Nematicidal and plant growth-promoting potential of the cell-free culture supernatant of Bacillus megaterium in tomato under Meloidogyne incognita infection: an in vitro and in vivo study.

This study evaluated the nematicidal efficacy and plant growth-promoting potential of the cell-free bacterial culture supernatant (BCS) of Bacillus megaterium in mitigating Meloidogyne incognita-induced stress in Solanum lycopersicum under both in vitro and in vivo conditions. Mortality bioassay revealed that cell-free BCS induced a clear dose- and time-dependent nematicidal response, with maximum juvenile mortality of 100% observed at 100% concentration within 96 h (p < 0.001). B. megaterium was characterized for plant growth-promoting and biocontrol traits including indole-3-acetic acid (IAA

Enhancement of simultaneous gold and copper recovery from discarded mobile phone PCBs using Bacillus megaterium: RSM based optimization of effective factors and evaluation of their interactions.

Bioleaching of Au from mobile phone printed circuit boards (MPPCBs) was studied, using Bacillus megaterium which is a cyanogenic bacterium. To maximize Au extraction, initial pH, pulp density, and glycine concentration were optimized via response surface methodology (RSM). Bioleaching of Cu, an important inhibitor on Au recovery, was also examined. To maximize Au recovery, the optimal condition suggested by the models was initial pH of 10, pulp density of 8.13g/l, and glycine concentration of 10g/l. Under the optimal condition, approximately 72% of Cu and 65g Au/ton MPPCBs, which is 7 times gr

17(S),18(R)-epoxyeicosatetraenoic acid generated by cytochrome P450 BM-3 from Bacillus megaterium inhibits the development of contact hypersensitivity via G-protein-coupled receptor 40-mediated neutrophil suppression.

Dietary intake of ω3 polyunsaturated fatty acids such as eicosapentaenoic acid and docosahexaenoic acid is beneficial for health control. We recently identified 17,18-epoxyeicosatetraenoic acid (17,18-EpETE) as a lipid metabolite endogenously generated from eicosapentaenoic acid that exhibits potent anti-allergic and anti-inflammatory properties. However, chemically synthesized 17,18-EpETE is enantiomeric due to its epoxy group-17(S),18(R)-EpETE and 17(R),18(S)-EpETE. In this study, we demonstrated stereoselective differences of 17(S),18(R)-EpETE and 17(R),18(S)-EpETE in amelioration of skin c

The Effect of Plant Growth Promoting Rhizobacteria (PGPRs) on Yield and Some Quality Parameters during Shelf Life in White Button Mushroom (Agaricus bisporus L.).

The use of different bacteria that increase yield and quality in plant production has become common since the 1990s. However, effects of plant growth promoting rhizobacterial (PGPR) treatments during the cultivation period of white button mushroom on quality during marketing duration are not known exactly. This study was carried out to determine the effects of different PGPRs in compost medium on mushroom yield and quality. For this reason, Azospillum lipoferum, Bacillus megaterium, Frateuria aurantia and Thiobacillus thiooxidans, for promoting nitrogen, phosphorus, potassium and sulfur transp

Priestia megaterium reduces hydrogen sulfide level in swine manure potentially through an assimilatory mechanism and microbiota modulation.

Noxious odors from swine farms pose significant health and environmental concerns. Hydrogen sulfide (H2S) is one of the major compounds causing noxious odors. Using biological inoculants offers a promising, efficient, and cost-effective method to reduce H2S emissions from these farms. Several studies have found that Priestia megaterium (formerly Bacillus megaterium) can help mitigate the production of H2S. However, detailed knowledge regarding its mechanisms of action is still limited. In this study, the potential of novel strain of P. megaterium, strain S188, for the reduction of H2S in swine

Complete genome sequence of Priestia megaterium S188, a hydrogen sulfide-degrading bacterium.

Priestia megaterium (formerly Bacillus megaterium) is a gram-positive, aerobic, spore-forming bacterium found in a wide range of environmental niches. Here, we report the complete genome sequence of P. megaterium S188 isolated from soil, which can decrease hydrogen sulfide (H2S) levels and help reduce malodor generation in livestock farms. Putative genes related to sulfide assimilation and conversion were found in the genome of P. megaterium S188; among these, one O-acetylhomoserine (O-AH) desulfhydrase, two cysteine synthases-primarily related to the biosynthesis of sulfur-containing amino ac

Germination of Bacillus spores by LiCl.

Spores of Bacillus subtilis have been found to germinate when incubated with LiCl, but not with other monovalent or divalent metal cations. Bacillus megaterium spores also germinated with LiCl, but B. cereus spores did not. In B. subtilis, the LiCl germination was via the activation of spores' GerA germinant receptor (GR), and in B. megaterium, it was the GerU GR. Notably, LiCl germination was much slower than normal physiological germinant triggered GR germination. In B. subtilis spores, rates of LiCl germination were increased in spores with a more fluid IM and decreased in spores with a les

Improvement of aquaculture water quality by mixed Bacillus and its effects on microbial community structure.

