Four cases of aniridia associated with subluxated lenses and microcornea are presented. The triad occurred in both eyes of the 4 affected members in one Ndebele family (one of the South African Negro tribes). No other ocular or systemic defects were noted, and intelligence was normal. Chromosomal studies on both parents showed no abnormality, and gene marker studies failed to reveal any linkage between the disease locus and a wide range of polymorphic loci.

This is the first report of a syndrome of aniridia and aplasia of the patella. The origin of the defect arises spontaneously in the grandmother of the proband and must be assumed to be a de novo mutation. There is no associated chromosomal abnormality or overt biochemical or other somatic defect. This is an extremely rare mutation exhibiting a dominant autosomal form of inheritance. The gene for aniridia is a dominant gene whose penetrance here is 100%. The combination of aniridia and bone malformations in general is very rare. This linked defect probably represents one of the most rare of hum

Congenital aniridia is a rare genetic eye disorder often associated with PAX6 gene mutations, leading to complex ocular manifestations, including aniridia-associated keratopathy (AAK) and cataracts. The surgical management of cataracts in these patients is particularly challenging due to severe corneal opacities, which obscure key surgical steps such as capsulorhexis and phacoemulsification. This report presents two cases of congenital aniridia with severe AAK, in which chandelier retroillumination-assisted cataract surgery was employed. This technique, typically used in vitreous surgery, prov

Aim: To describe our results with HumanOptics IOL-ArtificialIris complex in post traumatic aphakia and aniridia. Methods: Retrospective, single-surgeon chart review of cases in which aniridia and aphakia were corrected using HumanOptics IOL-ArtificialIris complex sutured to the sclera with Gore-Tex® sutures and coupled with the Akreos® IOL (Bausch&Lomb). Results: The authors present four cases of ocular trauma with globe rupture. For every patient, posterior vitrectomy was done and an artificial iris-lens diaphragm was sutured to the sclera. All patients had a good functional and cosmetic outc

Absence of part or all of the iris, aniridia, is a feature of several genetically distinct conditions. This review focuses on iris development and then the clinical features and molecular genetics of these iris malformations. Classical aniridia, a panocular eye malformation including foveal hypoplasia, is the archetypal phenotype associated with heterozygous PAX6 loss-of-function mutations. Since this was identified in 1991, many genetic mechanisms of PAX6 inactivation have been elucidated, the commonest alleles being intragenic mutations causing premature stop codons, followed by those causin

BACKGROUND: Aniridia is a congenital panocular malformation defined as iris aplasia or hypoplasia. It can be either isolated or be a part of multiple ocular anomalies such as cataracts, glaucoma, corneal pannus, optic nerve hypoplasia, absence of macular reflex or ectopia lentis. In the majority of cases the disease is caused by mutation in the PAX6 gene.
MATERIAL AND METHODS: A Polish family with aniridia was screened for the presence of genomic rearrangements in PAX6, WT1 and the flanking genes by means of multiplex ligation probe amplification (MLPA). MLPA reaction was performed using the

BACKGROUND: Aniridia typically appears as a familial condition with autosomal dominant inheritance but can present as an isolated disease or sporadically in association with several syndromes. In this report we describe the various ocular manifestations of aniridia as well as the association of familial aniridia with two different ocular and systemic abnormalities present across three generations in two different families.
METHODS: Descriptive case series of 33 patients (66 eyes) with aniridia. A full eye examination was performed at the beginning of the study, including past medical history,

A pathological study was conducted on four patients with nephroblastoma associated with aniridia. The age at diagnosis. was one year in three cases and 4 years in one case. Chromosomal analysis was performed in three cases, and showed 11p13 deletion in all. Each nephroblastoma consisted of metanephric blastemal, epithelial, and mesenchymal cells. All the tumors were centrilobar, and in one case an additional polypoid tumor grew into the renal pelvis. Histological examination showed conspicuous heterotopic cells, such as striated muscle and adipose tissue. Striated muscle was seen in all four c

