Novel antibody competition binding assay identifies distinct serological profiles associated with protection.

INTRODUCTION: Pre-erythrocytic malaria vaccines hold the promise of inducing sterile protection thereby preventing the morbidity and mortality associated with Plasmodium infection. The main surface antigen of P. falciparum sporozoites, i.e., the circumsporozoite protein (CSP), has been extensively explored as a target of such vaccines with significant success in recent years. Systematic adjuvant selection, refinements of the immunization regimen, and physical properties of the antigen may all contribute to the potential of increasing the efficacy of CSP-based vaccines. Protection appears to be

LncRNA TUG1 competitively binds to miR-340 to accelerate myocardial ischemia-reperfusion injury.

The aberrant expression of long noncoding RNA (lncRNA) taurine-upregulated gene 1 (TUG1) has been previously associated with myocardial ischemia-reperfusion injury (MIRI), but the underlying molecular mechanisms remain elusive. The current study aimed to clarify the functional role of TUG1/microRNA (miR)-340/histone deacetylase 4 (HDAC4)/β-catenin/glucose transporter type 1 (GLUT1) axes in MIRI. The database-based analyses performed predicted the downstream factors of lncRNA TUG1. In the MIRI mouse models and hypoxia/reoxygenation (H/R)-induced cardiomyocyte models, the expression of TUG1/miR-

Heterogeneous competition of Kv1 channel toxins with kaliotoxin for binding in rat brain: autoradiographic analysis.

The alpha-subunits of Kv1 channels display characteristic distributions and restricted co-assembly in mammalian brain. The heterogeneous composition of Kv1 channels has made it difficult to use specific toxins to label brain structures. We used autoradiography to analyse the competitive behaviour of three Kv1 channel toxins--alpha-dendrotoxin, kaliotoxin, and mast cell degranulating peptide--for binding to kaliotoxin binding sites in various brain structures. IC(50) varied considerably between brain regions (by up to three orders of magnitude) for each ligand. alpha-dendrotoxin and kaliotoxin

High-affinity dextromethorphan binding sites in guinea pig brain. II. Competition experiments.

Binding of dextromethorphan (DM) to guinea pig brain is stereoselective, since levomethorphan is 20 times weaker than DM in competing for DM sites. In general, opiate agonists and antagonists as well as their corresponding dextrorotatory isomers are weak competitors for tritiated dextromethorphan ([3H]DM) binding sites and display IC50 values in the micromolar range. In contrast, several non-narcotic, centrally acting antitussives are inhibitory in the nanomolar range (IC50 values for caramiphen, carbetapentane, dimethoxanate, and pipazethate are 25 nM, 9 nM, 41 nM, and 190 nM, respectively).

Competition between diftalone and other drugs for the binding site(s) on human serum albumin.

The influence of a series of drugs on the binding of diftalone to human serum albumin was investigated by equilibrium dialysis. The results indicated that among the different drugs tested phenylbutazone, indomethacin, warfarin and tolbutamide were, in the order listed, the most effective in displacing diftalone from its binding site(s). Additional investigation showed that there was no consistent inhibition by diftalone of warfarin and tolbutamide binding to albumin. The information obtained might prove useful in clinical therapy whenever a combination of drugs is to be administered.

Ubenimex synergizes with the PD-L1 blockade in gastric cancer by competitively binding LAP3 with UBE3A.

Immune checkpoint inhibitors (ICIs), particularly PD-1/PD-L1 blockade, represent a cornerstone of treatment for advanced gastric cancer (GC). However, their clinical efficacy is hampered by low response rates and the development of both primary and acquired resistance, underscoring the need for innovative combination therapies. In this study, we investigate the potential of Ubenimex, an immunomodulator and inhibitor of leucyl aminopeptidase 3 (LAP3), in enhancing the therapeutic efficacy of PD-L1 blockade in GC. Using a syngeneic GC mouse model, we demonstrate that Ubenimex significantly augme

FXYD3 enhances IL-17A signaling to promote psoriasis by competitively binding TRAF3 in keratinocytes.

