Pigs as large-animal models for translational behavioral neuroscience.

Model choice in behavioral neuroscience is often driven by convenience and established research pipelines, contributing to a persistent reliance on murine species. Although rodents are essential for discovery, their limited alignment with human brain architecture and clinical technologies can constrain translational inference. Here, we argue that the pig (Sus scrofa) is an underutilized large-animal model with substantial translational value due to its gyrencephalic brain, human-relevant physiology, and compatibility with clinical imaging and neurotechnology. We review evidence that pigs can s

Defining the threshold of birth: ethical introduction of artificial placenta and artificial womb technology in the neonatal intensive care unit.

Artificial amnion and placenta technology (AAPT), also colloquially referred to as artificial womb technology, will likely move into first-in-human trials in the next few years. The results of the AAPT animal trials inspired numerous commentaries in the ethics literature, with some ethicists and philosophers advocating for the creation of a new "status of being" to describe a patient supported by AAPT. Proposed states of being include terms such as: "gestateling" (a fetus outside the womb) or "fetonate" (a neonate with full human rights but retained fetal physiology). Adopting the "gestateling

Venlafaxine reduces sign-tracking in male Sprague-Dawley rats.

Sign-tracking, the behavior of approaching and interacting with cues for appetitive outcomes in classical conditioning, has been implicated in relapse to drug addiction. In this experimental study, we examined the serotonin/norepinephrine reuptake inhibitor venlafaxine to determine whether it was effective in reducing sign-tracking in an animal model (Sprague-Dawley male rats, N = 35) of sign-tracking using a retractable lever as a conditioned stimulus and sucrose pellets as an unconditioned stimulus. After training, male rats were administered venlafaxine (0, 30, or 60 mg/kg) intraperitoneall

Modeling impaired insight after drug use in rodents.

Impaired insight in substance use disorder has been argued to reflect a global deficit in using cognitive models to mentally simulate possible future outcomes. The process of mentally simulating outcomes allows us to understand our beliefs about their causes, that is, to have insight and thereby avoid potentially negative outcomes. However, work in humans cannot address whether impaired insight and its neural/neurochemical sequalae are present prior to the development of a substance use disorder, a consequence of substance use, or a combination of both. This is because baseline measurements pr

Distinguishing moral injury from burnout, secondary traumatic stress, and depression in animal care workers: A condition differentiation analysis.

OBJECTIVE: Animal care workers operate in conditions involving substantial occupational stressors that can significantly impact their mental health and well-being. Despite a growing awareness of these stressors, moral injury, a values-based condition arising from violations of deeply held moral beliefs, has not been empirically established in this population as a relevant condition distinct from burnout and compassion fatigue, the constructs that dominate the literature. The present study examined whether moral injury is empirically distinguishable from burnout, secondary traumatic stress, and

Coronary artery involvement early and late after radiofrequency current application in young pigs.

Radiofrequency current (500 kHz) was delivered by temperature guidance (75 degrees C) over a 30-second period in 10 young piglets with a steerable 6F electrode catheter equipped with a thermistor at the 4 mm tip electrode. Lesions were created at the right atrial aspect of the tricuspid valve anulus, at the left ventricular myocardium under the lateral mitral valve anulus, and at the left ventricular apex. After 48 hours, five animals were randomly sacrificed. Lesions in the five animals appeared as transmural gray-white coagulation necrosis. Lymphocytic infiltration around the right atrial le

Establishment of a canine model of pulmonary arterial hypertension induced by dehydromonocrotaline and ultrasonographic study of right ventricular remodeling.

OBJECTIVE: Pulmonary arterial hypertension (PAH) means high blood pressure in the lungs. We aimed to observe the right ventricular size, wall thickness and characteristic functional changes and their associations with PAH in an established model of beagle dogs, and to explore convenient, reliable and sensitive ultrasound indicators for assessing right ventricular remodeling. METHODS: Twenty healthy beagle dogs (8-10 kg) were randomly divided into control group (N-dimethylformamide, n = 10) and dehydromonocrotaline (DHMCT) group (DHMCT, n = 10). N-dimethylformamide or DHMCT was injected throug

Right Heart Diastology, Where Are We Now?

Right heart diastolic dysfunction is increasingly recognized as an early and prognostically relevant process in patients with pulmonary vascular disease, heart failure, and congenital heart disease, among others. Despite this, right heart diastolic dysfunction remains poorly defined and inconsistently measured. This review reframes right heart diastology as a disorder of a unified diastolic unit, integrating right atrial function, right ventricular compliance, and pulmonary arterial load. We synthesize invasive physiology, imaging, and human myocardial data to show that abnormalities in relaxa

Venous Shunt Versus Venous Ligation for Vascular Damage Control: The Immunohistochemical Evidence.

BACKGROUND: To evaluate the expression of immunohistochemical markers of tissue ischemia (iNOS, eNOS, and HSP70) in a vascular damage control experimental model to determine if a venous temporary vascular shunt insertion leads to a better limb perfusion when compared with the ligature of the injured vein. METHODS: Experimental study in male Sus Scrofa weighting 40 Kg. Animals were distributed into 5 groups: group 1 animals were submitted to right external iliac artery (EIA) shunting and right external iliac vein (EIV) ligation; group 2 animals were submitted to right EIA shunting and right EI

Specific Neural Mechanisms Underlying Humans' Processing of Information Related to Companion Animals: A Comparison with Domestic Animals and Objects.

