Molecular typing for epidemiological evaluation of Brucella abortus and Brucella canis isolated in Korea.

To investigate genotype relationships among regional groups of Brucella isolates, variable-number tandem-repeat (VNTR) analysis was conducted according to previously reported methods. Field strains of Brucella abortus and Brucella canis were isolated from 9 provinces in the Republic of Korea during the years 1996-2006 and each of the isolates was classified by eight loci of HOOF-Prints. On the basis of the alleles, the 33 B. abortus and 21 B. canis field strains were divided into 22 and 18 distinct genotypes, respectively. Phylogenetic cluster analysis of Brucella isolates could be discriminat

Substantial Protective Immunity Conferred by a Combination of Brucella abortus Recombinant Proteins against Brucella abortus 544 Infection in BALB/c Mice.

Chronic infection with intracellular Brucella abortus (B. abortus) in livestock remains as a major problem worldwide. Thus, the search for an ideal vaccine is still ongoing. In this study, we evaluated the protective efficacy of a combination of B. abortus recombinant proteins; superoxide dismutase (rSodC), riboflavin synthase subunit beta (rRibH), nucleoside diphosphate kinase (rNdk), 50S ribosomal protein (rL7/L12) and malate dehydrogenase (rMDH), cloned and expressed into a pMal vector system and DH5α, respectively, and further purified and applied intraperitoneally into BALB/c mice. Recombinant outer membrane protein 25c from Brucella abortus induces Th1 and Th2 mediated protection against Brucella abortus infection in mouse model.

Development of a safe and efficacious vaccine for brucellosis is a long standing challenge for scientists. Recognizing potential antigens towards developing vaccine candidate is crucial. Omp25c, a porin protein of Brucella, is a paralog of two previously identified promising vaccine candidates namely, Omp25 and Omp31, with notable sequence identity. Also, Omp25c is conserved in all major Brucella species. This highlights the possibility of employing this protein in multivalent subunit vaccine based approach of Brucella management. In this study, we were interested in examining the immunogenici

Genotyping of Brucella melitensis and Brucella abortus strains currently circulating in Xinjiang, China.

Brucellosis is a well-known zoonotic disease that can cause severe economic and healthcare losses. Xinjiang, one of the biggest livestock husbandry sectors in China, has gone through increasing incidence of brucellosis in cattle and small ruminants recently. In this paper, 50 B. melitensis strains and 9 B. abortus strains collected from across Xinjiang area (from 2010 to 2015) were genotyped using multiple locus variable-number tandem-repeat (VNTR) analysis (MLVA) and multi-locus sequence typing (MLST). Based on 8 loci (MLVA-8), 50 B. melitensis strains were classified into three genotypes. Ge

MLVA16 typing of Portuguese human and animal Brucella melitensis and Brucella abortus isolates.

To investigate the epidemiological relationship of isolates from different Portuguese geographical regions and to assess the diversity among isolates, the MLVA16(Orsay) assay (panels 1, 2A and 2B) was performed with a collection of 126 Brucella melitensis (46 human and 80 animal isolates) and 157 B. abortus field isolates, seven vaccine strains and the representative reference strains of each species. The MLVA16(Orsay) showed a similar high discriminatory power (HGDI 0.972 and 0.902) for both species but panel 1 and 2A markers displayed higher diversity (HGDI 0.693) in B. abortus compared to B

Comparative proteome analysis of laboratory grown Brucella abortus 2308 and Brucella melitensis 16M.

Brucella species are pathogenic agents that cause brucellosis, a debilitating zoonotic disease that affects a large variety of domesticated animals and humans. Brucella melitensis and Brucella abortus are considered major health threats because of their highly infectious nature and worldwide occurrence. The availability of the annotated genomes for these two species has allowed a comparative proteomics study of laboratory grown B. melitensis 16M and B. abortus 2308 by two-dimensional (2-D) gel electrophoresis and peptide mass fingerprinting. Computer-assisted analysis of the different 2-D gel

Prevalence study in Cameroon identifies Brucella abortus as the endemic Brucella species in livestock.

Brucellosis is a neglected zoonotic disease impacting agricultural economies, as well as animal and human health globally. In sub-Saharan Africa, brucellosis is considered endemic in many countries based on serological evidence, although the presence and distribution of specific Brucella species remain unverified due to limited bacteriological confirmation. Due to the economic importance and national/international trade routes, this cross-sectional study investigates Brucella prevalence in 4612 animals (cattle, sheep, goats, and pigs) from slaughterhouses in the Far-North, North, and West regi

Development and validation of immunoassay for whole cell detection of Brucella abortus and Brucella melitensis.

Brucella is alpha-2 Proteobacteria mainly responsible for multi-factorial bacterial zoonotic disease brucellosis with low concentration (10-100 CFU) required to establish the infection. In this study, we developed sandwich ELISA with detection range of 102 to 108 cells mL-1 and limit of detection at 103 cells mL-1 by employing polyclonal rabbit IgG (capture antibody, 10 µg mL-1) and mice IgG (detection antibody, 50 µg mL-1) antibody for its detection. Surface Plasmon Resonance evaluated the interaction of detection antibody with whole cell spiked serum samples at LOD of 102 cells mL-1 along wi

Characterization of Brucella abortus lipopolysaccharide macrodomains as mega rafts.

