BACKGROUND: Brucellosis, a disease of major veterinary and public health concern, remains widespread in several regions, including Egypt. This study reports the isolation and molecular confirmation of Brucella canis from dogs in Egypt and provides genetic relatedness of Brucella melitensis and Brucella canis in dogs to evaluate their public health significance. METHODS: A total of 785 dogs (both stray and owned) in Cairo and Giza (Feb–Dec 2024) were examined for Brucella spp.; all blood samples were examined using serological tests only (RBPT, SAT with 2-ME) however, 365 reproductive tissue sa

Brucella canis is a zoonotic pathogen of dogs that poses diagnostic challenges. While direct detection of B. canis by PCR or culture is ideal, serologic diagnosis is necessary for identification of carrier animals and can support a clinical diagnosis of brucellosis. Prior to 2022, B. canis seroscreening in the United States was primarily performed using a commercially available rapid slide agglutination test. However, the kit was discontinued by the manufacturer in early 2022, leaving a gap in the availability of commercial B. canis seroassays. The goal of this study was to compare the perform

A 2-year-old, spayed female, Bichon Frise dog was presented with reluctance to exercise, back pain, and frequent sitting down. Multiple osteolysis, periosteal proliferation, and sclerosis of the vertebral endplates of T11-13 were observed in the radiography, computed tomography, and magnetic resonance imaging. The bacterial culture of the urine specimen, the polymerase chain reaction (PCR) of the blood, and the antibody tests were positive for Brucella canis. Accordingly, discospondylitis caused by B. canis was diagnosed and doxycycline was administered. The clinical signs resolved and the cul

Introduction: The risk of Brucella canis infection in humans and dogs has increased due to the permanent exposure to asymptomatic carrier dogs. In Colombia, there is evidence of B. canis infection in humans living with dogs. In the case of Bogotá, an additional concern is the lack of updated information related to the prevalence of the infection in dogs. Objective: To determine the seroprevalence of infection by B. canis in dogs intended for adoption programs in Bogotá. Materials and methods: By means of a descriptive cross-sectional study carried out in a dog shelter in Bogotá, anti-B. canis

Brucella canis is a small intracellular Gram-negative bacterium whose primary host is the dog, but it also can cause mild human brucellosis. One of the main causes of an inefficient immune response against other species of Brucella is their interaction with dendritic cells (DCs), which affects antigen presentation and impairs the development of an effective Th1 immune response. This study analysed the cytokine pattern production, by RT-qPCR and ELISA, in human and canine DCs against whole B. canis or its purified LPS. Human and canine DCs produced different patterns of cytokines after stimulat

Brucella canis is a facultative intracellular pathogen responsible for canine brucellosis, a zoonotic disease that affects canines, causing abortions and reproductive failure; and the production of non-specific symptoms in humans. In 2005 the presence of B. canis in Antioquia was demonstrated and the strains were identified as type 2. The sequencing of the genome of a field strain denoted Brucella canis str. Oliveri, showed species-specific indel events, which led us to investigate the genomic characteristics of the B. canis strain isolated and to establish the phylogenetic relationships and t

The virB locus, which encodes the type IV secretion system, is a major component of virulence in Brucella. A non-polar virB10 mutant and a virB11 deletion mutant were constructed in Brucella canis. In the mouse model, both mutants were cleared at day 21 post-infection, indicating reduced virulence in mice. After challenging with wild-type B. canis, the amounts of CFU recovered at day 15 were significantly lower in the group previously vaccinated with the virB10 mutant. Levels of IgG1, IgG2a, IgG2b, and IgM, the induction of the cytokines IL-2, IL-4, IL-10, and the production of IFN-γ were meas

The multiple-locus VNTR analysis (MLVA) assay is a method frequently employed as a molecular epidemiological tool for Brucella genetic fingerprinting. The purpose of this study was to assess the genotyping of 77 B. canis isolates from 14 different dog breeding farms in Korea by the MLVA assay and to compare the epidemiological relationships between the Korean isolates and foreign ones. Simpson's diversity index for 17 loci showed a range from 0 to 0.846 in 77 B. canis isolates. B. canis isolates in Korea were observed to have high genetic diversity at the most variable loci and were divided in

