Impact of the three-fold channel substitution D131N on kinetics of translocation of Fe2+ across the protein coat is more severe for human cytosolic H-chain ferritin than for human mitochondrial ferritin.

Ferritins play a key role in iron management in organisms from all kingdoms of life. Excess iron is sequestered in mineral form within the hollow protein shell and can be liberated when supply becomes restricted. The protein consists of 24 isostructural monomeric units that pack with 4-, 3-, and 2-fold symmetry. Channels through the protein coat at the 3-fold axes of ferritins localised in the cytosol of animal cells contain a strictly conserved LCDFXEX 'twin carboxylate' motif, and have been shown to be the major iron entry route to animal ferritins, facilitating access to the H-chain intra-s

The iron matters: Aged microplastics disrupted the iron homeostasis in the liver organoids.

Plastic products undergo artificial and unintentional aging during daily use, causing the presence of aged microplastics (aMP). Humans are inevitably exposed to aMP. Liver is one of the critical target organs of MP through oral intake, however, limited research has focused on the hepatic toxicity of aMP compared to pristine MP (pMP). We utilized the human pluripotent stem cells-derived liver organoids (LOs) to compare the cytotoxicity of pristine polystyrene microplastics (pPS) (1 μm, carbonyl index 0.08) and aged polystyrene microplastics (aPS) (1 μm, carbonyl index 0.20) ranged from 20 to 20

Introducing a novel variant in the FTH1 gene; A closer look at FTH1-related neuroferritinopathy as a neurodegeneration with brain iron accumulation (NBIA) disorder.

BACKGROUND: Neurodegeneration with brain iron accumulation (NBIA) refers to a group of disorders with characteristic iron deposition in the basal ganglia. As an NBIA disorder, hereditary neuroferritinopathy is unique for its autosomal dominant inheritance pattern and the direct defect in iron metabolism. Caused by pathogenic variants in the FTL and FTH1 genes encoding ferritin light and heavy chains respectively, neuroferritinopathy presents clinically with a combination of dystonia, spasticity, parkinsonism, dysarthria, and cognitive decline. FTH1-related neuroferritinopathy is an exceedingly

A hemochromatosis allele that protects against Alzheimer's disease injury: A role for H-ferritin.

Recent epidemiological evidence showed that mutations to the HFE-63 allele of this hemochromatosis-associated iron-assimilation protein improve chances of avoiding Alzheimer's disease (AD). This is unexpected since increased brain ferroptosis in gray-matter increases the risk for vascular dementia and AD. However, diffusion tensor imaging from a key Alzheimer's Disease Neuroimaging Initiative biomarker study showed that the hemochromatosis H63D allele protected white matter tracts and improved cognitive performance in individuals when APOE4 accelerates AD. H63D-carrying individuals exhibit ele

Clinical and Molecular Clues to Diagnosing Hereditary Hyperferritinemia-Cataract Syndrome: Case Report and Literature Review.

Background: Hereditary hyperferritinemia-cataract syndrome (HHCS) is a rare autosomal dominant disorder characterized by persistently elevated serum ferritin and early-onset bilateral cataracts in the absence of systemic iron overload. It is caused by pathogenic variants in the iron-responsive element (IRE) of the FTL gene, leading to dysregulated L-ferritin synthesis. Methods: We evaluated a 12-year-old Czech girl with markedly elevated serum ferritin identified incidentally during workup for abdominal pain. Clinical assessment included biochemical, radiological, ophthalmological, and genetic

Ferritin light chain deficiency-induced ferroptosis is involved in preeclampsia pathophysiology by disturbing uterine spiral artery remodelling.

The proteomic analysis from samples of patients with preeclampsia (PE) displayed a low level of ferritin light chains (FTL), but we do not know what the significance of reduced FTL in PE pathophysiology is. To address this question, we first demonstrated that FTL was expressed in first- and third-trimester cytotrophoblasts, including extravillous trophoblasts (EVTs), of the human placenta. Furthermore, a pregnant rat model of FTL knockdown was successfully established by intravenously injecting adenoviruses expressing shRNA targeting FTL. In pregnant rats with downregulated FTL, we observed PE

Phosphate facilitates Fe(II) oxidative deposition in pea seed (Pisum sativum) ferritin.

