Role of antioxidant genes for the activity of artesunate against tumor cells.

The antimalaria drug, artesunate (ART), is very cytotoxic in tumor cell lines. The active moiety of ART is an endoperoxide bridge that generates carbon-centered free radicals and oxidative stress upon cleavage. Oxidative stress appears to be necessary for the antimalarial activity of ART. To test whether antioxidant gene expression affects the ART response in tumor cell lines we compared the baseline antioxidant mRNA gene expression in the 55 human tumor cell line panel from the National Cancer Institute Developmental Therapeutics Program to the ART IC50. Thioredoxin reductase expression showe

Artesunate protects against ischemic stroke through THBS2/CD36/TLR4 pathway.

Stroke remains a major global health challenge, causing irreversible neuronal damage with limited therapeutic options. Artesunate (ART), a derivative of artemisinin, has shown anti-inflammatory and neuroprotective properties, but its mechanisms in protecting against ischemic stroke are not fully elucidated. This study investigated the potential mechanisms of ART in mitigating experimental ischemic stroke using a transient middle cerebral artery occlusion (tMCAO) mouse model and cellular oxygen-glucose deprivation/reperfusion (OGD/R) models. The results demonstrated that ART significantly reduc

Delayed autoimmune haemolytic anaemia after artesunate therapy for severe malaria.

We report the case of a 51-year-old man with severe malaria, who developed delayed autoimmune haemolytic anaemia following artesunate therapy. Delayed autoimmune haemolytic anaemia following artesunate therapy has been previously described. Its diagnosis can be challenging in the setting of possible dengue coinfection. Clinicians should be vigilant of this potential side effect of artesunate therapy, especially in patients who later develop signs and symptoms of haemolytic anaemia.

Treatment for Severe Malaria: Post-Artesunate Delayed Haemolysis and Neutropenia.

Parenteral artesunate (AS) is the WHO first-line treatment recommended in adults and children for severe malaria. Post-artesunate delayed haemolysis (PADH) is an uncommon adverse reaction to AS with a mechanism that is not fully understood, occurring in adults and children. Neutropenia is another possible finding after AS treatment, albeit rare. We present the case of a child who experienced both effects after treatment with AS for imported severe Falciparum malaria with very high parasitaemia. In addition, thirty-five paediatric cases of PADH, five cases of delayed anaemia without known haemo

Declining efficacy of artesunate plus sulphadoxine-pyrimethamine in northeastern India.

BACKGROUND: Anti-malarial drug resistance in Plasmodium falciparum in India has historically travelled from northeast India along the Myanmar border. The treatment policy for P. falciparum in the region was, therefore, changed from chloroquine to artesunate (AS) plus sulphadoxine-pyrimethamine (SP) in selected areas in 2005 and in 2008 it became the first-line treatment. Recognizing that resistance to the partner drug can limit the useful life of this combination therapy, routine in vivo and molecular monitoring of anti-malarial drug efficacy through sentinel sites was initiated in 2009. FeS embedded bioreactor collaborate with artesunate for cascade-catalytic tumor ferroptosis.

Ferroptosis, a non-apoptotic programmed cell death modality, has been recognized as an emerging therapeutic target for cancer treatment, particularly with the rapid advancements in bionanotechnology. However, the insufficient intracellular Fe2+ ions and low reactive oxygen species (ROS) production severely restrict the efficacy of ferroptosis at tumor sites. Herein, a pH-responsive multifunctional nanoplatform (p-COF@GOx-FeS@HA/ART) was constructed to achieve efficient tumor ferroptosis through self-supplied Fe2+ ions and amplified ROS. In this system, the large specific surface area and mesop

Factors determining sensitivity or resistance of tumor cell lines towards artesunate.

Clinical oncology is still challenged by the development of drug resistance of tumors that result in poor prognosis for patients. There is an urgent necessity to understand the molecular mechanisms of resistance and to develop novel therapy strategies. Artesunate (ART) is an anti-malarial drug, which also exerts profound cytotoxic activity towards cancer cells. We first applied a gene-hunting approach using cluster and COMPARE analyses of microarray-based transcriptome-wide mRNA expression profiles. Among the genes identified by this approach were genes from diverse functional groups such as s

Artesunate misuse and Plasmodium falciparum malaria in traveler returning from Africa.

Plasmodium falciparum malaria developed in an African-born traveler who returned to Canada after visiting Nigeria. While there, she took artesunate prophylactically. Isolates had an elevated 50% inhibitory concentration to artemisinin, artesunate, and artemether, compared with that of other African isolates. Inappropriate use of artemisinin derivatives can reduce P. falciparum susceptibility.