Psoriasis is a common chronic inflammatory skin disease characterized by inflammatory cell infiltration and epidermal hyperplasia. However, the regulatory complexity of cytokine and cellular networks still needs to be investigated. Here, we show that the expression of FXYD3, a member of the FXYD domain-containing regulators of Na+/K+ ATPases family, is significantly increased in the lesional skin of psoriasis patients and mice with imiquimod (IMQ)-induced psoriasis. IL-17A, a cytokine important for the development of psoriatic lesions, contributes to FXYD3 expression in human primary keratinoc

Heavy Metals and Human Health: Possible Exposure Pathways and the Competition for Protein Binding Sites.

Heavy metals enter the human body through the gastrointestinal tract, skin, or via inhalation. Toxic metals have proven to be a major threat to human health, mostly because of their ability to cause membrane and DNA damage, and to perturb protein function and enzyme activity. These metals disturb native proteins' functions by binding to free thiols or other functional groups, catalyzing the oxidation of amino acid side chains, perturbing protein folding, and/or displacing essential metal ions in enzymes. The review shows the physiological and biochemical effects of selected toxic metals intera

Identification of mineralocorticoid binding sites in rat brain by competition studies and density gradient centrifugation.

A search for mineralocorticoid binding sites in neural tissue was carried out on the basis of competition by two mineralocorticoid antagonists (spironolactone and glycyrrhetinic acid), a mineralocorticoid agonist (9 alpha-fluorocortisol) and a pure glucocorticoid (RU 26988). The hippocampus was selected for these studies, in view of its preferential concentration of binding sites for [3H]-aldosterone (ALDO) and two glucocorticoids: [3H]-corticosterone (CORT) and [3H]-dexamethasone (DEX). Inhibition studies performed with 5 X 10(-4)-5 X 10(-9) M spironolactone and with 5 X 10(-4)-5 X 10(-8) M g

Competitive Binding of Gingerol Homologs to Fish Myofibrillar Protein Reduces the Retention of Aldehydic Off-Odor Compounds.

Persistent aldehydic off-notes in fish products are influenced not only by lipid oxidation but also by protein-associated retention of odor-active aldehydes. A remaining question is how gingerol side-chain length and aldehyde structure jointly alter this apparent retention. Mackerel myofibrillar protein (MP) was therefore combined with (E)-2-decenal (T2D), (E,E)-2,4-decadienal (DDE), or trans-4,5-epoxy-(E)-2-decenal (E2D), with or without 6-, 8-, or 10-gingerol. Blastomyces dermatitidis antigen detection in urine specimens from dogs with blastomycosis using a competitive binding inhibition ELISA.

A competitive binding inhibition enzyme linked immunosorbent assay (ELISA) was used to detect Blastomyces dermatitidis antigens in urine specimens from dogs with blastomycosis. Sera from rabbits immunized with B. dermatitidis killed whole yeast cells were used as the primary antibody in the competitive ELISA. This initial study was performed to determine if B. dermatitidis antigen detection was possible and to test the efficacy of the rabbit sera as a primary antibody. An indirect ELISA was also performed to compare antigen detection in urine to antibody detection in the sera of the infected d

Indole-3-carbinol attenuates cisplatin-induced premature ovarian failure by activating Nrf2 through competitive binding of Keap1.

BACKGROUND: Premature ovarian failure (POF) leads to female infertility and significantly increases long-term health risks. However, available drugs that can reverse this condition are lacking. PURPOSE: To elucidate the role of nuclear factor erythroid 2-related factor (Nrf2) in POF and the underlying molecular mechanism through which indole-3-carbinol (I3C) alleviates POF. METHODS: We established a POF model in wild type and Nrf2 knockout (Nrf2-KO) mice via cisplatin injection. ML385 and siRNA were used to inhibit and knockdown Nrf2 in vitro, respectively. Protein levels were detected by WB

Competitive binding of actin and SH3 domains at proline-rich regions of Las17/WASP regulates actin polymerisation.

Eukaryotic actin filaments bind factors that regulate their assembly and disassembly creating a self-organising system, the actin cytoskeleton. Despite extensive knowledge of signals that modulate actin organisation, significant gaps remain in our understanding of spatiotemporal regulation of de novo filament initiation. Yeast Las17/WASP is essential for actin polymerisation initiation supporting membrane invagination in Saccharomyces cerevisiae endocytosis and therefore its tight regulation is critical. The adaptor protein Sla1 inhibits Las17 but mechanisms underpinning Las17 activation remai

Ultrasensitive and facile electrochemical detection of hydrogen sulfide in rat brain microdialysate based on competitive binding reaction.

