Detection of pathogens in ixodid ticks collected from animals and vegetation in five regions of Ukraine.

The distribution and prevalence of zoonotic pathogens infecting ixodid ticks in Western Europe have been extensively examined. However, data on ticks and tick-borne pathogens in Eastern Europe, particularly Ukraine are scarce. The objective of the current study was, therefore, to investigate the prevalence of Anaplasma phagocytophilum, Anaplasmataceae, Rickettsia spp., Babesia spp., Bartonella spp., and Borrelia burgdorferi sensu lato in engorged and questing ixodid ticks collected from five administrative regions (oblasts) of Ukraine, namely Chernivtsi, Khmelnytskyi, Kyiv, Ternopil, and Vinny

Dynamics, co-infections and characteristics of zoonotic tick-borne pathogens in Hokkaido small mammals, Japan.

Many of the emerging infectious diseases originate in wildlife and many of them are caused by vector-borne pathogens. In Japan, zoonotic tick-borne pathogens (TBPs) are frequently detected in both ticks and wildlife. Here, we studied the infection rates of potentially zoonotic species, including Anaplasma, Ehrlichia, Neoehrlichia and Babesia spp., in Hokkaido's most abundant small mammals as they relate to variable extrinsic factors that might affect the infection rates of these pathogens. A total of 412 small mammals including 64 Apodemus argenteus, 219 Apodemus speciosus, 78 Myodes rufocanus

Potomac Horse Fever.

Potomac horse fever (PHF) is a common cause of equine colitis in endemic areas. Until recently, the only causative agent known to cause PHF was Neorickettsia risticii. However, N. findlayensis has been isolated from affected horses. Horses typically become infected upon ingestion of Neorickettsia spp.-infected trematodes within aquatic insects. The most common clinical signs include diarrhea, fever, anorexia, lethargy and colic. The diagnostic test of choice for PHF is PCR of blood and feces. Tetracyclines remain an effective treatment. Supportive care, including fluid therapy, colloid adminis

Neoehrlichia mikurensis infection associated with lymphoma or mimicking lymphoma in two cohorts of Swedish patients.

The tick-borne bacterium Neoehrlichia (N.) mikurensis causes latent infections that can become symptomatic when B-cell defenses are compromised. We investigated the prevalence of N. mikurensis in two cohorts of Swedish patients: a prospective cohort (n = 186) evaluated for lymphoma and a retrospective cohort (n = 198) diagnosed with seven lymphomas. Samples were tested for N. mikurensis by PCR. Nine of 116 patients (7.8%) diagnosed with lymphoma in the prospective cohort, and 7/198 (3.5%) in the retrospective cohort had a concomitant N. mikurensis infection. Indolent lymphomas, particularly ma

Dual detection of Neoehrlichia mikurensis and Anaplasma phagocytophilum: How biplex PCR led in France to the diagnosis of neoehrlichiosis in post-tick bite fever.

OBJECTIVES: Neoehrlichia mikurensis (NM) is an emerging intracellular bacterium member of the Anaplasmatacecae family. So far, only four cases have been described in France, all of them retrospectively. METHODS: This study describes two new cases of NM discovered following implementation of a biplex PCR targeting both Anaplasma and NM. RESULTS: Two immunocompetent patients developed a flu-like illness within 14 days after an identified tick bite. Both had negative Anaplasma PCR, but by using this biplex PCR NM, infection was diagnosed in both cases. One patient recovered spontaneously while

Molecular survey of Babesia microti, Ehrlichia species and Candidatus neoehrlichia mikurensis in wild rodents from Shimane Prefecture, Japan.

A significant number of patients are diagnosed with "fevers of unknown origin" (FUO) in Shimane Prefecture in Japan where tick-borne diseases are endemic. We conducted molecular surveys for Babesia microti, Ehrlichia species, and Candidatus Neoehrlichia mikurensis in 62 FUO cases and 62 wild rodents from Shimane Prefecture, Japan. PCR using primers specific for the Babesia 18S small-subunit rRNA (rDNA) gene and Anaplasmataceae groESL amplified products from 45% (28/62) and 25.8% (16/62) of captured mice, respectively. Of the 28 18S rDNA PCR positives, 23 and five samples were positive for Hobe

Zoonotic pathogen screening of striped field mice (Apodemus agrarius) from Austria.

The striped field mouse (Apodemus agrarius) is known to carry several zoonotic pathogens, including Leptospira spp. and Dobrava-Belgrade orthohantavirus (DOBV). Since its first detection in 1996 in south-east Austria, the striped field mouse has further expanded its range in Austria. Here, we screened 35 striped field mice collected in an Austrian region near the Hungarian border for DOBV, Leptospira spp. and seven vector-borne pathogens. Hantavirus RT-PCR screening and DOBV IgG ELISA analysis led to the detection of two DOBV-positive striped field mice. The complete coding sequences of all th

Vector-borne diseases in client-owned and stray cats from Madrid, Spain.

