including diagnoses, treatments, and outcomes. A total of 229 patients with UCDs were enrolled in this study: 73 males and 53 females with ornithine transcarbamylase deficiency (OTCD), 33 patients with carbamoylphosphate synthetase 1 deficiency, 4

Argininosuccinic aciduria: clinical and biochemical phenotype findings in Malaysian children.

Argininosuccinic aciduria is an inborn error of the urea cycle caused by deficiency of argininosuccinate lyase (ASL). ASL-deficient patients present with progressive intoxication due to accumulation of ammonia in the body. Early diagnosis and treatment of hyperammonemia are necessary to improve survival and prevent long-term handicap. Two clinical phenotypes have been recognized--neonatal acute and milder late-onset form. We investigated patients with hyperammonemia by a stepwise approach in which quantitative amino acids analysis was the core diagnostic procedure. Here, we describe the clinic

[Earthworm feces-induced systemic resistance of cucumber against anthracnose].

Pot experiment showed that mixing earthworm feces into soil induced a significantly lower (P < 0.05) cucumber anthracnose than mixing peat, but had no significant difference (P > 0.05) with the treatment of system acquired resistance. The activities of polyphenol oxidase, peroxidase and phenylalanine ammonia-lyase in cucumber leaves treated with and without pathogen were increased in different degrees by mixing earthworm feces, compared with by mixing peat. All these suggested that mixing with earthworm feces could induce the system resistance of cucumber and suppress the plant disease via sta

Culture filtrate of Lasiodiplodia theobromae restricts the development of natural resistance in Brassica nigra plants.

Culture filtrate of Lasiodiplodia theobromae increased respiration rate, phenylalanine ammonia lyase activity, and levels of hydrogen peroxide, lipid peroxides and salicylic acid in B. nigra plants. Salicylic acid (SA) level increased for 1 hr of interaction and reduced later. Development of systemic acquired resistance (SAR) was found restricted in plants infected with L. theobromae due to deficiency of SA, which is a major signal for development of SAR. Exogenously supplied SA did develop resistance and plant death was delayed. It was hypothesized that deficiency of SA could be due to jasmon

Biochemical responses of Camellia sinensis (L.) O. Kuntze to heavy metal stress.

Three heavy metals-mercury (II), copper (II) and nickel (II), each at a concentration of 10 and 100 micrograms/ml, were tested for their effects on various biochemical constituents of tea leaves. Both NI (II) and Hg (II) decreased the phenolic contents, while Cu (II) increased it to some extent. The metal treatments enhanced the activity of phenyl alanine ammonia lyase (PAL), while the activity of poly phenol oxidase (PPO) showed a decline. Heavy metal stress also decreased the chlorophyll content of the leaves, along with a significant reduction in Hill activity. Proline content increased sig

Farnesoid X receptor agonist tropifexor detoxifies ammonia by regulating the glutamine metabolism and urea cycles in cholestatic livers.

Hyperammonemia refers to elevated levels of ammonia in the blood, which is an important pathological feature of liver cirrhosis and hepatic failure. Preclinical studies suggest tropifexor (TXR), a novel non-bile acid agonist of Farnesoid X Receptor (FXR), has shown promising effects on reducing hepatic steatosis, inflammation, and fibrosis. This study evaluates the impact of TXR on hyperammonemia in a piglet model of cholestasis. We here observed blood ammonia significantly elevated in patients with biliary atresia (BA) and was positively correlated with liver injury. Targeted metabolomics and

Management of pegvaliase-related skin concerns: best practice recommendations using a modified Delphi approach.

INTRODUCTION: The approval of pegvaliase (PALYNZIQ®) represented a paradigm shift in the management of phenylketonuria (PKU) by enabling sustained reductions in blood phenylalanine levels and permitting an unrestricted diet. However, as it is a bacterially derived injectable therapy, individuals receiving pegvaliase may experience skin-related adverse events (AEs). Anticipating these AEs and other pegvaliase-related skin concerns and having best-practice strategies to manage them may be crucial to achieving optimal patient outcomes. METHODS: An expert panel comprising eight clinicians from th

Factors affecting aspartase activity.