The lipopolysaccharides (LPS) of intracellular Proteobacteria such as Brucella, Chlamydia, Legionella and Rickettsia, have properties distinct from enterobacterial LPSs. These properties include deficient LPS induction of host cell activation, low endotoxicity and resistance to macrophage degradation. Together these constitute key virulence mechanisms for intracellular survival and replication. We previously demonstrated that B. abortus LPS captured by macrophages was recycled back to the plasma membrane where it was found associated with macrodomains. Furthermore, this LPS interferes with the

Safety of Brucella abortus strain RB51 in black bears.

In two studies conducted from October 1999 to March 2000 and December 2000 to April 2001, adult black bears (Ursus americanus) were orally inoculated with 1.4-3.1 x 10(10) colony-forming units (CFU) of Brucella abortus strain RB51 (SRB51, n=12) or 2 ml of 0.15 M NaCl solution (saline, n=11). We did not detect a difference (P>0.05) in antibody titers to SRB51 in serum obtained before vaccination, at 8 wk after vaccination, or at necropsy at 21 or 23 wk after vaccination between SRB51-vaccinated and nonvaccinated bears. The SRB51 vaccine strain was recovered from tissues obtained at necropsy fro

[Persistent properties of Brucella abortus isolated from various sources].

Persistence factors in 70 B. abortus strains isolated from different sources were studied. Depending on the source of the isolation of B. abortus, the spread of their antilysozyme, anticomplementary, antiintercidal activity was determined and intraspecific differences in the manifestation of these signs were established. The highest spread and manifestation of persistence factors was registered in B. abortus populations isolated from brucellosis patients and from cattle.

Safety of Brucella abortus strain RB51 in bull elk.

Some of the elk (Cervus elaphus nelsoni) of the Greater Yellowstone Area (Wyoming, Idaho, Montana; USA) are infected with Brucella abortus, the bacterium that causes bovine brucellosis. Brucella abortus strain RB51 vaccine is being considered as a means to control B. abortus induced abortions in cow elk. However, the most probable vaccination strategies for use in free-ranging elk might also result in some bull elk being inoculated, thus, it is important to insure that the vaccine is safe in these animals. In the winter of 1995, 10 free-ranging bull elk calves were captured, tested for B. abor

Human exposure to Brucella abortus strain RB51--Kansas, 1997.

On May 26-27, 1997, nine persons (a farmer, four veterinary clinicians, and four veterinary students) in Manhattan, Kansas, participated in an attempted vaginal delivery, a cesarean delivery, and a necropsy on a stillborn calf that died because of Brucella abortus infection. The infection was confirmed by isolation of B. abortus from placental and fetal lung tissue cultures. The National Animal Disease Center, U.S. Department of Agriculture (USDA), identified the B. abortus isolate from the calf as the RB51 vaccine strain. RB51 is a live, attenuated strain that was licensed conditionally by th

Poor perinatal outcome associated with maternal Brucella abortus infection.

BACKGROUND: Reports suggest that perinatal infection with Brucella abortus does not cause poor obstetric outcomes, because of protective mechanisms in the human, not seen in animal species. CASE: We report a case of maternal brucellosis resulting in preterm labor, chorioamnionitis, placental abruption, and delivery of a live-born infant at 25 weeks' gestational age. Both maternal blood cultures and amniotic fluid cultures were positive for B abortus species, and delivery occurred despite aggressive antibiotic and tocolytic therapy. CONCLUSION: Maternal infection with B abortus during pregnan

The Genome Sequence of Brucella abortus vaccine strain A19 provides insights on its virulence attenuation compared to Brucella abortus strain 9-941.

BACKGROUND: Brucellosis is a widespread disease that affects animals and humans. The live attenuated Brucella abortus A19 strain is used for vaccination against brucellosis in China. In addition, the main mechanisms supporting the residual toxicity of A19 have not been elucidated. Here, we performed a comprehensive comparative analysis of the genome-wide sequence of A19 against the whole genome sequences of the published virulent reference strain 9-941. The primary objective of this study was to identify candidate virulence genes by systematically comparing the genomic sequences between the tw

Completion of the genome sequence of Brucella abortus and comparison to the highly similar genomes of Brucella melitensis and Brucella suis.

Brucellosis is a worldwide disease of humans and livestock that is caused by a number of very closely related classical Brucella species in the alpha-2 subdivision of the Proteobacteria. We report the complete genome sequence of Brucella abortus field isolate 9-941 and compare it to those of Brucella suis 1330 and Brucella melitensis 16 M. The genomes of these Brucella species are strikingly similar, with nearly identical genetic content and gene organization. However, a number of insertion-deletion events and several polymorphic regions encoding putative outer membrane proteins were identifie

Diagnosis and treatment of infection with Brucella abortus, biotype 5.

A case of brucellosis due to Brucella abortus, biotype 5, occurred in a bulk milk-tanker driver who collected milk from refrigerated tanks on 23 farms. The organism was isolated from blood cultures by using a serum dextrose broth containing antibiotics. Serological investigations indicate that both abortus and melitensis antigen suspensions should be used in the investigation of cases of suspected brucellosis. As Brucella abortus is an intracellular parasite heavy, prolonged, antibiotic therapy is necessary in the treatment of the disease. The danger of inadequate treatment of the acute diseas
