Intravesical mycobacteria reshape bladder and gut microbiota in a murine bladder cancer model.

The bladder microbiota may influence tumor progression and responses to intravesical therapies, yet the effects of Mycobacterium bovis BCG instillations on bladder and gut microbiota remain unexplored. This study evaluates, for the first time, the impact of M. bovis BCG and Mycobacterium brumae, a non-pathogenic immunomodulator, on microbiota composition in bladder tissue and cecal stool of healthy and tumor-bearing mice. Mycobacterial instillations increased gut microbial richness but reduced Actinobacteria. In tumor-bearing mice, Muribaculaceae and Bacteroidaceae were enriched, while M. bovi

Human microbiota in dengue infection: A narrative review.

Dengue fever, a widespread mosquito-borne viral infection in tropical regions, typically manifests fever and gastrointestinal symptoms, including nausea, vomiting, and diarrhea. However, the human gut microbiota's role in dengue pathogenesis remains incompletely understood. Studies have demonstrated dysbiosis during dengue virus infection, characterized by increased abundance of potentially pathogenic bacteria like Bacteroidaceae and Proteobacteria, particularly during the critical phase. Furthermore, microbial translocation and leaky gut syndrome, characterized by the translocation of intesti

Polysaccharides from Sargassum fusiforme after UV/H2O2 degradation effectively ameliorate dextran sulfate sodium-induced colitis.

In this study, degraded polysaccharides from Sargassum fusiforme (PSF-T2) were prepared by UV/H2O2 treatment for 2 h, and its effects on ameliorating dextran sulfate sodium-induced colitis were evaluated using a mouse model. Results showed that PSF-T2 relieved colitis symptoms, characterized by increasing the colon length and body weight, decreasing disease activity index and relieving colon damage. In addition, PSF-T2 decreased the secretion and expression of IL-1β, IL-6 and TNF-α, and increased the expression of MUC-2, ZO-1 and occludin. Besides, PSF-T2 promoted the production of short-chain

An HRP-integrated CRISPR/Cas12a biosensor towards chair-side diagnosis for Porphyromonas gingivalis.

Rapid diagnostic tools for Porphyromonas gingivalis (Pg), the primary microorganism responsible for the development of periodontitis, particularly those designed for chair-side applications, could provide substantial health benefits to patients. To address this issue, we developed a CRISPR/Cas12a-based rapid Pg detection method. Dual-gRNA and hairpin reporter strategies were employed to enhance CRISPR/Cas12a reaction efficiency. By modifying the hairpin reporter with HRP, the pre-amplification-free HRP-CRISPR/Cas12a reaction was enabled to produce a colorimetric output, amplifying the detectio

Porphyromonas gingivalis Periodontal Infection and Its Putative Links with Alzheimer's Disease.

Periodontal disease (PD) and Alzheimer's disease (AD) are inflammatory conditions affecting the global adult population. In the pathogenesis of PD, subgingival complex bacterial biofilm induces inflammation that leads to connective tissue degradation and alveolar bone resorption around the teeth. In health, junctional epithelium seals the gingiva to the tooth enamel, thus preventing bacteria from entering the gingivae. Chronic PD involves major pathogens (Porphyromonas gingivalis, Treponema denticola, and Tannerella forsythia) which have an immune armoury that can circumvent host's immune surv

Delineation of global, absolutely essential and conditionally essential pangenomes of Porphyromonas gingivalis.

Porphyromonas gingivalis is a Gram-negative, anaerobic oral pathobiont, an etiological agent of periodontitis and the most commonly studied periodontal bacterium. Multiple low passage clinical isolates were sequenced, and their genomes compared to several laboratory strains. Phylogenetic distances were mapped, a gene absence-presence matrix generated, and core (present in all genomes) and accessory (absent in one or more genomes) genes delineated. Subsequently, a second pangenome delineating the prevalence of inherently essential genes was generated. The prevalence of genes conditionally essen

[Role of Porphyromonas gingivalis in rheumatoid arthritis and inflammatory spondyloarthropathies].