A retrospective study of 135 dogs with diskospondylitis revealed 14 dogs with concurrent Brucella canis infection. Sexually intact male dogs and dogs in the southeastern United States appeared to be at higher risk. Results of bacteriologic culturing of blood were less likely to be positive for dogs with diskospondylitis caused by B canis infection than for dogs with diskospondylitis caused by other organisms. Follow-up evaluation of 13 of the 14 dogs revealed complete remission of clinical signs in nine, but serologic test results continued to be positive for B canis infection long after resol

Brucella canis is a Gram-negative, facultative intracellular bacterium and the causative agent of canine brucellosis, a highly contagious disease of dogs that can be transmitted to humans. Unfortunately, no vaccine is available to prevent infection. We recently characterized the kinetics of B. canis infection in the mouse model, establishing the required dose necessary to achieve systemic infection. The objective of this study was to investigate the utility of the mouse model in assessing canine brucellosis vaccine candidates and to subsequently investigate the safety and efficacy of a live at

Brucella canis is a zoonotic pathogen and the main causative agent of canine brucellosis. In the Netherlands, B. canis had previously only been detected in individual cases of imported dogs. However, an outbreak of B. canis occurred for the first time in a cohort of autochthonous dogs in a breeding kennel in 2019. The outbreak began with a positive serological test result of an imported intact male dog showing clinical symptoms of brucellosis. Consequently, urine and blood samples were collected and tested positive for B. canis by culture, matrix-assisted laser desorption/ionization - time of

The golden jackal Canis aureus occurs in south-eastern Europe, Asia, the Middle East, the Caucasus, and Africa. In Serbia, jackals neared extinction; however, during the last 30 years, the species started to spread quickly and to increase in number. Few studies in the past have revealed their potential role as carriers of zoonotic diseases. Animal samples were collected over a three-year period (01/2010-02/2013) from 12 sites all over Serbia. Of the tissue samples collected, spleen was chosen as the tissue to proceed; all samples were tested for Leishmania species and Brucella species by real-

OBJECTIVES: The aim of this study was to illustrate and help address a growing need for regulatory or molecular tools to track and control the spread of canine brucellosis. Our study objectives were to first characterize Brucella canis outbreaks in Wisconsin kennels in the context of the dog trade in the USA, and then to identify a molecular technique that may be useful for strain differentiation of B. canis isolates.
METHODS: Wisconsin Veterinary Diagnostic Laboratory (WVDL) B. canis serology data from 1995 to 2005 were reviewed, three canine brucellosis outbreaks in Wisconsin dog kennels we

The establishment of infection by Brucella ovis and Brucella canis in J774.A1 macrophages was found to be dependent upon cholesterol and ganglioside GM(1), two components of lipid rafts. This process also required a class A scavenger receptor of macrophages, and was not inhibited by smooth and rough lipopolysaccharides from Brucella spp. In response to infection, both bacteria induced a weak degree of macrophage activation. These results demonstrate that B. ovis and B. canis use cell surface receptors common to smooth Brucella spp. for macrophage infection, thus limiting macrophage activation

In the present study, the validation of an enzyme-linked immunosorbent assay (ELISA) for serodiagnosis of canine brucellosis is described. Two different antigenic extracts, obtained by heat or ultrasonic homogenization of microbial antigens from a wild isolate of Brucella canis bacteria, were compared by ELISA and Western blot (WB). A total of 145 canine sera were used to define sensitivity, specificity and accuracy of the ELISA as follows: (1) sera from 34 animals with natural B. canis infection, confirmed by blood culture and PCR, as well as 51 sera samples from healthy dogs with negative re

Human serological assays designed to detect brucellosis will miss infections caused by Brucella canis, and low levels of periodic bacteremia limit diagnosis by blood culture. Recent B. canis outbreaks in dogs and concomitant illnesses in caretakers suggest that unapparent human infections may be occurring. With more than a quarter of a million persons in occupations involving dogs, and nearly 80 million dog owners in the United States, this pathogen is an under-recognized human health threat. To investigate occupational exposure to B. canis, we adapted a commercial canine serological assay and
