Characteristic of Uterine Rhabdomyosarcoma by Algorithm of Potential Biomarkers for Uterine Mesenchymal Tumor.

BACKGROUND/AIM: Patients with uterine sarcoma comprise 2-5% of all patients with uterine malignancies; however, the morbidity of uterine sarcoma is low compared with that of other gynecological cancers. For many cases, malignant uterine tumors are diagnosed during follow-up of benign uterine leiomyoma. Of the uterine sarcomas, rhabdomyosarcoma is considered a mixed tumor containing components of epithelial cells and mesenchymal cells. Therefore, the onset of primary uterine rhabdomyosarcoma during follow-up of uterine leiomyoma is extremely rare. Rhabdomyosarcoma is a relatively common maligna

Small molecule metabolite biomarkers for hepatocellular carcinoma with bile duct tumor thrombus diagnosis.

Hepatocellular carcinoma with bile duct tumor thrombus (BDTT) is a malignant disease. The most commonly used diagnosis methods for BDTT are MRCP/ERCP, ultrasonic diagnosis or CT scan. However, BDTT is often misdiagnosed as other bile duct diseases, such as extrahepatic cholangiocarcinoma (EHCC), choledochal cyst (Cyst) and common bile duct stone (Stone). Diagnostic methods, which are more accurate and less destructive, are urgently needed. In this paper, we analyzed the small molecule metabolites in the serum of BDTT, Stone, Cyst and EHCC patients and normal people using untargeted GC-MS, and

Tumor-induced osteoclast miRNA changes as regulators and biomarkers of osteolytic bone metastasis.

Understanding the mechanism by which tumor cells influence osteoclast differentiation is crucial for improving treatment of osteolytic metastasis. Here, we report broad microRNA (miRNA) expression changes in differentiating osteoclasts after exposure to tumor-conditioned media, in part through activation of NFκB signaling by soluble intracellular adhesion molecule (sICAM1) secreted from bone-metastatic cancer cells. Ectopic expression of multiple miRNAs downregulated during osteoclastogenesis suppresses osteoclast differentiation by targeting important osteoclast genes. Intravenous delivery of

Predictive and Prognostic Biomarkers and Tumor Antigens for Targeted Therapy in Urothelial Carcinoma.

Urothelial carcinoma (UC) is the fourth most prevalent cancer amongst males worldwide. While patients with non-muscle-invasive disease have a favorable prognosis, 25% of UC patients present with locally advanced disease which is associated with a 10-15% 5-year survival rate and poor overall prognosis. Muscle-invasive bladder cancer (MIBC) is associated with about 50% 5 year survival when treated by radical cystectomy or trimodality therapy; stage IV disease is associated with 10-15% 5 year survival. Current therapeutic modalities for MIBC include neoadjuvant chemotherapy, surgery and/or chemor

Impact of Preanalytical Factors on the Measurement of Tumor Tissue Biomarkers Using Immunohistochemistry.

Analysis of formalin-fixed paraffin-embedded (FFPE) tissue by immunohistochemistry (IHC) is commonplace in clinical and research laboratories. However, reports suggest that IHC results can be compromised by biospecimen preanalytical factors. The National Cancer Institute's Biospecimen Preanalytical Variables Program conducted a systematic study to examine the potential effects of delay to fixation (DTF) and time in fixative (TIF) on IHC using 24 cancer biomarkers. Differences in IHC staining, relative to controls with a DTF of 1 hr, were observed in FFPE kidney tumor specimens after a DTF of ≥

Rules of tumor cell development and their application to biomarkers for ovarian cancer.

PURPOSE/OBJECTIVES: To apply the Hanahan and Weinberg conceptual framework for tumor development to the specific biomarkers observed or expressed in ovarian cancer. DATA SOURCES: Data-based publications, topical reviews, and book chapters. DATA SYNTHESIS: Articles specific to ovarian cancer were reviewed to examine whether the six rules from the Hanahan and Weinberg conceptual framework were applicable to biomarkers of ovarian cancer. This approach allows for the application of a general framework for the development of solid tumors to the development of ovarian cancer. CONCLUSIONS: The six

CYR61, a potential biomarker of tumor inflammatory response in epithelial ovarian cancer microenvironment of tumor progress.

