Proteomic and virus-induced gene silencing (VIGS) Analyses reveal that gossypol, brassinosteroids, and jasmonic acid contribute to the resistance of cotton to Verticillium dahliae.

Verticillium wilt causes massive annual losses of cotton yield, but the mechanism of cotton resistance to Verticillium dahliae is complex and poorly understood. In this study, a comparative proteomic analysis was performed in resistant cotton (Gossypium barbadense cv7124) on infection with V. dahliae. A total of 188 differentially expressed proteins were identified by mass spectrometry (MALDI-TOF/TOF) analysis and could be classified into 17 biological processes based on Gene Ontology annotation. Most of these proteins were implicated in stimulus response, cellular processes and metabolic proc

A mutation in the brassinosteroid biosynthesis gene CpDWF5 disrupts vegetative and reproductive development and the salt stress response in squash (Cucurbita pepo).

A Cucurbita pepo mutant with multiple defects in growth and development has been identified and characterized. The mutant dwfcp displayed a dwarf phenotype with dark green and shrinking leaves, shortened internodes and petioles, shorter but thicker roots and greater root biomass, and reduced fertility. The causal mutation of the phenotype was found to disrupt gene Cp4.1LG17g04540, the squash orthologue of the Arabidopsis brassinosteroid (BR) biosynthesis gene DWF5, encoding for 7-dehydrocholesterol reductase. A single nucleotide transition (G > A) causes a splicing defect in intron 6 that lead

Brassinosteroid enhances salt tolerance via S-nitrosoglutathione reductase and nitric oxide signaling pathway in mangrove Kandelia obovata.

Brassinosteroid (BR) has been shown to modulate plant tolerance to various stresses. S-nitrosoglutathione reductase (GSNOR) is involved in the plant response to environment stress by fine-turning the level of nitric oxide (NO). However, whether GSNOR is involved in BR-regulated Na+ /K+ homeostasis to improve the salt tolerance in halophyte is unknown. Here, we firstly reported that high salinity increases the expression of BR-biosynthesis genes and the endogenous levels of BR in mangrove Kandelia obovata. Then, salt-induced BR triggers the activities and gene expressions of GSNOR and antioxida

Genes encoding plant-specific class III peroxidases are responsible for increased cold tolerance of the brassinosteroid-insensitive 1 mutant.

We previously reported that one of the brassinosteroidinsensitive mutants, bri1-9, showed increased cold tolerance compared with both wild type and BRI1-overexpressing transgenic plants, despite its severe growth retardation. This increased tolerance in bri1-9 resulted from the constitutively high expression of stress-inducible genes under normal conditions. In this report, we focused on the genes encoding class III plant peroxidases (AtPrxs) because we found that, compared with wild type, bri1-9 plants contain higher levels of reactive oxygen species (ROS) that are not involved with the activ

Overexpression of GmMYB14 improves high-density yield and drought tolerance of soybean through regulating plant architecture mediated by the brassinosteroid pathway.

MYB transcription factors (TFs) have been reported to regulate the biosynthesis of secondary metabolites, as well as to mediate plant adaption to abiotic stresses, including drought. However, the roles of MYB TFs in regulating plant architecture and yield potential remain poorly understood. Here, we studied the roles of the dehydration-inducible GmMYB14 gene in regulating plant architecture, high-density yield and drought tolerance through the brassinosteroid (BR) pathway in soybean. GmMYB14 was shown to localize to nucleus and has a transactivation activity. Stable GmMYB14-overexpressing (GmM

Brassinosteroid regulates thermotolerance through affecting the antioxidant capacity and the auxin pathway under heat stress in Brassica rapa ssp. pekinensis.

Heat stress threatens the yield and quality of plants. Brassinosteroid (BR) is widely used in agricultural production and plays important roles in regulating plant growth and stress response, but it is rarely reported on its function in thermotolerance in Chinese cabbage. In this study, we found that BR improves thermotolerance by alleviating the inhibition of leaf growth, and the BR biosynthesis inhibitor brassinazole (BRZ) treatment significantly reduced thermotolerance and plant growth in Chinese cabbage. Under heat stress, the accumulation of malondialdehyde and H2O2 was inhibited, and ant

Genetic manipulations of brassinosteroid-related genes improve various agronomic traits and yield in cereals enabling new biotechnological revolution: Achievements and perspectives.

