Food preparation with mucoralean fungi: A potential biosafety issue?

Mucorales have been used for production of fermented food in Asia and Africa since time immemorial. Particularly Rhizopus species are rapidly growing, active producers of lipases and proteases and occur naturally during the first stages of soybean fermentation. Two biosafety issues have been raised in recent literature: (1) pathogenicity, Rhizopus species being prevalent opportunists causing erosive infections in severely compromised patients, and (2) toxicity, strains harbouring endosymbiotic Burkholderia producing toxic secondary metabolites. At the molecular level, based on different gene m

Clinical and microbiological features of obstructive cholangitis with bloodstream infection caused by Pandoraea apista identified by MALDI-TOF mass spectrometry and ribosomal RNA sequencing in a cancer patient.

BACKGROUND: Pandoraea species are multidrug-resistant glucose-nonfermenting gram-negative bacilli that are usually isolated from patients with cystic fibrosis (CF) and from water and soil. Reports of diseases, including bloodstream infections, caused by Pandoraea spp. in non-CF patients are rare, and the clinical and microbiological characteristics are unclear. The identification of Pandorea spp. is limited by conventional microbiological methods and may be misidentified as other species owing to overlapping biochemical profiles. Here, we report the first case of obstructive cholangitis with b

Differential gut microbiota and inflammatory cytokines contribute to IgA vasculitis.

OBJECTIVES: Immunoglobulin A vasculitis (IgAV) is the most common form of vasculitis in childhood. Emerging evidence indicates that gut microbiota plays a key role in the pathogenesis of IgAV. However, the factors linking gut microbiota to the onset and progression of IgAV are poorly understood. We aimed to demonstrate that the presence of a specific dysbiosis in patients with IgAV contributes to the onset of IgAV. METHODS: We transplanted gut microbiota from human donors with IgAV or healthy controls (HCs). The changes in gut microbiota and serum indexes of the recipient mice were detected,

Comparative microbiome analysis in cystic fibrosis and non-cystic fibrosis bronchiectasis.

BACKGROUND: Bronchiectasis is a condition characterized by abnormal and irreversible bronchial dilation resulting from lung tissue damage and can be categorized into two main groups: cystic fibrosis (CF) and non-CF bronchiectasis (NCFB). Both diseases are marked by recurrent infections, inflammatory exacerbations, and lung damage. Given that infections are the primary drivers of disease progression, characterization of the respiratory microbiome can shed light on compositional alterations and susceptibility to antimicrobial drugs in these cases compared to healthy individuals. METHODS: To ass

16S rRNA sequencing in chronic dacryocystitis.

CLINICAL RELEVANCE: The pathogenesis of chronic dacryocystitis (CDC) is associated with a variety of bacteria. Investigating microflora has the potential to provide a theoretical basis for preventing and treating CDC. BACKGROUND: 16S rRNA sequencing is a sequence-based bacterial analysis. The application of 16S rRNA sequencing in CDC is rarely reported. METHODS: A case-control study of infected and healthy eyes diagnosed as CDC patients was conducted. Seventy-eight patients were divided into A (conjunctival sac secretions from healthy eyes), B (conjunctival sac secretions from affected eyes)

The oral cavity and intestinal microbiome in children with functional constipation.

Constipation is a widespread problem in paediatric practice, affecting almost 30% of children. One of the key causal factors of constipation may be disturbances in the homeostasis of the gastrointestinal microbiome. The aim of the study was to determine whether the oral and fecal microbiomes differ between children with and without constipation. A total of 91 children over three years of age were included in the study. Of these, 57 were qualified to a group with constipation, and 34 to a group without. The saliva and stool microbiomes were evaluated using 16S rRNA gene amplicon sequencing. Fun

Identification of genes involved in the biosynthesis of the cytotoxic compound glidobactin from a soil bacterium.

Glidobactins (syn. cepafungins) are a family of structurally related cytotoxic compounds that were isolated from the soil bacterial strain K481-B101 (ATCC 53080; DSM 7029) originally assigned to Polyangium brachysporum and, independently, from an undefined species related to Burkholderia cepacia. Glidobactins are acylated tripeptide derivatives that contain a 12-membered ring structure consisting of the two unique non-proteinogenic amino acids erythro-4-hydroxy-l-lysine and 4(S)-amino-2(E)-pentenoic acid. Here we report the cloning and functional analysis of a gene cluster (glbA-glbH) involved

Robust design of LAMP assays for in-field detection of major bacterial vascular diseases of banana.

Bacterial diseases of banana are a growing global threat, causing yield losses and increased management costs. Major diseases include Moko, banana blood disease (BBD), and banana Xanthomonas wilt (BXW), caused by Ralstonia solanacearum, Ralstonia syzygii subsp. celebesensis, and Xanthomonas vasicola pv. musacearum, respectively. Effective surveillance requires point-of-care diagnostics such as loop-mediated isothermal amplification (LAMP) for on-site use. We aimed to develop three LAMP assays to specifically detect the bacteria responsible for Moko, BBD and BXW, directly from banana tissues, u

Comparative Analysis of Tumor Microbiota Identifies a Metastatic-Specific Bacterial Signature, Highlighting Streptococcus spp. As the Predominant Hub across Cancers.

