Human beings can be parasitized by various species of intestinal amoebae. Entamoeba histolytica is the only intestinal amoeba recognized to be pathogenic, while other amoeba species, E. dispar, E. moshkovskii, E. hartmanni, E. coli, E. polecki, Endolimax nana and Iodamoeba buetschlii are considered to be non-pathogenic. The aim of this review is to synthesize the main morphological characteristics of the trophozoite and cyst stages of each amoeba as the basis for precise microscopical diagnosis. The difficulty of morphological differentiation among species included in the so-called "Entamoeba

Since the discovery that Legionella pneumophila can survive and grow within free-living amoebae, there has been an increasing number of microbial species shown to have similar relationships. These include many bacterial species, fungi, other protozoa (e.g. Cryptosporidium) and viruses. Among bacteria, mycobacteria are of particular importance because of their role in human and animal infections. This review will consider the progress made in understanding the relationships between mycobacteria and amoebae, and their consequences in terms of ecology and epidemiology.

The acquisition of invasiveness of mammalian tissue by free living amoebae (FLA) is an extremely noteworthy phenomenon. Many factors may be responsible for this evolutionary change in their physiology. The present work aimed to clarify the host factors in this problem. Albino mice were the laboratory animals used in this work. Non-pathogenic strains of FLA were isolated from ear, nose and pharyngeal swabs and from contact lenses of patients attending out patient clinics in Benha University Hospitals. Isolation was done by cultivation of non-nutrient agar at 37 & 43 degrees C and identification

Environmental mycobacteria are a common cause of human infections. Recently, contaminated domestic water supplies have been suggested as a potential environmental source of several mycobacterial diseases. Since many of these mycobacterial species replicate best intracellularly, environmental hosts have been sought. In the present study, we examined the interaction of Mycobacterium avium with a potential protozoan host, the water-borne amoeba Acanthamoeba castellanii. We found that M. avium enters and replicates in A. castellanii. In addition, similar to that shown for mycobacteria within macro

A patient wearing an intrauterine contraceptive device (IUD) was diagnosed as being colonized by Actinomyces and amoebae by examination of a routine Papanicolaou smear. The patient received vaginal treatment with metronidazole, which temporarily eliminated the protozoa; however, it was necessary to remove the IUD to definitively eradicate the microorganisms. Follow-up Papanicolaou smears taken after removal of the IUD have been normal and free of both microorganisms.

Legionella pneumophila, the causative agent of Legionnaires' disease, interacts in the environment with free-living amoebae that serve as replicative niches for the bacteria. Among these amoebae, Acanthamoeba castellanii is a natural host in water networks and a model commonly used to study the interaction between L. pneumophila and its host. However, certain crucial aspects of this interaction remain unclear. One such aspect is the role of oxidative stress, with studies focusing on reactive oxygen species (ROS) production by the host and putting less emphasis on the involvement of the host's

Predation by amoebae can induce adaptive traits in microorganisms, enhancing their resilience and pathogenic potential. This study investigated whether interaction with Acanthamoeba castellanii modulates the virulence of environmental fungi Candida haemulonii complex species. Our findings reveal that adaptation to an amoebic host induces significant phenotypic changes in C. duobushaemulonii isolated from a beetle's gut, leading to four distinct phenotypes: smooth-pink, rough-pink, intense-pink, and hyphae-white. These variants exhibited increased aggregation and filamentation. Amoeba-adapted i

INTRODUCTION: There are 12 protozoan genera that provoke zoonotic disease in humans and animals. We discuss the most common ones with a highlight on Babesia spp and Entamoeba histolytica, also mentioning Toxoplasma gondii, Trypanosoma cruzi, and Leishmania spp.
AREAS COVERED: The complex life cycle of pathogenic protozoans is deeply understood but this did not contribute to the discovery of new drugs. The clinical armamentarium is poor and includes antiinfectives originally proposed as antibacterial (azithromycin, clindamycin, paromomycin, sulfadrugs), antifungals (amphotericin B), or they ar

Vibrio cholerae is a human pathogen and the causative agent of cholera. The persistence of this bacterium in aquatic environments is a key epidemiological concern, as cholera is transmitted through contaminated water. Predatory protists, such as amoebae, are major regulators of bacterial populations in such environments. Therefore, we investigated the interaction between V. cholerae and the amoeba Acanthamoeba castellanii at the single-cell level. We observed that V. cholerae can resist intracellular killing. The non-digested bacteria were either released or, alternatively, established a repli

