64Cu-Labeled Boronic-Acid-Conjugated Porphyrin: A Novel Agent for PET Imaging and Boron Neutron Capture Therapy.

Radiometal-labeled porphyrin derivatives have emerged candidates for targeted brain tumor imaging and therapy. In this study, we synthesized a boronic-acid-conjugated porphyrin and radiolabeled it with 64Cu to evaluate its dual-function potential for PET imaging and boron neutron capture therapy (BNCT). The 64Cu-labeled 5,10,15,20-(tetra-4-dihydroxyborylphenyl)-porphyrin ([64Cu]-Cu-TDBP) exhibited excellent serum stability and significant uptake in U87MG cells. PET imaging in both xenograft and orthotopic models demonstrated time-dependent tumor accumulation, with selective localization in the

A DNA damage calculation method for large-scale tumor models in boron neutron capture therapy.

Objective.Boron neutron capture therapy (BNCT) is considered a binary, targeted radiotherapy modality that can effectively eradicate tumor cells via the10B(n,α)7Li nuclear reaction. However, because conventional Monte Carlo approaches suffer from severe computational inefficiency in DNA damage calculations, most studies have been confined to single-cell models, which are inadequate for characterizing the spatial distribution of DNA damage in clinically relevant tumor tissues. To address this limitation, we propose a method that quantitatively evaluates DNA damage yields induced byαand7Li parti

Salvage boron neutron capture therapy for recurrent intracranial malignant meningioma: Case report and literature review.

RATIONALE: Meningiomas are the most common central nervous system tumors in adults, accounting for approximately one-third of the total. Although only 1% to 3% of meningiomas are malignant, these cases exhibit a high degree of aggression and frequent recurrence, resulting in a poor prognosis. There are no accepted treatment standards for salvage re-irradiation. Therefore, detailed case descriptions are essential for treatment guidance. PATIENT CONCERNS: A 56-year-old female with malignant meningioma underwent curative resection and adjuvant radiotherapy (66 Gy in 33 fractions). However, the t

Advanced Boron Neutron Capture Therapy Targeting Cancer Stem Cells by Selective Induction of LAT1 Overexpression.

This study conducted fundamental research to develop a more effective BNCT targeting cancer stem cells. We constructed plasmids that induced the overexpression of L-type amino acid transporter 1 (LAT1) tagged with tdTomato on the cytoplasmic membranes of CD133 expressing cancer cells. After transfection of the plasmids into a glioblastoma cell line (T98G), several clones overexpressing LAT1-tdTomato in the hypoxic microenvironment of the spheroids formed from each clone were obtained. Confocal laser microscopic observation confirmed that signals from LAT1-tdTomato overlapped with immunofluores

A novel pH sensitive theranostic PLGA nanoparticle for boron neutron capture therapy in mesothelioma treatment.

This study aims to develop poly lactic-co-glycolic acid (PLGA) nanoparticles with an innovative imaging-guided approach based on Boron Neutron Capture Therapy for the treatment of mesothelioma. The herein-reported results demonstrate that PLGA nanoparticles incorporating oligo-histidine chains and the dual Gd/B theranostic agent AT101 can successfully be exploited to deliver a therapeutic dose of boron to mesothelioma cells, significantly higher than in healthy mesothelial cells as assessed by ICP-MS and MRI. The selective release is pH responsive taking advantage of the slightly acidic pH of

Synthesis of Cobalt Bis(Dicarbollide)-Curcumin Conjugates for Potential Use in Boron Neutron Capture Therapy.

A series of novel cobalt bis(dicarbollide)-curcumin conjugates were synthesized. Two conjugates were obtained through the nucleophilic ring-opening reaction of the 1,4-dioxane and tetrahydropyran derivatives of cobalt bis(dicarbollide) with the OH group of curcumin, and using two equiv. of the oxonium derivatives, two other conjugates containing two cobalt bis(dicarbollide) units per molecule were obtained. In contrast to curcumin, the conjugates obtained were found to be non-cytotoxic against both tumor and normal cell lines. The analysis of the intracellular accumulation of the conjugates by

Boron neutron capture therapy for vulvar melanoma and genital extramammary Paget's disease with curative responses.

BACKGROUND: Although the most commonly recommended treatment for melanoma and extramammary Paget's disease (EMPD) of the genital region is wide surgical excision of the lesion, the procedure is highly invasive and can lead to functional and sexual problems. Alternative treatments have been used for local control when wide local excision was not feasible. Here, we describe four patients with genital malignancies who were treated with boron neutron capture therapy (BNCT). METHODS: The four patients included one patient with vulvar melanoma (VM) and three with genital EMPD. They underwent BNCT a

Suppression of Tumor Growth in a Rabbit Hepatic Cancer Model by Boron Neutron Capture Therapy With Liposomal Boron Delivery Systems.

BACKGROUND/AIM: Tumor cell destruction by boron neutron capture therapy (BNCT) is attributed to the nuclear reaction between 10B and thermal neutrons. The accumulation of 10B atoms in tumor cells without affecting adjacent healthy cells is crucial for effective BNCT. We previously reported that several types of liposomal boron delivery systems (BDS) delivered effective numbers of boron atoms to cancer tissues, and showed tumor-growth suppression after thermal neutron irradiation. In the present study, we examined the effects of BNCT after intra-arterial infusion of 10B-borono-dodecaborate (10B

Design, Synthesis, and Biological Evaluation of Boron-Containing Macrocyclic Polyamines and Their Zinc(II) Complexes for Boron Neutron Capture Therapy.

