Clinical significance of CD146 and latexin during different stages of thyroid cancer.

Molecular mechanisms underlying thyroid tumorigenesis and identifying new therapeutic targets are still under investigation. We aim to investigate the role of CD146 and latexin (Lxn) and examine whether they have any clinical significance in thyroid cancer. Human thyroid papillary (PTC), follicular (FTC), anaplastic (ATC) cancer cells, and other control cells were used in this study. Western blot, cell proliferation, invasion assay, and shRNA were applied to study the expression levels and functional significances of CD146 and Lxn in thyroid cells. The protein expression was evaluated by immun

Cell adhesion molecules in the development and progression of malignant melanoma.

Cell adhesion molecules belonging to the integrin, cadherin and immunoglobulin superfamilies have been implicated in tumor progression in cutaneous melanoma. Expression of the alpha v beta 3 integrin first appears with the change from radial to vertical growth, a step which is associated with the development of metastatic potential. VLA-4 expression is characteristic of advanced primary tumors and may mediate interaction of the tumor cells with VCAM-1 on vascular endothelium. Expression of these integrins is a marker of poor prognosis in patients and can confer invasive (alpha v beta 3) and me

Neurite extension of DRG neurons by gicerin expression is enhanced by nerve growth factor.

Gicerin, a cell adhesion molecule, is expressed in dorsal root ganglion (DRG) and sciatic nerves during chick development. This molecule re-appears in these tissues after an injury to the sciatic nerve. In the present study, we investigated the expression of nerve growth factor (NGF) in the regenerating sciatic nerve of chicks and the effects of NGF on the expression and neurite activities of gicerin in DRG. In the sciatic nerve after a crush injury, the expression of NGF and gicerin increased in the Schwann cells and in the nerve fibers, respectively. NGF promoted the neurite projections from

Influence of age on stem cells depends on the sex of the bone marrow donor.

Ageing is often accompanied by an increase in bone marrow fat together with reduced bone volume and diseases of the bone such as osteoporosis. As mesenchymal stem cells (MSCs) are capable of forming bone, cartilage and fat tissue, studying these cells is of great importance to understand the underlying mechanisms behind age-related bone diseases. However, inter-donor variation has been found when handling MSCs. Therefore, the aim of this study was to investigate the effects of donor age and sex by comparing in vitro characteristics of human bone marrow-derived MSCs (hBMSCs) from a large donor

Hyperglycemia in obese type 2 diabetes mellitus rats affects pulp wound healing.

OBJECTIVE: Hyperglycemia in individuals with type 2 diabetes mellitus is associated with delayed wound healing, although the underlying mechanism remains unclear. Macrophages play a crucial role in the initiation, maintenance, and resolution of inflammation by polarizing into proinflammatory and anti-inflammatory subtypes. Hyperglycemia may interfere with odontoblast-like cell differentiation and cause persistent proinflammatory polarization in individuals with type 2 diabetes mellitus. METHODS: Eight-week-old spontaneously diabetic Torii fatty rats (model rats for type 2 diabetes mellitus) a

Measurement of telomere length in cells from pleural effusion: Asbestos exposure causes telomere shortening in pleural mesothelial cells.

We estimated the telomere lengths of neoplastic and non-neoplastic mesothelial cells and examined their correlation with asbestos exposure and the expression of markers of mesothelial malignancy. Cell blocks of pleural effusion obtained from 35 cases of non-neoplastic disease (NN), 12 cases of malignant mesothelioma (MM) and 12 cases of carcinomatous effusion due to lung adenocarcinoma (LA) were examined. Fifteen of the 35 NN cases had pleural plaques (NNpp+) suggestive of asbestos exposure, and the other 20 cases had no pleural plaques (NNpp-). Telomere length was measured using the tissue qu

CD146+ CAFs mediate immunosuppression in gastric cancer via COL4A1: potential therapeutic targets.

BACKGROUND: The immunomodulatory roles of cancer-associated fibroblasts (CAFs) in gastric cancer have been increasingly recognized. This study aimed to identify specific CAF subpopulations and to elucidate the mechanisms underlying their immunosuppressive effects. METHODS: We developed a single-cell RNA sequencing (scRNA-seq) protocol to enrich CAFs and matched normal fibroblasts (NFs) from stomach tissues. Bioinformatics analyses, along with in vitro and in vivo experiments, were performed to functionally validate a distinct CAF subset. RESULTS: We profiled 8,322 high-quality fibroblasts fr

Characterizing the Long Non-Coding RNA Profile of Endometrial Mesenchymal Stem/Stromal Cell-Derived Extracellular Vesicles and Their Anti-Inflammatory Role in Osteoarthritis.

Endometrial tissue-derived mesenchymal stem/stromal cells (eMSCs) have potential therapeutic properties partially exerted via their secreted extracellular vesicles (EVs). eMSC-EVs contain cargos with regenerative and immunomodulatory properties. Specifically, the miRNA profile of CD146High eMSC-EVs has been shown to promote anti-inflammatory M2 macrophage polarization in vitro. Herein, we aimed to characterize the lncRNA profile of CD146High and CD146Low eMSC-EVs and further assess their immunomodulatory and anabolic therapeutic function in osteoarthritis (OA). We hypothesized that the CD146Hi

An immune sandwich electrochemical biosensor based on triple-modified zirconium derivatives for detection of CD146 in serum.