BACKGROUND: Recent studies have found that inflammatory response is involved in the pathogenesis of ovarian cancer. Advanced ovarian cancer is often presented with ascites that is rich in cytokines, inflammatory factors or cancer cells. Therefore, it is important to study the microenvironment of ascites in order to further clarify the occurrence and progression of ovarian cancer. As a pro-inflammatory factor, the Cyr61 expression patterns are inconsistent in human tumors. Although it has been reported that Cyr61 is related to the progression of ovarian cancer, its specific mechanism is not yet

Integrating tumor-immune mechanistic modeling with transcriptomics reveals pattern-driven prognostic biomarkers in tumors.

The spatial patterns of immune cells within the tumor microenvironment hold profound prognostic implications, yet a mechanistic understanding of their formation and functional impact remains lacking. Current mechanistic models operate in a theoretical vacuum, while spatial transcriptomics (ST) provides static snapshots without dynamic insight. To bridge this gap, we present an integrative framework that couples a reaction-diffusion model of tumor-immune kinetics with multicohort ST data. We theoretically explore the spatiotemporal conditions for Turing spatial patterns. Stability analysis iden

Circulating tumor DNA as a prognostic and monitoring biomarker in urothelial cancers.

The application of circulating tumor DNA (ctDNA) has advanced in the treatment of multiple cancer types, including urothelial cancer (UC). While ctDNA currently plays a limited role in non-muscle-invasive bladder cancer (NMIBC), its relevance is expanding in muscle-invasive bladder cancer (MIBC), as evidenced by recent positive results from the IMvigor011 trial. This study demonstrated that a ctDNA-guided approach to adjuvant atezolizumab therapy enhances disease-free survival. Furthermore, ctDNA is well-established as a tool for monitoring treatment response and assessing minimal residual dis

Biomarker-directed aptamer panels for diagnosis and differentiation of neuroendocrine tumors.

Neuroendocrine tumors (NETs) are rare tumors that may arise in the small intestine (siNET) or pancreas (pNET). The tumors have heterogeneous clinical characteristics, and diagnosis may be challenging. Current non-invasive biomarkers perform suboptimally, and novel biomarkers are highly needed. We aimed to investigate the use of RNA aptamer panels, selected by the APTASHAPE technology, in distinguishing healthy individuals from individuals with NET as well as discriminating siNET from pNET. Plasma samples from 68 individuals with NET and 24 healthy individuals were collected and randomly divide

Potential Biomarkers in Thymic Epithelial Tumors: Thymoma and Thymic Carcinoma.

Thymic epithelial tumors (TETs) are rare malignancies originating from thymic epithelial cells, comprising thymomas and thymic carcinomas. The heterogeneity of TETs poses significant diagnostic and therapeutic challenges, necessitating the identification of reliable biomarkers to improve classification and treatment stratification. The integration of multiomic approaches and artificial intelligence are transforming biomarker discovery, offering new insights into TET pathogenesis and therapeutic vulnerabilities. However, clinical translation of these biomarkers remains limited due to the rarity

PTPRC functions as a prognosis biomarker in the tumor microenvironment of cutaneous melanoma.

Cutaneous melanoma is one of the most malignant types of skin cancer, with an extremely poor prognosis. Immune cells infiltrated in the tumor microenvironment (TME) affects melanoma initiation, progression, prognosis and immunotherapy strategies in melanoma. The potential utility of TME-related genes as a prognostic model for melanoma and as a predictor of immunotherapeutic response merits further exploration. In this study, we determined that an immune-related gene, protein tyrosine phosphatase receptor type C (PTPRC), was positively correlated with the positive prognosis of melanoma patients

Circulating Tumor Cells: A Promising Biomarker in the Management of Nasopharyngeal Carcinoma.

Nasopharyngeal carcinoma (NPC) is a malignancy that arises from the mucosal epithelium of the nasopharynx, and its prognosis is relatively favorable. The 5-year overall survival rate in patients with locally advanced NPC currently exceeds 80%, but the development of individualized diagnosis and treatment at the molecular level is relatively lacking. Circulating tumor cells (CTCs) is the generic term for tumor cells that are present in the peripheral blood circulation. As a new biomarker with good clinical application prospects, the detection of CTCs has the advantages of being non-invasive, si

circMAN1A2 could serve as a novel serum biomarker for malignant tumors.

Novel diagnostic and prognostic biomarkers of cancers are needed to improve precision medicine. Circular RNAs act as important regulators in cancers at the transcriptional and posttranscriptional levels. The circular RNA circMAN1A2 is highly expressed in nasopharyngeal carcinoma according to our previous RNA sequencing data; however, the expression and functions of circMAN1A2 in cancers are still obscure. Therefore, in this study, we evaluated the expression of circMAN1A2 in the sera of patients with nasopharyngeal carcinoma and other malignant tumors and analyzed its correlations with clinica
