Analysis of function, specificity and T cell receptor expression of cloned mucosal T cell lines in Crohn's disease.

Monoclonal populations of mucosal T cells were established from the earliest visible lesions in eight patients with well defined Crohn's disease. The FACS phenotype of all the mucosal derived clones to date are TCR alpha/beta+, CD3+, CD4+, and CD45RO+ memory cells. TCR variable region Beta chain analysis revealed predominantly V beta families 1, 2, 5.1, 5.2, 6, 7 and 8, with V beta family analysis supporting antigen expansion in the diseased mucosa. Putative autoreactivity was evaluated by stimulating individual clones with a battery of antigens and determining proliferation and IL-2 productio

HLA-DR antigens and phenotypes in Dutch coeliac children and their families.

In a study on the HLA-DR antigens and phenotypes in a series of Dutch coeliac children and their first-degree relatives, the B-cell antigens of 36 unrelated coeliac children, 110 first-degree relatives of 33 of them, and 201 controls were typed with the two-colour fluorescence test. The most frequent antigen was HLA-DR3 (69%), followed by DR7 (36%). The distribution of DR phenotypes showed that the most frequent was DR3/other DR (25%), followed by DR3/DR7 (17%), DR3/DR4 (14%), and DR3/DR3 (14%). However, due to the frequency of certain antigens in the controls, only phenotypes DR3/DR3 (relativ

Variant sublines of the human B-lymphoma cells Namalwa are at different stages of differentiation.

Variant sublines of the Burkitt's lymphoma cell line Namalwa, previously shown to differ in MHC class II antigen and immunoglobulin expression, have been tested with monoclonal antibodies defining B-cell specific clusters of differentiation (CD). The Namalwa sublines express antigens recognized by the CD antibodies in patterns that indicate that the cells are arrested at different stages of differentiation.

Production of antiendomysial antibodies after in-vitro gliadin challenge of small intestine biopsy samples from patients with coeliac disease.

BACKGROUND: Diagnosis of many immune-mediated diseases is helped by detection of antibodies directed against the pathogenetic (self or foreign) antigen. In coeliac disease (CD), we have a situation in which antiendomysial antibodies (EMA), which are not specific for the pathogenetic antigen, reach a specificity and sensitivity of detection of CD approaching 100%, whereas detection of antibodies against gliadin (AGA), the pathogenetic antigen, is far less specific and sensitive in diagnosis. No direct evidence of a relation between gluten/gliadin challenge and EMA production exists. We tried to

T-helper cell activation in bullous pemphigoid.

In 10 untreated patients suffering from acute bullous pemphigoid the number of peripheral blood T cells (CD 3), T suppressor (CD 8) and T helper cells (CD 4) as well as activation antigens (DR and Tac) bearing lymphocytes was evaluated by monoclonal antibodies. While the pan-T cell population (CD 3) and T suppressor subpopulation (CD 8) were normal, the T helper subpopulation (CD 4) and the number of DR and Tac positive lymphocytes were significantly increased in the acute stage of bullous pemphigoid when compared to the age- and sex-matched controls. Phenotypically those DR positive cells bel

Disease-specific B cell clones are shared between patients with Crohn's disease.

B cells have important functions in gut homeostasis, and dysregulated B cell populations are frequently observed in patients with inflammatory bowel diseases, including both ulcerative colitis (UC) and Crohn's disease (CD). How these B cell perturbations contribute to disease remains largely unknown. Here, we perform deep sequencing of the B cell receptor (BCR) repertoire in four cohorts of patients with CD, together with healthy controls and patients with UC. We identify BCR clones that are shared between patients with CD but not found in healthy individuals nor in patients with UC, indicatin

Cutaneous T cell lymphoma: immunocytochemical study on activation/proliferation and differentiation associated antigens in lymph nodes, skin, and peripheral blood.

The expression of activation/proliferation antigens (CD 25, CD 30, Ki 67, transferrin receptor) in lymph nodes and skin were compared in nine patients with mycosis fungoides (MF) with patients with erythroderma not related to MF, and patients with reactive lymphofollicular hyperplasias (a total of 14 patients). A panel of differentiation antigens was analyzed in addition. Reactivities were revealed by the APAAP technique. Activation/proliferation antigen scores in lymph nodes were related to the clinical stages of MF in most instances (low scores in cases of MF stage I/II, high scores in 3/4 c

Intracellular Localization of Microbial Transglutaminase and Its Influence on the Transport of Gliadin in Enterocytes.

OBJECTIVE: Celiac disease (CD) is a systemic inflammatory disorder, characterized by the destruction of duodenal epithelium. The CD8 T cells involved are associated with cross-presentation. In addition to other factors, the rising prevalence of CD might be induced by microbial transglutaminase (mTG) an enzyme frequently used in food production that shares enzymatic and antigenic properties of tissue transglutaminase (TG2), the autoantigen in CD. We hypothesized that mTG and gliadin are transported into the endoplasmic reticulum (ER), indicating cross-presentation of both antigens. METHODS: Ap

Human intestinal dendritic cell and macrophage subsets in coeliac disease.