Agricultural crops are investments that can be exploited by others. Farmer clones of the social amoeba Dictyostelium discoideum carry bacteria to seed out new food populations but they also carry other non-food bacteria such as Burkholderia spp. Here we demonstrate that these farmer-carried Burkholderia inhibit the growth of non-farmer D. discoideum clones that could exploit the farmers' crops. Using supernatants, we show that inhibition is due to molecules secreted by Burkholderia. When farmer and non-farmer amoebae are mixed together at various frequencies and allowed to complete the social

BACKGROUND: Patients admitted to intensive care units are frequently exposed to pathogenic microorganisms present in their environment. Exposure to these microbes may lead to the development of hospital-acquired infections that complicate the illness and may be fatal. Amoeba-associated microorganisms (AAMs) are frequently isolated from hospital water networks and are reported to be associated to cases of community and hospital-acquired pneumonia.
METHODOLOGY/PRINCIPAL FINDINGS: We used a multiplexed immunofluorescence assay to test for the presence of antibodies against AAMs in sera of intens

Acanthamoeba polyphagia was found in the cooling towers of the air conditioning systems. This finding directs the attention to more studies on water and air conditioning systems of hospitals and dwellings and the relation to outbreaks of legionnaire's disease be caused by Legionella bacilli which complete its natural life cycle inside the Acanthamoeba.

The effects of the metabolic inhibitors KCN and SHAM on phototaxis and photophobic response in Amoeba proteus have been studied. Both drugs neither change amoebae photophobic response nor the phototactic reaction. The results indicate clearly that the negative phototactic orientation is not impaired by impediment of respiration thus it is not directly coupled to the differences in energy production in different parts of A. proteus body.

Ultrastructural changes in mitochondrial morphology were observed in amoebae exposed to the uncoupling agents dinitrophernol (DNP), pentachlorophenol (PCP), and m-chlorocarbonyl cyanide phenylhydrazone (CCCP). These alterations occurred with rapidity and were present before whole-cell activity changes could be detected. They included changes in profile shape and overall dimensions, matrix density changes, and alterations to the cristal membranes, so that distinction between control Type I and Type II conformations was eliminated and a form with intermediate characters generated. It is proposed

Free-living amoebae (FLA), including Naegleria fowleri, Acanthamoeba castellanii, Balamuthia mandrillaris, and Sappinia pedata, are ubiquitous protozoa capable of causing severe infections such as primary amoebic meningoencephalitis (PAM), granulomatous amoebic encephalitis (GAE), and Acanthamoeba keratitis (AK). Early diagnosis remains challenging due to disease rarity, nonspecific clinical presentation, and limited access to specialized laboratory methods. Rapid and accurate detection is critical for patient management and public health response, particularly amid changing environmental expo

Amoebae, fascinatingly diverse protists, showcase a dual nature that positions them as both friends and foes in our world. These organisms, defined by their distinctive pseudopodia, span a spectrum from harmful to helpful. On the darker side, species like Entamoeba histolytica pose serious health risks, causing intestinal and liver diseases, while the infamous "brain-eating" Naegleria fowleri leads to fatal primary amoebic meningoencephalitis (PAM), with a daunting 97% mortality rate. Other free-living amoebae, including Acanthamoeba castellanii and Balamuthia mandrillaris, also threaten the h

Naegleria fowleri, Balamuthia mandrillaris, and Acanthamoeba spp. can cause devastating brain infections in humans which almost always result in death. The symptoms of the three infections overlap, but brain inflammation and the course of the disease differ, depending on the amoeba that is responsible. Understanding the differences between these amoebae can result in the development of strategies to prevent and treat these infections. Recently, numerous scientific advancements have been made in the understanding of pathogenicity mechanisms in general, and the basic biology, epidemiology, and t

Primary amoebic meningoencephalitis (PAM) caused by Naegleria fowleri is a rare and deadly disease that requires prompt treatment with multiple therapies. Although N. fowleri previously was only found in warmer areas, climate change appears to be contributing to its geographic spread. Clinicians should consider PAM when faced with a patient with meningitis, especially if the patient participates in outdoor water activities or practices nasal rinsing.
