Aspergillus oryzae fermentation modifies the composition of Massa Medicata Fermentata and its effects on the gut microbiota in mice with spleen-deficiency constipation.

This study investigated the effects of Aspergillus oryzae fermentation on the chemical composition of Massa Medicata Fermentata (MMF) and the gut microbiota in mice with spleen-deficiency constipation by directly comparing unfermented MMF (UFL) with A. oryzae-fermented MMF (AOFL). We examined whether A. oryzae fermentation altered the small-molecule chemical profile of MMF and whether, compared with UFL, AOFL was associated with more pronounced changes in selected host functional indices and gut microbial profiles. The model was induced using Folium Sennae decoction, restricted water access, a

Iron-enriched Aspergillus oryzae as an alternative to iron sulphate to limit iron accumulation, growth and motility of the enteric pathogen S. Typhimurium.

Excess unabsorbed iron in the gastrointestinal tract may select for enteric pathogens and increase the incidence and severity of infectious disease. Aspergillus oryzae (Ao) is a filamentous fungus that has the ability to accumulate and store large amounts of iron, and when used as a supplement or fortificant, has similar absorption to ferrous sulphate (FeSO4) in humans. The objective of this study was to determine the effect of iron-enriched Ao (Ao iron) compared with FeSO4 on iron accumulation, growth and motility of the Gram-negative enteric pathogen, S. Typhimurium. S. Typhimurium was cultu

Studies on the respiratory system of Aspergillus oryzae. V. Some properties of the respiratory system of mitochondria from mycelia grown in the presence of chloramphenicol.

Presence of chloramphenicol in the growth medium for mycelia of Aspergillus oryzae was without effect on the oxidative activity, respiratory control, or P/O ratio of isolated mitochondria. The mitochondria oxidized Krebs cycle intermediates even in the presence of cyanide at the concentration markedly inhibiting the normal mitochondrial oxidation. However, the P/O ratio during the mitochondrial oxidation decreased by about 1.0 on addition of cyanide. The c-type cytochromes, shown to occur in large amounts than in normal mitochondria (Wakiyama and Ogura, 1972), were suggested to act as electron

A robust whole-cell biocatalyst that introduces a thermo- and solvent-tolerant lipase into Aspergillus oryzae cells: characterization and application to enzymatic biodiesel production.

To develop a robust whole-cell biocatalyst that works well at moderately high temperature (40-50°C) with organic solvents, a thermostable lipase from Geobacillus thermocatenulatus (BTL2) was introduced into an Aspergillus oryzae whole-cell biocatalyst. The lipase-hydrolytic activity of the immobilized A. oryzae (r-BTL) was highest at 50°C and was maintained even after an incubation of 24-h at 60°C. In addition, r-BTL was highly tolerant to 30% (v/v) organic solvents (dimethyl carbonate, ethanol, methanol, 2-propanol or acetone). The attractive characteristics of r-BTL also worked efficiently o

Assessment of Acute Serum Iron, Non-Transferrin-Bound Iron, and Gastrointestinal Symptoms with 3-Week Consumption of Iron-Enriched Aspergillus oryzae Compared with Ferrous Sulfate.

BACKGROUND: Iron deficiency anemia (IDA) is a widespread nutritional deficiency, and iron supplementation, especially with ferrous sulfate (FeSO4), is the most common strategy to treat IDA; however, compliance is often poor with daily FeSO4 owing to negative side effects. In a previous study, iron from iron-enriched Aspergillus oryzae [Ultimine® Koji Iron (ULT)] was absorbed similarly to FeSO4. OBJECTIVES: The main objective of this study was to assess the safety of consuming ULT in terms of increasing non-transferrin-bound iron (NTBI) and gastrointestinal distress. METHODS: Young female par

Synchronous production of bioethanol and short-chain fatty acids associated with microbial mechanisms via the short-term cultivation of waste molasses inoculated with Aspergillus oryzae.

This study proposes a novel approach for the concurrent production of short-chain fatty acids (SCFAs) and bioethanol, during a 96 h cultivation of waste molasses (with direct air exposure) inoculated with Aspergillus oryzae using vermiculite as a carrier. Results showed that fungal-bacterial symbiotes were formed by enriching acidogens for the bioconversion of SCFAs, such as Enterococcus, Bacillus and Pseudomonas and bioethanol producers, like Klebsiella, Candida tropicalis, Aspergillus oryzae and Barnettozyma californica. The Aspergillus oryzae was found to play various diverse roles during t

A combination of acid lactase from Aspergillus oryzae and yogurt bacteria improves lactose digestion in lactose maldigesters synergistically: A randomized, controlled, double-blind cross-over trial.