The most recent studies confirm the link between rheumatoid arthritis (RA) and periodontal disease. RA patients have higher prevalence of chronic periodontitis and periodontal disease is often more severe in these patients. Both RA and PD show similar pathophysiological mechanisms and risk factors. Autoimmunity to citrullinated peptides is the primary element in the pathogenesis of RA, not found in other diseases. Porphyromonas gingivalis, the major periodontal pathogen associated with the etiology of chronic periodontitis, is the only bacterium currently known to produce the enzyme peptidylar

Gut dysbiosis in Down syndrome: A potentially unexplored culprit for early Alzheimer's disease.

INTRODUCTION: Gut dysbiosis has been found to play a role in sporadic Alzheimer's disease (AD) but has not been explored in Down syndrome (DS), despite the strong relationship between DS and early AD. Here, we compared the gut microbiomes of 20 adults with DS, either cognitively stable or with mild cognitive impairment. METHODS: DNA from stool samples was profiled using 16S rRNA sequencing. RESULTS: Cognitive status was associated with a significant difference in overall microbiome composition (p < 0.01) and with significant differences in the abundance of Bacteroidaceae, Enterobacteriaceae,

Perinatal antibiotics in a db/db mouse model impact obesity and hyperglycemia in a microbiome-dependent manner.

Obesity affects over one billion people globally; however, the role of the gut microbiome and host genetics in its manifestation is poorly understood. We demonstrate that genetic obesity in leptin-receptor-deficient db/db mice requires a permissive gut microbiome, which is established during early life. Using perinatally-administered antibiotic cocktail treatment until pups were 8-weeks-old, we demonstrate that microbiome perturbation substantially reduces weight gain and significantly reduces hyperglycemia in homozygous, leptin-receptor-deficient db/db (Hom) mice without altering caloric inta

Association of gut microbiota composition and microbe-host interactions with response to chemoimmunotherapy in gastric cancer.

Immune checkpoint inhibitors combined with chemotherapy have improved outcomes for advanced gastric cancer, yet only a subset of patients derive benefit, and the contribution of the gut microbiome remains incompletely defined. To identify microbial signatures associated with chemoimmunotherapy response, we performed 16S rRNA gene sequencing on 90 fecal samples collected from 30 patients at baseline, one month after treatment initiation, and at the end of therapy. Patients were stratified by RECIST1.1 criteria into partial response (PR), stable disease (SD), and progressive disease (PD). While

Comparison of gut viromes across captive mammals reveals extensive genetic diversity in bacteriophage dark matter and mammalian viruses.

Comprehensive characterization of mammalian gut viromes is essential for early detection of commensal and potentially zoonotic viruses and for reducing the risk of cross-species transmission. Viral metagenomics was applied to profile gut viral communities from zoo mammals maintained across multiple zoological institutions in China. Viral communities differed markedly among host dietary guilds, with herbivores exhibiting the highest viral species diversity. In total, 1 027 viral sequences representing five major viral groups were recovered, including multiple mammal-associated astroviruses, pic

Auricularia auriculaPolysaccharides Exert Anti-inflammatory Effects in Hepatic Fibrosis by the Gut-Liver Axis and Enhancing SCFA Metabolism.

Auricularia auricula, esteemed in Chinese culture for their culinary and medicinal properties, exhibits notable metabolic and immunomodulatory effects. The principal active constituents are indigestible fermentable polysaccharides, which not only exhibit anti-inflammatory activities but also facilitate the proliferation of beneficial gut microbiota. However, the influence of gut-derived components on liver-regulated metabolic products remains insufficiently understood. This item offers insights into the therapeutic potential of wood ear mushrooms for treating hepatic fibrosis and the associate

Alleviation of Helicobacter pylori- or aspirin-induced gastritis and neuroinflammation in mice by Lactococcus lactis and Bifidobacterium longum.

