Butyrate prevents muscle atrophy after sciatic nerve crush.

INTRODUCTION: Histone deacetylases (HDACs) have been implicated in neurogenic muscle atrophy, but the mechanisms by which HDAC inhibitors might have beneficial effects are not defined. METHODS: We used sciatic nerve crush to determine the effect of butyrate on denervation-induced gene expression and oxidative stress. RESULTS: Butyrate treatment initiated 3 weeks before injury and continued 1 week after injury increases histone acetylation and reduces muscle atrophy after nerve crush. Butyrate delivered only after nerve crush similarly prevented muscle atrophy. Butyrate had no effect on the i

An Acute Butyr-Fentanyl Fatality: A Case Report with Postmortem Concentrations.

In this case report, we present an evaluation of the distribution of postmortem concentrations of butyr-fentanyl in a fatality attributed principally to the drug. A man who had a history of intravenous drug abuse was found unresponsive on the bathroom floor of his home. Drug paraphernalia was located on the bathroom counter. Toxicology testing, which initially screened positive for fentanyl by enzyme-linked immunosorbent assay, subsequently confirmed butyr-fentanyl, which was then quantitated by gas chromatography-mass spectrometry-specific ion monitoring (GC-MS SIM) analysis following liquid-

Behavioral and neurochemical effects of sodium butyrate in an animal model of mania.

The present study investigated the effect of the histone deacetylase inhibitor, sodium butyrate (SB), on locomotor behavior and on mitochondrial respiratory-chain complexes activity in the brain of rats subjected to an animal model of mania induced by d-amphetamine (d-AMPH). In the reversal treatment, Wistar rats were first treated with d-AMPH or saline (Sal) for 14 days. Thereafter, between days 8 and 14, rats were administered SB or Sal. In the prevention treatment, rats were treated with SB or Sal for 14 days and received d-AMPH or Sal between days 8 and 14. The d-AMPH treatment increased l

Butyrate enemas improve intestinal anastomotic strength in a rat model.

INTRODUCTION: Anastomotic leakage is a common clinical complication with incidences up to 10% to 17% in colorectal surgery, leading to high morbidity and mortality. Butyrate is the product of colonic fermentation of indigestible carbohydrates and is considered beneficial to gastrointestinal healing. The aim of this study was to investigate the effect of intraluminal supplementation of butyrate on colonic anastomotic strength in a rat model. METHODS: Wistar rats were randomly assigned to one of 3 groups (18 animals each). All rats underwent a 1-cm left colonic resection and end-to-end anastomo

Butyrate modulates oxidative stress in the colonic mucosa of healthy humans.

BACKGROUND & AIMS: Butyrate, a short-chain fatty acid produced by colonic microbial fermentation of undigested carbohydrates, has been implicated in the maintenance of colonic health. This study evaluates whether butyrate plays a role in oxidative stress in the healthy colonic mucosa. METHODS: A randomized, double blind, cross-over study with 16 healthy volunteers was performed. Treatments consisted of daily rectal administration of a 60 ml enema containing 100 mM sodium butyrate or saline for 2 weeks. After each treatment, a blood sample was taken and mucosal biopsies were obtained from the

Topical butyrate for acute radiation proctitis: randomised, crossover trial.

BACKGROUND: No available therapy has, as yet, proven effective to treat acute radiation proctitis (ARP) following radiation therapy for malignant pelvic disease. We assessed whether sodium butyrate enemas, at a dose of 80 mmol/L (80 mL/24 h), might offer effective treatment for this condition. METHODS: 20 patients presenting with ARP after completing a cycle of 35-52 Gy external-beam radiation therapy for pelvic malignant disease, were treated for 3 weeks with topical sodium butyrate and saline enemas according to a randomised, double-blind, crossover protocol. Clinical, endoscopic, and histo

Effect of sodium butyrate on the expression of genes transduced by retroviral vectors.

We have studied the effects of sodium butyrate (NaBu) on the expression of genes transduced by retroviral vectors and stably expressed in two salivary gland-derived cell lines, A5-DAP and A5-BAG, established earlier. These cell lines were obtained by infecting A5 cells with the retroviral vectors DAP and BAG, respectively, and by selecting neomycin-resistant transduced cells. A5-DAP cells express human placental alkaline phosphatase (PLAP) and A5-BAG cells bacterial beta-galactosidase, both under the control of the viral long terminal repeat (LTR) enhancer-promoter. NaBu in the concentration o

In vivo butyrate metabolism and colonic permeability in extensive ulcerative colitis.

BACKGROUND & AIMS: Impaired short-chain fatty acid metabolism by the colonocyte has been suggested as a pathogenic factor in ulcerative colitis (UC). The aim of this study was to measure in vivo butyrate metabolism in UC and to correlate butyrate oxidation with colonic permeability. METHODS: Butyrate oxidation was measured by means of a 14CO2-breath test after rectal instillation of 14C-butyrate. 51Cr-ethylenediaminetetraacetic acid (EDTA) was added to the enema, and the urinary % dose excretion of 51Cr-EDTA after 6 hours was a measure for permeability. RESULTS: Patients with active extensiv

Gamma amino-butyric acid and the control of GnRH secretion in sheep.

The release of GnRH from nerve terminals in the median eminence into the portal vessels is influenced by factors in the internal and external environment of the animal. In the former category are the gonadal steroid hormones oestrogen and progesterone which alter the characteristics of GnRH secretion during the oestrous and seasonal cycles. These cannot exert their actions directly on the GnRH neurones as they do not possess hormone receptors. Therefore, some other steroid-sensitive neuronal system must relay this information to the GnRH neurones. Gamma amino-butyric acid (GABA) neurones are g

Effect of butyrate enemas on the colonic mucosa in distal ulcerative colitis.