The iron core within phytoferritin interior usually contains the high ratio of iron to phosphate, agreeing with the fact that phosphorus and iron are essential nutrient elements for plant growth. It was established that iron oxidation and incorporation into phytoferritin shell occurs in the plastid(s) where the high concentration of phosphate occurs. However, so far, the role of phosphate in iron oxidative deposition in plant ferritin has not been recognized yet. In the present study, Fe(II) oxidative deposition in pea seed ferritin (PSF) was aerobically investigated in the presence of phospha

Gold Electron Microscopy Grids with Anisotropic Foil Geometry Enable On-Grid Contact Guidance.

All-gold electron microscopy (EM) grids reduce beam-induced motion relative to conventional holey carbon supports and provide biocompatible substrates for cellular cryo-EM. However, placing customizable all-gold grid fabrication in the hands of researchers requires accessible processes based on standard microfabrication tools. We report a wafer-scale process using microfabrication techniques available in most academic cleanrooms, such as lift-off metallization, electroplating, and sacrificial layer release, to fabricate 594 all-gold grids per 4-inch wafer without individual grid handling. EphB1-Mediated Transient Blood-Brain Barrier Opening Facilitates a Ferritin-Based Nanotherapeutic for Alzheimer's Disease.

The treatment of Alzheimer's disease (AD) is severely hampered by the blood-brain barrier (BBB), which limits the delivery of therapeutic agents like donepezil (DPZ), an acetylcholinesterase inhibitor. While DPZ has multi-faceted benefits, its clinical efficacy is constrained by poor BBB penetration, requiring high doses that lead to significant side effects. To overcome this, we developed a brain-targeted nanotherapeutic utilizing apoferritin (AFn) nanoparticles loaded with DPZ (AFn-DPZ). We demonstrate that this platform, by binding to the EphB1 receptor on the blood-brain barrier, enables t

Bimodally Porous WO3 Microbelts Functionalized with Pt Catalysts for Selective H2S Sensors.

Bimodally meso- (2-50 nm) and macroporous (>50 nm) WO3 microbelts (MBs) functionalized with sub-3 nm Pt catalysts were fabricated via the electrospinning technique followed by subsequent calcination. Importantly, apoferritin (Apo), tea saponin and polystyrene colloid spheres (750 nm) dispersed in an electrospinning solution acted as forming agents for producing meso- and macropores on WO3 MBs during calcination. Particularly, mesopores provide not only numerous reaction sites for effective chemical reactions, but also facilitate gas diffusion into the interior of the WO3 MBs, dominated by Knud

Aspirin increases ferritin synthesis in endothelial cells: a novel antioxidant pathway.

Aspirin has recently been shown to increase endothelial resistance to oxidative damage. However, the mechanism underlying aspirin-induced cytoprotection is still unknown. Using cultured cells, the present study investigates the effect of aspirin on the expression of ferritin, a cytoprotective protein that sequesters free cytosolic iron, the main catalyst of oxygen radical formation. In bovine pulmonary artery endothelial cells, aspirin at low antithrombotic concentrations (0.03 to 0.3 mmol/L) induced the synthesis of ferritin protein in a time- and concentration-dependent fashion up to 5-fold

TDP-43 transports ferritin heavy chain mRNA to regulate oxidative stress in neuronal axons.

Amyotrophic lateral sclerosis (ALS) is characterized by the mislocalization and abnormal deposition of TAR DNA-binding protein 43 (TDP-43). This protein plays important roles in RNA metabolism and transport in motor neurons and glial cells. In addition, abnormal iron accumulation and oxidative stress are observed in the brain and spinal cord of patients with ALS exhibiting TDP-43 pathology and in animal models of ALS. We have previously demonstrated that TDP-43 downregulation significantly affects the expression of ferritin heavy chain (Fth1) mRNA in the axonal regions of neurons. Nevertheless

Ferritin-Nanocaged ATP Traverses the Blood-Testis Barrier and Enhances Sperm Motility in an Asthenozoospermia Model.

