Human testis-specific TTRP regulates circadian clock as an evolutionarily young natural antisense transcript of PER2.

The circadian clock orchestrates a variety of physiological and behavioral processes, endowing the ability to adapt to the cycling environment. Here, we report the identification of TTRP, a TDP-43 pseudogene that emerged in higher Hominoidea species ~8.6 Mya. TTRP is a human testis-specific gene that overlaps with the clock gene PER2 and interferes with PER2 transcription. TTRP encodes a protein that interacts with the clock proteins PER2, BMAL1, and CRY2, and promotes PER2 and CRY1 turnover. The expression of gorilla and bonobo TTRP proteins changes the period and amplitude of circadian rhyth

Effect of monochromatic light on the circadian clock of cultured chick retinal tissue.

The central biological clock system of bird is formed by hypothalamus suprachiasmatic nucleus, pineal gland and retina thereby interacting with each other in a neuroendocrine loop. Previous results have confirmed that monochromatic light can influence the clock genes in the pineal gland, hypothalamus and retina of chicks in vivo. The present work was conducted to study whether the cultured retinal tissue of chick could maintain the circadian oscillation and whether the monochromatic light affect the expression level of cultured retinal circadian clock in vitro. Retinal tissues of 0-day-old chi

Melatonin refines ovarian mitochondrial dysfunction in PCOS by regulating the circadian rhythm gene Clock.

Mitochondrial dysfunction is present in the ovaries of patients with polycystic ovary syndrome (PCOS). Melatonin (MT) has shown promise in treating PCOS by improving mitochondrial dysfunction, though the underlying mechanisms remain unclear. In this study, we first assessed the levels of proteins associated with mitochondrial autophagy and dynamics in ovary granulosa cells (GCs) of PCOS patients and in the ovaries of DHEA-induced PCOS mice. We found abnormal expression of these proteins, indicating the presence of mitochondrial dysfunction in PCOS ovaries. Notably, the expression of the circad

Hypoxia up-regulated 1 (HYOU1) is involved in hepatic steatosis via N6-methyladenosine (m6A)-dependent stabilization and modulation of circadian clock genes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is characterized by profound disturbances in hepatic lipid metabolism. However, the role of N6-methyladenosine (m6A)-mediated dysregulation of specific stress-responsive proteins in governing metabolically critical gene expression, particularly the clock system, in this context remains largely unexplored. Notably, there is no prior experimental evidence linking Hypoxia Up-Regulated 1 (HYOU1) to MASLD pathology. Combining bioinformatics with validation, we identify HYOU1 as aberrantly upregulated in MASLD. Elevated HYOU1 promotes

Circadian clock and output genes are rhythmically expressed in extratesticular ducts and accessory organs of mice.

Circadian clocks regulate multiple rhythms in mammalian tissues. In most organs core clock gene expression is oscillatory, with negative components Per and Cry peaking in antiphase to Bmal1. A notable exception is the testis, where clock genes seem nonrhythmic. Earlier mammalian studies, however, did not examine clock expression patterns in accessory ductal tissue required for sperm maturation and transport. Previous studies in insects demonstrated control of sperm maturation in vas deferens by a local circadian system. Sperm ducts express clock genes and display circadian pH changes controlle

Acute hypoxia induced dysregulation of clock-controlled ovary functions.

High altitudes or exposure to hypoxia leads to female reproductive disorders. Circadian clocks are intrinsic time-tracking systems that enable organisms to adapt to the Earth's 24-h light/dark cycle, which can be entrained by other environmental stimuli to regulate physiological and pathological responses. In this study, we focused on whether ovarian circadian clock proteins were involved in regulating female reproductive dysfunction under hypoxic conditions. Hypobaric hypoxia was found to induce a significantly prolonged estrous cycle in female mice, accompanied by follicular atresia, pituita

HSP90 affects the stability of BMAL1 and circadian gene expression.

The mammalian circadian clock comprises a system of interconnected transcriptional and translational feedback loops. Proper oscillator function requires the precisely timed synthesis and degradation of core clock proteins. Heat shock protein 90 (HSP90), an adenosine triphosphate (ATP)-dependent molecular chaperone, has important functions in many cellular regulatory pathways by controlling the activity and stability of its various client proteins. Despite accumulating evidence for interplay between the heat shock response and the circadian system, the role of HSP90 in the mammalian core clock

The molecular circadian clock of eosinophils: a potential therapeutic target for asthma.

