Cerebrospinal fluid concentrations of atracurium, laudanosine and vecuronium following clinical subarachnoid hemorrhage.

BACKGROUND: Neuromuscular blocking agents may exert central nervous system effects when they reach the brain. This study assessed the concentrations and the time course of passage of vecuronium, atracurium, and its metabolite laudanosine in the cerebrospinal fluid (CSF) of patients undergoing intracranial aneurysm clipping. METHODS: Twenty-five patients with subarachnoid hemorrhage were randomly allocated to receive an intravenous infusion of vecuronium (n=13) or atracurium (n=12). Arterial blood and lumbar CSF were sampled before and 1, 2, 3, 4 and 8 h after the start of the relaxant infusio

[The hemodynamic effects of atracurium in geriatric patients during intravenous anesthesia using propofol].

OBJECTIVE: Neuromuscular blocking drugs may produce cardiovascular effects by virtue of drug-induced release of histamine or other vasoactive substances from circulating mast cells, effects at cardiac muscarinic receptors or effects at nicotinic receptors at autonomic ganglia. These haemodynamic reactions may be more pronounced in elderly patients, since cardiovascular compensating mechanisms are attenuated with advancing age. The present study was designed to investigate the cardiovascular effects of atracurium with special regard to the reactions in younger adults and in elderly patients. M

Effects of adding atracurium to Lidocaine solution during intravenous regional anesthesia in dogs.

BACKGROUND: Acute pain management and the efficacy of analgesic therapies are essential in orthopedic surgery on the distal extremities of dogs' forelimbs. This is due to the manipulation of both soft and orthopedic tissues. Therefore, this study aimed to compare the antinociceptive, akinesia, cardiovascular, and plasma-level effects of adding atracurium to intravenous regional anesthesia (IVRA) with lidocaine in dogs. METHODS: Fifty male Mongrel dogs weighing 15 ± 5 kg and aged 2.5 ± 0.6 years were premedicated with 0.045 mg/kg of atropine sulfate and 0.05 mg/kg of acepromazine. While under

Seizure activity following atracurium continuous rate infusion in three mechanically ventilated juvenile dogs.

OBJECTIVE: To describe patient recovery in dogs that had undergone mechanical ventilation (MV) and received atracurium as a continuous rate infusion (CRI). CASE SERIES SUMMARY: Three dogs were treated with atracurium CRI while mechanically ventilated and were successfully weaned. All were pediatric patients (2-3 months old) ventilated due to respiratory fatigue and hypoxemia. All dogs experienced seizure activity in the initial 24 h following extubation and were treated with various anticonvulsant protocols; no patients experienced documented seizures after 48 h of discontinuation from MV. Tetanus-induced changes in apparent recovery after bolus doses of atracurium or vecuronium.

The current study evaluated the duration and magnitude of post-tetanic effects in 56 patients recovering from a bolus dose of nondepolarizing relaxant to assess the impact of tetanus on monitoring in a common clinical setting. After induction of general anesthesia (thiopental, fentanyl, oxygen, nitrous oxide, and isoflurane), a baseline response to train-of-four (TOF) stimulation was recorded using an adductor pollicis force transducer, and the ratio of the fourth response (T4) to the first (T1) was calculated. Patients then received a bolus dose of either atracurium 0.50 mg.kg-1 (n = 28) or v

The effect of dexmedetomidine vs. atracurium on intubation condition in children - a randomized clinical trial.

BACKGROUND: Using neuromuscular blocking agents (NMBA) in pediatric induction protocol is a challenging matter. Therefore, in this study, we aimed to find a safer way for anesthesia in children. We compared the effects of dexmedetomidine with atracurium on intubation conditions in children aged 6-12 years under general anesthesia. If dexmedetomidine has the similar effect as atracurium, we can use it for intubation and anesthesia of children in situations where the use of atracurium is challenging for any reason. METHODS: This clinical trial was carried out on children between 6 and 12 years

Equipotent Dose and Cost Comparison of Atracurium and Rocuronium in Laboratory Pigs Anesthetized with Propofol.