Sperm motility can be enhanced by adding ATP exogenously during in vitro fertilization. However, administering exogenous ATP to the testis to improve sperm motility for in vivo asthenozoospermia treatment has not been investigated yet. Inspired by the recent advances in nanomedicine, we investigated whether the capability of drug delivery nanocarriers to traverse the blood-testis barrier (BTB) can facilitate ATP-dependent asthenozoospermia treatment. We found that the human H-ferritin (HFn) nanocarrier possesses the capability to traverse the BTB and specifically targets the head of elongated

Disruption of neuronal CXCR4 function by opioids: preliminary evidence of ferritin heavy chain as a potential etiological agent in neuroAIDS.

The chemokine CXCL12 and its receptor, CXCR4, regulate neuronal migration, differentiation, and survival. Alterations of CXCL12/CXCR4 signaling are implicated in different neuropathologies, including the neurological complications of HIV infection. Opiates are important co-factors for progression to neuroAIDS and can disrupt the CXCL12/CXCR4 axis in vitro and in vivo. This paper will review recently identified mechanisms of opiate-induced CXCR4 impairment in neurons and introduce results from pilot studies in human brain tissue, which highlight the role of the protein ferritin heavy chain in H

Targeting iron-associated protein Ftl1 in the brain of old mice improves age-related cognitive impairment.

Understanding cellular and molecular drivers of age-related cognitive decline is necessary to identify targets to restore cognition at old age. Here we identify ferritin light chain 1 (FTL1), an iron-associated protein, as a pro-aging neuronal factor that impairs cognition. Using transcriptomic and mass spectrometry approaches, we detect an increase in neuronal FTL1 in the hippocampus of aged mice, the levels of which correlate with cognitive decline. Mimicking an age-related increase in neuronal FTL1 in young mice alters labile iron oxidation states and promotes synaptic and cognitive feature

Species-Specific Isotope Dilution Methodology for the SI Traceable Quantification of Serum Human Ferritin Light Chain (FTL).

There is an urgent need for reference methodologies to assign SI traceable values to quality control (QC) materials for protein biomarkers to validate biochemical tests currently used in disease diagnosis and patient-specific therapeutics. Here we describe a novel methodology specifically developed for the SI traceable quantification of human serum ferritin light chain (FTL), an established biomarker for the diagnosis of iron-related disorders such as hemochromatosis. Natural and 34S isotopically enriched human FTL standards were recombinantly expressed using the HEK-293 mammalian cell system.

Ferritin Light Chain Alleviates Cerebral Ischemic-Reperfusion Injury-Induced Neuroinflammation via the HIF1α Mediated NF-κB Signaling Pathways.

Ferritin light chain (FtL) is a complex formed by apoferritin and iron core and is one of the main storage forms of iron. Currently, the precise role of FtL in cerebral ischemia/reperfusion injury (CIRI) remains undetermined. This investigation aimed to elucidate the roles and underlying mechanisms of FtL in CIRI. To induce CIRI, an oxygen-glucose deprivation (OGD) model in microglia and middle cerebral artery occlusion (MCAO) model were established using C57BL/6 J mice. The in vivo and in vitro FtL expression patterns were assessed. Furthermore, the potential regulatory mechanism of FtL at th

Ferritin heavy chain supports stability and function of the regulatory T cell lineage.

Regulatory T (TREG) cells develop via a program orchestrated by the transcription factor forkhead box protein P3 (FOXP3). Maintenance of the TREG cell lineage relies on sustained FOXP3 transcription via a mechanism involving demethylation of cytosine-phosphate-guanine (CpG)-rich elements at conserved non-coding sequences (CNS) in the FOXP3 locus. This cytosine demethylation is catalyzed by the ten-eleven translocation (TET) family of dioxygenases, and it involves a redox reaction that uses iron (Fe) as an essential cofactor. Here, we establish that human and mouse TREG cells express Fe-regulat
