An oligonucleotide probe specific for Onchocerca volvulus.

A genomic DNA library of a Liberian strain of Onchocerca volvulus was prepared in the vector bacteriophage lambda gt10. The library was differentially screened by hybridisation with radiolabelled total DNA from the homologous parasite, two heterologous Onchocerca parasites (Onchocerca gibsoni and Onchocerca gutturosa) and human liver cells. A clone (C1A1) was isolated whose binding to O. volvulus DNA was at least 50 times stronger than to the other parasite DNA samples. No binding was observed with human DNA. The insert of C1A1 was subcloned into the filamentous phage vector M13 mp18 and seque

Development of a receptor-based inhibitory penta-unit-conjugated peptide to enhance anthrax toxin neutralization.

Anthrax toxin is a key virulence factor for Bacillus anthracis. The cell-binding component of anthrax toxin, protective antigen (PA), mediates the entry of the toxin into cells by first binding to the extracellular von Willebrand factor A (VWA) domain of the cellular anthrax toxin receptor (ATR). Herein, we targeted the VWA domain of the cellular receptor to develop a more effective antitoxin agent for neutralization of anthrax toxin. We selected ATR-binding peptides by using a phage display: among these, we identified two novel peptides binding to the ATR with high affinity and specificity, a

Four-in-one multifunctional CoCu-NC@AuPt nanozyme integrated M13 phage-displayed nanobody based multimodal lateral flow immunoassay for bovine lactoferrin detection.

Bovine lactoferrin (LF) is widely used as a nutritional additive in infant formula and functional foods, but is also a milk allergen. Therefore, rapid and sensitive detection of LF is crucial for ensuring food quality and protecting public health. Here, we report a multimodal lateral flow immunoassay (LFIA) for LF detection that integrates an M13 phage-displayed nanobody (M13-Nb) with a "four-in-one" multifunctional nanozyme, namely an AuPt nanoparticle-loaded CoCu-embedded nitrogen-doped carbon nanocomposite (CoCu-NC@AuPt). The specific M13-Nb against LF was isolated from a naïve nanobody lib

Bacteriophage-based particles carrying the TNF-related apoptosis-inducing ligand (TRAIL) gene for targeted delivery in hepatocellular carcinoma.

The TRAIL (Tumour Necrosis Factor-Related Apoptosis-Inducing Ligand) is a promising candidate for cancer treatment due to its unique ability to selectively induce programmed cell death, or apoptosis, in cancer cells while sparing healthy ones. This selectivity arises from the preferential binding of the TRAIL to death receptors on cancer cells, triggering a cascade of events that lead to their demise. However, significant limitations in using the TRAIL for cancer treatment are the administration of the TRAIL protein that can potentially lead to tissue toxicity (off-target) and the short half-l

A new biochromatography model based on DNA origami assembled PPARγ: construction and evaluation.

As drug targets, receptors have potential to screen drugs. Silica is an attractive support to immobilize receptors; however, the lack of biocompatibility makes it easier for receptors to lose bioactivity, which remains an obstacle to its widespread use. With the advantage of biocompatibility, DNA origami can be used as a biological carrier to improve the biocompatibility of silica and assemble receptors. In this study, a new biochromatography model based on DNA origami was constructed. A large quantity of M13ssDNA was used as a scaffold, leading to significant costs, so M13ssDNA was self-produ

Quencher-enhanced specificity of psoralen-photosensitized virus inactivation in platelet concentrates.

BACKGROUND: Treatment with psoralens and UVA (PUVA) has been shown to be efficacious in eliminating the risk of virus transmission by platelet concentrates (PCs). It has previously been demonstrated that, during the inactivation of cell-free vesicular stomatitis virus (VSV) by aminomethyltrimethylpsoralen (AMT) and UVA in PCs, platelet function could be protected either by oxygen removal before irradiation or by inclusion of a type I free radical quencher, such as mannitol. STUDY DESIGN AND METHODS: Under previous PUVA treatment conditions for PCs (25 micrograms/mL AMT; 30 min UVA at 7 mW/cm2

Characterization of phage that bind plastic from phage-displayed random peptide libraries.

