Goose astrovirus (GoAstV) is an emerging pathogen responsible for severe gout in goslings, causing substantial economic losses to the waterfowl industry, with no commercially available vaccines to date. In this study, we developed a novel mRNA vaccine expressing the capsid protein of GoAstV, delivered via lipid nanoparticles (LNPs). The vaccine elicited robust humoral and cellular immune responses in mice, including high titers of GoAstV-specific IgG, IgM, and neutralizing antibodies, significantly increased lymphocyte proliferation capacity, and a Th2-biased cytokine profile. In goslings, the

Gosling gout, caused by goose astrovirus 2 (GAstV-2), poses a significant threat to the goose industry due to its high morbidity and mortality, while effective control measures remain unavailable. The development of specific countermeasures is hindered by a limited characterization of monoclonal antibodies (mAbs) and epitopes against the major ORF2 antigen. To address this, we generated two mAbs (A12G1 and A10D12) against GAstV-2 ORF2 using hybridoma technology. Epitope mapping identified two novel linear B-cell epitopes, 1MADRA5 (A12G1) and 36YKPQKLPMKA45 (A10D12), residing in the conserved S

The first evidence of avian nephritis virus (ANV) in ducks is described. A diagnostic investigation was performed on three duck farms in Croatia. Samples from dead-in-shell ducklings and ducklings aged 30 days were collected and prepared for molecular and histopathological examination. Intestinal and liver samples were tested by polymerase chain reaction (PCR) for the presence of ANV, duck enteritis virus, duck hepatitis virus 1 and Derzsy's disease virus. Multiple tissues were collected for histological examination and lesions were found to be confined to the kidney and intestine. Moderate fo

The inducible nitric oxide synthase (iNOS) enzyme has long been recognized as a key mediator of innate immune responses to infectious diseases across the phyla. Its role in killing or inactivating bacterial, parasitic, and viral pathogens has been documented in numerous host systems. iNOS, and its innate immune mediator NO has also been described to have negative consequence on host tissues as well; therefore understanding the pathogenesis of any infectious agent which induces iNOS expression requires a better understanding of the role iNOS and NO play in that disease. Previous studies in our

Several outbreaks of gout were reported in commercial broilers in India during 2011 and 2012, causing up to 40% mortality in the birds. Gross and histopathological observations confirmed gout. Quantitative reverse transcriptase polymerase chain reaction (qRT-PCR) analysis from kidney samples of gout-affected birds indicated the presence of chicken astrovirus (CAstV) in 41.7% of cases and a mixed infection of CAstV and avian nephritis virus (ANV) in 36.4% of cases. CAstV isolated from gout-affected kidneys by inoculating embryonated specific pathogen free (SPF) eggs showed dwarfing in embryos a
