Transgenic approach to increase artemisinin content in Artemisia annua L.

Artemisinin, the endoperoxide sesquiterpene lactone, is an effective antimalarial drug isolated from the Chinese medicinal plant Artemisia annua L. Due to its effectiveness against multi-drug-resistant cerebral malaria, it becomes the essential components of the artemisinin-based combination therapies which are recommended by the World Health Organization as the preferred choice for malaria tropica treatments. To date, plant A. annua is still the main commercial source of artemisinin. Although semi-synthesis of artemisinin via artemisinic acid in yeast is feasible at present, another promising

Discovery and characterization of a novel marafivirus from Artemisia argyi plants.

Artemisia argyi is a significant medicinal plant in China. To date, no viruses have been reported to infect A. argyi. In this study, using the RNA-seq technique, we identified a novel marafivirus in A. argyi plants showing leaf mottle and albinism symptoms, which we have tentatively named “Artemisia argyi albinism-associated virus” (AAAaV). The AAAaV genome is a positive-sense single-stranded RNA with a length of 6749 nt, excluding the poly(A) tail, containing a single open reading frame (ORF) encoding a large polyprotein. Phylogenetic analysis based on the full-length genome and polyprotein a

Effects of phosphorus stress on the growth and secondary metabolism of Artemisia argyi.

Phosphorus is essential in critical plant processes such as signaling, photosynthesis, energy metabolism, and enzyme activity during respiration. Phosphorus stress therefore has a significant impact on plant growth and metabolism. Here, we characterized the biochemical responses of Artemisia argyi Level. et Vant to low phosphorus (LP) and high phosphorus (HP) stress. Plants were treated with 0 g (LP), 1.5 g (control), or 3 g (HP) P per 10 kg of soil. The results demonstrated that CK encouraged the most plant growth, as quantified by leaf size and plant biomass. We also found that the total amo

Allelopathic effect of Artemisia argyi on the germination and growth of various weeds.

Allelopathy means that one plant produces chemical substances to affect the growth and development of other plants. Usually, allelochemicals can stimulate or inhibit the germination and growth of plants, which have been considered as potential strategy for drug development of environmentally friendly biological herbicides. Obviously, the discovery of plant materials with extensive sources, low cost and markedly allelopathic effect will have far-reaching ecological impacts as the biological herbicide. At present, a large number of researches have already reported that certain plant-derived alle

Artemisia santolinifolia enhances glutamatergic neurotransmission in the nucleus of the solitary tract.

Artemisia extracts have been used as remedies for a variety of maladies related to metabolic and gastrointestinal control. Because the vagal afferent-nucleus of the solitary tract (NST) synapse regulates the same homeostatic functions affected by Artemisia, it is possible that these extracts may have activity at the synaptic level in the NST. Therefore, we evaluated how extracts of three common medicinal Artemisia species, Artemisia santolinifolia (SANT), Artemisia scoparia (SCO), and Artemisia dracunculus L (PMI-5011), modulate the excitability of the glutamatergic vagal afferent-NST synapse.

Ethnopharmacology in overdrive: the remarkable anti-HIV activity of Artemisia annua.

ETHNOPHARMACOLOGICAL RELEVANCE: Artemisia annua contains the well-known antimalarial compound artemisinin, which forms the backbone of the global malaria treatment regime. In African countries a tea infusion prepared from Artemisia annua has been used for the treatment of malaria only for the past 10-20 years. Several informal claims in Africa exist that the Artemisia annua tea infusions are also able to inhibit HIV. Since HIV is a relatively newly emerged disease, the claims, if substantiated, could provide a very good example of "ethnopharmacology in overdrive". The objective of this study w

AabHLH140 simultaneously enhances artemisinin and polymethoxylated flavonoid accumulation in Artemisia annua.

Plants provide a wide array of specialized metabolites, which serve as important sources for pharmaceuticals, food additives, and cosmetic ingredients. The biosynthesis of these metabolites is orchestrated by transcription factors. Parallel regulation of multiple metabolites can enhance metabolite content and reduce production costs from a metabolic engineering perspective. However, such coordinated regulation remains rarely reported. In this study, we characterized a bHLH transcription factor, AabHLH140, from Artemisia annua. Artemisinin and polymethoxylated flavonoid content were strongly pr

Hepatorenal protective activity of Artemisia against diclofenac toxicity in male rats.

INTRODUCTION: Artemisia is one of the important alternative treatments for many diseases, as well as the prevention of the effect of oxidizing substances that cause damage to the various organs of the body, including the liver and kidneys. The kidney and the liver are considered the body's most critical organs, and their functions in storage, metabolism, detoxification and elimination of medications, and their metabolic products make them target structures for "drug-induced" harm. The goal of this investigation was to see if Artemisia extract might protect hepatic and renal tissues from diclof

Expression of key genes affecting artemisinin content in five Artemisia species.

Artemisinin, an effective anti-malarial drug is synthesized in the specialized 10-celled biseriate glandular trichomes of some Artemisia species. In order to have an insight into artemisinin biosynthesis in species other than A. annua, five species with different artemisinin contents were investigated for the expression of key genes that influence artemisinin content. The least relative expression of the examined terpene synthase genes accompanied with very low glandular trichome density (4 No. mm-2) and absence of artemisinin content in A. khorassanica (S2) underscored the vast metabolic capa

Dried-leaf Artemisia annua: A practical malaria therapeutic for developing countries?

Artemisinin from the plant Artemisia annua (A. annua) L, and used as artemisinin combination therapy (ACT), is the current best therapeutic for treating malaria, a disease that hits children and adults especially in developing countries. Traditionally, A. annua was used by the Chinese as a tea to treat "fever". More recently, investigators have shown that tea infusions and oral consumption of the dried leaves of the plant have prophylactic and therapeutic efficacy. The presence of a complex matrix of chemicals within the leaves seems to enhance both the bioavailability and efficacy of artemisi