During routine screenings of random peptide libraries displayed at the N terminus of the pIII coat protein of M13 bacteriophage, clones were isolated that bound directly to the polystyrene (PS) surface used to immobilize the target protein. The plastic-binding phage (P-b phi) bind to both unblocked plastic (PS and polyvinyl chloride, PVC) and plastic blocked with bovine serum albumin (BSA) but require non-ionic detergent to bind to plastic blocked with milk. Comparison of the P-b phi to antibody-binding phage (Ab-b phi) indicates that similar numbers of phage particles are bound, but fewer P-b

A Biological Approach for the Synthesis of Bismuth Nanoparticles: Using Thiolated M13 Phage as Scaffold.

We report the synthesis of Bi nanoparticles (Bi NPs) using the M13 phage as scaffold. The p8 protein of the phage is functionalized with thiol groups of different lengths, and these thiolated regions act as nucleation centers for Bi(3+) ions. The size distribution, shape, and resilience to oxidation of the Bi NPs depend on the length of the thiol group used. The NPs are characterized by high resolution transmission electron microscopy, Raman, and IR spectroscopies, matrix assisted laser desorption/ionization, and optical absorption. These results show that the nanoparticles are crystalline and

Phage-displayed peptides as biosensor reagents.

This study investigated the potential to utilize phage-displayed peptides as reagents in sensor applications. A library of random 12-mers displayed on phage was panned against staphylococcal enterotoxin B (SEB), a causative agent of food poisoning. Nine SEB binding phage clones were isolated, all of which share the consensus sequence Trp His Lys at their amino terminus. Binding of several of these phage was shown to be inhibited when they were assayed in a competitive enzyme-linked immunosorbent assay (ELISA) format with synthesized peptide corresponding to the peptide-encoding region of one o

Silver Nanoparticles Templated by the M13 Phage Exhibit High Antibacterial Activity against Gram-Negative Pathogens and a Reduced Rate of Bacterial Resistance In Vitro.

Silver and silver nanoparticles (AgNPs) are well-known for their antibacterial properties. However, their low potency and the emergence of resistance are major barriers to the use of AgNPs in systemic therapy. Biological products can be used to reduce and cap AgNPs during synthesis from silver salts, but the structure and properties of biotemplated AgNPs are not understood well. We observed that AgNPs templated by the Escherichia coli phage M13 showed unusually high potency as well as activity against multiple Gram-negative pathogens, including E. coli, Pseudomonas aeruginosa, and Vibrio chole

Nucleotides bearing a cleavable genotoxic group on the phosphate.

A new class of nucleotide derivatives in which N-nitroso-pyrrolidine (NPYR) is linked to the phosphate group through an ester linkage at the alpha-carbon of the nitrosamine were prepared by a reaction between a nucleoside monophosphate and alpha-acetoxy-NPYR. The NPYR derivatives were prepared from dpT, dpC, dpA, dpG, pA and dpCpT. Treatment of the NPYR-pX with acid, snake venom phosphodiesterase, or near ultraviolet light caused cleavage of the NPYR group, regenerating pX. All of these nucleotide derivatives were directly mutagenic towards Salmonella typhimurium TA1535. When phage M13mp2 doub

Characterization of phage peptide interaction with antibody using phage mediated immuno-PCR.

Real-time immuno-PCR (RT-IPCR) is a powerful technique that combines ELISA with the specificity and sensitivity of PCR. RT-IPCR of phage-displayed peptides exploits the unique physical associations between phenotype (the displayed peptide) and genotype (the encoding DNA) within the same phage particle. Previously, we identified phage peptides specific for recombinant antibodies (rAbs) prepared from clonally expanded plasma cells in multiple sclerosis (MS) cerebrospinal fluid (CSF) and subacute sclerosing panencephalitis (SSPE) brain. Herein, we applied phage-mediated RT-IPCR to study reactivit

Remediation of surface water contaminated by pathogenic microorganisms using calcium peroxide: Matrix effect, micro-mechanisms and morphological-physiological changes.