Humans show neural specificity in processing animal-related information, especially regarding companion animals. However, the underlying cognitive mechanisms remain poorly understood. This study's main objective is to investigate human neural specificity in processing companion animal-related information, compared to other animal types and inanimate objects. Forty participants viewed four image types (companion animals, neutral animals, positive objects, neutral objects) during functional magnetic resonance imaging (fMRI) scans and judged image categories. T-test results showed: 1. Processing

Infant biases for detecting speech in complex scenes.

How do infants learn the sounds of their native language when there are many simultaneous sounds competing for their attention? Adults and children detect when speech sounds change in complex scenes better than when other sounds change. We examined whether infants have similar biases to detect when human speech changes better than nonspeech sounds including musical instruments, water, and animal calls in complex auditory scenes. We used a change deafness paradigm to examine whether 5-month-olds' change detection is biased toward certain sounds within high-level categories (e.g., biological or

Novel Therapeutic Targets for the Treatment of Right Ventricular Remodeling: Insights from the Pulmonary Artery Banding Model.

Right ventricular (RV) function is the main determinant of the outcome of patients with pulmonary hypertension (PH). RV dysfunction develops gradually and worsens progressively over the course of PH, resulting in RV failure and premature death. Currently, approved therapies for the treatment of left ventricular failure are not established for the RV. Furthermore, the direct effects of specific vasoactive drugs for treatment of pulmonary arterial hypertension (PAH, Group 1 of PH) on RV are not fully investigated. Pulmonary artery banding (PAB) allows to study the pathogenesis of RV failure sole

Resurgence of punishment-suppressed cocaine seeking in rats.

Alternative reinforcement-based treatments are among the most effective for reducing substance abuse. However, relapse often occurs when alternative reinforcement ends. Relapse following the loss of alternative reinforcement is called resurgence. An animal model has been used to study basic factors that may ultimately reduce resurgence but uses drug unavailability (i.e., extinction) to reduce drug seeking. In humans, drug abstinence is thought to be a product of aversive consequences associated with drug use rather than extinction. This discrepancy is important because the environmental and ne

Left Pulmonary Artery Ligation and Chronic Pulmonary Artery Banding Model for Inducing Right Ventricular-Pulmonary Hypertension in Sheep.

Pulmonary hypertension (PH) is a progressive disease that leads to cardiopulmonary dysfunction and right heart failure from pressure and volume overloading of the right ventricle (RV). Mechanical cardiopulmonary support has theoretical promise as a bridge to organ transplant or destination therapy for these patients. Solving the challenges of mechanical cardiopulmonary support for PH and RV failure requires its testing in a physiologically relevant animal model. Previous PH models in large animals have used pulmonary bead embolization, which elicits unpredictable inflammatory responses and has

Propelling and perturbing appendages together facilitate strenuous ground self-righting.

Terrestrial animals must self-right when overturned on the ground, but this locomotor task is strenuous. To do so, the discoid cockroach often pushes its wings against the ground to begin a somersault which rarely succeeds. As it repeatedly attempts this, the animal probabilistically rolls to the side to self-right. During winged self-righting, the animal flails its legs vigorously. Here, we studied whether wing opening and leg flailing together facilitate strenuous ground self-righting. Adding mass to increase hind leg flailing kinetic energy increased the animal's self-righting probability.

CILP1 as a biomarker for right ventricular maladaptation in pulmonary hypertension.

The aim of our study was to analyse the protein expression of cartilage intermediate layer protein (CILP)1 in a mouse model of right ventricular (RV) pressure overload and to evaluate CILP1 as a biomarker of cardiac remodelling and maladaptive RV function in patients with pulmonary hypertension (PH).Pulmonary artery banding was performed in 14 mice; another nine mice underwent sham surgery. CILP1 protein expression was analysed in all hearts using Western blotting and immunostaining. CILP1 serum concentrations were measured in 161 patients (97 with adaptive and maladaptive RV pressure overload

Cardiac Contractility Modulation in a Model of Repaired Tetralogy of Fallot: A Sheep Model.

The onset of right ventricular dysfunction in patients presenting with congenital heart disease is associated with a dismal long-term outcome and often represents a therapeutic dead end. Our study had several objectives: (1) to analyse the anatomical, functional, histological and cellular characteristics of an animal model of repaired tetralogy of Fallot with right ventricular dysfunction (2) to test the new electrical treatment known as cardiac contractility modulation in this animal model. Seven sheep underwent a first surgery at the age of three weeks aiming to mimic the characteristics of

Experimental and clinical tests of FDA-approved kinase inhibitors for the treatment of neurological disorders (update 2024).

Since our previous summary of the 74 FDA-approved kinase inhibitors in clinical and preclinical trials for non-cancerous neurological treatment, the US FDA has approved 13 additional kinase inhibitors since early 2022. This update incorporates new evidence for the now 87 FDA-approved kinase inhibitors in clinical and preclinical trials for the treatment of non-cancerous neurological disorders. By the end of October 2024, nearly all 87 FDA-approved kinase inhibitors have been tested in various animal models of non-cancerous neurological disorders, with twenty entered into clinical trials and si

Autism Spectrum Disorder presents one of the most complex challenges in contemporary neuroscience. This review adopts an unconventional narrative structure, drawing inspiration from song titles by The Beatles to explore the multifaceted biological, developmental, and social dimensions of autism. Spanning historical perspectives to embryonic origins and adult cognition, we examine critical topics including cortical folding, sensory processing, and the contributions of various brain regions such as the cerebellum and brainstem.
