Conditional Antimicrobial Peptide Therapeutics.

Antimicrobial peptides (AMPs) constitute a promising class of alternatives to antibiotics to curb antimicrobial resistance. Nonetheless, their utility as a systemic agent is hampered by short circulation time and toxicity. Infection sites, analogous to tumors, harbor an aberrant microenvironment that has the potential to be exploited to develop conditionally activated therapeutics with an improved therapeutic index. In particular, we identified strategies to prolong systemic circulation of small, cationic AMPs in a mouse model of bacterial pneumonia. Specifically, we report an albumin-binding

Small Peptides with a Big Role: Antimicrobial Peptides in the Pregnant Female Reproductive Tract.

There is a growing interest in the role of antimicrobial peptides (AMPs) in the female reproductive tract during pregnancy. This commentary highlights recent advances in the field including those of Itakoa and colleagues who have demonstrated elafin and secretory leukocyte protease inhibitor (SLPI) expression in cervical cells from pregnant women during pregnancy. They suggest that these specific AMPs may be raised in women in true preterm labour. This complements other studies exploring the use cervico-vaginal fluid elafin and other antimicrobial peptides as biomarkers to predict risk of spon

Antimicrobial peptide-based strategies to overcome antimicrobial resistance.

Antibiotic resistance has emerged as a global threat, rendering the existing conventional treatment strategies ineffective. In view of this, antimicrobial peptides (AMPs) have proven to be potent alternative therapeutic interventions with a wide range of applications in clinical health. AMPs are small peptides produced naturally as a part of the innate immune responses against a broad range of bacterial, fungal and viral pathogens. AMPs present a myriad of advantages over traditional antibiotics, including their ability to target multiple sites, reduced susceptibility to resistance development

Structure of the antimicrobial peptide tachystatin A.

The solution structure of antimicrobial peptide tachystatin A from the Japanese horseshoe crab (Tachypleus tridentatus) was determined by two-dimensional nuclear magnetic resonance measurements and distance-restrained simulated annealing calculations. The correct pairs of disulfide bonds were also confirmed in this study. The obtained structure has a cysteine-stabilized triple-stranded beta-sheet as a dominant secondary structure and shows an amphiphilic folding observed in many membrane-interactive peptides. Interestingly, tachystatin A shares structural similarities with the calcium channel

Antimicrobial Peptides: Bringing Solution to the Rising Threats of Antimicrobial Resistance in Livestock.

Antimicrobial therapy is the most applied method for treating and preventing bacterial infection in livestock. However, it becomes less effective due to the development of antimicrobial resistance (AMR). Therefore, there is an urgent need to find new antimicrobials to reduce the rising rate of AMR. Recently, antimicrobial peptides (AMPs) have been receiving increasing attention due to their broad-spectrum antimicrobial activity, rapid killing activities, less toxicity, and cell selectivity. These features make them potent and potential alternative antimicrobials to be used in animals. Here, we

Antimicrobial peptides with symmetric structures against multidrug-resistant bacteria while alleviating antimicrobial resistance.

In response to the dramatically increasing antimicrobial resistance, a series of new symmetric peptides were designed and synthesized in this study by a "WWW" motif as the symmetric center, arginine as the positive charge amino acid and the terminus symmetrically tagged with hydrophobic amino acids. Amongst the new symmetric peptide FRRW (FRRWWWRRF-NH2) presented the highest cell selectivity for bacteria over mammalian cell and exerted excellent antimicrobial potential against a broad of bacteria, especially difficult-to-kill multidrug-resistant strains clinical isolates. FRRW also displayed p

Beyond Antimicrobial Defense: Insect Antimicrobial Peptides as Neuroimmune Effectors and Insect-Derived Peptide Resources.

Insect antimicrobial peptides (AMPs) are classically viewed as terminal effectors of innate immunity, but emerging evidence suggests that some can also shape defined neural states. In this Review, we argue that insect systems provide a powerful framework for resolving immune-brain communication at the level of individual peptide effectors, because genetically tractable innate-immune pathways allow pathway activation to be distinguished from peptide-specific effector function. Rather than surveying AMP families exhaustively, we focus on representative cases in which peptide identity, source, an

Optimization of 68Ga-DOTA radiolabeling conditions for disulfide-directed multicyclic peptides and amphiphilic antimicrobial peptides.

Gallium-68 labeled peptide radiotracers have attracted significant attention in positron emission tomography (PET) imaging of various diseases. While 68Ga-DOTA radiolabeling protocols are well-established, two classes of peptides, disulfide-directed multicyclic peptides (DDMPs) and amphiphilic antimicrobial peptides (AAMPs), presented challenges in achieving satisfactory radiolabeling efficiency using existing methods. This study therefore aimed to establish optimized 68Ga-DOTA radiolabeling protocols specifically tailored to these peptides, in order to achieve high radiochemical purity (RCP)

Combating Bacterial Infections with Vitamin D-Induced Antimicrobial Peptides.