Asthma is a chronic inflammatory airway disease exhibiting time-of-day variability in symptoms and severity. Eosinophils, pivotal players and biomarkers in asthma, are regulated by the molecular circadian clock. This study aimed to investigate the impact of the molecular circadian clock on eosinophil effector function and its potential as a diagnostic biomarker and therapeutic target. We monitored clock proteins by flow cytometry in peripheral blood eosinophils from participants with mild asthma over a 24-h period. The observed decreased protein levels were confirmed in a cohort of patients wi

Circadian regulators of intestinal lipid absorption.

Among all the metabolites present in the plasma, lipids, mainly triacylglycerol and diacylglycerol, show extensive circadian rhythms. These lipids are transported in the plasma as part of lipoproteins. Lipoproteins are synthesized primarily in the liver and intestine and their production exhibits circadian rhythmicity. Studies have shown that various proteins involved in lipid absorption and lipoprotein biosynthesis show circadian expression. Further, intestinal epithelial cells express circadian clock genes and these genes might control circadian expression of different proteins involved in i

Necdin regulates BMAL1 stability and circadian clock through SGT1-HSP90 chaperone machinery.

Circadian clocks are endogenous oscillators that control ∼24-hour physiology and behaviors in virtually all organisms. The circadian oscillator comprises interconnected transcriptional and translational feedback loops, but also requires finely coordinated protein homeostasis including protein degradation and maturation. However, the mechanisms underlying the mammalian clock protein maturation is largely unknown. In this study, we demonstrate that necdin, one of the Prader-Willi syndrome (PWS)-causative genes, is highly expressed in the suprachiasmatic nuclei (SCN), the pacemaker of circadian c

Phase-resetting mechanism of the circadian clock in Chlamydomonas reinhardtii.

Although the circadian clock is a self-sustaining oscillator having a periodicity of nearly 1 d, its period length is not necessarily 24 h. Therefore, daily adjustment of the clock (i.e., resetting) is an essential mechanism for the circadian clock to adapt to daily environmental changes. One of the major cues for this resetting mechanism is light. In the unicellular green alga Chlamydomonas reinhardtii, the circadian clock is reset by blue/green and red light. However, the underlying molecular mechanisms remain largely unknown. In this study, using clock protein-luciferase fusion reporters, w

Role of circadian clock in oral squamous cell carcinoma pathogenesis and treatment.

The circadian clock is an endogenous timing mechanism that enables cellular physiological activities to adapt to environmental changes. It is primarily composed of circadian clock genes and their encoded proteins, which form a transcription-translation negative feedback loop, generating circadian rhythmic oscillations that regulate cellular function. Circadian clock genes interact with multiple molecular pathways that regulate physiological processes such as the cell cycle and metabolism; therefore, the circadian clock plays a vital role in maintaining cellular homeostasis. Studies have shown

Casein kinase 1δ activity: a key element in the zebrafish circadian timing system.

Zebrafish have become a popular model for studies of the circadian timing mechanism. Taking advantage of its rapid development of a functional circadian clock and the availability of light-entrainable clock-containing cell lines, much knowledge has been gained about the circadian clock system in this species. However, the post-translational modifications of clock proteins, and in particular the phosphorylation of PER proteins by Casein kinase I delta and epsilon (CK1δ and CK1ε), have so far not been examined in the zebrafish. Using pharmacological inhibitors for CK1δ and CK1ε, a pan-CK1δ/ε inh

Circadian regulator BMAL1::CLOCK promotes cell proliferation in hepatocellular carcinoma by controlling apoptosis and cell cycle.

Hepatocellular carcinoma (HCC) remains a global health challenge whose incidence is growing worldwide. Previous evidence strongly supported the notion that the circadian clock controls physiological homeostasis of the liver and plays a key role in hepatocarcinogenesis. Despite the progress, cellular and molecular mechanisms underpinning this HCC-clock crosstalk remain unknown. Addressing this knowledge gap, we show here that although the human HCC cells Hep3B, HepG2, and Huh7 displayed variations in circadian rhythm profiles, all cells relied on the master circadian clock transcription factors

A clockwork organ.

The vertebrate circadian clock was thought to be highly localized to specific anatomical structures: the mammalian suprachiasmatic nucleus (SCN), and the retina and pineal gland in lower vertebrates. However, recent findings in the zebrafish, rat and in cultured cells have suggested that the vertebrate circadian timing system may in fact be highly distributed, with most if not all cells containing a clock. Our understanding of the clock mechanism has progressed extensively through the use of mutant screening and forward genetic approaches. The first vertebrate clock gene was identified only a