BACKGROUND & AIMS: Lactose digestion can be improved in subjects with impaired or completely absent intestinal lactase activity by administration of lactase preparations and particularly of acid lactase, which is active in the stomach, or by yogurt containing live lactic acid bacteria. It is the question, if lactose digestion can be further enhanced by combining these two approaches. METHODS: We investigated in a randomised, placebo-controlled, double-blind, 5-arm crossover study on 24 lactose malabsorbers with variable degrees of lactase deficiency if different lactase preparations and freez

Comparison of the aflR gene sequences of strains in Aspergillus section Flavi.

Aflatoxins are polyketide-derived secondary metabolites produced by Aspergillus parasiticus, Aspergillus flavus, Aspergillus nomius and a few other species. The toxic effects of aflatoxins have adverse consequences for human health and agricultural economics. The aflR gene, a regulatory gene for aflatoxin biosynthesis, encodes a protein containing a zinc-finger DNA-binding motif. Although Aspergillus oryzae and Aspergillus sojae, which are used in fermented foods and in ingredient manufacture, have no record of producing aflatoxin, they have been shown to possess an aflR gene. This study exami

The potential of Monascus purpureus pigments of red yeast rice for skin-lightening cosmetics: an in silico pharmacology approach towards human tyrosinase-related protein 1 and Aspergillus oryzae active-tyrosinase and in vivo validation in UVB-exposed Wistar rats.

Monascus purpureus red yeast rice (RYR) was reported to exhibit photoprotective and anti-melanogenic activities in vitro. This study aims to predict the pharmacokinetic properties of Monascus pigments, their binding modes, and complex stability towards tyrosinase enzymes using in silico techniques, validate them in an animal model, and determine the protein and monacolin K in the extract. All the pigments exhibit low absorption, low distribution, low skin permeability, and are unlikely to alter CYP2D6, but may be metabolised by CYP3A4. They interact with tyrosinases, showing a binding mode sim

A case of occupational allergic bronchopulmonary aspergillosis unique to Japan.

A 15-year-old female was diagnosed in 1980 as having allergic bronchopulmonary aspergillosis (ABPA) due to Aspergillus fumigatus based on Rosenberg and Patterson's criteria for the disease. The patient is the eldest daughter of a family of domestic brewers of soy sauce and bean paste in a small village, an occupation unique to Japan. The brewing process involved the use of Aspergillus oryzae as a fermenting agent. The patient had experienced episodic wheezing and pulmonary infiltrates during the same seasons in the previous three years, corresponding to the time of the highest A oryzae spore c

Molecular Confirmation of the Linkage between the Rhizopus oryzae CYP51A Gene Coding Region and Its Intrinsic Voriconazole and Fluconazole Resistance.

Rhizopus oryzae is the most prevalent causative agent of mucormycosis, an increasingly reported opportunistic fungal infection. These Mucorales are intrinsically resistant to Candida- and Aspergillus-active antifungal azole drugs, such as fluconazole (FLC) and voriconazole, respectively. Despite its importance, the molecular mechanisms of its intrinsic azole resistance have not been elucidated yet. The aim of this work was to establish if the Rhizopus oryzaeCYP51 genes are uniquely responsible for intrinsic voriconazole and fluconazole resistance in these fungal pathogens. Two CYP51 genes were

A Comparison of Static and Shake-Flask Cultures for Filamentous Fungal Biomass and L-Carnitine Production in Rum Stillage.

This study investigated the potential of Bundaberg rum stillage as a low-cost medium for cultivating two food-related filamentous fungi: Aspergillus oryzae and Rhizopus oligosporus. Traditional shake-flasks and a novel static-liquid culture fermentation approach were used to assess biomass and L-carnitine synthesis. Stillage was assessed to determine the optimal concentration for use, and this was applied throughout. Growth was benchmarked against a complex synthetic medium containing glucose, yeast extract and malt extract. The highest biomass yields were obtained in the stillage supplemented
