A murine hybridoma antibody, 6C2A.4.2, previously characterized as an immunoglobulin G class 2a that binds in radioimmunoassay to bluetongue virus serotype 17 (BTV-17) but not the other 19 BTV serotypes, neutralizes BTV-17, inhibits hemagglutination with BTV-17, and precipitates viral polypeptides 2 and 3 from BTV-17-infected cells, was produced as an ascites in the peritoneal cavities of hybridoma-inoculated mice. This ascitic fluid, but not those containing other, non-neutralizing anti-BTV-17 antibodies of the same isotype, provided serotype-specific passive protection against BTV-17-induced

A survey of South Carolina diary cattle in 12 commercial herds was conducted, using serologic testing to determine the frequency and impact on reproduction of bluetongue (BT) and bovine parvovirus (BFV) infections. Results of the study of serum antibodies titers and dairy cattle health records indicated that the majority of the cattle surveyed was adequately protected against the major reproductive tract diseases through vaccination programs. The frequency of reproductive dysfunction was common in vaccinated herds, however. Six of the 12 herds in the survey had cattle which were BT serotest re

Bluetongue virus (BTV) is an economically important arbovirus of domestic and wild ruminants transmitted primarily by Culicoides biting midges. The Middle East represents a key ecological corridor linking African, Asian, and European transmission systems, yet entomovirological evidence of BTV infection in regional Culicoides populations remains fragmented. This narrative review synthesizes available literature on Culicoides species distribution and published evidence of BTV detection in field-collected midges across Middle Eastern countries. Major bibliographic databases were searched to ident

INTRODUCTION: bluetongue virus (BTV) infection triggers dramatic and complex changes in the host's transcriptional profile to favor its own survival and reproduction. However, there is no whole-transcriptome study of susceptible animal cells with BTV infection, which impedes the in-depth and systematical understanding of the comprehensive characterization of BTV-host interactome, as well as BTV infection and pathogenic mechanisms.
METHODS: to systematically understand these changes, we performed whole-transcriptome sequencing in BTV serotype 1 (BTV-1)-infected and mock-infected sheep embryoni

UNLABELLED: Bluetongue virus (BTV) is endemic in many parts of the world, often causing severe hemorrhagic disease in livestock. To date, at least 27 different serotypes have been recognized. Vaccination against all serotypes is necessary to protect susceptible animals and to prevent onward spread of the virus by insect vectors. In our previous studies, we generated replication-deficient (disabled infectious single-cycle [DISC]) virus strains for a number of serotypes and reported preliminary data on their protective efficacy in animals. In this report, to advance the DISC vaccines to the mark

Bluetongue (BT) is an infectious, non-contagious, arthropod-borne viral disease of ruminants, and has a severe impact on livestock. It is caused by Bluetongue virus (BTV), a double-stranded (ds) RNA virus with a segmented genome (10 segments), belonging to the Seoreoviridae family, Orbivirus genus. Over the last 25 years, Europe has suffered multiple incursions of different BTV serotypes with serious consequences, which have mainly been controlled thanks to vaccination. In the case of Spain, from 2000 to 2023, BTV serotypes 1, 2, 4 and 8 have caused epidemics, and, sporadically, BTV-1 and -4 w

Bluetongue virus (BTV) reverse genetics (RG), available since 2007, has allowed the dissection of the virus replication cycle, including discovery of a primary replication stage. This information has allowed the generation of Entry-Competent-Replication-Abortive (ECRA) vaccines, which enter cells and complete primary replication but fail to complete the later stage. A series of vaccine trials in sheep and cattle either with a single ECRA serotype or a cocktail of multiple ECRA serotypes have demonstrated that these vaccines provide complete protection against virulent virus challenge without c

Bluetongue (BT) is an arbovirus-borne disease of ruminants caused by bluetongue virus (BTV) that has the potential to have a serious economic impact. Currently available commercial vaccines include attenuated vaccines and inactivated vaccines, both of which have achieved great success in the prevention and control of BTV. However, these vaccines cannot distinguish between infected animals and immunized animals. To control outbreaks of BTV, the development of labeled vaccines is urgently needed. In this study, we used the plasmid-based reverse genetics system (RGS) of BTV to rescue four recombi

Bluetongue (BT) and epizootic hemorrhagic disease (EHD) are noncontagious, insect-transmitted diseases of domestic and wild ruminants caused by related but distinct viruses. There are significant gaps in our scientific knowledge and available countermeasures to control an outbreak of orbivirus-induced disease, whether BT or EHD. Both BT virus (BTV) and EHD virus (EHDV) cause hemorrhagic fevers in susceptible ruminants; however, BT is principally a disease of domestic livestock whereas EHD is principally a disease of certain species of wild, non-African ungulates, notably white-tailed deer. Bluetongue (BT) is a non-contagious disease of sheep and other domestic and wild ruminants caused by the bluetongue virus (BTV). Currently 26 serotypes of the virus have been identified. In South Africa, 22 serotypes have been identified and BT is controlled mainly by annual vaccinations using a freeze-dried live attenuated polyvalent BTV vaccine. The vaccine is constituted of 15 BTV serotypes divided into three separate bottles and the aim is to develop a vaccine using fewer serotypes without compromising the immunity against the disease. This study is based on previously reported cross-neutr

BACKGROUND: Bluetongue is a non-contagious viral disease that affects both domestic and wild ruminants. It is transmitted primarily by small hematophagous Diptera belonging to the genus Culicoides (Diptera: Ceratopogonidae). The current study represents the first molecular investigation into the potential role of Culicoides imicola, Culicoides paolae, Culicoides newsteadi, Culicoides spp., and Culicoides circumscriptus as bluetongue virus (BTV) vectors in Morocco. Additionally, the study aimed to evaluate the vectorial activity of midges during the survey seasons.

