Deficient arsenic methylation and global proteomic reprogramming in human keratinocytes during arsenic-induced skin carcinogenesis.

Chronic inorganic arsenic (iAs) exposure affects > 220 million people worldwide and skin cancer is a hallmark of long-term iAs exposure. Limited information exists regarding arsenic methylation by human keratinocytes and how methylation influences skin carcinogenesis. Inorganic arsenite (iAsIII) and its methylated metabolites disrupt diverse zinc finger proteins, leading to differential toxicity patterns. We examined arsenic methylation capacity in non-malignant human keratinocytes and interrogated proteomic remodeling across three stages of iAsIII induced malignant transformation using the we

Unveiling the molecular basis of arsenic resistance in Dietzia maris isolated from arsenic-contaminated soil.

BACKGROUND: The highly toxic nature and uncontrolled release of arsenic from anthropogenic activities into ecosystems create a huge health concern globally. Being a class 1 carcinogen, arsenic is affecting billions of people in both developed and developing countries. RESULTS: Dietzia maris strain sha9 was isolated from industrially contaminated soil, screened phenotypically and genetically, and investigated for its potential in arsenic resistance and bioremediation. Minimum inhibitory concentration assay showed that it can tolerate < 36 mM arsenite and < 185 mM arsenate along with other mult

Cotreatment strategy for hazardous arsenic-calcium residue and siderite tailings via arsenic fixation as scorodite.

Siderite tailings is a potentially cost-free iron (Fe) source for arsenic (As) fixation in hazardous arsenic-calcium residues (ACR) as stable scorodite. In this study, a pure siderite reagent was employed to investigate the mechanism and optimal conditions for As fixation in ACR via scorodite formation, while the waste siderite tailings were used to further demonstrate the cotreatment method. The cotreatment method starts with an introduction of sulfuric acid to the ACR for As extraction and gypsum precipitation, and is followed by the addition of H2O2 to oxidize As(III) in the extraction solu

Consequences of Arsenic Contamination on Plants and Mycoremediation-Mediated Arsenic Stress Tolerance for Sustainable Agriculture.

Arsenic contamination in water and soil is becoming a severe problem. It is toxic to the environment and human health. It is usually found in small quantities in rock, soil, air, and water which increase due to natural and anthropogenic activities. Arsenic exposure leads to several diseases such as vascular disease, including stroke, ischemic heart disease, and peripheral vascular disease, and also increases the risk of liver, lungs, kidneys, and bladder tumors. Arsenic leads to oxidative stress that causes an imbalance in the redox system. Mycoremediation approaches can potentially reduce the

Quantifying Inorganic Arsenic and Other Water-Soluble Arsenic Species in Human Milk by HPLC/ICPMS.

Because the toxicity of arsenic depends on its chemical form, risk assessments of arsenic exposure must consider the type of arsenic compound, and hence they require sensitive and robust methods for their determination. Furthermore, the assessment should include studies on the most vulnerable people within a population, such as newborns and infants, and thus there is a need to quantify arsenic species in human milk. Herein we report a method for the determination of arsenic species at low concentrations in human milk by HPLC/ICPMS. Comparison of single and triple quadrupole mass analysers show

Monitoring of arsenic exposure with speciated urinary inorganic arsenic metabolites for ion implanter maintenance engineers.

For wafer fabrication in the semiconductor industry, maintenance engineers are potentially exposed to hazards during their work of disassembling machine components for cleanup. One special concern is the presence of arsenic or arsenic compounds in the working environment. This study analyzed speciated urinary inorganic arsenic metabolites of the maintenance engineers using high-performance liquid chromatography-hydride generation atomic absorption spectrometry to study the potential arsenic exposure during their maintenance work. In total, from six wafer fabrication facilities, 30 maintenance

Impacts of arsenic contamination on plants and the role of microalgae in arsenic stress mitigation for sustainable agriculture.

Arsenic (As) is a toxic metalloid that is accumulating in agricultural soils and irrigation water. As is occurring from both natural geochemical processes and various anthropogenic activities. Consequently, arsenic contamination is a serious global concern, negatively affecting plant productivity, food safety, and human health. Plants absorb As from the soil through their roots via various transport mechanisms. The absorbed As is further translocated to the shoots and other tissues, where it involved in several metabolic activities and hinders in various physiological and biochemical processes

Associations of arsenic exposure and arsenic metabolism with the risk of non-alcoholic fatty liver disease.