Boron neutron capture therapy (BNCT) is a binary therapeutic method for cancer treatment based on the use of a combination of a cancer-specific drug containing boron-10 (10B) and thermal neutron irradiation. For successful BNCT, 10B-containing molecules need to accumulate specifically in cancer cells, because destructive effect of the generated heavy particles is limited basically to boron-containing cells. Herein, we report on the design and synthesis of boron compounds that are functionalized with 9-, 12-, and 15-membered macrocyclic polyamines and their Zn2+ complexes. Their cytotoxicity, i

Uptakes of boronophenylalanine in the in vitro and in situ glioblastoma and the potential efficacy of boron neutron capture therapy.

Human glioblastoma (GBM) is the most malignant brain tumor. Boron neutron capture therapy (BNCT) is proved to be a new technology for the effective treatment of GBM. We previously reported that boronophenylalanine (BPA), the boron drug used in BNCT prefers to the tumor of a mouse subcutaneous tumor model injected U87 and GL261, two GBM cell lines. The present study was designed to investigate the distribution of BPA in the tumor of a mouse in situ brain tumor model injected U87 and GL261 in the brain. The tumor model was evaluated by using small animal magnetic resonance imaging (MRI) and conf

Biodistribution of p-boronophenylalanine in patients with glioblastoma multiforme for use in boron neutron capture therapy.

OBJECTIVE: The success of boron neutron capture therapy depends on the safety and specificity of the boron delivery agent. As a preface to clinical boron neutron capture therapy of glioblastoma multiforme, a biodistribution study of intravenous p-boronophenylalanine (BPA) in patients undergoing craniotomy for resection of glioblastoma was performed. METHODS: Varying doses of intravenously administered BPA-fructose (130-250 mg BPA per kilogram of body weight) were given to patients 2 to 3 hours prior to the start of craniotomy for either suspected or known glioblastoma multiforme. Blood sample

A method for delivering the required neutron fluence in an accelerator-based boron neutron capture therapy system employing a lithium target.

Accelerator-based boron neutron capture therapy (BNCT) systems employing a solid-state lithium target indicated the reduction of neutron flux over the lifetime of a target, and its reduction could represent the neutron flux model. This study proposes a novel compensatory approach for delivering the required neutron fluence and validates its clinical applicability. The proposed approach relies on the neutron flux model and the cumulative sum of real-time measurements of proton charges. The accuracy of delivering the required neutron fluence for BNCT using the proposed approach was examined in f

Maximizing nuclear capture and fission reactions: The innovative potential of boron delivery agents for neutron capture therapy in cancer treatment.

Neutron capture therapy (NCT) is a binary radiotherapeutic modality that relies on the nuclear capture and fission reactions of boron-10 for the selective destruction of cancer cells. Boron delivery agents (BDAs) are used to selectively deliver boron-10 to cancer cells, where it can be activated by neutron irradiation to produce highly cytotoxic alpha particles and recoiling lithium-7 nuclei that cause localized DNA damage and cell death. The development of BDAs for NCT has been an area of active research for several decades, with significant progress made in recent years. Polyhedral boranes,

Novel Drug Delivery Particles Can Provide Dual Effects on Cancer "Theranostics" in Boron Neutron Capture Therapy.

Boron (B) neutron capture therapy (BNCT) is a novel non-invasive targeted cancer therapy based on the nuclear capture reaction 10B (n, alpha) 7Li that enables the death of cancer cells without damaging neighboring normal cells. However, the development of clinically approved boron drugs remains challenging. We have previously reported on self-forming nanoparticles for drug delivery consisting of a biodegradable polymer, namely, "AB-type" Lactosome® nanoparticles (AB-Lac particles)- highly loaded with hydrophobic B compounds, namely o-Carborane (Carb) or 1,2-dihexyl-o-Carborane (diC6-Carb), and

Recurrent meningioma treated with boron neutron capture therapy: a feasibility study with dosimetric and clinical correlates.

BACKGROUND: Boron neutron capture therapy (BNCT) is a targeted radiotherapy modality that has shown promise in the treatment of recurrent gliomas and head and neck cancers. Although meningiomas are generally slow-growing, recent studies have demonstrated favorable uptake of boron-containing compounds, particularly boronophenylalanine (BPA), suggesting a potential role for BNCT in recurrent meningioma. METHODS: We retrospectively analyzed 13 patients with recurrent meningiomas treated with salvage BNCT at the Tsing Hua Open-Pool Reactor between August 2020 and May 2024. Tumor uptake was assess

Development and in vitro studies of epidermal growth factor-dextran conjugates for boron neutron capture therapy.

A delivery molecule for directed boron neutron capture therapy against epidermal growth factor (EGF) receptor-rich tumors, such as gliomas, squamous carcinomas, and breast cancers, is presented. EGF and sulfhydryl boron hydride (BSH) were covalently coupled to an allylated 70 kDa dextran chain to form a conjugate. Conjugates with low and high substitution rates of BSH, as well as without BSH, were investigated. The conjugate with a low amount of boron had approximately 6 BSH (72 boron atoms) per dextran, while the conjugates with higher amounts had an average substitution of 55 BSH (660 boron

Boron neutron capture therapy combined with immunotherapy: insights from radiotherapy, BNCT-specific evidence, and future directions.

Boron neutron capture therapy (BNCT), a precision radiotherapy modality, has shown significant promise in targeted cancer treatment. Like conventional radiotherapy, the development of radioresistance hinders the efficacy of BNCT. Nevertheless, the physical parameters in BNCT induce the massive anti-tumor response compared to low-LET radiotherapy. Recent advancements have highlighted its synergistic potential with immunotherapy. In this review, we explore how the unique physical-to-biological trajectory of BNCT drives a potent "Cold-to-Hot" transformation of the tumor microenvironment. We descr