Bluetongue virus (BTV) causes the hemorrhagic disease bluetongue (BT) in ruminants. The best way to control outbreaks is vaccination. Currently, conventionally modified-live and inactivated vaccines are commercially available, which have been successfully used to control BT, but nonetheless have their specific shortcomings. Therefore, there is a need for improved BT vaccines. The ideal BT vaccine is efficacious, safe, affordable, protective against multiple serotypes and enables the differentiation of infected from vaccinated animals. Different field situations require specific vaccine profile

In summer 2024, Western Europe experienced extensive outbreaks of bluetongue virus Serotype 3 (BTV-3), a Culicoides-borne orbivirus. Clinical disease was first detected in August 2024 in one of two Belgian zoos, predominantly affecting European bison (Bison bonasus) and American bison (Bison bison). These species showed high morbidity, two fatalities, and severe multisystemic lesions at necropsy. This outbreak coincided with reemergence of BTV-8 and increasing reports of emerging BTV-12 and epizootic hemorrhagic disease virus (EHDV) circulation in Europe. Following the outbreak, samples from 1

INTRODUCTION: Bluetongue virus (BTV) is a species of genus Orbivirus belonging to the Sedoreoviridae family. Bluetongue (BT) is endemic in India and responsible for causing significant economic losses to livestock farmers. In India, antibodies to BTV serotype 24 (BTV-24) have been reported in 2005; it was first isolated in 2010, and it caused several outbreaks in sheep during 2012-2014. The in vivo studies investigating the pathogenetic potential of various BTV serotypes in the susceptible host sheep are scarce. Furthermore, detailed investigations to elucidate the pathogenetic mechanisms of B

Bluetongue virus (BTV) is a non-enveloped virus and causes substantial morbidity and mortality in ruminants such as sheep. Fashioning a receptor-binding protein (VP2) and a membrane penetration protein (VP5) on the surface, BTV releases its genome-containing core (VP3 and VP7) into the host cell cytosol after perforation of the endosomal membrane. Unlike enveloped ones, the entry mechanisms of non-enveloped viruses into host cells remain poorly understood. Here we applied single-particle cryo-electron microscopy, cryo-electron tomography and structure-guided functional assays to characterize i

Clinical and immunologic responses of sheep to vaccination and subsequent bluetongue virus (BTV) challenge exposure were studied and compared with those of non-vaccinated sheep. Sheep were vaccinated with inactivated BTV administered with aluminum hydroxide and cimetidine or levamisole. After sheep were vaccinated, precipitating group-specific antibodies to BTV were detected, but serotype-specific neutralizing antibodies were not detected. Cellular immune responses (lymphocyte blastogenesis) to BTV were not detected. After virulent BTV challenge exposure, vaccinated and nonvaccinated sheep dev

In 2021, a pregnant Rottweiler dog living on a sheep farm was diagnosed with clinical bluetongue (BT) infection. This study reports on the investigation of this farm where bluetongue virus (BTV) infection was diagnosed in this atypical host species. Samples were collected during farm visits 14, 28, 60, and 89 days after the onset of clinical signs in the pregnant Rottweiler. Blood was collected from all farm dogs (n = 6) and tested for BTV genome using a reverse-transcriptase quantitative PCR (RT-qPCR) assay and BTV antibodies with the competitive ELISA (cELISA) and dogs positive by RT-qPCR we

Bluetongue virus (BTV), a nonenveloped double-stranded RNA virus, is a potent inducer of type Ι interferons in multiple cell systems. In this study, we report that BTV16 treatment of primary human macrophages induced both type I and III IFN expression, resulting in the production of multiple antiviral factors, including myxovirus resistance protein A, 2',5'-oligoadenylate synthetase, and the IFN-stimulated gene 56. Additionally, BTV-treated macrophages expressed increased HIV restriction factors (apolipoprotein B mRNA-editing enzyme catalytic polypeptide 3 G/F/H) and CC chemokines (macrophage

An EDTA-blood sample from a cow without clinical signs, which gave early birth to a newborn calf that died soon after delivery, was shown to be positive for bluetongue virus (BTV)-RNA using a group-specific real-time RT-PCR (RT-qPCR). In-house serotype-specific RT-qPCR assays for bluetongue virus serotype 1 (BTV-1), -6 and -8 all gave negative results. Subsequent assays were carried out using conventional (gel-based) RT-PCR primers for all 25 BTV serotypes and only two primer sets, both specific for BTV-11, gave bands of the expected size. The cDNAs generated were sequenced and comparisons of

A number of viruses have the capacity to cross the placenta and infect the fetus to cause, among other potential outcomes, developmental defects (teratogenesis), fetal death and abortion. Bluetongue virus (BTV) infection of fetal ruminants provides an excellent model for the study of virus-induced teratogenesis. This model has shown that only viruses modified by passage in cell culture, such as modified live virus vaccine strains, readily cross the ruminant placenta, and that the timing of fetal infection determines the outcome. Thus, cerebral malformations only occur after fetal infection at