Dendritic cells (DC) and macrophages (Mϕ) constitute the most abundant antigen presenting cells in the human intestinal mucosa. In resting conditions, they are essential to maintain the mechanisms of immune tolerance toward food antigens and commensals, at the time that they keep the capacity to initiate and maintain antigen-specific pro-inflammatory immune responses toward invading pathogens. Nevertheless, this delicate equilibrium between immunity and tolerance is not perfect, like in coeliac disease (CD), where DC and Mϕ drive the development of antigen-specific immune responses toward diet

Adherence of gliadin fractions to lymphocytes in coeliac disease.

Mice were immunised with a mixture of low-molecular-weight peptides prepared from beta-gliadin by digestion and ultrafiltration and found to be toxic to cultures of intestinal mucosa from patients with coeliac disease (CD). The antiserum obtained was used to detect the adherence of the same peptides to white blood cells of CD patients and healthy controls. 43% of B lymphocytes of CD patients exhibited binding of gliadin peptides compared with only 12.5% of their T lymphocytes and 17% of B lymphocytes of healthy controls. Adherence of gliadin fractions seemed to have no relation to the HLA anti

HLA antigens in patients with juvenile diabetes mellitus, coeliac disease and both of the diseases.

We determined the prevalence of 24 antigens controlled by the HLA-A and B loci in twenty patients with juvenile diabetes mellitus (JDM), in twenty patients with coeliac disease (CD), and in eight patients with both of these diseases. The prevalence of HLA-B8 was increased in JDM chi2 = 12.52, p = 0.00040) and in CD (chi2 = 26.47, p less than 0.000001) as compared to 900 controls. There was only a modest increase of Bw15 in JDM (chi2 = 8.86, p = 0.0029) and in patients with both diseases (chi2 = 2.72). The observed prevalence of phenotype HLA-B8, Bw15 was enhanced in JDM (chi2 = 16.03, p = 0;00

Strong association between the HLA-Dw3-related B cell alloantigen -DRw3 and coeliac disease.

The frequency of HLA-A, -B, and -C antigens, of -DRw3 and the associated B-lymphocyte alloantigen -DRw3 has been studied in 68 children with coeliac disease (CD). CD was found to be primarily associated with the B-lymphocyte alloantigen, the relative risk being calculated to 54. The possible relation between the B-lymphocyte alloantigen DRw3 and -Dw3 is discussed. The high relative risk and the fact that 95% of the patients with probable or definite CD had the B-lymphocyte alloantigen DRw3, underline the diagnostic value of B-lymphocyte typing in coeliac disease.

Human glycoprotein-2 expressed in Brunner glands - A putative autoimmune target and link between Crohn's and coeliac disease.

Glycoprotein 2 (GP2) is an autoantigen in Crohn's (CD) and coeliac disease (CeD). We assessed GP2-isoform (GP21-4)-expression in intestinal biopsies of paediatric patients with CD, CeD, ulcerative colitis (UC), and healthy children (HC). Transcription of GP21-4 was elevated in proximal small intestine in CeD and CD patients (only GP22/4) compared to jejunum (CeD/CD) and large bowel (CD). CeD patients demonstrated higher duodenal GP22/4-mRNA levels compared to HC/UC patients whereas CD patients showed higher GP24-mRNA levels compared to UC patients. Duodenal synthesis of only small GP2 isoforms

Overview of Celiac Disease in Russia: Regional Data and Estimated Prevalence.

Celiac disease (CD) is an autoimmune enteropathy triggered by the ingestion of dietary gluten from some cereals mainly in individuals carrying the HLA-DQ2 and/or HLA-DQ8 haplotypes. As an autoimmune disease, CD is manifested in the small intestine in the form of a progressive and reversible inflammatory lesion due to immune response to self-antigens. Indeed, CD is one of the most challenging medicosocial problems in current gastroenterology. At present, the global CD prevalence is estimated at approximately 1% based on data sent from different locations and available CD screening strategies us

Biosensors for the Diagnosis of Celiac Disease: Current Status and Future Perspectives.

Celiac disease (CD) is an autoimmune enteropathy initiated and sustained by the ingestion of gluten in genetically susceptible individuals. It is caused by a dysregulated immune response toward both dietary antigens, the gluten proteins of wheat, rye, and barley, and autoantigens, the enzyme tissue transglutaminase (TG2). The small intestine is the target organ. Although routine immunochemical protocols for a laboratory diagnosis of CD are available, faster, easier-to-use, and cheaper analytical devices for CD diagnosis are currently unavailable. This review focuses on biosensors, consisting o
