Immunocytochemical localization of salivary peptide P-C in human submandibular gland.

Human saliva contains a proline-rich polypeptide, salivary peptide P-C, which potentiates insulin release and reduces glucagon release from perfused rat pancreas to decrease blood glucose level. To elucidate the process of secretion into humoral fluid of this peptide morphologically, we investigated ultrastructural localization of P-C in human submandibular gland by immunogold technique with anti-peptide P-C whose specificity to P-C was confirmed by immunoblotting. The labeling with gold particles which represents the distribution of P-C-like-immunoreactivity (P-C-LI) was detected in the secre

Expression of C-type natriuretic peptide in the bovine pineal gland.

Recent studies have pointed to membrane-bound guanylyl cyclases (GCs) type A and type B in the rat pineal gland, which augment levels of cyclic GMP (cGMP) in response to atrial natriuretic peptide (ANP), brain-type natriuretic peptide (BNP), and C-type natriuretic peptide (CNP). The present report demonstrates for the first time the expression of CNP in the bovine pineal gland. The CNP prohormone transcript (unlike pre-pro-ANP) was found by reverse transcriptase (RT)-PCR in bovine pineal extracts. CNP immunoreactivity (ir) was revealed in a subpopulation of pinealocytes in situ and in nearly a

Increased urinary C-peptide and albumin excretion in juvenile borderline hypertensives.

The study was designed to evaluate the urinary excretion of C-peptide and albumin, and urinary N-acetyl-beta-D-glucosaminidase (NAG) activity in juvenile borderline hypertensives. The second aim was to examine the relationship between these variables and ambulatory blood pressure level and variability. The study group consisted of 21 non-obese males consecutively chosen from patients with borderline hypertension, defined by sphygmanometer readings, examined in our outpatient clinic. All subjects collected separately their day-time and night-time urines during the period of ambulatory blood pre

Evaluating C-peptide Levels in Children with Type 1 Diabetes and Its Association with Anthropometric and Clinical Variables : C-peptide Levels and Clinical Variables in Pediatric T1DM.

BACKGROUND: C-peptide, a byproduct of insulin production, plays significant physiological roles, including stabilizing blood glucose levels and protecting tissues. Its potential therapeutic role in Type 1 diabetes mellitus (T1DM) management remains underexplored, particularly in preserving β-cell function and improving glycemic control. MATERIALS AND METHODS: This cross-sectional study included 69 pediatric patients with T1DM at Bo Ali Diabetes Clinic, Shiraz, Iran, from December 2022 to April 2023. Patients with a disease duration less than 2 years or metabolic comorbidities were excluded. A

Characterization of an insulin-like growth factor-I/somatomedin-C radioimmunoassay specific for the C-peptide region.

Insulin-like growth factor-I (IGF-I) and somatomedin-C )SM-C) have been shown to be functionally identical by a number of criteria. We have synthesized the 12 amino acid C-peptide region of IGF-I (Gly-Tyr-Gly-Ser-Ser-Ser-Arg-Arg-Ala-Pro-Glu-Thr) and developed a RIA based on antibodies against this synthetic peptide. IGF-I and SM-C were indistinguishable in this RIA. No other peptides competed for this antiserum. The SM-C/IGF-I values of acid-chromatographed serum were strongly age dependent. The mean of children 1-5 yr old was 0.67 +/- 0.033 U/ml (mean +/- SD; n = 23), whereas the mean of chil

C-type natriuretic peptide mediates the hypothalamic actions of the natriuretic peptides to inhibit luteinizing hormone secretion.

Both A- and C-type natriuretic peptides (ANP and CNP, respectively) significantly reduce LH secretion when injected into the third cerebral ventricle of conscious rats. To establish which natriuretic peptide receptor subtype transduces these inhibitory messages, we have employed novel cytotoxin cell targeting techniques to selectively destroy cells in the hypothalamus that respond to ANP or CNP. Rats pretreated with ANP conjugated to the toxic A-chain of the plant cytotoxin ricin failed 1 week later to respond to central injection of ANP with the normal inhibition of LH secretion. These rats d

Screening and rational identification of a novel angiotensin-converting enzyme C-domain inhibitory peptide from Fabaceae food peptide library.

Angiotensin-converting enzyme (ACE), consisting of N-domain and C-domain, is a key regulator of blood pressure. The use of cACE-specific inhibitors helps minimize side effects in clinical applications. Legumes are a good source of proteins containing ACE inhibitory peptides; however, no studies have reported the identification of cACE-specific inhibitory peptides from Fabaceae. In this study, thermal hydrolysates from seeds, sprouts, pods, seedlings, and flowers of legumes were analyzed. Flowers of legumes exhibited a C-domain-preference ACE inhibition and anti-hypertensive effect in rats. Scr

C-Terminal Functionalization Methods of Amino Acids, Peptides, and Proteins.

The functionalization of the C-terminus of amino acids, peptides, and proteins has been extensively studied for more than 150 years and is central to modern peptide synthesis. While most functionalization methods target the N-terminus or reactive residues, such as lysine, cysteine, or tyrosine, the chemo- and regioselective modification of the C-terminus remains relatively underdeveloped. In this review, we discuss strategies for C-terminal activation and functionalization, including classical coupling reagents, enzymatic modifications, photo- and electrochemical decarboxylation, and emerging

The role of C-peptide in diabetes and its complications: an updated review.

