Hydralazine potentiates the antiproliferative effect of photodynamic therapy on Candida albicans and Candida parapsilosis in vitro.

The incidence of drug-resistant Candida infections is increasing globally, emphasizing the need for novel antifungal strategies. Photodynamic therapy (PDT), which generates reactive oxygen species through appropriate light-activated photosensitizers, represents a promising alternative for superficial infections. This study investigated the effect of the epigenetic drugs sodium butyrate (NaBut) and hydralazine (HLZ) combined with PDT on the growth of Candida spp. The minimum inhibitory concentrations (MICs) for NaBut, HLZ and PDT alone were assessed against C. albicans, C. parapsilosis, C. glab

Catechin Augments the Antifungal Efficacy of Fluconazole Against Candida parapsilosis.

The rising global incidence of Candida parapsilosis infections is increasingly complicated by antifungal resistance, resulting in frequent therapeutic failure. This study investigated the potential of the natural compound catechin to enhance the efficacy of fluconazole through synergistic interaction. We evaluated the susceptibility of C. parapsilosis clinical isolates and a reference strain to combinations of catechin and fluconazole using standardized microbiological assays and molecular techniques. In vivo efficacy was assessed using the Galleria mellonella infection model. Mechanistic stud

Small pangenome of Candida parapsilosis reflects overall low intraspecific diversity.

Candida parapsilosis is an opportunistic yeast pathogen that can cause life-threatening infections in immunocompromised humans. Whole-genome sequencing studies of the species have demonstrated remarkably low diversity, with strains typically differing by about 1.5 single nucleotide polymorphisms (SNPs) per 10 kb. However, SNP calling alone does not capture the full extent of genetic variation. Here, we define the pangenome of 372 C. parapsilosis isolates to determine variation in gene content. The pangenome consists of 5,859 genes, of which 48 are not found in the genome of the reference strai

Hospitalizations involving Candida parapsilosis bloodstream infection, United States, 2016-2024.

Candidemia is a common healthcare-associated infection associated with substantial morbidity and mortality. Candida parapsilosis is among the most common causative species; it has a notable ability to form biofilms and colonize skin, facilitating patient-to-patient spread, and it is increasingly associated with fluconazole resistance. We aimed to describe temporal trends, clinical characteristics, and antifungal resistance patterns among C. parapsilosis candidemia hospitalizations in the United States during 2016-2024. We used the Premier Healthcare Database (which represents >500 hospitals co

Structural characterization and inhibition of carbonic anhydrase from Candida parapsilosis.

Fungal carbonic anhydrases (CAs) are metalloenzymes that catalyze the reversible hydration of carbon dioxide and play an essential role in fungal adaptation to environments with fluctuating CO2 concentrations. The opportunistic pathogen Candida parapsilosis expresses a single β-class CA, CpNce103p, which is structurally distinct from human α-class CAs and therefore may represent a promising antifungal target. Here, we report the biochemical and structural characterization of CpNce103p, including its crystal structure in complex with the classical CA inhibitor acetazolamide. Mass photometry and

Clinical impact of fluconazole-resistant Candida parapsilosis: a narrative review.

Fluconazole resistance in Candida parapsilosis is an increasing global concern, with resistance rates varying widely and reaching up to 80% in some regions. This trend has led to hospital outbreaks, primarily driven by mutations in the ERG11 gene, especially the Y132F substitution. The clinical relevance of fluconazole resistance remains controversial, as studies have yielded conflicting results regarding its impact on mortality. While some studies described an increased mortality associated with resistant strains, others reported no significant difference. Treatment options are limited: echin

Candida parapsilosis keratitis treated successfully with topical and oral fluconazole.

A 73-year-old male patient presented with ocular pain, redness, and blurred vision in the left eye, which had been ongoing for more than 2 months. An oval-shaped paracentral corneal ulcer with stromal infiltration and a mild anterior chamber reaction were found. Despite treatment with empiric antibiotics, the lesion progressed and corneal thinning in the middle area was noted. The culture yielded Candida parapsilosis. We therefore prescribed topical 0.2% fluconazole (FCZ) in combination with oral FCZ as an antifungal treatment, following which the stromal infiltration gradually subsided. Compl

Intravital Imaging Reveals Divergent Cytokine and Cellular Immune Responses to Candida albicans and Candida parapsilosis.