Growing evidences supported that arsenic exposure contributes to non-alcoholic fatty liver disease (NAFLD) risk, but findings were still inconsistent. Additionally, once absorbed, arsenic is methylated into monomethyl and dimethyl arsenicals. However, no studies investigated the association of arsenic metabolism with NAFLD. Our objectives were to evaluate the associations of arsenic exposure and arsenic metabolism with NAFLD prevalence. We conducted a case-control study with 1790 participants derived from Dongfeng-Tongji cohort and measured arsenic species (arsenite, arsenate, monomethylarsona

Synthesis and characterization of arsenate-intercalated layered double hydroxides (LDHs): prospects for arsenic mineralization.

The arsenate-intercalated layered double hydroxide (LDH) of Mg and Al is synthesized by coprecipitation. The higher thermodynamic stability and the consequent lower solubility of the unitary arsenates preclude the formation of arsenate-intercalated LDHs of other metals directly from solution. However other M/Al-AsO(4) (M=Co, Ni, Zn) LDHs could be prepared by anion exchange, showing that arsenate intercalation proceeds topotactically. The intercalation of various species of As(V) into the interlayer of LDHs and the subsequent arsenate carrying capacity are dependent upon the pH of the solution.

Diastereoisomerism of thiol complexes of arsenic acids and pseudoasymmetry of arsenic: a 1H and 13C NMR study.

Diastereoisomeric complexes of methylphenylarsinic acid and (L)-glutathione could be partially separated by HPLC, but the separated compounds rapidly racemized, presumably by pyramidal inversion at the arsenic atom. Hydrolysis of the diastereoisomeric complexes yielded methylphenylarsinous acid as a pair of enantiomers revealed by a 1H NMR study with an asymmetric lanthanide shift reagent. Methylphenylarsinous acid was also synthesized as an enantiomeric pair, shown by an asymmetric shift reagent experiment, by the hydrolysis of iodomethylphenylarsine. 1H and 13C NMR spectroscopy were used to

[Hygienic evaluation of the effects of vibration and arsenic in miners engaged in extraction of arsenic-containing ore].

Hygienic evaluation covered work conditions on 4 gold mines including 3 ones for gold and arsenic and one "arsenic-free". The mines demonstrated similar occupational factors, but differed in arsenic content of the ore extracted and therefore in arsenic concentrations in air of the workplaces. Miners engaged into extraction of the arsenic-containing ore, especially those exposed to vibration and arsenic concentrations over the MAC, manifest extensive arsenic accumulation in the hair and excretion with the urine, higher morbidity with transitory disablement, more prevalent vibration disease and

Effect of Levofloxacin and moxifloxacin on arsenic metabolism and cardiotoxicity in APL patients treated with arsenic trioxide.

Arsenic trioxide (ATO) is the hallmark therapy for acute promyelocytic leukemia (APL); however, its clinical utility is limited by severe cardiotoxicity. APL patients develop neutropenia and immunodeficiency due to bone marrow suppression, increasing susceptibility to community-acquired pneumonia (CAP). Levofloxacin (LFX) and moxifloxacin (MFX) serve as first-line antimicrobial agents for treating CAP in patients with APL. This study quantitatively detected plasma concentrations of inorganic arsenic (iAs), monomethylarsonic acid (MMAV), and dimethylarsinic acid (DMAV) in 96 APL patients underg

Competitive adsorption of arsenate and phosphate on hematite facets: Molecular insights for enhanced arsenic retention.

Understanding the competition for adsorption between arsenate and other common oxyanions at mineral-water interfaces is critical for enhancing arsenate retention in the subsurface environment and mitigating exposure risks. This study investigated the competitive adsorption between arsenate and phosphate on hematite facets using batch experiments, together with in-situ infrared spectroscopy, two-dimensional correlation spectroscopy (2D-COS), and ab initio molecular dynamic (AIMD) simulations. This study's findings revealed that hematite exhibited notable selectivity for arsenate over phosphate

Toxicity, arsenic speciation and characteristics of hyphenated techniques used for arsenic determination in vegetables. A review.