Helicobacter pylori (HP) causes gastritis and peptic ulcer. Therefore, we examined whether probiotics Lactococcus lactis P135 and Bifidobacterium longum P142, which inhibited HP growth by 37.9% and 35.3%, respectively, and HP-induced IL-8 expression in KATO III cells by 68.6% and 63.1%, respectively, compared to those of normal controls, could mitigate HP-induced gastritis and psychiatric disorder in mice. Oral administration of P135 and/or P142 alleviated HP- or aspirin-induced gastritis, colitis, neuroinflammation, and depression/cognitive impairment-like behavior. They also suppressed HP in

Gastrointestinal digestion, probiotic fermentation behaviors and immunomodulatory effects of polysaccharides from Sanghuangporus vaninii.

In this study, the crude polysaccharides (CSVP) and the preliminary purified polysaccharides (PSVP) from Sanghuangporus vaninii were obtained. The physicochemical properties, gastrointestinal digestion, and probiotic fermentation behaviors of CSVP and PSVP as well as the immunomodulatory effects of PSVP in cyclophosphamide-treated mice were investigated. The results showed that PSVP had higher total polysaccharides content and solubility, but lower radical scavenging activity than CSVP. Moreover, PSVP showed lower hydrolysis degree and better probiotic effects than CSVP. In immunosuppression m

Effects of Moderate Alcohol Consumption in Non-Alcoholic Fatty Liver Disease.

Alcoholic liver disease (ALD) and non-alcoholic fatty liver disease (NAFLD) have emerged as leading causes of chronic liver diseases worldwide. ALD and NAFLD share several pathophysiological patterns as well as histological features, while clinically, they are distinguished by the amount of alcohol consumed daily. However, NAFLD coexists with moderate alcohol consumption in a growing proportion of the population. Here, we investigated the effects of moderate alcohol consumption on liver injury, lipid metabolism, and gut microbiota in 30 NAFLD-patients. We anonymously assessed drinking habits,

Pathogenesis of Porphyromonas gingivalis peptidylarginine deiminase: from periodontal microenvironment to systemic diseases.

Periodontitis is a globally prevalent chronic inflammatory disease that exerts profound effects on both oral and systemic health. Porphyromonas gingivalis is a key pathogenic bacterium in periodontitis. Its secreted peptidylarginine deiminase (PPAD) is a unique protein-citrullinating enzyme. It catalyzes the citrullination of arginine residues in proteins, thereby altering protein structure and function. This review systematically integrates current evidence to elucidate the enzymatic properties, secretion regulatory mechanisms, and pathogenic roles of PPAD in the periodontal microenvironment

There's no place like OM: Vesicular sorting and secretion of the peptidylarginine deiminase of Porphyromonas gingivalis.

The oral pathogen Porphyromonas gingivalis is one of the major periodontal agents and it has been recently hailed as a potential cause of the autoimmune disease rheumatoid arthritis. In particular, the peptidylarginine deiminase enzyme of P. gingivalis (PPAD) has been implicated in the citrullination of certain host proteins and the subsequent appearance of antibodies against citrullinated proteins, which might play a role in the etiology of rheumatoid arthritis. The aim of this study was to investigate the extracellular localization of PPAD in a large panel of clinical P. gingivalis isolates.

Porphyromonas gingivalis: a potential trigger of neurodegenerative disease.

Porphyromonas gingivalis (P. gingivalis) is a gram-negative bacterium and the main causative agent of periodontitis, a disease closely associated with the development of periodontal disease. The progression of periodontitis, a chronic infectious disease, is intricately linked to the inflammatory immune response. Inflammatory cytokines act on periodontal tissues via immunomodulation, resulting in the destruction of the periodontal tissue. Recent studies have established connections between periodontitis and various systemic diseases, including cardiovascular diseases, tumors, and neurodegenerat