BACKGROUND: Cancer remains one of the leading global health challenges, with lung cancer (LC), breast cancer (BC), and colorectal cancer (CRC) among the most prevalent and deadly malignancies. The intratumoral microbiota (IM), a distinct microbial ecosystem within tumor tissues, has recently emerged as a potential modulator of carcinogenesis, immune responses, and metastatic progression. However, comparative cross-cancer analyses remain limited. Therefore, this study aimed to compare the IM across these cancer types, with particular emphasis on distinguishing metastatic from non-metastatic mal

Successful salvage of a PICC in catheter-related bloodstream infection caused by multidrug-resistant Ralstonia mannitolilytica using topical minocycline: a case report.

BACKGROUND: Ralstonia mannitolilytica is a Gram-negative bacillus belonging to the genus Ralstonia in the family Burkholderiaceae. As an opportunistic pathogen, it is widely distributed in water sources and oligotrophic environments, but cases of catheter-related bloodstream infection (CRBSI) caused by this bacterium are relatively rare. Clinical guidelines recommend catheter removal combined with systemic antibiotic therapy as the standard treatment for CRBSI. However, catheter removal may not be an ideal option for critically ill patients with limited vascular access. CASE PRESENTATION: A 47

Consistent microbial responses during the aerobic thaw of Alaskan permafrost soils.

Arctic systems are warming at four times the global average, causing permafrost-permanently frozen soil, ice, organic matter, and bedrock-to thaw. Permafrost thaw exposes previously unavailable soil carbon and nutrients to decomposition-a process mediated by microbes-which releases greenhouse gases such as carbon dioxide and methane into the atmosphere. While it is well established that thaw alters the composition and function of the permafrost microbiome, patterns revealing common responses to thaw across different permafrost soil types have not yet emerged. In this study, we address how perm

Enhancement of bio-S0 recovery and revealing the inhibitory effect on microorganisms under high sulfide loading.

Biodesulfurization is a mature technology, but obtaining biosulfur (S0) that can be easily settled naturally is still a challenge. Increasing the sulfide load is one of the known methods to obtain better settling of S0. However, the inhibitory effect of high levels of sulfide on microbes has also not been well studied. We constructed a high loading sulfide (1.55-10.86 kg S/m3/d) biological removal system. 100% sulfide removal and 0.56-2.53 kg S/m3/d S0 (7.0 ± 0.09-16.4 ± 0.25 μm) recovery were achieved at loads of 1.55-7.75 kg S/m3/d. Under the same load, S0 in the reflux sedimentation tank, w

Microbial Community Profile and Water Quality in a Protected Area of the Caatinga Biome.

The Caatinga is a semi-arid biome in northeast Brazil. The Paraguaçú River is located in the Caatinga biome, and part of its course is protected by the National Park of Chapada Diamantina (PNCD). In this study we evaluated the effect of PNCD protection on the water quality and microbial community diversity of this river by analyzing water samples obtained from points located inside and outside the PNCD in both wet and dry seasons. Results of water quality analysis showed higher levels of silicate, ammonia, particulate organic carbon, and nitrite in samples from the unprotected area compared wi

The roles of extracellular polymeric substances of Pandoraea sp. XY-2 in the removal of tetracycline.

In this study, the roles of extracellular polymeric substances (EPSs) excreted by Pandoraea sp. XY-2 in the removal of tetracycline (TC) were investigated. In the early stage, TC in the solution was mainly removed by the adsorption of EPSs, which accounted for 20% of TC. Thereafter, large amount of TC was transported into the intracellular and biodegraded. EPSs was extracted and the contents of polyprotein and polysaccharides reached their maximum values (30.84 mg/g and 11.15 mg/g) in the first four days. Fourier transform infrared spectroscopy analysis revealed that hydroxyl, methylidyne, met

Phage communities in household-related biofilms correlate with bacterial hosts.

The average American spends 93% of their time in built environments, almost 70% of that is in their place of residence. Human health and well-being are intrinsically tied to the quality of our personal environments and the microbiomes that populate them. Conversely, the built environment microbiome is seeded, formed, and re-shaped by occupant behavior, cleaning, personal hygiene and food choices, as well as geographic location and variability in infrastructure. Here, we focus on the presence of viruses in household biofilms, specifically in showerheads and on toothbrushes. Bacteriophage, virus

Antibiotic resistance genes are more abundant in microplastic textile biofilms than natural cotton biofilms in freshwater.

UNLABELLED: Microplastic fibers (MPFs) are widespread pollutants in freshwater systems, providing artificial surfaces that facilitate microbial attachment and the potential spread of antibiotic resistance genes (ARGs). We compared bacterial colonization on natural cotton fibers with that on synthetic MPFs (Kevlar, acrylonitrile, polyester, and nylon) incubated in river and lake water. Bacterial biomass and community composition were analyzed using epifluorescence microscopy, scanning electron microscopy, and 16S rRNA sequencing, while the presence and relative abundance of key ARGs (blaNDM-1,
