Previously, we hypothesized that hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) can be biotransformed by anaerobic sludge via three different routes: (1) direct ring cleavage via alpha-hydroxylation of a-CH(2) group, (2) reduction of one of the -NO(2) groups to -NO, (3) N-denitration prior to ring cleavage. The present study describes biotransformation of RDX via route 3 by a diaphorase (EC 1.8.1.4) from Clostridium kluyveri using NADH as electron donor. The removal of RDX was accompanied by the formation and accumulation of nitrite ion (NO(2)(-)), formaldehyde (HCHO), ammonium (NH(4)(+)), and

The biotransformation of trichloroethylene (TCE) under various electron acceptor conditions was investigated by using enrichment cultures developed from the anaerobic digester sludge of Thibodaux sewage treatment plant. The results indicated that TCE was biotransformed under sulfate reducing, methanogenic, nitrate reducing, iron reducing, and fermenting conditions. However, the rates of TCE removal varied among the conditions studied. The fastest removal of TCE (100% removal in 9 days) was observed under mixed electron acceptor conditions, followed in order by methanogenic, fermenting, iron re

Chlorinated per- and polyfluoroalkyl substances (Cl-PFAS) represent an important subgroup in replacement chemicals for legacy PFAS, and they have been detected in various environments including water bodies, soil, as well as air particles. However, the biodegradability of these chemicals is largely unknown. A recent study reported dechlorination triggered anaerobic biodefluorination of Cl-PFCAs by activated sludge communities. In this study, we further investigated the biodefluorination of Cl-PFCAs by pure cultures. Six of ten selected Cl-PFCAs displayed significant defluorination by Acetobact

The biodegradation of dissolved organic matter (DOM) regulates riverine carbon dioxide emissions, yet the specific molecular biotransformation pathways driving dissolved organic carbon (DOC) degradation kinetics remain poorly constrained. Using incubation experiments coupled with FT-ICR MS in the Yangtze River, we show that biodegradation reshapes DOM via a mass-difference-based molecular network dominated by oxidation (26.4%) and carboxylic acid reactions (20.9%), yielding smaller, highly oxidized, and heteroatom-depleted products. These pathways exhibit distinct compositional selectivity, as

Biodegradable plastics can enhance food waste utilization in anaerobic digestion (AD) units, but their fate under thermophilic conditions remains unclear. Previous studies using methane production, calorimetry, or spectroscopic analyses often report inconsistent results. This study tracks the biotransformation of polybutylene adipate co-terephthalate (PBAT) in thermophilic AD (55 °C) using 1H NMR to quantify monomers. While 1,4-butanediol degraded quickly, adipic (AA) and terephthalic acid (TPA) accumulated over time. Monomer analysis estimated PBAT biotransformation at 11.1 ± 1.9 % (TPA) and

Tetracycline antibiotics are used worldwide in human and veterinary medicine. On the basis of low metabolization and through organic fertilizers, tetracyclines enter the environment in a biologically active form. This can have toxic effects on microbial communities and promote the selection of resistant strains. The use of fungi could be a promising approach to deactivate tetracyclines by degradation or derivatization as a result of their particular enzyme endowment. Here, we highlight the current analytical and biotechnological challenges associated with the bioconversion of tetracyclines by

The assessment of the bioaccumulation potential of chemicals is an essential and mandatory part of their regulatory environmental risk and hazard assessment. So far, in vitro data on fish metabolism is rarely available for biocidal active substances such as anticoagulant rodenticides. In this case study we present in vitro biotransformation rates of eight biocidal and one pharmaceutical anticoagulants in rainbow trout (Oncorhynchus mykiss) liver subcellular S9 fraction (RT-S9) determined following the Organisation for Economic Co-operation and Development test guideline 319B method at two diff

Indole, an important signaling molecule as well as a typical N-heterocyclic aromatic pollutant, is widespread in nature. However, the biotransformation mechanisms of indole are still poorly studied. Here, we sought to unlock the genetic determinants of indole biotransformation in strain Cupriavidus sp. SHE based on genomics, proteomics and functional studies. A total of 177 proteins were notably altered (118 up- and 59 downregulated) in cells grown in indole mineral salt medium when compared with that in sodium citrate medium. RT-qPCR and gene knockout assays demonstrated that an indole oxygen

The tremorgenic mycotoxin Thomitrem A is a secondary metabolite produced mainly by the fungus Penicillium crustosum that is frequently found on spoiled stored food and feed. Typical signs of intoxication observed in dogs after the consumption of food waste are emesis, tremors, seizures progressing to ataxia and lack of coordinated movements. How uptake of Thomitrem A relates to exposure is unknown so far since data on biotransformation and toxicokinetics are missing. In this study the toxin was therefore metabolised in an exploratory in vitro experiment by rat hepatocytes, and substrate deplet