Neuromuscular blocking agents such as atracurium and rocuronium are commonly used during anesthetic procedures in laboratory pigs. However, species-specific dosing guidelines remain limited, leading to reliance on data extrapolated from other species. This prospective, blinded study aimed to determine the equipotent dose for atracurium (A) and rocuronium (R) in laboratory pigs receiving propofol and to compare their cost-effectiveness. Twelve healthy animals were randomly distributed according to the drug administered (n = 6 per group). For both drugs, the infusion rate was adjusted following

The effects of d-tubocurarine, pancuronium and atracurium on the responses of gastrocnemius and soleus muscles in the cat.

In vivo, the effects of d-tubocurarine (0.20 mg kg-1), pancuronium (0.015 mg kg-1) and atracurium (0.15 mg kg-1) on the responses of the indirectly stimulated cat gastrocnemius (fast) and soleus (slow) muscles to a twitch, train-of-four and tetanic stimuli were studied. The soleus muscle demonstrated a greater degree of fade than the gastrocnemius in response to tetanic stimuli (50 Hz). There was no difference between the responses of the two muscles to twitch or train-of-four stimuli with any of the drugs. Recovery of train-of-four ratio occurred more rapidly than did the tetanic fade ratio.

Atracurium-Induced Bronchospasm With Flat Capnograph at Induction of General Anaesthesia: A Case Report.

Benzylisoquinolinium neuromuscular blocking agents can precipitate bronchospasm either through allergy/anaphylaxis or isolated stimulation of mast cell histamine release. This report presents a 75-year-old female who attended the day surgery unit for a rigid cystoscopy under general anaesthesia. She had a hyper-reactive airway history of mild historic asthma and sensitivity to aerosols. After administration of atracurium at induction of anaesthesia, ventilation became challenging with no chest rise and a flat CO2 trace. Repeat video laryngoscopy confirmed correct endotracheal tube position. Th

Efficacy of Atracurium-Vecuronium Combination in Patients Undergoing Laparoscopic Surgery: A Randomized Controlled Study.

INTRODUCTION: Deep neuromuscular blockade (d-NMB) is an essential requirement for carboperitoneum during laparoscopy surgery. However, sustaining d-NMB till the completion of surgery delays the reversal of the residual block. Therefore, there is a merit in exploring the effect of synergistic vecuronium-atracurium combination on the duration-of-action of d-NMB during "laparoscopic" surgery when we compare intubating bolus non-depolarizers (atracurium, vecuronium) administered alone. This study aims to evaluate whether the synergistic effect atracurium-vecuronium combination increases duration-o

Inhibition of the neuromuscular acetylcholine receptor with atracurium activates FOXO/DAF-16-induced longevity.

Transcriptome-based drug screening is emerging as a powerful tool to identify geroprotective compounds to intervene in age-related disease. We hypothesized that, by mimicking the transcriptional signature of the highly conserved longevity intervention of FOXO3 (daf-16 in worms) overexpression, we could identify and repurpose compounds with similar downstream effects to increase longevity. Our in silico screen, utilizing the LINCS transcriptome database of genetic and compound interventions, identified several FDA-approved compounds that activate FOXO downstream targets in mammalian cells. Echocardiography and cardiorespiratory changes post ketofol or atracurium/ketofol on acepromazine-methadone premedicated dogs.

Cardiovascular failure has been recognized as the predominant cause of perioperative mortality in small animals, particularly dogs. This study was designed to evaluate the effectiveness of adding intravenous atracurium to a ketofol infusion during anesthesia in dogs. Thirty male mongrel dogs were premedicated with an intramuscular injection containing 0.02 mg/kg of acepromazine and 0.2 mg/kg of methadone. Thirty minutes later, the dogs were equally and randomly divided into two groups (n = 15): the Ketofol Group (KFG), in which anesthesia was induced using IV administration of 0.5 ml/kg of ket