Calcium peroxide (CaO2), a common solid peroxide, has been increasingly used in contaminated site remediation due to its ability to release oxygen (O2) and hydrogen peroxide (H2O2) and its environmental friendliness. Our present study is first to explore micromechnisms of CaO2 to efficaciously inactivate pathogen indicators including gram-negative bacterium of Escherichia coli (E. coli), gram-positive bacterium of Staphylococcus aureus (S. aureus), and virus of Escherichia coli-specific M13 bacteriophage (VCSM13) under low concentration (≤ 4 mmol L-1 (mM)). Sunlight mutagenesis: changes in mutational specificity during the irradiation of phage M13mp2.

We reported previously that the mutations in phage M13mp2, a single-stranded DNA phage, induced by sunlight exposure are predominated by G-to-C transversions. We have now made an unexpected observation that an exposure to sunlight for a short period of time results in induction mainly of C-to-T transitions while a longer exposure results in the induction of G-to-C transversions. This peculiar phenomenon suggests that DNA damage formed by initial sunlight exposure can be transformed during an elongated exposure. 7, 8-Dihydro-8-oxoguanine (8-oxoG) in DNA might be involved in the shift of the mut

Mapping of epitopes recognized by PM/Scl autoantibodies with gene-fragment phage display libraries.

Sera from patients suffering from the polymyositis/scleroderma overlap syndrome (PM/Scl) recognize two antigenically non-related proteins with apparent molecular masses of 100 kDa and 75 kDa respectively. The two proteins are part of a particle termed PM/Scl localized in the granular component of the nucleolus. The predominant immunoreactivity of the PM/Scl sera was shown to be directed against the 100 kDa protein. The cDNA of the 100 kDa protein has been cloned recently and its immunogenic regions have been partially mapped using recombinant proteins. Thus far the localization of antigenic de

Identification of peptides specific for antibodies in vitiligo using a phage library.

Patients with vitiligo produce specific autoantibodies that can be detected in their sera. These antibodies are believed to play a role in the pathogenesis of this disease. A random peptide library displayed on phage is a technique that can be used to identify the epitopes that react with monoclonal and polyclonal antibodies. We used this technique to identify the epitopes that react specifically with the vitiligo autoantibodies. By screening the random peptide phage library and using ELISA, two clones that showed a higher frequency of reactivity with the antibodies in the sera of patients wit

Helicobacter pylori-antigen-binding fragments expressed on the filamentous M13 phage prevent bacterial growth.

Colonization of the human stomach by Helicobacter pylori is associated with the development of gastritis, duodenal ulcer, mucosa-associated lymphoid tissue (MALT) lymphoma, and gastric cancer. H. pylori-antigen-binding single-chain variable fragments (ScFv) were derived from murine hybridomas producing monoclonal antibodies and expressed as a g3p-fusion protein on a filamentous M13 phage. The recombinant ScFv-phage reacted specifically with a 30-kDa monomeric protein of a H. pylori surface antigen preparation and by means of immunofluorescence microscopy the phage was shown to bind to both the

An intrastrand d(GpG) platinum crosslink in duplex M13 DNA is refractory to repair by human cell extracts.

We have examined the ability of human cell extracts to repair the most frequent DNA adduct caused by the cancer chemotherapeutic agent cis-diamminedichloroplatinum(II). A circular DNA duplex with an intrastrand d(GpG) crosslink positioned at a specific site was synthesized. Human cell extracts were unable to induce repair synthesis in a 29-base-pair region encompassing the adduct or in adjacent regions. The same extracts could repair a single defined 2-acetylaminofluorene lesion in a similar location. When molecules containing the platinum adduct were cleaved by Escherichia coli UvrABC enzyme,

Targeting CDK4/6 in Combination with Phage-Based Anti-HER2 Vaccination Overcomes Immune Evasion and Enhances the Anticancer Response in Breast Cancer.

Background/Objectives: Cancer vaccines represent the next frontier in immunotherapy, aiming to elicit long-lasting protective anti-tumor immune responses. Human epidermal growth factor receptor 2 (HER2) is a well-established therapeutic target in breast cancer. Active immunization with HER2-displaying M13 bacteriophages can induce a therapeutic immune response against HER2-positive breast cancer, offering a promising alternative to trastuzumab. However, the duration of anticancer immune protection triggered by anti-HER2 phage-based vaccines is limited by tumor-immune suppressive mechanisms. Me
