β, β-Dimethylacrylshikonin (DMAS), a natural naphthoquinone derivative isolated from the root of a biennial herb belonging to the family Boraginaceae, has been demonstrated to exhibit anti-cancer and anti-inflammatory features. Yet, the effects of DMAS on restraining the progression of prostate cancer (PC) remain mostly elusive. Here, we attempted to examine whether DMAS hampers PC progression and subsequently explored the underlying mechanisms. Our results demonstrate that DMAS elicited cytotoxicity to PC cell lines, DU-145 and PC-3 cells, along with promotion of apoptosis and cell cycle arre

Shikonin, a naphthoquinone secondary metabolite derived from the dried roots of Lithospermum erythrorhizon Sieb. et Zucc. (family Boraginaceae), has garnered significant attention in natural drug development due to its diverse pharmacological activities, including anti-inflammatory, immunomodulatory, and anti-tumor effects. Its anti- inflammatory and immunomodulatory properties are primarily mediated through the inhibition of key signaling pathways, such as NF-κB and MAPK, leading to the reduced expression of pro-inflammatory cytokines like TNF-α and IL-8, and the modulation of Th17/Treg cell

Shikonin, a 1,4-naphthoquinone derivative produced by some limited Boraginaceae species, exhibits unique pharmacological properties and is also used as a natural dye. The regulatory factors of shikonin production have been demonstrated using a cell culture system of Lithospermum erythrorhizon. Among these factors, copper is known to be the strongest enhancer of shikonin production, and the copper-requiring biosynthetic step is shikonin-specific, unlike the byproduct benzoquinones. Despite the long history of shikonin biosynthesis studies for over 40 years, the copper-involved reaction has been