The role of various vector-borne pathogens as a cause of disease in cats has not been clearly determined. The current study evaluated risk factors, clinical and laboratory abnormalities associated with Ehrlichia spp., Anaplasma spp., Neorickettsia spp., Leishmania spp., and Bartonella spp. infection or exposure in 680 client-owned and stray cats from Madrid, Spain. Our results indicate that a large portion (35.1%) of the cat population of Madrid, Spain, is exposed to at least one of the five vector-borne pathogens tested. We found seroreactivity to Bartonella henselae in 23.8%, to Ehrlichia ca

Detection of relapsing fever Borrelia spp., Bartonella spp. and Anaplasmataceae bacteria in argasid ticks in Algeria.

BACKGROUND: Argasid ticks (soft ticks) are blood-feeding arthropods that can parasitize rodents, birds, humans, livestock and companion animals. Ticks of the Ornithodoros genus are known to be vectors of relapsing fever borreliosis in humans. In Algeria, little is known about relapsing fever borreliosis and other bacterial pathogens transmitted by argasid ticks. METHODOLOGY/PRINCIPAL FINDINGS: Between May 2013 and October 2015, we investigated the presence of soft ticks in 20 rodent burrows, 10 yellow-legged gull (Larus michahellis) nests and animal shelters in six locations in two different

Prevalence of Borrelia burgdorferi sensu lato and Anaplasmataceae members in Ixodes ricinus ticks in Alsace, a focus of Lyme borreliosis endemicity in France.

Due to the high Lyme borreliosis incidence in Alsace, in northeastern France, we investigated in 2003-2004 three cantons in this region in order to determine the density of Ixodes ricinus ticks infected by Borrelia burgdorferi sensu lato and Anaplasmataceae. The peak density of nymphs infected by B. burgdorferi sensu lato at Munster and Guebwiller, where the disease incidence was high, was among the highest reported in Europe (105 and 114 per 100 m(2), respectively). In contrast, the peak density of infected nymphs was low in the canton of Dannemarie (5/100 m(2)), where the disease incidence w

Anaplasma phagocytophilum invasin AipB interacts with the β2 integrin β-subunit CD18 to orchestrate infection.

Anaplasma phagocytophilum is an obligate intracellular bacterium that invades neutrophils to cause the potentially severe infection, human granulocytic anaplasmosis. The A. phagocytophilum invasin-host cell receptor interactions that facilitate bacterial uptake and hence disease are incompletely defined. Here, we show that EPHNCH_1611, which we refer to as A. phagocytophilum invasion protein B (AipB), mediates bacterial entry but not adhesion. The AipB host cell-interacting domain was narrowed down to amino acids 109 to 129 and is conserved among Anaplasma and Ehrlichia species. AipB109-123 an

Detection and phylogenetic relationship of Neorickettsia risticii in horses from Southern Rio Grande do Sul, Brazil.

BACKGROUND: Neorickettsia risticii, an obligate intracellular bacterium, is the causative agent of Potomac Horse Fever (PHF) - a systemic, acute, and potentially fatal diarrheal disease. This pathogen is an endosymbiont of digenetic trematodes, with freshwater snails playing a role in its biological cycle. The disease exhibits endemic and seasonal patterns, primarily affecting horses in wetlands during warmer months, and has been documented in the United States, Canada, Europe, and South America. Previous studies confirmed the pathogen's presence in herds from southern Brazil via serology and

Listeriosis in sheep. Eperythrozoon ovis infection used as a model to study predisposing factors.

Three groups of 9 months old lambs, each group consisting of 5 animals, were infected experimentally with Eperythrozoon ovis (Eo), Listeria monocytogenes (Lm) and Eo/Lm, respectively. The animals infected with Eo developed haemolytic anaemia, but otherwise no clinical symptoms were seen. The animals infected with Lm had a period with fever and reduced appetite after infection. These symptoms lasted longer and were more pronounced in the group with the dual infection (Eo/Lm). None of the lambs developed clinical meningo-encephalitis during the experiment. Group Lm developed the highest reciproc

The prevalence of selected vector-borne diseases in dromedary camels (Camelus dromedarius) in the United Arab Emirates.

Vector-borne diseases (VBDs) affecting dromedary camels (Camelus dromedarius) have considerable importance in the United Arab Emirates (UAE) because of the consequences associated with production decline and economic losses. Our study aimed to determine the prevalence of selected VBDs in camels in the UAE and identify risk factors. This research is currently affected by the low number of epidemiological molecular surveys addressing this issue. Blood samples were obtained from 425 dromedary camels from different locations across the UAE. Whole genomic DNA was isolated, and PCR screening was don

Genetic diversity of Anaplasma bacteria: Twenty years later.

The genus Anaplasma (family Anaplasmataceae, order Rickettsiales) includes obligate intracellular alphaproteobacteria that multiply within membrane-bound vacuoles and are transmitted by Ixodidae ticks to vertebrate hosts. Since the last reclassification of Anaplasmataceae twenty years ago, two new Anaplasma species have been identified. To date, the genus includes eight Anaplasma species (A. phagocytophilum, A. marginale, A. centrale, A. ovis, A. bovis, A. platys, A. odocoilei, and A. capra) and a large number of unclassified genovariants that cannot be assigned to known species.