Owing to emerging global threats such as viral pandemics and the rise of antibiotic resistance, there is an urgent need for novel approaches to combat drug-resistant pathogens. Antimicrobial peptides (AMPs), also known as cationic host defense peptides (CHDPs), are considered promising candidates because of their ability to target various pathogenic microorganisms both directly and indirectly. These functionally versatile molecules modulate host immune responses by activating multiple signaling pathways simultaneously. The unique mechanisms of interaction within the pathogen-host system, combi

Fungal expression strategies for heterologous production of antimicrobial peptides.

Antimicrobial peptides (AMPs) are promising candidates for next-generation therapeutics due to their broad-spectrum activity and reduced propensity for resistance, making them valuable in medicine, agriculture, and biotechnology. However, traditional AMP production methods including isolation from natural sources and chemical synthesis are costly, inefficient, and environmentally unsustainable, particularly for longer or post-translationally modified peptides. While heterologous expression has emerged as a scalable and versatile alternative, its success depends strongly on host selection and t

Microbe-Derived Antimicrobial Peptides: Immunomodulation and Gut Microbiota Homeostasis.

Antimicrobial peptides (AMPs) are key components of the innate immune system across a wide range of organisms. In addition to their direct antimicrobial activity, AMPs exert immunomodulatory effects that differ from those of conventional antibiotics. This review summarizes recent evidence on the roles of microbe-derived AMPs in regulating immune cell function, including neutrophil activation and NET formation, macrophage polarization, and lymphocyte proliferation and differentiation. It also examines their organ-specific roles, particularly in intestinal immunity through reinforcement of the i

Antimicrobial peptides for bacterial infections and their biomedical applications.

Due to the rapid development of multidrug-resistant (MDR) bacteria due to the inappropriate use and misuse of antibiotics and the ineffective performance of antibiotics against refractory biofilm-associated infections (BRI), there is an urgent need for novel alternative antimicrobials and strategies to combat bacterial infections. Antimicrobial peptides (AMPs) have attracted considerable interest due to their potent activity against MDR pathogens and biofilm-associated infections, coupled with a substantially reduced risk of driving antimicrobial resistance-especially when employed as alternat

Antimicrobial Peptides: A Promising Strategy for Anti-tuberculosis Therapeutics.

The high global burden of tuberculosis (TB) and the increasing emergence of the drugresistant (DR) strain of Mycobacterium tuberculosis (Mtb) emphasize the urgent need for novel antimycobacterial agents. Antimicrobial peptides (AMPs) are small peptides widely existing in a variety of organisms and usually have amphiphilic cationic structures, which have a selective affinity to the negatively charged bacterial cell wall. Besides direct bactericidal mechanisms, including interacting with the bacterial cell membrane and interfering with the biosynthesis of the cell wall, DNA, or protein, some AMP

Taming the Devil: Antimicrobial Peptides for Safer TB Therapeutics.

Tuberculosis (TB) is a highly contagious infection with extensive mortality and morbidity. The rise of TB-superbugs (drug-resistant strains) with the increase of their resistance to conventional antibiotics has prompted a further search for new anti-mycobacterial agents. It is difficult to breach the barriers around TB bacteria, including mycolic cell wall, granuloma, biofilm and mucus, by conventional antibiotics in a short span of time. Hence, there is an essential need for molecules with an unconventional mode of action and structure that can efficiently break the barriers around mycobacter

Antimicrobial Peptides and Copper(II) Ions: Novel Therapeutic Opportunities.

The emergence of new pathogens and multidrug resistant bacteria is an important public health issue that requires the development of novel classes of antibiotics. Antimicrobial peptides (AMPs) are a promising platform with great potential for the identification of new lead compounds that can combat the aforementioned pathogens due to their broad-spectrum antimicrobial activity and relatively low rate of resistance emergence. AMPs of multicellular organisms made their debut four decades ago thanks to ingenious researchers who asked simple questions about the resistance to bacterial infections o

Membrane Interactions of Latarcins: Antimicrobial Peptides from Spider Venom.

A group of seven peptides from spider venom with diverse sequences constitute the latarcin family. They have been described as membrane-active antibiotics, but their lipid interactions have not yet been addressed. Using circular dichroism and solid-state 15N-NMR, we systematically characterized and compared the conformation and helix alignment of all seven peptides in their membrane-bound state. These structural results could be correlated with activity assays (antimicrobial, hemolysis, fluorescence vesicle leakage). Functional synergy was not observed amongst any of the latarcins. The revitalization of antimicrobial peptides in the resistance era.

The antibiotic resistance crisis is becoming incredibly thorny due to the indiscriminate employment of antibiotics in agriculture and aquaculture, such as growth promoters, and the emergence of bacteria that are capable of enduring antibiotic treatment in an endless stream. Hence, to reverse this situation, vigorous efforts should be made in the process of identifying other alternative strategies with a lower frequency of resistance. Antimicrobial peptides (AMPs), originated from host defense peptides, are generally produced by a variety of organisms as defensive weapons to protect the host fr