Congenital aniridia is a panocular disorder that is typically characterized by iris hypoplasia and aniridia-associated keratopathy (AAK). AAK results in the progressive loss of corneal transparency and thereby loss of vision. Currently, there is no approved therapy to delay or prevent its progression, and clinical management is challenging because of phenotypic variability and high risk of complications after interventions; however, new insights into the molecular pathogenesis of AAK may help improve its management. Here, we review the current understanding about the pathogenesis and managemen

OBJECTIVE: To describe the findings in a family with hereditary keratitis.
DESIGN: Case series.
SETTING: Eye genetics clinic at a university-affiliated hospital in Edmonton.
PATIENTS: Fifteen affected members, nine female and six male, of a four-generation family with hereditary keratitis.
RESULTS: The pattern of transmission was consistent with autosomal dominant inheritance. The disorder was characterized by the presence of a circumferential band of opacification and vascularization at the level of Bowman's membrane adjacent to the limbus. Progression toward the central cornea occurred i

The developmental birth eye disorder of iris is known as aniridia. Heterozygous PAX6 gene, which causes human aniridia and small eye in mice, is located on chromosome 11p13. The variability had been documented between the affected individuals within the families, is due to genotypic variation. Haploinsufficiency renders PAX6 allele non-functional or amorphic, however it presents hypomorphic or neomorphic alleles. India is not a well-studied ethnic group, hence the focus on congenital aniridia gene analysis supports the literature and the phenotypic association were analysed both in sporadic as

INTRODUCTION: Phacoemulsification and falls are both common in the elderly population. We present a case of acquired total aniridia and vitreous haemorrhage occurring as a result of trauma in a pseudophakic eye.
METHODS: Interventional case report with history, clinical photograph and discussion with literature review.
RESULTS: A previously healthy 74-year-old Caucasian female was referred with a painful left eye and poor vision following a fall and trauma to the left side of her face. Initial examination confirmed visual acuity of perception of light with total hyphaema and vitreous haemorr

Beginning in June 1991, we implanted a newly designed, black diaphragm intraocular lens (IOL) into 13 eyes with congenital aniridia after cataract surgery. To the best of our knowledge, this is the first group of patients to receive a black diaphragm IOL. With the first prototype we experienced various difficulties, which prompted four design modifications. The latest IOL with a diaphragm diameter of 10 mm can be guided safely into the ciliary sulcus. IOL implantation improved visual acuity in 9 of 13 eyes; 4 of these exhibited considerably less nystagmus than was observed preoperatively. Duri

Three additional cases of keratolenticular adhesion and aniridia have been reported. A review of the embryology and terminology in the description of complex lesion of the anterior segment might suggest the more general phrase, anterior segment dysgenesis, to have advantage. Finally, the simultaneous occurrence of anterior segment anomalies and hemophilia is reported.

PURPOSE: To report on the incidence, characteristics, and treatment of glaucoma in association with aniridia in a population of Egyptian children.
METHODS: A retrospective chart review was conducted of children (<18 years of age) presenting with aniridia between 2007 and 2022. Diagnosis of glaucoma was based on cup:disk ratio (>0.3) and IOP (>16 mm Hg). Success of glaucoma surgery was defined as IOP ≤16 mm Hg and stable or regressing cup:disk ratio.
RESULTS: A total of 93 eyes of 47 children were included. The mean patient age at presentation was 41.2 ± 44.7 months. The mean intraocular pres

Heterozygous mutation of PAX6 in humans leads to congenital aniridia (OMIM 106210) which is typified by congenital iris and foveal defects, and later onset glaucoma, aniridic keratopathy, and cataract. Mice heterozygous for Pax6 mutations phenocopy many aspects of aniridia including the iris defects, keratopathy and cataract, although Pax6 mutant mice have small lenses, a phenotype which is not typically reported in human aniridia, perhaps due to difficulties in measuring lens diameter during typical ophthalmic examinations as the lens periphery is shielded by the iris.