Arsenic (As) speciation is an interesting topic because it is well recognized that the toxicity of this metalloid ultimately depends on its chemical form. More than 300 arsenicals exist naturally. However, As can be present in four oxidation states: As-III, As0, AsIII and AsV. Long-term exposure to As from different sources, such as anthropogenic processes, or water, fauna and flora contaminated with As, has put human health at risk for decades. There are many side-effects correlated with exposure to InAs species, such as skin problems, respiratory diseases, kidney problems, cardiovascular dis

Arsenic trioxide and other arsenical compounds inhibit the NLRP1, NLRP3, and NAIP5/NLRC4 inflammasomes.

Inflammasomes are large cytoplasmic multiprotein complexes that activate caspase-1 in response to diverse intracellular danger signals. Inflammasome components termed nucleotide-binding oligomerization domain-like receptor (NLR) proteins act as sensors for pathogen-associated molecular patterns, stress, or danger stimuli. We discovered that arsenicals, including arsenic trioxide and sodium arsenite, inhibited activation of the NLRP1, NLRP3, and NAIP5/NLRC4 inflammasomes by their respective activating signals, anthrax lethal toxin, nigericin, and flagellin. These compounds prevented the autopro

Evidence against the nuclear in situ binding of arsenicals--oxidative stress theory of arsenic carcinogenesis.

A large amount of evidence suggests that arsenicals act via oxidative stress in causing cancer in humans and experimental animals. It is possible that arsenicals could bind in situ close to nuclear DNA followed by Haber-Weiss type oxidative DNA damage. Therefore, we tested this hypothesis by using radioactive (73)As labeled arsenite and vacuum filtration methodology to determine the binding affinity and capacity of (73)As arsenite to calf thymus DNA and Type 2A unfractionated histones, histone H3, H4 and horse spleen ferritin. Arsenicals are known to release redox active Fe from ferritin. At c

Secondary Mobility of Arsenic Due to In Situ-Generated Iron-Arsenic Colloids upon Iron Oxide Amendments.

Applying iron (Fe) amendments to arsenic (As)-contaminated groundwater to form As-laden Fe precipitates represents a promising in situ remediation approach. However, less is known about the secondary risk of As mobilization due to formation of Fe-As colloids released from these precipitates. Here, we systematically explored the nanoscale formation, composition, and stability of Fe-As colloids in high-As contaminated groundwater following the addition of Fe salts under oxic conditions. Batch experiments show that total aqueous As is incorporated into settled particles upon adding Fe(II) and Fe(

LCA of Disposal Practices for Arsenic-Bearing Iron Oxides Reveals the Need for Advanced Arsenic Recovery.

Iron (Fe)-based groundwater treatment removes carcinogenic arsenic (As) effectively but generates toxic As-rich Fe oxide water treatment residuals (As WTRs) that must be managed appropriately to prevent environmental contamination. In this study, we apply life cycle assessment (LCA) to compare the toxicity impacts of four common As WTR disposal strategies that have different infrastructure requirements and waste control: (i) landfilling, (ii) brick stabilization, (iii) mixture with organic waste, and (iv) open disposal. The As disposal toxicity impacts (functional unit = 1.0 kg As) are compare

Attenuation of DNA damage-induced p53 expression by arsenic: a possible mechanism for arsenic co-carcinogenesis.

Inhibition of DNA repair processes has been suggested as one predominant mechanism in arsenic co-genotoxicity. However, the underlying mode of action responsible for DNA repair inhibition by arsenic remains elusive. To further elucidate the mechanism of repair inhibition by arsenic, we examined the effect of trivalent inorganic and methylated arsenic metabolites on the repair of benzo(a)pyrene diol epoxide (BPDE)-DNA adducts in normal human primary fibroblasts and their effect on repair-related protein expression. We observed that monomethylarsonous acid (MMA(III)) was the most potent inhibito

Arsenic and other heavy metals in soils from an arsenic-affected area of West Bengal, India.

Domkal is one of the 19, out of 26 blocks in Murshidabad district where groundwater contains arsenic above 0.05 mg/l. Many millions of cubic meters of groundwater along with arsenic and other heavy metals are coming out from both the hand tubewells, used by the villagers for their daily needs and shallow big diameter tubewells, installed for agricultural irrigation and depositing on soil throughout the year. So there is a possibility of soil contamination which can moreover affect the food chain, cultivated in this area. A somewhat detailed study was carried out, in both micro- and macrolevel,