Depue, Robert H. (Hahnemann Medical College, Philadelphia), and Albert G. Moat. Factors affecting aspartase activity. J. Bacteriol. 82:383-386. 1961.-Cells of Escherichia coli grown in a glucose-mineral salts medium contain about one-fifth the amount of aspartase activity observed in cells grown in a yeast extract-peptone medium. The aspartase activity of the cells grown in glucose-salts medium would appear to be too low to provide a mechanism for synthesis of amino groups. Aspartase was purified approximately eightfold by ammonium sulfate precipitation and column chromatography of cell-free e

Revisiting SA biosynthesis: new post-PAL route in plant immunity.

Salicylic acid (SA), the active component of aspirin, is a crucial defense phytohormone that coordinates both local and systemic acquired resistance. For decades, SA biosynthesis was attributed to two pathways: (i) the isochorismate synthase pathway, which is dominant in arabidopsis (Arabidopsis thaliana) and other Brassicaceae, and (ii) the phenylalanine ammonia-lyase (PAL) pathway, common in most other plants. However, the PAL pathway remained biochemically incomplete due to the lack of a molecularly identified benzoic acid 2-hydroxylase. Recent findings fill this gap by identifying a conser

Moving Protein PEGylation from an Art to a Data Science.

PEGylation is a well-established and clinically proven half-life extension strategy for protein delivery. Protein modification with amine-reactive poly(ethylene glycol) (PEG) generates heterogeneous and complex bioconjugate mixtures, often composed of several PEG positional isomers with varied therapeutic efficacy. Laborious and costly experiments for reaction optimization and purification are needed to generate a therapeutically useful PEG conjugate. Kinetic models which accurately predict the outcome of so-called "random" PEGylation reactions provide an opportunity to bypass extensive wet la

Urea cycle defects in adulthood: clinical presentation, diagnosis and treatment in genetically encoded hepatic metabolic disorders with a potential for encephalopathy.

Hyperammonaemia is an important cause for encephalopathy. Ammonia is the waste product of amino acid degradation and cannot be excreted via urine. Ammonia is metabolized to water-soluble urea via the urea cycle. Hyperammonaemia not only occurs during acute liver failure, but also in rare genetically determined defects of enzymes or transporters involved in the urea cycle resulting in elevated ammonia concentrations. Enzyme defects include deficiency of carbamylphosphate synthase, N-acetylglutamate synthase, ornithine transcarbamylase, argininosuccinate lyase and arginase, transporter defects a

[The reactions in ginseng cells induced by elicitor derived from digesting ginseng cell walls by cellulase].

After treatment of the elicitor derived from digesting ginseng cell walls by cellulase, the starch grains were degraded, and a large number of spherosomes were produced in Ginseng cells. In the mean time, the activity of amylase in the cells was increased and the activities of esterases in the cells were reduced; in addition, the content of soluble sugar and the content of lipoid were increased. These indicate that the emergency reactions and the adjustment of metabolism of lipoid were taken placed in the cells elicited. Besides, after the treatment with certain concentrations of elicitor, the

A neonatal screening test for argininosuccinic acid lyase deficiency and other urea cycle disorders.

A simple enzyme-multiple auxotroph assay has been developed for the identification of newborn infants with several of the inherited metabolic defects in the Krebs cycle for the detoxification of ammonia and in the ornithine metabolic pathway. This mass screening test is used with dried filter paper blood specimens and can easily be added to existing multiple testing programs presently used in screening for phenylketonuria or congenital hypothyroidism. This assay can be used to detect patients with citrullinemia, argininosuccinic acid lyase deficiency, and argininemia. In addition to these urea