Short-chain fatty acid irrigation has been shown to ameliorate inflammation in diversion colitis. In this study the effect of butyrate enemas was tested in 10 patients with distal ulcerative colitis who had been unresponsive to or intolerant of standard therapy for 8 weeks. They were treated for 2 weeks with sodium butyrate (100 mmol/L) and 2 weeks with placebo in random order (single-blind trial). Before and after treatment, clinical symptoms were noted and the degree of inflammation was graded endoscopically and histologically. Rectal proliferation was assessed by autoradiography. After buty

Gas-permeable cellulose acetate butyrate (CAB) contact lenses.

Gas-permeable cellulose acetate butyrate (CAB) contact lenses may often be worn succesfully by aphakic and other patients who are unable to wear hard contact lenses. The comfort characteristics of the CAB lenses are betweeen those of hard and soft contact lenses. They are much more permeable to O2 and CO2 than soft lenses and thus are less apt to cause edema. They are more flexible and more wettable than hard lenses. This study presents 50 patients who, having had to discontinue wearing hard contact lenses because of discomfort, diffuse central corneal edema, or visual problems, were fitted wi

Butyrate-Producing Bacteria in Intestinal Disease Therapy: Potential and Challenges.

Butyrate-producing bacteria have emerged as keystone species whose metabolic activity orchestrates host-microbial homeostasis in the human gut. This review synthesizes current understanding of how these anaerobic Firmicutes, including Faecalibacterium prausnitzii, Roseburia spp., and Eubacterium rectale, function as key contributors to intestinal health through convergent mechanisms: serving as the primary energy source for colonocytes, enforcing mucosal hypoxia that excludes facultative pathogens, and modulating immunity via histone deacetylase inhibition and G-protein-coupled receptor signal

EndD mediates butyrate-dependent toxin release in Clostridioides difficile.

Clostridioides difficile is an urgent threat to human health. Current treatments for C. difficile infections (CDIs) are antibiotics and microbiome restoration therapy (MRT) for recurrent cases. However, antibiotics contribute to antibiotic resistance and recurrent CDIs and the long-term sustainability and accessibility of MRTs remains to be determined. Since a dysbiotic gut microbiome is the primary risk factor for CDI, a better understanding of the interactions between C. difficile, the microbiome, and the host will aid development of treatments with improved precision. Emerging evidence supp

Butyrate improves dextran sulfate sodium-induced enterotoxicity in broilers.

BACKGROUND: Intestinal inflammation has emerged as a significant global concern in poultry farming, particularly due to its strong association with macrophage imbalance. Research has identified macrophages as critical target cells for butyrate, which collaboratively maintains intestinal immune homeostasis. This study delves into the regulatory effects of butyrate on macrophages to ameliorate enteritis in broiler chickens. RESULTS: Our findings indicate that broilers treated with dextran sulfate sodium (DSS) exhibit reduced body weight and compromised intestinal mucosal integrity, characterize

Gut-derived butyrate as a key regulator in the pathogenesis of multiple sclerosis.

Multiple sclerosis (MS) is an autoimmune condition that affects the central nervous system (CNS) and leads to neuroinflammation. The chronic inflammatory cascades of the immune system damage the functions of the nerves and myelin tissue. The heterogeneity of MS symptoms depends on the anatomical location of the lesion within the CNS. The identification of the gut microbiota as a crucial modulator of neuroinflammation via the microbiota-gut-brain axis has been the subject of new studies. Butyrate, one of the well-known short-chain fatty acids (SCFAs) produced by gut bacteria through the ferment

Butyrate: a key mediator of gut-brain communication in Alzheimer's disease.

Alzheimer's disease (AD), a prevalent neurodegenerative disorder, represents a significant global health challenge, characterized by cognitive decline and neuroinflammation. Recent investigations have highlighted the critical role of the gut-brain axis in the pathogenesis of AD, particularly focusing on the influence of short-chain fatty acids (SCFAs), metabolites produced by the gut microbiota through the fermentation of dietary fiber. Among SCFAs, butyrate has emerged as a crucial mediator, positively impacting various pathological processes associated with AD, including epigenetic regulatio

Optimizing butyrate production from methanol and CO2 in microbial electrosynthesis.

Microbial electrosynthesis (MES) enables the conversion of carbon dioxide (CO2) into valuable chemicals utilizing renewable electricity. Acetate is often the main product but supplying a soluble electron donor facilitates upgrading acetate to butyrate via chain elongation. Compared to ethanol as the electron donor, methanol is a promising alternative as its production avoids the competition with food production. However, the optimal operation conditions for maximizing butyrate production rates in methanol assisted MES have not yet been determined. In this study, methanol assisted chain elongat

Intratumor microbiome-derived butyrate promotes lung cancer metastasis.

Most recurrences of lung cancer (LC) occur within 3 years after surgery, but the underlying mechanism remains unclear. Here, we collect LC tissues with shorter (<3 years, recurrence group) and longer (>3 years, non-recurrence group) recurrence-free survival. By using 16S sequencing, we find that intratumor microbiome diversity is lower in the recurrence group and butyrate-producing bacteria are enriched in the recurrence group. The intratumor microbiome signature and circulating microbiome DNA can accurately predict LC recurrence. We prove that intratumor injection of butyrate-producing bacter