Brassinosteroids (BRs) are steroid phytohormones which regulate various developmental and physiological processes throughout plant life cycle, from seed development and germination, up to modulation of reproduction and senescence. Importantly, mutants defective in the BR biosynthesis or response show various degree of plant height reduction (dwarfism or semi-dwarfism). This agronomic trait is of particular importance considering that in contrast to tall cereal varieties, semi-dwarf cereal plants are more tolerant to lodging which occurs during unfavorable weather conditions and constitutes a s

Single nucleotide substitution in the CsDET2 gene leads to complex alterations in plant architecture and affects brassinosteroid profile in cucumber (Cucumis sativus L.)0.

Most cucumber (Cucumis sativus L.) cultivars are characterized by indeterminate and vigorous growth, which requires a large cultivation area and increased labor for crop maintenance. Manipulating plant architecture may help optimize cucumber production in certain cultivation systems. The objective of this study was to identify the gene associated with phenotypic changes of the short petiole-2 (sp-2) mutant, which was discovered from EI (ethylenimine)-induced mutagenesis population. The line carrying the sp-2 gene was characterized by dwarf growth and dark-green wrinkled leaves with short petio

Differential Modulation of Brassinosteroid and Ethylene Signalling Systems by Native and Constitutively Active Forms of the AtCPK1 Gene in Transgenic Tobacco Plants Under Heat Stress.

Among other calcium decoders, Ca2+-dependent protein kinases (CDPK) stands out for its ability, depending on calcium levels, to activate key components of the defence system. However, calcium dependence prevents the effective use of CDPKs in comprehensive investigations of their functions. Previously, we showed that a modified constitutively active form of AtCPK1 improved heat tolerance in tobacco plants. At present, the role of calcium ions and their decoders in the regulation of heat tolerance is not fully understood. The response of plant cells to excessive temperature increases is regulate

Brassinosteroid-induced S-nitrosylation of fructose-1,6-bisphosphate aldolase increased ATP synthesis under low temperatures in mini Chinese cabbage seedlings.

Low temperature causes an imbalance in plant energy metabolism homeostasis. Brassinosteroids (BRs) are a class of hormones with important regulatory functions in plant responses to low temperature. S-nitrosylation of proteins is a key pathway for nitric oxide-mediated regulation of plant stress. In this study, we investigated whether S-nitrosylation proteins are involved in regulating energy metabolism in mini Chinese cabbage seedlings at low temperature. Low temperature inhibited the transcriptional accumulation of BrFBA2, BrGAPDH, BrPGK, and BrPK. However, applying exogenous BR and S-nitroso

STay tunEd: mutational analysis of the HvSTE1 gene in barley provides insight into the balance between semi-dwarfism and maintenance of grain size in brassinosteroid biosynthesis-dependent manner.

Brassinosteroids (BRs) are steroid phytohormones which regulate various physiological and developmental processes throughout plant life cycle. The BR biosynthesis has been studied mainly in the dicot model species - Arabidopsis thaliana. However, our current understanding of the BR biosynthesis and its regulation in other species, including cereal crops, is limited. Functions of enzymes which catalyze early stages of the BR biosynthesis in cereals remain poorly understood. Moreover, mechanisms regulating expression of genes encoding these enzymes remain obscure. One of the genes which particip

Multiple origins of Indian dwarf wheat by mutations targeting the TREE domain of a GSK3-like kinase for drought tolerance, phosphate uptake, and grain quality.

Multiple origins of Indian dwarf wheat were due to two mutations targeting the same TREE domain of a GSK3-like kinase, and these mutations confer to enhanced drought tolerance and increased phosphate and nitrogen accumulation for adaptation to the dry climate of Indian and Pakistan. Indian dwarf wheat, featured by the short stature, erect leaves, dense spikes, and small, spherical grains, was a staple crop in India and Pakistan from the Bronze Age until the early 1900s. These morphological features are controlled by a single locus Sphaerococcum 1 (S1), but the genetic identity of the locus and

ZmSSRP1-mediated chromatin accessibility modulates maize development via regulation of ZmBRD1 transcription.

The FAcilitates Chromatin Transcription (FACT) complex, comprising structure-specific recognition protein 1 (SSRP1) and suppressor of Ty 16 (SPT16), plays a key role in regulating gene transcription by facilitating ATP-independent nucleosome assembly. SSRP1 contains an HMG domain that binds to DNA and regulates cell proliferation, apoptosis, and DNA repair. However, its role in plant development remains poorly understood. In this study, we identified a mutant, small kernel 301 (smk301), characterized by reduced kernel size and semi-dwarfism. Map-based cloning revealed that the mutation affects

Membrane oestrogen receptor alpha signalling to cell functions.