In this study, a sensitive and facile method with wide linear range and low detection limit for detecting hydrogen sulfide in rat brain microdialysate was developed. The design of the sensor is based on the competitive binding reaction principle, in which cysteine was self-assembly immobilized on the surface of gold electrode, and then the Cu2+ as the electrochemical probe was anchored to the cysteine film through coordination bonding with carboxyl (-COOH) and amino group (-NH2) to form the Cu2+/Cys/Au electrode. The Cu2+/Cys/Au electrode can serve as an electrochemical H2S sensor through a li

Identification of a soluble Fc gamma-binding molecule (annexin II) in human serum using a competitive ELISA.

We have previously produced a monoclonal antibody (mAb), B1D6, reactive with a 37 kD placental IgG Fc-binding molecule (FcR), recently identified as annexin II. Annexin II is an intracellular molecule found in several cell types, including endothelium and monocytes. Since soluble Fc-binding molecules are of importance in the regulation of the immune response, we have now used B1D6 in a competitive ELISA to study levels of soluble annexin II in human sera. Soluble annexin II was detected in all sera studied. The highest levels were observed in patients with infectious mononucleosis. Gel filtrat

Porcine Deltacoronavirus M Protein Binds NLRP3 to Promote Inflammasome Assembly via Competition with TRIM31.

Porcine deltacoronavirus (PDCoV) infection induces severe intestinal inflammation and acute diarrhea in piglets, resulting substantial economic losses. The NLR family pyrin domain containing 3 (NLRP3) inflammasome, a key component of the innate immune system, drives interleukin (IL)-1β maturation, and its excessive activation is closely linked to viral pathogenesis. However, its role in PDCoV infection remains unclear. Here, we report that PDCoV infection induces IL-1β maturation through NLRP3 inflammasome activation both in vivo and in vitro, and the M protein promotes NLRP3 inflammasome asse

Low-density lipoproteins in human serum competitively inhibit the binding and entry of vesicular stomatitis virus.

Vesicular stomatitis virus (VSV) is a promising anticancer agent but rapidly loses its infectivity in human serum. Here, we demonstrate that low-density and very low-density lipoproteins (LDLs and VLDLs) compete with VSV-G-displaying vectors for occupancy of cellular LDL receptors (LDLRs). Infectivity of VSV-G-displaying vectors was reduced two- to three-logs in the presence of heat-inactivated human serum but not if the serum was depleted of lipoproteins. The inhibitory effect was replicated in serum-free media supplemented with physiological concentrations of purified lipoprotein particles t

Evaluation of the solvent competitive binding in halogen and chalcogen bonding: a theoretical theory study for supramolecular applications.

Noncovalent interactions such as halogen bonding (HaB) and chalcogen bonding (ChB) are increasingly utilized in host-guest chemistry and supramolecular catalysis due to their high directionality and tuneable strength. However, the choice of solvent remains a critical challenge, as solvent molecules can compete with the intended substrates for the electrophilic σ-hole sites of the catalysts or receptors. In this work, we present a comprehensive DFT study (PBE0-D3/def2-TZVP) analysing the competitive binding of nine common solvents (acetonitrile, ether, water, methanol, DMF, DMSO, dichloromethan

PCMTD1-127aa suppresses osteosarcoma progression by competitively binding to USP10 to promote c-MYC degradation.

Osteosarcoma (OS) is a highly malignant tumor prone to distant metastasis and is associated with a poor prognosis. To date, therapeutic outcomes for this disease remain unsatisfactory. In this study, we found that circPCMTD1 encodes a 127-amino acid protein, termed PCMTD1-127aa in OS. The expression of PCMTD1-127aa was significantly lower in OS tissues than in normal paratumoral tissues. Clinical data analysis revealed that low expression of PCMTD1-127aa was associated with poor OS prognosis. Functionally, overexpression of PCMTD1-127aa not only inhibited the proliferation and invasion of oste