CD146, also known as melanoma cell adhesion molecule (MCAM), is overexpressed in various cancer patients, making it a valuable predictor for early diagnosis. In this work, an immune sandwich electrochemical biosensor is proposed for sensitive and non-invasive quantitative detection of CD146 in serum. Zirconium-based MOF (UIO-66) was modified by simultaneous copper atom doping, in situ growth carbon-based support and physical embedding of platinum nanoparticles (PtNPs). Triple-modified Cu-UIO-66@SWCNT/PtNPs nanocomposites with high stability and excellent electrochemical properties, serve as su

Microcirculation surrounding end-stage human chronic skin wounds is associated with endoglin/CD146/ALK-1 expression, endothelial cell proliferation and an absence of p16Ink4a.

Angiogenesis is an essential part of normal skin healing, re-establishing blood flow in developing granulation tissue. Non-healing skin wounds are associated with impaired angiogenesis and although the role of re-establishing macroscopic blood flow to limbs to prevent wound chronicity is well investigated, less is known about vascular alterations at the microcirculatory level. We hypothesised that significant phenotypic changes would be evident in blood vessels surrounding chronic skin wounds. Wound edge tissue, proximal to wound (2 cm from wound edge) and non-involved skin (>10 cm from wound

Soluble CD146 reflects altered endothelial and metabolic homeostasis in peripheral artery disease.

BACKGROUND: Peripheral Arterial Disease (PAD) is a prevalent but underdiagnosed pathology. Soluble CD146 (sCD146) was described as a marker of endothelial dysfunction and vascular congestion. OBJECTIVE: We hypothesize that sCD146 may represent a novel biomarker of PAD. Our objective was to evaluate the association between plasma sCD146 levels and the occurrence and severity of PAD. METHODS: In this case-control study, 184 Caucasian men with symptomatic PAD were compared to 163 age-matched healthy control patients. PAD diagnosis was confirmed using ankle-brachial index (ABI) and imaging. Plas

CD146: a promising target in respiratory diseases.

Respiratory diseases are major causes of chronic disorders and death worldwide, involving inflammatory, tumoral or infectious processes. It has been proven that vascular mechanisms are key contributors to the pathogenesis of these diseases. For that purpose, it is essential to describe and validate new biomarkers and/or therapeutic targets responsible for lung vascularisation and/or angiogenesis. CD146 is an endothelial cell adhesion molecule also expressed on mesenchymal stem cells, epithelial cells and T-helper 17 lymphocytes. A soluble form of CD146 exists, sCD146, which can be detected in

Comparison of different culture conditions for smooth muscle cell differentiation of human umbilical cord vein CD146+ perivascular cells.

Pericytes are CD146+ perivascular cells (PCs) that have multipotential differentiation capacity as mesenchymal stem cells. Beside their crucial roles in vascular development and blood flow regulation, they have ability to differentiate into vascular cell types in vivo. These properties make pericytes preferred cells in the field of vascular tissue engineering. Culture medium for in vitro differentiation of pericytes to vascular smooth muscle cells (SMCs) has not been defined yet. The aim of this study is to try different culture media for SMC differentiation of CD146+ PCs. For this purpose, CD

Specific lipid magnetic sphere sorted CD146-positive bone marrow mesenchymal stem cells can better promote articular cartilage damage repair.

BACKGROUND: The characteristics and therapeutic potential of subtypes of bone marrow mesenchymal stem cells (BMSCs) are largely unknown. Also, the application of subpopulations of BMSCs in cartilage regeneration remains poorly characterized. The aim of this study was to explore the regenerative capacity of CD146-positive subpopulations of BMSCs for repairing cartilage defects. METHODS: CD146-positive BMSCs (CD146 + BMSCs) were sorted by self-developed CD146-specific lipid magnetic spheres (CD146-LMS). Cell surface markers, viability, and proliferation were evaluated in vitro. CD146 + BMSCs we

CD146 + interstitial cells contribute to the dystrophic skeletal muscle phenotype in vitro.

CD146 + interstitial cells are myogenic progenitors which contribute to skeletal muscle regeneration. However, their role in Duchenne muscular dystrophy (DMD) remains unclear. We compared CD146 + cells from wild-type and mdx mice, revealing distinct molecular and functional differences. Mdx CD146 + differentiated less efficiently into myoblasts and multinucleated myotubes but more efficiently into fibroblasts and adipocytes in vitro. Conditioned medium from mdx CD146 + cells impaired endothelial cell differentiation, through dysregulation of Sdf-1 and Angpt2 secretion. Transcriptomic analyses

Galectin-3/CD146 interaction promotes renal damage and systemic inflammation after acute kidney injury.

Galectin-3 (Gal-3) expression is abnormally increased after renal ischemic injury, in various studies, further promoting tissue damage. Gal-3 may interact with the endothelial layer to induce inflammation during renal injury, linking the systemic and tissue compartments. However, this mechanism has not been investigated after acute kidney injury (AKI). To this end, wild-type (WT) and Gal-3-/- mice underwent renal ischemia followed by reperfusion (rIR). Bone marrow transplantation was used to differentiate Gal-3 expression in renal tissue from systemic bone marrow cells. WT mice were injected w