Conservation of steroid hormone action outside the nucleus occurs from plants that make brassinosteroids to higher metazoans (primates). In plants, steroid hormone action occurs when the brassinosteroids bind a membrane tyrosine kinase receptor. Ligated receptors for all sex steroids exist at the plasma membrane and rapidly signal through G proteins to second messengers including calcium, cAMP and cGMP, activating proximal and more distal kinases. These signal cascades impact many functions of steroid hormones, responsible for the biological actions of these molecules. Support also exists for

GSK3/shaggy-like kinase 1 ubiquitously regulates cell growth from Arabidopsis to Moso bamboo (Phyllostachys edulis).

Moso bamboo (Phyllostachys edulis) is one of the fastest growing species with a maximum growth rate of 1 m/day. However, the regulator genes for this explosive growth phenomenon have not been functionally studied. Here, we found that Moso bamboo GSK3/shaggy-like kinase 1 (PeGSK1) acts as a negative regulator of cell growth. Over-expression of PeGSK1 in Arabidopsis showed significant growth arrest phenotypes, including dwarfism, small leaves, reduced cell length, and disturbed cell elongation of petiole. Furthermore, Overexpression of PeGSK1 fully inhibited the longer hypocotyl phenotype of Ara

An oomycete effector that induces shade avoidance-like growth and suppresses plant defences targets the AUX/IAA protein IAA11.

Filamentous pathogens, such as oomycetes, employ multiple strategies to suppress plant immunity, including manipulation of phytohormone signalling pathways. The effector HaRxL106 from the oomycete Hyaloperonospora arabidopsidis (Hpa), whose expression in planta induces shade avoidance syndrome (SAS)-like growth, has previously been shown to interact with BIM1, a transcription factor involved in brassinosteroid signalling. Here, we present evidence that HaRxL106 interacts with another key host target, the AUX/IAA protein IAA11, a negative regulator of auxin responses. Our data suggest that HaRx

Variation in Splicing Efficiency Underlies Morphological Evolution in Capsella.

Understanding the molecular basis of morphological change remains a central challenge in evolutionary-developmental biology. The transition from outbreeding to selfing is often associated with a dramatic reduction in reproductive structures and functions, such as the loss of attractive pheromones in hermaphroditic Caenorhabditis elegans and a reduced flower size in plants. Here, we demonstrate that variation in the level of the brassinosteroid-biosynthesis enzyme CYP724A1 contributes to the reduced flower size of selfing Capsella rubella compared with its outbreeding ancestor Capsella grandifl

CRISPR/dCas9-Mediated BRL3 Activation Enhances Growth and Metabolic Resilience Under Osmotic Stress in Nicotiana tabacum.

Brassinosteroids (BRs) are crucial plant hormones that influence growth and stress adaptation. However, the specific function of the BR receptor BRL3 under osmotic stress remains largely unexplored outside Arabidopsis thaliana. In this study, we used a CRISPR/dCas9-based transcriptional activation (CRISPRa) system to upregulate the Nicotiana tabacum BRASSINOSTEROID INSENSITIVE-LIKE 3 receptor (NtBRL3) and assessed its impact on osmotic stress tolerance. Synthetic activation vectors were constructed using Loop Assembly, featuring dCas9-6TAL-VP128 modules driven by either a constitutive (CaMV35S

Structure modification of nonsteroidal brassinolide-like compound, NSBR1.

Brassinolide (BL) is a possible plant growth regulator in agriculture, but the presence of a steroid skeleton hampers the field application of BL in agriculture because of its high synthetic cost. We discovered NSBR1 as the first nonsteroidal BL-like compound using in silico technology. Searching for more potent BL-like compounds, we modified the structure of NSBR1 with respect to 2 benzene rings and the piperazine ring. The activity of synthesized compounds was measured in Arabidopsis hypocotyl elongation. The propyl group of butyryl moiety of NSBR1 was changed to various alkyl groups, such a

Overexpression of a BR inactivating enzyme gene GhPAG1 impacts eggplant fruit development and anthocyanin accumulation mainly by altering hormone homeostasis.

Brassinosteroids (BRs) function importantly in plant growth and development, but the roles in regulating fruit development and anthocyanin pigmentation remain unclear. Eggplant (Solanum melongena L.) is an important Solanaceae vegetable crop rich in anthocyanins. The fruit size and coloration are important agronomic traits for eggplant breeding. In this study, transgenic eggplant exhibiting endogenous BRs deficiency was created by overexpressing a heterologous BRs-inactivating enzyme gene GhPAG1 driven by CaMV 35 S promoter. 35 S::GhPAG1 eggplant exhibited severe dwarfism, reduced fruit size,
