Decisional Capacity in Amyotrophic Lateral Sclerosis.

The cognitive and behavioral changes that can be observed in the neurodegenerative terminal disease amyotrophic lateral sclerosis (ALS), once characterized as purely a motor neuron disease, have become increasingly recognized over the past century. Detecting cognitive deficits earlier and identifying continued changes at regular intervals can lead to improved care, proactive treatments, and earlier discussions about end-of-life wishes. Although medical decisional capacity is required for every treatment decision made, its importance becomes paramount when making decisions on complex medical tr

Behavioral profile in a Dctn1G71A knock-in mouse model of Perry disease.

Perry disease (Perry syndrome) is a rare, rapidly progressive, autosomal dominant neurodegenerative disease characterized by parkinsonism, depression/apathy, weight loss, and respiratory symptoms including central hypoventilation. It is caused by missense mutations (e.g. p.G71A) in the DCTN1 gene. We previously generated transgenic mice that expressed human DCTN1G71A mutant protein under the control of Thy1 promoter. These mice exhibited apathy-like behavior and parkinsonism. However, it is possible that this phenotype was due to a gene-dosage imbalance or transgene insertion position. To circ

Behavioral satiety sequence (BSS) for the diagnosis of drug action on food intake.

The Behavioral Satiety Sequence (BSS) is the name given to the orderly transitions of eating, activity grooming and resting measured during the postingestive period. Because the BSS is considered to reflect the operations of natural physiological processes underlying satiety, the sequence can be used to discriminate between different drugs (and other manipulations) that reduce food intake via these natural physiological mechanisms or those that do so by interference. The BSS is only produced by the presence of a caloric load in the gut, and the preabsorptive satiety factors (such as CCK) the c

Trichobilharzia regenti (Schistosomatidae): 3D imaging techniques in characterization of larval migration through the CNS of vertebrates.

Migration of parasitic worms through the host tissues, which may occasionally result in fatal damage to the internal organs, represents one of the major risks associated with helminthoses. In order to track the parasites, traditionally used 2D imaging techniques such as histology or squash preparation do not always provide sufficient data to describe worm location/behavior in the host. On the other hand, 3D imaging methods are widely used in cell biology, medical radiology, osteology or cancer research, but their use in parasitological research is currently occasional. Thus, we aimed at the ev

Longitudinal In Vivo Imaging of Retinal Cells with Tailored Adeno-Associated Virus Serotypes via Confocal Scanning Laser Ophthalmoscopy.

The dynamic nature of retinal cellular processes necessitates advancements in gene delivery and live monitoring techniques to enhance the understanding and treatment of ocular diseases. This study introduces an optimized adeno-associated virus (AAV) approach, utilizing specific serotypes and promoters to achieve optimal transfection efficiency in targeted retinal cells, including retinal ganglion cells (RGCs) and Müller glia. Leveraging the precision of confocal scanning laser ophthalmoscopy (CSLO), this work presents a non-invasive method for in vivo imaging that captures the longitudinal exp

Dual diagnoses. Psychiatric disorders in developmental disabilities.

A high prevalence of psychiatric illness exists in persons with mental retardation. Among children with mild to moderate retardation, psychiatric illnesses resemble those seen in the general population. Major affective disorders, ADHD, and conduct disorder are common and respond to the same interventions used in children without mental retardation. Persons with severe to profound retardation are more likely to engage in stereotypies and self-injurious behaviors. In addition, certain specific syndromes associated with mental retardation present with particular neurocognitive, behavioral, and ps

Assessment of behavioral tasks performed by hemiplegic patients with impaired dexterity post stroke.

Disability continues to be one of the leading reasons individuals affected by stroke are left incapable of performing daily activities. Due to the staggering number of disabled adults suffering post-stroke neurological damage, there is a critical need for creating and monitoring effects of successful, intensive stroke therapies. Behavioral assessments are useful tools by which to examine the effectiveness of these stroke therapies as they allow for the investigation of multiple variables, including task performance time, performance quality, and degree of motor function. The purpose of this re

A clinical investigation of the possible effects of long-term habituation of sexual arousal in assisted covert sensitization.

Behavior principles are naturally implicated in most of the behavior therapy techniques used in the treatment of sexual dysfunctions and sexual disorders. Hypotheses differ, however, in the extent to which behavior therapy procedures emphasize the classical conditioning of sexual arousal (e.g. penile responses) or the operant conditioning of deviant sexual behavior patterns, or other often ignored behavior principles such as habituation. The purpose of the present case study is to investigate the possibility that at least part of the clinical effects of a widely used behavior therapy technique

Long-Term Biodistribution of Fe3O4@Au Core-Satellite Nanoparticles Assessed by CT and MRI In Vivo.

Nanoparticles combining gold and iron oxide are promising for a wide range of biomedical applications, including photothermal therapy, radiotherapy enhancement, drug delivery, and diagnostic imaging. However, their long-term biodistribution and safety profile remain largely unexplored. Here, we synthesized Fe3O4@AuNP core-satellite nanoparticles (Fe3O4@AuNPs) using femtosecond laser ablation, functionalized them with 15 kDa polyethylene glycol (PEG), and characterized them using a panel of physicochemical techniques. Healthy C57BL/6 mice received an intravenous injection of Fe3O4@AuNPs at 730

Some indicators of nutritional status are associated with activity and exploration in infants at risk for vitamin and mineral deficiencies.