Label-free proteomics and single-cell mass cytometry analysis of 5 immune tissues from mice with artesunate administration reveal immunosuppression of artesunate via BST2 and MDSCs.

Artesunate is a worldwide prevalent antimalarial medicine, which has also been demonstrated for anti-tumor, anti-virus and anti-autoimmune diseases. Yet the underlying molecular and cellular mechanisms remain largely unexplored. Here, we examined immunological features of 5 immune tissues (spleen, bone marrow, thymus, lymph nodes and peritoneal cavity) of mice with artesunate administration through label-free proteomics and high-dimensional single-cell mass cytometry (CyTOF). Label-free proteomics uncovered dramatic alterations for the interferon-I (IFN-I) induced proteins, especially BST2 and

Effect of artesunate and artemether against Opisthorchis felineus in rodent model.

A comparative in vivo assessment of the artemisinin derivatives artesunate and artemether (150 and 300 mg/kg) and praziquantel (400 mg/kg) was conducted in a hamster model of opisthorchiasis caused by Opisthorchis felineus. Physiological, biochemical, and hematological parameters, including complete blood counts and peripheral blood cell composition, were evaluated in all experimental groups. The animals were infected with 100 metacercariae of O. felineus and 1.5 months post-infection received a single oral dose of the tested compounds. Food consumption was monitored before the experiment ende

Artesunate-induced anaemia: A case report of a pharmacological complication to consider.

We present the case of a 52-year-old male, with a recent history of travel to Ghana, who was admitted to the ICU due to septic shock caused by severe malaria, with a parasitaemia index of 50 %. He received treatment with intravenous artesunate followed by dihydroartemisinin (40 mg) plus piperaquine (320 mg). Despite presenting with multiorgan failure upon admission, the patient demonstrated significant clinical improvement and a rapid clearance of parasitaemia, with a parasitaemia index of 0 % at 72 h after treatment's initiation. However, he developed severe anaemia with haemoglobin levels as

Synthesis of Artesunic Acid-Coumarin Hybrids as Potential Antimelanoma Agents.

Current therapy against melanoma relies on surgical treatment or, in alternative, on conventional drug therapy. Often these therapeutic agents are ineffective due to the development of resistance phenomena. For this purpose, chemical hybridization emerged as an effective strategy to overcome the development of drug resistance. In this study, a series of molecular hybrids were synthesized combining the sesquiterpene artesunic acid with a panel of phytochemical coumarins. Cytotoxicity, antimelanoma effect, and cancer selectivity of the novel compounds were evaluated by MTT assay on primary and m

Artesunate in glioblastoma therapy: Case reports and review of clinical studies.

BACKGROUND: Artesunate, a derivative of the active ingredient artemisinin from Artemisia annua L. used for centuries in the traditional Chinese medicine, is being applied as front-line drug in malaria treatment. As it is cytotoxic for cancer cells, trials are ongoing to include this drug as supplement in cancer therapy. In glioblastoma cells, artesunate was shown to induce oxidative stress, DNA base damage and double-strand breaks (DSBs), apoptosis, and necroptosis. It also inhibits DNA repair functions and bears senolytic activity. Compared to ionizing radiation, DNA damages accumulate over t

Degradation kinetics of artesunate for the development of an ex-tempore intravenous injection.

BACKGROUND: Artesunate is recommended by the World Health Organization (WHO) for parenteral treatment of severe Plasmodium falciparum malaria. However, artesunate is inherently unstable in an aqueous solution and hydrolyses rapidly after its preparation for injection. Therefore, the aim of the study was to evaluate the stabilizing effects of phosphate buffer and mannitol against short-term (ex-tempore) artesunate hydrolysis. METHODS: A HPLC-UV isocratic method was developed using a reversed-phase fused core column (HALO RP-C18) and a mobile phase consisting of a mixture of 45% ammonium format

Targeted liposomes for combined delivery of artesunate and temozolomide to resistant glioblastoma.

The effective treatment of glioblastoma (GBM) is a great challenge because of the blood-brain barrier (BBB) and the growing resistance to single-agent therapeutics. Targeted combined co-delivery of drugs could circumvent these challenges; however, the absence of more effective combination drug delivery strategies presents a potent barrier. Here, a unique combination ApoE-functionalized liposomal nanoplatform based on artesunate-phosphatidylcholine (ARTPC) encapsulated with temozolomide (ApoE-ARTPC@TMZ) was presented that can successfully co-deliver dual therapeutic agents to TMZ-resistant U251