Microbial remediation, especially the application of probiotics, has recently gained popularity in improving water quality and maintaining aquatic animal health. The efficacy and mechanism of mixed Bacillus for improvement of water quality and its effects on aquatic microbial community structure remain unknown. To elucidate these issues, we applied two groups of mixed Bacillus (Bacillus megaterium and Bacillus subtilis (A0 + BS) and Bacillus megaterium and Bacillus coagulans (A0 + BC)) to the aquaculture system of Crucian carp. Our results showed that the improvement effect of mixed Bacillus A

[Dissociative phases and pathogenicity of different species of the Bacillus genus].

The previously postulated hypothesis, according to which different species of the genus Bacillus show strictly similar morphological and biological properties when the same variants are considered, has been confirmed by the present research. The "S" (smooth) variants of the five studied species (B. anthracis, B. subtilis, B. cereus, B. megaterium, B. mesentericus) are all lethal, at the experimented dose, for mice, whereas the "R" (rough, "star-shaped" colonies) variant of the same strains of the same species are all not pathogenic for the same animals. Likewise the "S" variants of three speci

Occurrence of a barbiturate-inducible catalytically self-sufficient 119,000 dalton cytochrome P-450 monooxygenase in bacilli.

In a recent publication (Narhi, L.O. and Fulco, A.J.[1986] J. Biol. Chem. 261, 7160-7169) we described the characterization of a catalytically self-sufficient 119,000 Dalton cytochrome P-450 fatty acid monooxygenase (P-450BM-3) induced by barbiturates in Bacillus megaterium ATCC 14581. We have now examined cell-free preparations from 12 distinct strains of B. megaterium and from one or two strains each of B. alvei, B. brevis, B. cereus, B. licheniformis, B. macerans, B. pumilis and B. subtilis for the presence of this inducible enzyme. Determination of the X-ray structure of the snake venom protein omwaprin by total chemical synthesis and racemic protein crystallography.

The 50-residue snake venom protein L-omwaprin and its enantiomer D-omwaprin were prepared by total chemical synthesis. Radial diffusion assays were performed against Bacillus megaterium and Bacillus anthracis; both L- and D-omwaprin showed antibacterial activity against B. megaterium. The native protein enantiomer, made of L-amino acids, failed to crystallize readily. However, when a racemic mixture containing equal amounts of L- and D-omwaprin was used, diffraction quality crystals were obtained. The racemic protein sample crystallized in the centrosymmetric space group P2(1)/c and its struct

Molecular Epidemiological Analysis of Bacillus Pseudo-Outbreak due to Contaminated Culture Tubes Containing Stuart Medium.

BACKGROUND: It is challenging to determine whether Bacillus species other than Bacillus anthracis cause infections. Pseudo and true outbreaks of Bacillus spp. have been noted. Here, we present a molecular analysis of a Bacillus spp. pseudo-outbreak caused by contaminated culture tubes containing Stuart medium. METHODS: Between January and March 2015, a high percentage of Bacillus spp. was isolated from the wound samples of inpatients at the Karabuk University Hospital, and an outbreak was suspected. Environmental and staff nasal samples were cultured aerobically, and Bacillus spp. were isolat

Cr(VI) removal performance from wastewater by microflora isolated from tannery effluents in a semi-arid environment: a SEM, EDX, FTIR and zeta potential study.

Hexavalent chromium removal from the environment remains a crucial worldwide challenge. To address this issue, microbiological approaches are amongst the straightforward strategies that rely mainly on the bacteria's and fungi's survival mechanisms upon exposure to toxic metals, such as reduction, efflux system, uptake, and biosorption. In this work, scanning electron microscopy, energy-dispersive X-ray spectrophotometry, Fourier transform infrared spectroscopy, and zeta potential measurements were used to investigate the ability of chromium adsorption by Bacillus licheniformis, Bacillus megate

A closed-loop biorefinery approach for polyhydroxybutyrate (PHB) production using sugars from carob pods as the sole raw material and downstream processing using the co-product lignin.

A novel closed-loop biorefinery model using carob pods as the feed material was developed for PHB production. The carob pods were delignified, and as the second step, sugars present in the delignified carob pods were extracted using water. Ralstonia eutropha and Bacillus megaterium were cultivated on the carob pod extract and its performance was evaluated using Taguchi experimental design. R. eutropha outperformed the B. megaterium in terms of its capability to grow at a maximum initial sugar concentration of 40 g L-1 with a maximum PHB production of 12.2 g L-1. Finally, the concentrated ligni