Candida yeasts are common commensals that can cause mucosal disease and life-threatening systemic infections. While many of the components required for defense against Candida albicans infection are well established, questions remain about how various host cells at mucosal sites assess threats and coordinate defenses to prevent normally commensal organisms from becoming pathogenic. Using two Candida species, C. albicans and C. parapsilosis, which differ in their abilities to damage epithelial tissues, we used traditional methods (pathogen CFU, host survival, and host cytokine expression) combi

Evaluation of the IR Biotyper for Candida parapsilosis typing in hospital surveillance.

The IR Biotyper (IR-BT) was evaluated for its performance in typing Candida parapsilosis strains, in comparison to a short tandem repeat analysis. Although the IR-BT offers advantages such as speed and ease of use for routine applications, it demonstrated lower discriminatory power and concordance with our short tandem repeat analysis. These results suggest that IR-BT needs further optimization for the characterization of C parapsilosis strains.

[Investigation of ERG11 and MRR1 Resistance Genes in Candida parapsilosis Isolates].

Candida parapsilosis is an important fungal pathogen that is particularly isolated from blood cultures. In recent years, although there are regional differences between centres, resistance to fluconazole which is the most commonly preferred drug for candidemia, has increased in many regions. This increase complicates infection control and adversely affects treatment. In C.parapsilosis, codon 132 Y/F change in ERG11 is accepted as the dominant mechanism and it is reported that some amino acid changes in MRR1 may also contribute to resistance. In this study, we aimed to elucidate the azole resis

The transcription factor Ndt80 is a repressor of Candida parapsilosis virulence attributes.

Candida parapsilosis is an emergent opportunistic yeast among hospital settings that affects mainly neonates and immunocompromised patients. Its most remarkable virulence traits are the ability to adhere to prosthetic materials, as well as the formation of biofilm on abiotic surfaces. The Ndt80 transcription factor was identified as one of the regulators of biofilm formation by C. parapsilosis; however, its function in this process was not yet clarified. By knocking out NDT80 (CPAR2-213640) gene, or even just one single copy of the gene, we observed substantial alterations of virulence attribu

Aneuploidy Underlies Tolerance and Cross-Tolerance to Drugs in Candida parapsilosis.

Candida species are the most common human fungal pathogens worldwide. Although C. albicans remains the predominant cause of candidiasis, infections caused by non-albicans Candida species, including C. parapsilosis, are increasing. In C. albicans, genome plasticity has been shown to be a prevalent strategy of adaptation to stresses. However, the role of aneuploidy in C. parapsilosis is largely unknown. In this study, we found that six different aneuploid karyotypes conferred adaptation to the endoplasmic reticulum stress inducer tunicamycin (TUN) in C. parapsilosis. Interestingly, a specific an

Candida parapsilosis meningitis associated with shunt infection in an adult male.

Candida parapsilosis is a very rare cause of meningitis. Though several cases have now been reported in neonates and children, only one has been described in an adult. We report on a 55-year-old male that was admitted due to altered mental status. He had recent sinus drainage and polypectomy, craniotomy with drainage of brain abscess, and ventriculo-peritoneal shunt placement. On admission, imaging studies showed no evidence of shunt dysfunction but did reveal extensive white matter decreased attenuation. Microscopic examination of the first 10 daily cerebrospinal fluid (CSF) cultures revealed

Insight Into the Properties and Immunoregulatory Effect of Extracellular Vesicles Produced by Candida glabrata, Candida parapsilosis, and Candida tropicalis Biofilms.

Currently, non-albicans Candida species, including C. tropicalis, C. glabrata, and C. parapsilosis, are becoming an increasing epidemiological threat, predominantly due to the distinct collection of virulence mechanisms, as well as emerging resistance to antifungal drugs typically used in the treatment of candidiasis. They can produce biofilms that release extracellular vesicles (EVs), which are nanometric spherical structures surrounded by a lipid bilayer, transporting diversified biologically active cargo, that may be involved in intercellular communication, biofilm matrix production, and in

Synergistic effect of nikkomycin Z with caspofungin and micafungin against Candida albicans and Candida parapsilosis biofilms.

Antifungal lock therapy has received significant interest in the last few years because the frequently usage of intravascular devices is associated with an increasing number of catheter-related bloodstream infections caused by Candida species. Antifungal combinations with synergistic interaction can be a good choice for antifungal lock therapy; therefore, interactions were examined between two echinocandins (caspofungin and micafungin) and the chitin synthesis inhibitor nikkomycin Z against Candida albicans and C. parapsilosis biofilms. Susceptibility was evaluated using the XTT-based checkerb