Melatonin, the predominant product of the pineal gland, is involved in the maintenance of diurnal rhythms. Nocturnal blood concentrations of melatonin have been shown to be enhanced by fluvoxamine, but not by other serotonin reuptake inhibitors. Because fluvoxamine is an inhibitor of several cytochrome P450 (CYP) enzymes, the authors studied the biotransformation of melatonin and the effects of fluvoxamine on the metabolism of melatonin in vitro using human liver microsomes and recombinant human CYP isoenzymes. Melatonin was found to be almost exclusively metabolized by CYP1A2 to 6-hydroxymela

1. To aid in the prediction of drug interactions with alprazolam, the human CYP involved in the 1'- and 4-hydroxylation of alprazolam were characterized using human liver microsomes, expressed enzymes and selective chemical inhibitors. 2. The formation of 4-hydroxyalprazolam and 1'-hydroxyalprazolam at an alprazolam concentration of 62.5 microM were reduced by the prototypic CYP3A inhibitor, troleandomycin (50 microM), by 97 and 9900 respectively. Only microsomes from B-lymphoblastoid cells expressing CYP3A4 were capable of catalysing the 1'- and 4-hydroxylation of alprazolam. 3. The formation

Volcanic fermentation is an innovative technique in post-harvest coffee processing that involves forming conical mounds, called "volcanoes", to create specific biotransformation conditions. This study investigates the impact of different volcano fermentation methods on the chemical composition and sensory attributes of coffee. Four methods were evaluated: asphalt patio (E1), on pallets (E2), in steel containers under the sun (E3), and in steel containers in the shade (E4). The chemical composition was analyzed in terms of sugars (sucrose, glucose, fructose), organic acids (citric, malic, succi

Alcohol and nicotine (tobacco smoke) are often used together, and taking both addictive substances is associated with an increased risk of certain diseases. It is extremely important to understand the pharmacodynamic and pharmacokinetic mechanisms of the interaction between nicotine and ethanol, which are still not fully understood. The study aimed to evaluate the influence of chronic alcohol consumption on nicotine biotransformation in ethanol-preferring and non-preferring male and female rats. Rats were divided into four groups depending on their alcohol preferences and gender. Nicotine, nor

The fungus Cunninghamella elegans is recognised as a microbial model of mammalian drug metabolism owing to its ability to catabolise xenobiotic compounds in an analogous fashion to animals. Its ability to produce phase I (oxidative) metabolites of drugs is associated with cytochrome P450 (CYP) activity; however, almost nothing is known about these enzymes in the fungus. In this paper we report the in silico analysis of the genome sequence of C. elegans B9769, which contains 32 genes putatively coding for CYPs. Based on their predicted amino acid sequences these were classified as belonging to

Pharmaceutically active compounds are of great concern due to their detection frequency in the environment and the unexpected risks. In this study, the simultaneous removal of mixed pharmaceuticals by microalgae was explored using a typical freshwater diatom Navicula sp. Results showed that Navicula sp. could efficiently remove atenolol, carbamazepine, ibuprofen and naproxen with the efficiencies of >90% after 21 d of exposure. As compared to the removal efficiencies of each pharmaceutical in the individual pharmaceutical treatments, the degradation of sulfamethoxazole, bezafibrate, and naprox

Chirality is a critical topic in the medicinal and agrochemical fields. One quarter of all agrochemicals was chiral in 1996, and this proportion has increased remarkably with the introduction of new compounds over time. Despite scientists have made great efforts to probe the enantiomeric selectivity of chiral chemicals in the environment since early 1990s, the different behaviours of individual enantiomers in biologically mediated processes are still unclear. In the present review, we highlight state-of-the-knowledge on the stereoselective biotransformation and accumulation of chiral contamina

Flubendazole (FLBZ) is a broad-spectrum benzimidazole anthelmintic used in pigs, poultry, and humans. It has been proposed as a candidate for development for use in elimination programmes for lymphatic filariasis and onchocerciasis in humans. Moreover, FLBZ has shown promise in cancer chemotherapy, particularly for neuroblastoma. This work investigated the hepatic carbonyl-reducing pathway of FLBZ in different species, including humans. Microsomal and cytosolic fractions were obtained from sheep, cattle, pig, hen, rat, and human liver. Both subcellular fractions of each species converted FLBZ
