A novel functional screening method for generation of a synthetic microbial community: Case study with control of Fusarium wilt in pigeonpea.

Conventional methods to combat phytopathogens have ecological implications: chemical fertilizers pollute the environment, while bioinoculants are often inconsistent under field conditions. Microbiome-assisted rhizosphere engineering aims to re-structure the rhizosphere microbiome to promote plant growth and/or mitigate stress. This study employs a strategy based on rhizosphere engineering to combat stress caused by Fusarium udum in Cajanus cajan, by generating synthetic microbial communities (SMCs). We used a culture bank of indigenous bacterial strains belonging to the family Bacillaceae, iso

Cajanolactone A alleviates high-fat diet-induced MAFLD by modulating liver de novo lipogenesis, inflammatory signaling, and bile acid composition in the gut-liver axis.

Metabolic dysfunction-associated fatty liver disease (MAFLD) is a common chronic liver disease worldwide, affecting more than a quarter of the adult population. Cajanolactone A (CLA), a stilbenoid derived from Cajanus cajan, has been shown to significantly reduce hepatic lipid accumulation. However, its molecular mechanisms in MAFLD remain unclear. In this high-fat diet (HFD)-induced mice model of MAFLD, CLA significantly improved dyslipidemia, suppressed liver de novo lipogenesis by downregulating the expression of core genes and proteins involved in glycolysis and the tricarboxylic acid (TCA

Protection of acetaminophen induced mitochondrial dysfunctions and hepatic necrosis via Akt-NF-kappaB pathway: role of a novel plant protein.

Oxidative stress is a major cause of drug induced hepatic diseases. The present study aims to investigate the antioxidative signaling mechanism of a protein isolated from the herb, Cajanus indicus against acetaminophen induced necrotic cell death. We found that incubation of hepatocytes with the protein prevented acetaminophen-induced loss in cell viability, reduction in glutathione level and enhancement of reactive oxygen species generation. Treatment of mice with the protein before administration of acetaminophen also reduced serum nitrite and TNF-alpha formation. Moreover, it counteracted a

Effect of detergents and hexafluoroisopropanol on the conformation of a non-helical and a helical plant protease inhibitor.

We have earlier reported the purification of a non-helical proteinase inhibitor from Cajanus cajan and a helical proteinase/amylase inhibitor from Phaseolus aureus. The effect of detergents, viz. sodium dodecyl sulfate (SDS), sodium deoxycholate (DOC) and 3-[(3-cholamidopropy) dimethylammonio]-1-propane sulfonate (CHAPS) and hexafluoroisopropanol on the conformation of these proteinaceous inhibitors was investigated using circular dichroism spectroscopy. The present report focuses on changes in the polypeptide backbone conformation with respect to induction of helical structure. SDS causes min

Cajaninstilbene acid in macrophage immunometabolism and angiogenesis: Mechanisms and therapeutic potential.

Macrophage immunometabolism is an important regulator of inflammatory resolution and angiogenic responses. Aberrant macrophage polarization, accompanied by metabolic disturbances such as enhanced glycolysis and mitochondrial dysfunction, has been implicated in impaired angiogenesis in chronic inflammatory conditions. Cajaninstilbene acid (CSA), a natural stilbene derived from Cajanus cajan, exhibits anti-inflammatory, antioxidant, and metabolic regulatory activities. In non-diabetic inflammatory settings, including liver injury, ischemia-reperfusion injury, and inflammatory bowel disease, CSA

Transmission of antibiotic resistance through organic amendments in arable land: A 3-year field study with pigeonpea-wheat cropping system.

The worldwide emergence of antimicrobial resistance (AMR) poses a substantial risk to human health and environmental stability. In agriculture, organic amendments (derived from organic sources such as manure, and plant residues) are beneficial in restoring soil properties and providing essential nutrients to crops but raise concerns about harboring antibiotic resistance, which emphasizes the need for vigilant monitoring and strategic interventions in their application. The current study assessed the impact of farming practices (organic and conventional) in a three-year field experiment with pi

Activity of the antiestrogenic cajanin stilbene acid towards breast cancer.

Antiestrogenic therapy is a mainstay for estrogen receptor (ERα)-positive breast cancer. Due to the development of resistance to established antihormones such as tamoxifen, novel compounds are required. The low abundant cajanin stilbene acid (CSA) recently isolated by us from Pigeon Pea (Cajanus cajan) has structural similarities with estrogen. We analyzed the cytotoxic and anticancer activity of CSA in ERα-positive and -negative human breast cancer cells in vitro, in vivo and in silico. CSA exerts anticancer and antiestrogenic activities towards ERα-positive breast cancer, and it showed cytot

Antidepressant-like effects of cajaninstilbene acid and its related mechanisms in mice.

Cajaninstilbene acid (CSA), a bioactive constituent isolated from pigeon pea leaves, exhibited neuroprotective activities in previous studies. The present study aims to evaluate the antidepressant effects of CSA by using behavioral despair models of tail suspension test (TST) and forced swimming test (FST), and a chronic unpredictable mild stress (CUMS) model. CSA (30 or 60 mg/kg), intragastrically administrated for 7 days, could significantly reduce the immobility time of mice in TST and FST. CSA treatment (15 or 30 mg/kg) significantly reversed the depressive-like behavioral changes of mice

Waterlogging tolerance in pigeonpea: current understanding of physiological responses, genetic resources, and genomic prospects.

Waterlogging is an escalating threat to pigeonpea (Cajanus cajan L. Millsp.) production in tropical and subtropical regions, where increasingly erratic monsoon rainfall and poorly drained Vertisols frequently expose crops to root-zone hypoxia during germination and early vegetative growth. Waterlogging in pigeonpea has historically been associated with substantial production losses in affected areas; however, the magnitude of these losses depends on stress duration, developmental stage, genotype, environmental conditions, and post-stress recovery. Oxygen deprivation disrupts root respiration,

Cajaninstilbene Acid targets TNF-α-mediated mitochondrial translocation to restore osteogenic-osteoclastic coupling equilibrium and ameliorate estrogen-deficient osteoporosis.

BACKGROUND: Oestrogen-deficient osteoporosis features an imbalanced bone formation-resorption coupling with excessive osteoclast activation and impaired osteoblast function. Compounds that bidirectionally regulate bone metabolism remain rare among natural small molecules. Cajaninstilbene acid (CSA) from Cajanus cajan leaves exerts regulatory effects on bone metabolism and anti-inflammatory activity, but its mechanism in restoring bone homeostasis via intercellular mitochondrial transfer remains unclear. PURPOSE: To investigate the therapeutic effect of CSA on oestrogen-deficient osteoporosis