Worldwide, diabetes and its complications have seriously affected people's quality of life and become a serious public health problem. C-peptide is not only an indicator of pancreatic β-cell function, but also a biologically active peptide that can bind to cell membrane surface signaling molecules and activate downstream signaling pathways to play antioxidant, anti-apoptotic and inflammatory roles, or regulate cellular transcription through internalization. It is complex how C-peptide is related to diabetic complications. Both deficiencies and overproduction can lead to complications, but thei

Ketoacidosis Onset of Diabetes on a Patient with Normal C-Peptide Value.

Background:Diabetic ketoacidosis is an acute major life-threatening complication of diabetes, characterized by hyperglycemia, ketoacidosis and ketonuria, which can be life threatening if it is not promptly recognized and treated. This occurs mainly in patients with type 1 diabetes, but stressors like trauma and infection can increase the risk of ketoacidosis in other forms of diabetes such as type 2 diabetes. Type 2 diabetes mellitus is a complex metabolic disorder of heterogeneous etiology with behavioral, social, and environmental risk factors that unmask the effects of genetic susceptibilit

The potential of signal peptide peptidase as a therapeutic target for hepatitis C.

Chronic infection with hepatitis C virus (HCV) causes liver steatosis, cirrhosis, metabolic syndrome with inflammation, and eventually leads to hepatocellular carcinoma. HCV core protein is a well-known capsid protein and pathogenic factor related to lipid accumulation, type 2 diabetes mellitus, and carcinogenesis. Cleavage of the C-terminal transmembrane region by signal peptide peptidase (SPP) is required for maturation of the core protein. Areas covered: Herein, this review details the general aspects of the structure, lifecycle, pathogenesis, and maturation of the HCV core protein, the fun

The effect of oral and intravenous dextrose on C-peptide secretion in ponies.

Managing equine hyperinsulinemia is crucial for preventing laminitis, but our understanding of the mechanisms involved in insulin dysregulation in this species is incomplete. C-peptide is co-secreted with insulin but is resistant to hepatic metabolism and can be used to study insulin dysregulation. This study examined C-peptide secretion in serial blood samples collected after oral and i.v. dextrose (0.75 g/kg) administration to 9 ponies (BCS, 7.1 ± 0.5). The ponies were designated as hyperinsulinemic (HI) or normoinsulinemic (NI) responders before the study, using oral glucose tests and faste

The effect of C-peptide on diabetic nephropathy: A review of molecular mechanisms.

C-peptide is a small peptide connecting two chains of proinsulin molecule and is dissociated before the release of insulin. It is secreted in an equimolar amount to insulin from the pancreatic beta-cells into the circulation. Recent evidence demonstrates that it has other physiologic activities beyond its structural function. C-peptide modulates intracellular signaling pathways in various pathophysiologic states and, could potentially be a new therapeutic target for different disorders including diabetic complications. There is growing evidence that c-peptide has modulatory effects on the mole

Synthetic anti-lipopolysaccharide peptides and hepatitis C virus infection.

INTRODUCTION: Hepatitis C virus (HCV) infection is a leading cause of cirrhosis and hepatocellular carcinoma. Although antiviral therapy has been markedly improved by the licensing of direct-acting antivirals, safety, resistance, high costs and difficult-to-treat patients remain important challenges. AREAS COVERED: This article focuses and comments on the recent development of synthetic anti-lipopolysaccharide peptides (SALPs) which bind to highly sulfated glycosaminoglycan/heparan sulfate (HS) on cell surface. HS serves as a primary docking site for several viruses to their respective host c

Non-hydrolytic functions of acetylcholinesterase. The significance of C-terminal peptides.

This review explores the possibility that acetylcholinesterase may play a pivotal, non-hydrolytic role in neurodegeneration. More specifically, C-terminal sequences of acetylcholinesterase may act as signalling molecules in key brain regions characteristically vulnerable to Alzheimer's, Parkinson's and motor neuron disease.

Foramen magnum stenosis and midface hypoplasia in C-type natriuretic peptide-deficient rats and restoration by the administration of human C-type natriuretic peptide with 53 amino acids.

C-type natriuretic peptide (CNP)-knockout (KO) rats exhibit impaired skeletal growth, with long bones shorter than those in wild-type (WT) rats. This study compared craniofacial morphology in the CNP-KO rat with that in the Spontaneous Dwarf Rat (SDR), a growth hormone (GH)-deficient model. The effects of subcutaneous administration of human CNP with 53 amino acids (CNP-53) from 5 weeks of age for 4 weeks on craniofacial morphology in CNP-KO rats were also investigated. Skulls of CNP-KO rats at 9 weeks of age were longitudinally shorter and the foramen magnum was smaller than WT rats. There we

Teneurin C-terminal associated peptide-1 attenuates chronic stress in male rats.

Hyperactivity of the hypothalamic-pituitary-adrenal axis core stress response system can lead to anxiety, chronic stress, and other neuropsychiatric disorders, such as major depressive disorder and posttraumatic stress disorder. Mitigating this response requires novel interventions to overcome the delayed efficacy and nonspecificity that characterize the currently available, suboptimal chronic stress treatments. Teneurin C-terminal Associated Peptide (TCAP)-1, a naturally occurring neuropeptide, is an established regulator of corticotropin-releasing factor (CRF) mediated stress responses. Alth