Severe malnutrition, both protein-energy and micronutrient deficiency, results in decreased activity, but the results regarding mild-to-moderate malnutrition are equivocal. Our objective in this investigation was to describe the activity and exploratory behavior of Mexican infants and describe the relationship among nutritional status, activity, and exploration in this population at high risk for mild-to-moderate micronutrient deficiency, but at low risk for severe malnutrition. The participants were infants, 4-12 mo old, of low socioeconomic status from 3 states in southern Mexico. We measure

Striatal microinfusion of Tourette syndrome and PANDAS sera: failure to induce behavioral changes.

Rodent striatal microinfusions have been suggested as a model for assessing the behavioral effects induced by antineuronal antibodies. We used this approach to evaluate the proposed autoimmune etiology for Tourette syndrome (TS) and pediatric autoimmune neuropsychiatric disorders associated with streptococcal infection (PANDAS). Sera were assessed from patients with TS (n = 9) preselected based on the presence of elevated enzyme-linked immunosorbent assay optical densities against putamen homogenate and sera from patients with PANDAS (n = 8), selected from a larger group assayed for antibodies

Repeated mild traumatic brain injuries induce persistent changes in plasma protein and magnetic resonance imaging biomarkers in the rat.

A single mild traumatic brain injury (mTBI) typically causes only transient symptoms, but repeated mTBI (RmTBI) is associated with cumulative and chronic neurological abnormalities. Clinical management of mTBI is challenging due to the heterogeneous, subjective and transient nature of symptoms, and thus would be aided by objective biomarkers. Promising biomarkers including advanced magnetic resonance imaging (MRI) and plasma levels of select proteins were examined here in a rat model of RmTBI. Rats received either two mild fluid percussion or sham injuries administered five days apart. Rats un

Enhanced Interfacial Bonding of Graft Copolymers.

To understand how thermoplastic welding strength can be tuned through chemical modifications and macromolecular topology, we combined coarse-grained molecular dynamics (MD) simulations with experimental validation. Our simulations examined the diffusion dynamics of both linear and graft polymers across representative interfaces, revealing that diffusion-controlled interdigitation follows a power law, with the exponent decreasing from 0.34 to 0.11 as grafting density increases from 7.5 to 196% (with side chains grafted to both sides of a monomer unit). The addition of side chains enhances weldi

Structural and magnetic properties of mixed-metal (Bi2O2)(Fe1-xMxF4) Aurivillius oxyfluorides.

A series of mixed-metal Aurivillius oxyfluorides of the ideal formula [Bi2O2][Fe1-xMxF4] was synthesized by hydrothermal synthesis with M = Mn, Co, and Ni. We first re-examined the Fe-only compound and deduced that despite the observation of inhomogeneous lattice parameters between batches, the iron valence remains constant around Fe∼2.5+ in all samples measured using Mössbauer spectroscopy. The mixed valency charge compensation is mainly assigned to the formation of Bi vacancies. For the mixed Fe/M phases, the most common observation, using various diffraction techniques, of long-range orderi

Activity in grafted human iPS cell-derived cortical neurons integrated in stroke-injured rat brain regulates motor behavior.

Stem cell transplantation can improve behavioral recovery after stroke in animal models but whether stem cell-derived neurons become functionally integrated into stroke-injured brain circuitry is poorly understood. Here we show that intracortically grafted human induced pluripotent stem (iPS) cell-derived cortical neurons send widespread axonal projections to both hemispheres of rats with ischemic lesions in the cerebral cortex. Using rabies virus-based transsynaptic tracing, we find that at 6 mo after transplantation, host neurons in the contralateral somatosensory cortex receive monosynaptic

Non-Invasive Techniques Reveal Heifer Response to Fescue Endophyte Type in Grazing Studies.

Cattle grazing tall fescue (Schedonorus arundinaceous) infected with wild-type endophytes (WE) leads to a syndrome commonly known as fescue toxicosis. Replacing WE tall fescue with a novel endophyte-infected (NE) tall fescue can mitigate this problem but adoption of this technology has been limited. This study measured and determined the physiological and behavioral responses of heifers that grazed either WE or NE tall fescue, utilizing relatively non-invasive techniques including hair cortisol, thermography (for extremity temperatures), small loggers for intravaginal temperature, and remote o

A Task Analysis of Client Re-engagement: Therapeutic De-escalation of High-Conflict Coparents.

Parents who are engaged in protracted conflict following a divorce are often referred to coparenting therapy. Episodes of intense conflict are common during these therapy sessions and often result in coparents disengaging from the therapist while they engage in escalating conflict with each other, potentially disrupting their progress in therapy. The purpose of this study was to identify how therapists successfully re-engage clients in the session. To understand this process, 24 disengagement events (12 successful and 12 unsuccessful) from 13 cases were analyzed using a task analytic approach.

[Delirium in Palliative Care: Evidence and Practice].

Delirium in Palliative Care: Evidence and Practice Abstract. Delirium is a frequent condition in Palliative Care. For patients, their families and the formal caregivers it is associated with substantial burden, fears and challenges. It is also associated with increased morbidity and mortality and often irreversible. Of utmost importance is the identification of patients and threat for delirium and prophylactic measures to avoid delirium whenever possible. For this, risk factors should be identified and eliminated whenever possible. The correct identification of delirium is challenging and espe
